New Columnar Apple Variety 'Tinkerbell' for Pollenizer (사과 무측지성 수분수용 신품종 '팅커벨')

  • This research was performed to breed new columnar-typed apple variety for pollinator in apple orchard. We cross-fertilized 'Hongro' that was an ovary parent and 'Maypole' a pollen parent in 2003. We gathered seeds in 2003 and sowed after breaking of dormancy in 2004. Seedlings was been growth and development till flowering. After fruit setting, We investigated characteristics of growth and fruit including mating compatibility from 2010 to 2012, then finally selected '03-2-95' named 'Tinkerbell' in 2012, registered as new variety on Korea Seed & Variety Service in 2015. The full bloom stage of 'Tinkerbell' was later 2 days then of 'Maypole'. Tree habit of 'Tinkerbell' was alike to of 'Maypole', for example weak tree vigor, columnar type. But there were many different characteristics between 'Tinkerbell' and 'Maypole'. Fruit shape of 'Tinkerbell' was ovoid whereas of 'Maypole' was ellipsoid. Fruit of 'Tinkerbell' was bigger, smoother skin, no waxy bloom than of 'Maypole'. Fruit of 'Tinkerbell' was more delicious than of 'Maypole' because of having higher soluble sugar content and lower titratable acidity. 'Tinkerbell' was excellent pollinator in apple orchard. Because of having high fruit setting rate with major apple varieties and a narrow tree width that was advantageous as pollinator in apple high density system.

Assessment of gene flow from insect-resistant genetically modified rice (Agb0101) to non-GM rice (해충저항성 유전자변형 벼(Agb0101) 유전자 이동성 평가)

  • Genetically modified (GM) crops have been developed worldwide through the recombinant DNA technology and commercialized by global agricultural companies. Until now, GM crops have not been cultivated commercially in Korea. Commercialization of GM crops requires a compulsory assessment of environmental risk associated with the release of GM crops. This study was conducted to evaluate the frequency of pollen mediated gene flow from Bt transgenic rice (Agb0101) to japonica non-GM rice (Nakdongbyeo), indica non-GM rice (IR36), and weedy rice (R55). A total of 729,917, 596,318 and 230,635 seeds were collected from Nakdongbyeo, IR36, and R55, respectively, which were planted around Agb0101. Selection of the hybrids was determined by repeated spraying of herbicide and Cry1Ac1 immunostrip assay. Finally, the hybrids were confirmed by PCR analysis using specific primer. The hybrids were found in all non-GM rice and out-crossing ranged from 0.0005% at IR36 to 0.0027% at Nakdongbyeo. All of hybrids were located within 1.2 m distance from the Agb0101 rice plot. The meteorological elements including rainfall and temperature during rice flowering time were found to be important factors to determine rice out-crossing rate. Consideration should be taken for many factors like the meteorological elements of field and physiological condition of crop to set up the safety management guideline to prevention of GM crops gene flow.

Embryological Characteristics on Seed Sterility of Ligusticum chuanxiong Hoit. (천궁의 종자불임에 관한 발생학적 특성)

  • Ligusticum chuanxiong is receiving much attentions as one of the important medicinal crops with the increasement of the crude drug demands. This study was conducted to obtain the basic informations of breeding of Ligusticum chuanxiong. Embryological characteristics were examined to elucidate the process of male and female gametophytic development and fertilization. Meiosis and nucleus division of megaspore and microspore were proceeded normally. With regard to the formation of female gametophyte, only a half of female gametophyte developed to normal egg apparatus. While another 50% showed abnormal egg apparatus with poly-nuclei or non-nuclei ovule. The pollens developed from the micros pore were formed more than 90 % of normal pollen. It was difficult to observe fertilization because ovule tissue was very compact and cell was extremely tiny, but could be easely observed proembryo and embryo formation. Only 30 percent developed into proembryo and subsequently into embryo, and the others were degenerated.

  • To characterize active cells during microspore culture of Angelica gigas, flow cytometric and epifluorescent techniques were applied. The knowledge obtained from these types of studies will give us insight into early stage in plant development and may lead to the application of microspore-derived from haploid plants for breeding in recalcitrant species. Viability of cultured microspore differed depending on the developmental stages. Frequencies of active cells from tetrad, uni-nucleate, bi-nucleate and matured pollen were 12.8, 49.3, 42.3 and 31.7%, respectively. Alive microspores have luminescent the green fluorescence stained with FDA and blue fluorescence stained with DAPI.

Enhanced Seed Development in the Progeny from the Interspecific Backcross (Fagopyrum esculentum ${\times}$ F. homotropicum) ${\times}$ F. esculentum

  • To facilitate the introgression of F. esculentum into the traits of F. homotropicum, several accessions of the hybrids between these two species were pollinated with F. esculentum as the recurrent parent. The embryo in vitro rescue was performed to increase the recovery of backcross progenies. The $F_{2}$ generation was more amenable than $F_{1}$ hybrids to produce backcross progenies. The $F_{1}$ hybrids were backcrossed twice with common buckwheat (pin-type F. esculentum) (recurrent backcrossing). Also, alternate backcrosses with common buckwheat and F. homotropicum (congruity backcrossing) were carried out. Pollen tube growth of BC$F_{1}$ ${\times}$ F. esculentum (thrum) and F. homotropicum ${\times}$ BC$F_{1}$ was the disturbed penetration exceeded for all initial interspecific hybrids, and its requirement was proportionally lower when the common buckwheat was used as the recurrent parent and as the last parent of congruity hybrids. Effects of both common buckwheat and F. homotropicum on seed success rate for hybridization were observed. Growth of hybrid embryos before rescue, regeneration of mature hybrids all increased recurrent and congruity backcrosses and inter-crosses between $F_{1}$ plants and selected fertile plants of the second congruity backcrosses.

  • The indica ${\times}$ japonica rice (Oryza sativa L.) cultivars released in Japan since 1980 are high-yielding. However, occasionally in these cultivars their high yield potentials cannot be realized, particularly depending on climate conditions. The objectives of this study were to identify the reason for yield instability and the critical yield component factor causing lower grain yield in these high-yielding cultivars. Standard-yield japonica, high-yielding japonica-dominant, and indica-dominant cultivars were grown in western Japan. Rough grain yield (RY) in these high-yielding cultivars ranged from 450 to $980g\;m^{-2}$, and was positively correlated with potential grain yield (PRY). By fluctuations of solar radiation, RY changed with spikelet number (SNO), and SNO was correlated with cumulative radiation during the panicle formation period of 30 days before heading. Even if higher SNO was achieved under higher radiant conditions, RY was lower than PRY. The lower grain-filling in plants bearing higher SNO resulted from a lower filling percentage of spikelets (%F, RY/PRY), and %F was strongly correlated with spikelet fertility (%S) across all cultivars. %S was highly influenced by cumulative radiation per PRY during pollen development and establishment around heading. Inhibition of assimilation by leaf removal lowered %S. Conversely, stem thinning and removal of upper panicles around heading increased %S in spikelets of the lower part of the panicle in which sterility was higher. These results suggest that limitation of assimilate-supply due to low irradiance at the spikelet-formation and flowering stages restrains the potential of the high-yielding cultivars, depending on reduction of SNO and %S, respectively.

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed.   • In these proceedings, we will address the problem of air pollution on the premises of the Cement factory SHARRCEM L.L.C. in Hani Elezit in the Republic of Kosovo respectively around the clinker cooler, rotary kiln, and raw material mill. By air pollution, we mean the introduction of chemicals, particles, or other harmful materials into the atmosphere which in one way or another causing damage to the development of plants and organisms. Air pollution occurs when certain substances are released into the air, which depending on the quantitative level, can be harmful to human health, animals, and the environment in general. The analysis of air shows the influence of the extractive and processing industry on the chemical composition of air. Parameters analyzed though under control such as the case of carbon dioxide, due to the increasing production capacity of cement, the production of hundreds of thousands of cubic meters of CO2 gas made CO2 production a concern. With the purchase of the latest technology by the SHARCEM Factory in Hani Elezit, the amount of air pollution has been reduced and the allowed parameters of environmental pollution have been kept under control. Air pollutants are introduced into the atmosphere from various sources which change the composition of the atmosphere and affect the biotic environment.The concentration of air pollutants depends not only on the quantities that are emitted from the sources of air pollution but also on the ability of the atmosphere to absorb or disperse these emissions. Sources of air pollutants include vehicles, industry, indoor sources, and natural resources. There are some natural pollutants, such as natural fog, particles from volcanic eruptions, pollen grains, bacteria, and so on.

Effects of Plant Growth Regulators on Haploid Production of Wheat (밀 반수체 생산에서 식물생장조절제의 영향)

  • The introduction of doubled haploid (DH) approach into breeding programs has reduced the times and population sizes required for the production of pure lines. We carried out the experiment for development on effective method of producing haploid in wheat. Emasculated spikelets of wheat were pollinated with maize pollen and cultured in the solution containing 40 g/L sucrose and 2,4-D, NAA, 2,4,5-T and dicamba 24 h after pollination, and then incubated until embryo rescue. Fourteen to sixteen days after pollination, the embryos are excised and cultured in half-strength MS basal medium supplemented with 20 g/L sucrose and 1 mg/L NAA. The type of plant growth regulators was found to be most significant in production of haploid plants. The application of synthetic auxins to pollinated florets, stimulates haploid embryo development to a stage where the embryos can be rescued onto nutrient media. The percentage of seeded florets was significantly affected by 100 mg/L 2,4-D, 150 mg/L 2,4,5-T and 50 mg/L dicamba. The percentage of embryos formed was significantly increased by treatment with 2,4-D and 2,4,5-T at 100 mg/L, and dicamba at 50 mg/L, but the treatments with 150 mg/L 2,4-D inhibited embryo development and plant regeneration. The optimum application time of plant growth regulators was 24 hrs after pollination.

Analysis of Honeybee-Keeping Management Types and Skills Level in Korea (양봉농가의 경영형태와 기술수준 분석)

  • For the improvement of productivity of beekeepers and to develop new management strategy through the analysis of current honeybee-keeping management types and evaluation of related skills and basis in Korea were surveyed. The serial surveys were carried out in 2008 for a total of 253 beekeepers from different localities in Korea. The results revealed that 145.9 colonies of honeybee were managed by each beekeeper in Korea. The beekeepers group who were younger than 55 years possessed 191.7 colonies. The older age group of beekeepers kept rather smaller number of colonies. The career groups over 15 years in apiculture were keeping 176.0 colonies in average. The numbers of bee colonies were not significantly different among localities and educational backgrounds. The migratory apiaries with 145.9 hives were dominant than those fixed ones with 83.0 (p<0.01). It was observed that 57.8% of apiaries were located in the vicinity of farmer's house, whereas 30.3% were managed in mountain area; however older group (65.4%) preferred to keep it near their houses (p<0.05). The proportion (35.7%) of part-time fixed apiary was the highest among four apiary management types. The younger group and high career group operated full-time migratory apiary (38.8% and 40.3%) (p<0.05), respectively. However, the older group and low career group preferred as part-time fixed one (43.2% and 46.6%), respectively (p<0.001). There was no significant difference among localities. The surveyed beekeepers produced propolis, commercial colonies, pollen, royal jelly, and pollination hives in addition to honey. The group of less than 15 years career and mountain apiary mainly produced propolis and pollen. However, those who dwell in small and middle areas mainly produced propolis and commercial colonies. The levels related to beekeeping skills and basis were evaluated as intermediate from 3.11 and 2.86 points on 5 points scale, respectively. More careered group having skill level of 3.33 point and basis level of 3.02 showed higher levels than less careered one. The levels of skill and basis evaluated from age groups and localities were similar with each other with no significance.
