Effect of Matrix Viscosity on Clay Dispersion in Preparation of Polymer/Organoclay Nanocomposites

  • The viscosity effect of matrix polymer on melt exfoliation behavior of an organoclay in poly($\varepsilon$-caprolactone) (PCL) was investigated. The viscosity of matrix polymer was controlled by changing the molecular weight of poly($\varepsilon$-eaprolactone), the processing temperature, and the rotor speed of a mini-molder. Applied shear stress facilitates the diffusion of polymer chains into the gallery of silicate layers by breaking silicate agglomerates down into smaller primary particles. When the viscosity of PCL is lower, silicate agglomerates are not perfectly broken into smaller primary particles. At higher viscosity, all of silicate agglomerates are broken down into primary particles, and finally into smaller nano-scale building blocks. It was also found that the degree of exfoliation of silicate layers is dependent upon not only the viscosity of matrix but thermodynamic variables.

랫드의 두개골결손부 모델에서 HA/PCL 지지체를 사용한 골이식 시 Matrigel의 효과 (Effect of Matrigel for Bone Graft using Hydroxyapatite/Poly $\varepsilon$-caprolactone Scaffold in a Rat Calvarial Defect Model)

  • 본 연구에서는 hydroxyapatite/poly $\varepsilon$-caprolactone composite (HA/PCL) 지지체와 matrigel을 랫드의 두개골 결손부 모델에 함께 이식 시의 골형성 정도를 평가하였다. 두개골결손부는 Sprague Dawley rat (n = 18)에서 수술적으로 형성하였으며 실험군은 Matrigel과 함께 HA/PCL 지지체를 이식한 군(M-HA/PCL group, n = 6)과 HA/PCL 지지체단독이식군(HA/PCL group, n = 6)으로 나누었고 대조군(CD group, n = 6)에는 아무 것도 이식하지 않았다. 수술 4주 후, 골형성은 방사선촬영, micro CT 및 조직검사를 통해 평가되었다. 방사선상에서 CD군의 골형성은 관찰되지 않았으나 HA/PCL과 M-HA/PCL군에서는 관찰되었고 골과 유사한 방사선비투과성이 M-HA/PCL군에서 더 많이 관찰되었다. Micro CT 평가에서 골부피는 HA/PCL군보다 M-HA/PCL군에서 더 높았으나 두 군 사이의 유의적 차이는 관찰할 수 없었다. 그러나 골밀도에서는 HA/PCL군보다 M-HA/PCL군이 더 유의적으로 높음을 확인할 수 있었다(p < 0.05). 조직학적 검사에서는 CD군에서 새로운 골은 원래 존재하던 골로부터만 형성되었으며 두개골결손부 내의 골형성은 보이지 않았다. HA/PCL군에서 새로운 골형성은 원래 존재하던 골로부터만 유래되었으나 M-HA/PCL군은 가장 많은 골형성을 보여주었으며 새로운 골이 원래 존재하던 골과 HA/PCL지지체 주변에서도 관찰되었다. 이러한 결과로 미루어볼 때 HA/PCL 지지체와 matrigel을 함께 사용하는 것이 골의 임계결손부에서 골형성을 증대시키는 효과적인 방법이 될 수 있을 것으로 생각된다.

Refractive Indexes of Porous Thin Films Prepared From Organic-templated Polymethylsilsesquioxanes

  • Organosilyl-modified and star-shaped poly (${\varepsilon}$-caprolactone) (m-PCL) was prepared, and added to polymethylsilsesquioxane (PMSSQ), to make composites. The end groups of m-PCL are chemically similar to PMSSQ, and m-PCL mixed well with PMSSQ in the composite. Porous PMSSQ film was made by further calcination of the composite film at elevated temperature. m-PCL-templated PMSSQ and the as-prepared porous PMSSQ were structurally, optically, and thermally characterized in thin films. The chemical binding of m-PCL and PMSSQ effectively suppressed the phase separation of PMSSQ and m-PCL during the curing process. After calcination at elevated temperature, there remained many pores in the PMSSQ matrix. The refractive indices of the resulting porous PMSSQ thin films decreased with increase of the film porosities, depending on the initial m-PCL loadings.

전기방사공정과 발포제를 이용한 Polycaprolactone 나노섬유 지지체 제작 (Polycaprolactone Nanofiber Mats Fabricated Using an Electrospinning Process Supplemented with a Chemical Blowing Agent)

  • 생체재생용 지지체는 높은 다공구조와 적당한 기계적인 강도를 필요로 한다. 높은 다중성과 적당한 다공크기는 지지체와 주변 환경 사이에 영양분의 공급을 원활하게 하여 셀의 지지체에 대한 초기 집착력과 성장을 가능하게 하는 구조를 제공한다. 본 논문에서는 polycaprolactone(PCL) 나노섬유를 화학 발포제와 전기방사공정을 이용하여 다양한 조건하에서 제조하였다. PCL 용액의 농도가 8wt%, 발포제의 함량 0.5wt%, 발포온도 $100^{\circ}C$ 및 체류시간 2-3초에서 가공성 측면과 다공성 측면에서 우수한 발포된 나노섬유를 얻을 수 있었다. 또한 세포의 성장성을 측정하기 위하여 인체피부세포를 셀 켤츄어링하여, 발포되지 않은 나노섬유와 비교하였다.

Fabrication and characterization of PCL/TCP-coated PHBV composite multilayer as a bone plate

  • In this work, Poly($\varepsilon$-Caprolactone)(PCL) andpoly(3-hydroxybutyrate-co-3-hydroxyvalerate)(PHBV) mats were fabricated usingelectrospinning process. The electrospinning process is a simple and efficient method to fabricate the nanofibrous mats. PCL and PHBV is a kind of biodegradable polymer but their mechanical properties aren't good. For improving mechanical properties, PHBV mats were coated by TCP. Using PCL mats and TCP-coated PHBV composite mats, a bio-resorbablebone plate were made by pressing. Detailed micro-structural characterization was done by SEM techniques. Tensile strength and bending strength were also evaluated for mechanical properties. The cytotoxicity evaluation ofPCL/TCP-coated PHBV composite multilayer was done by MTT assay. The evidence obtained in this work implies the potential for use as a biodegradable boneplate.

Quercetin과 Rutin을 함유하는 PCL-b-PEG 고분자 미셀의 특성 및 피부 흡수에 관한 In vitro 연구 (Physical Characteristic and In vitro Transdermal Delivery of PCL-b-PEG Micelles Containing Quercetin and Rutin)

  • 본 연구에서는 항산화 물질로 잘 알려진 quercetin과 그 배당체인 rutin을 함유하는 poly(${\varepsilon}$-caprolactone)-b-poly(ethylene glycol) 미셀을 제조하여, 활성물질(quercetin, rutin)의 in vitro 피부 흡수 증진에 관한 연구를 수행하였다. 입자크기는 PCL-b-PEG 고분자의 농도가 증가함에 따라 미셀의 초기 입자 크기가 증가하는 경향을 보였다. 고분자 미셀의 표면 전위(Zeta potential)는 비교적 일정함을 확인하였다. 제조한 고분자 미셀의 피부 흡수력을 평가하기 위하여, 용액 상태의 활성물질을 미셀의 대조군으로 하여 Franz cell을 이용한 투과실험을 진행한 결과 용액 상태보다 미셀에서 더 높게 나타났음을 확인하였다. 또한 화장품 소재로서의 안전성 평가를 위한 인체 피부 일차자극 실험(patch test) 결과 어떠한 피부 자극도 관찰되지 않았다.

[ $W_{1}/O/W_{2}$ ] Multi-emulsion Capsules Containing Ascrobic Acid-2-Glucoside Prepared Using PCL-based Amphiphilic Di- and Tri-block Copolymers

  • Biodegradable and amphiphilic di-block and tri-block copolymers, prepared with monomethoxy poly ethylene glycol (MPEG) and ${\varepsilon}-caprolactone\;({\varepsilon}-CL)$, were used for the application of W1/O/W2 multi- emulsion capsules. The effects of topology and the ratio of hydrophilic moiety of PCL-based polymers on the encapsulation efficiency of the $W_{1}/O/W_{2}$ multi-emulsion capsules containing Ascorbic Acid-2-Glucoside (AA-2-G) were investigated. The ratio of PEG and PCL was 1:0.5, 1:0.75, 1:1, and 1:1.25. PEG-PCL block copolymers were added to the first step of the preparation of $W_{1}/O$ emulsions. The dispersion stability, the particle size, the morphology of the $W_{1}/O/W_{2}$ multi-emulsion capsules were observed using an on-line turbidity meter, dynamic light scattering (DLS), a confocal microscopy (with FITC) and an optical microscopy. Biodegradable behavior of the PEG-PCL block copolymers and release behavior of AA-2-G were also observed by Gel Permeation Chromatography (GPC) and High Performance Liquid Chromatography (HPLC).

Biotin-Conjugated Block Copolymeric Nanoparticles as Tumor-Targeted Drug Delivery Systems

  • To achieve targeted drug delivery for chemotherapy, a ligand-mediated nanoparticulate drug carrier was designed, which could identity a specific receptor on the surfaces of tumor cells. Biodegradable poly(ethylene oxide)/poly$({\varepsilon}-caprolactone)$ (PEG/PCL) amphiphilic block copolymers coupled to biotin ligands were synthesized with a variety of PEG/PCL compositions. Block copolymeric nanoparticles harboring the anticancer drug paclitaxel were prepared via micelle formation in aqueous solution. The size of the biotin-conjugated PEG/PCL nanoparticles was determined by light scattering measurements to be 88-118 nm, depending on the molecular weight of the block copolymer, and remained less than 120 nm even after paclitaxel loading. From an in vitro release study, biotin-conjugated PEG/PCL nanoparticles containing paclitaxel evidenced sustained release profiles of the drug with no initial burst effect. The biotin-conjugated PEG/PCL block copolymer itself evidenced no significant adverse effects on cell viability at $0.005-1.0{\mu}g/mL$ of nanoparticle suspension regardless of cell type (normal human fibroblasts and HeLa cells). However, biotin-conjugated PEG/PCL harboring paclitaxel evidenced a much higher cytotoxicity for cancer cells than was observed in the PEG/PCL nanoparticles without the biotin group. These results showed that the biotin-conjugated nanoparticles could improve the selective delivery of paclitaxel into cancer cells via interactions with over-expressed biotin receptors on the surfaces of cancer cells.

Mechanical Behavior of Shape Memory Fibers Spun from Nanoclay-Tethered Polyurethanes

  • This study examined the effect of nanoclays on the shape memory behavior of polyurethane (PU) in fibrous form. A cation was introduced into the PU molecules to disperse the organo-nanoclay (MMT) into poly($\varepsilon$-caprolactone) (PCL)-based PU (PCL-PU). The MMT/PCL-PU nanocomposites were then spun into fibers through melt-processing. The shape memory performance of the spun fibers was examined using a variety of thermo-mechanical tests including a new method to determine the transition temperature of shape memory polymers. The MMTs showed an improved the fixity strain rate of the MMT /PCL- PU fibers but a slight decrease in their recovery strain rate. This was explained by the limited movement of PU molecules due to the presence of nanoclays. The shape memory performance of the MMT/PCL-PU fibers was not enhanced significantly by the nanoclays. However, their recovery power was improved significantly up to a strain of approximately 50%.

Amphotericin B Aggregation Inhibition with Novel Nanoparticles Prepared with Poly(${\varepsilon}$-caprolactone)/Poly(N,N-dimethylamino-2-ethyl methacrylate) Diblock Copolymer

  • Diblock copolymers composed of poly(${\varepsilon}$-caprolactone) (PCL) and poly(N,N-dimethylamino-2-ethyl methacrylate) (PDMAEMA), or methoxy polyethylene glycol(PEG), were synthesized via a combination of ring-opening polymerization and atom-transfer radical polymerization in order to prepare polymeric nanoparticles as an antifungal drug carrier. Amphotericin B (AmB), a natural antibiotic, was incorporated into the polymeric nanoparticles. The physical properties of AmB-incorporated polymeric nanoparticles with PCL-b-PDMAEMA and PCL-b-PEG were studied in relation to morphology and particle size. In the aggregation state study, AmB-incorporated PCL-b- PDMAEMA nanoparticles exhibited a monomeric state pattern of free AmB, whereas AmB-incorporated PCL-b- PEG nanoparticles displayed an aggregated pattern. In in vitro hemolysis tests with human red blood cells, AmBincorporated PCL-b-PDMAEMA nanoparticles were seen to be 10 times less cytotoxic than free AmB (5 ${\mu}g$/ml). In addition, an improved antifungal activity of AmBincorporated polymeric nanoparticles was observed through antifungal activity tests using Candida albicans, whereas polymeric nanoparticles themselves were seen not to affect activity. Finally, in vitro AmB release studies were conducted, proving the potential of AmB-incorporated PCL-b-PDMAEMA nanoparticles as a new formulation candidate for AmB.
