Infestation Characteristics of Parasite (Ligula intestinalis) in Abdominal Cavity of Zacco platypus in the Small Stream of Korea (국내 중소하천에서 피라미 (Zacco platypus)의 복강 기생충 감염특성)

  • In freshwater ecosystems, the types of fish parasites are diverse and routes of infection are complicated because they maintain the life cycle in a variety of host animals, The present study investigated the infestation characteristics of parasites occurring in abdominal cavity of Zacco platypus in Seom River from February to October 2007. At the same time, four streams (Gam Stream, Daepo Stream, Yangsan Stream, and Yulha Stream) were also surveyed for the reference of fish infection. Aquatic insects and shellfish predominated benthic macroinvertebrate assemblages by 90.4% and 7.7% of relative abundance, respectively. Aquatic insects were composed mostly of Ephemeroptera (46.8%) and Trichoptera (29.8%), and shellfish was Gastropods. Freshwater fish was most abundant Cyprinid and Z. platypus by 85.7% and 48.8%, respectively. Among waterfowl, Anas poecilorhyncha and Egretta garzetta were dominant in August. Fish parasites observed in the abdominal cavity of Z. platypus was classified as Ligula intestinalis, belonging to Phylum Platyhelminthes Cestoda Pseudophyllidae. L. intestinalis was observed from Z. platypus in October, as the number of one individual in the Seom River and fifteen individuals in the Yangsan Stream. Infection rate of plerocercoid ranged 1.4~15.8%. We discussed the relationship between the life cycle of the parasite and the host, and hope our results to be useful for understanding the ecology of the fish abdominal parasites in Korean river ecosystem.

Biological Characteristics of Spirometra erinacei and S. mansonoides by Developmental Stages

  • To clarify the species validity of the genus Spirometra, the biological characteristics of Spirometra erinacei and S. mansonoides by developmental stages were compared. Their experimental life cycles were maintained under the same laboratory conditions, and the biological characteristics were experimentally observed in vivo and in vitro conditions. Eggs of S. erinacei and S. mansonoides were $59.6\pm35.6{\mu}m\;and\;61.4\pm35.8{\mu}m$ in each average size. Both of them became fully matured and hatched in 8 days after incubation at $29^{\circ}C$. The coracidium of S. erinacei was $43.6\times35.8{\mu}m$ in average size, and retained a oncosphere of $39.3\times31.0{\mu}m$. That of S. mansonoides was $43.0\times36.3{\mu}m$ in average size, and retained a oncosphere of $38.3\times30.8{\mu}m$. Procercoids of S. erinacei were somewhat larger than those of S. mansonoides. Both species of procercoids older than 7 days in cyclops had minute spines at the anterior end, calcium corpuscles in the parenchyme and a cercomer at the posterior end. The procercoids older than 4 days in cyclops were infective to tadpoles. The procercoids older than 8 days revealed the infectivity to mice. Plerocercoids of S. erinacei were somewhat lager than those of S. mansonoides when they were compared by age of worms in tadpoles. Both species of plerocercoids older than 5 days were infective to mice. Among 138 plerocercoids of S. erinacei recovered from the experimental mice, $55(39.9\%)$ were detected in the neck portion, $35 (25.4\%)$ in the back portion, $25(18.1\%)$ in the anterior legs, and $23 (16.7\%)$ were found in the abdomen. In case of S. mansonoides plerocercoids, $42.0\%$ were found in the neck portion, $23.8\%$ in the back portion, $14.4\%$ in the abdomen, $13.3\%$ in the anterior legs, and $6.1\%$ were found in the posterior legs. From the above results, it was confirmed that the biological characteristics of S. erinacei and S. mansonoides are almost same when their life cycles are mainteined under the same laboratory condition. Accordingly, these findings suggest that S. erinacei and S. mansonoides may be the same species.

A Survey and Morphological Study of Plerocercoidal Nybelinia surmenicola (Cestoda: Tentaculariidae) in the Common Squid from the Korean Coastal Waters (한반도 연안 오징어의 Nybelinia surmenicola 충미충 역학조사 및 형태학적 연구)

  • Trypanorhych plerocercoids of Nybelinia surmenicola are described in the common squid, Todarodes pacificus, through the use of light and scanning electron microscopy. Upon dissection, out of 200 common squid from the Korean coastal water, 54 (27%) were found infected with N. surmenicola plerocercoids. Mean N. surmenicola abundance was 2.28 per squid. Body length and width ranged from $6.1-11.1(8.05){\times}2.0-2.2(2.1)mm$. Bothria are broad, bean-shaped, measured $2.4-2.8{\times}0.62-0.69(2.65{\times}0.64)mm$. The short and long tentacular armatures which are spirally arranged are homeoacanthous and homeomorphous uncinate hooks and consist of compact rose-thorn-shaped tentacular hooks. The tentacle sheaths are coiled in a spiral near the bulbs. Four large bothria are separated by filiform microtriches along the bothridal margins. This is the first description of the plerocercoids of N. surmenicola in the common of squid from the Korean coastal waters.

Genetic and Morphologic Identification of Spirometra ranarum in Myanmar

  • In the present study, we identified a Spirometra species of Myanmar origin (plerocercoid) by molecular analysis using mitochondrial cox1 and nad1 genes, as well as by morphological observations of an adult tapeworm. Spargana specimens were collected from a paddy-field in Taik Kyi Township Tarkwa Village, Yangon, Myanmar in December 2017. A total of 5 spargana were obtained from 20 frogs Hoplobatrachus rugulosus; syn: Rana rugulosa (Wiegmann, 1834) or R. tigrina (Steindachner, 1867). The plerocercoids were used for experimental infection of a dog. After 4 weeks of infection, an adult tapeworm was recovered from the intestine of the dog. Morphologically, the distinct features of Spirometra sp. (Myanmar origin) relative to S. erinaceieuropaei and S. decipiens include a uterine morphology comprising posterior uterine coils that larger than the terminal uterine ball and coiling of the uteri diagonally (swirling) rather than spirally. The cox1 sequences (1,566 bp) of the Myanmar-origin Spirometra species showed 97.9% similarity to a reference sequence of S. decipiens (GenBank no. KJ599679) and 90.5% similarity to a reference sequence of S. erinaceieuropaei (GenBank no. KJ599680). Phylogenetic tree topologies were identical and presented high confidence level of values for the 3 major branches of the 3 Spirometra species in cox1 and nad1 genes. These results indicated that Myanmar-origin Spirometra species coincided with those of S. ranarum and may be considered as a valid species.

  • Spirometru erinocei의 유충인 sparganum에서 추출한 조항원 단백질을 항원으로 하여 sparganosis, cysticercosis, hydatidosis환자의 IgG 항체와 정상인의 IgG 항체를 혈청반응시켜 ELISA와 EITB에 의해 교차반응을 일으키는 비 특이항원 성분과 종특이항원 성분을 추구하였다. 1. sparganum에서 0.01 M PBS (PH 7.4)로 추출한 조항원 단백질을 SDS-PAGE로 전개하여 290 Kd에서 23 Kd범위의 분자량을 가진 25개의 단백질이 분획되었다. 2. sparganum의 조항원을 항원으로하여 ELISA로 sparganosis, cysticercosis, hydatidosis 환자의 190 항체와의 혈청 반응치는 sparganosis환자에게서는 0.44 $\pm$ 0.07에서 1.90 $\pm$ 0.03으로 negative control normal sera의 0.D (0.15 $\pm$ 0.03)를 기준으로 하였을 때 모두 양성이며 민감도(sensitivity)가 100 %이었으며 cysticercosis, hydatidosis환자혈청에서 양성반응이 나타났으며 교차반응도 있었다. 3. EITB에서는 spargancsis환자의 IgG항체에 의해 16개의 항원성분이 인지되었으며 이 중 6개의 항원성분이 정상인의 혈청에서도 인지되어 교차반응을 일으키는 항원성분이었으며 cysticercosis환자혈청에서 인지된 4개의 항원성분 중 2개의 항원성분이 sparganosis 환자혈청에서 인지된 것과 같았으며 hydatidosis환자의 190항체에 의해 인지된 19개의 항원성분 중 12개의 항원성분이 sparganosis환자혈청에서 인지된 항원 성분과 같았다. 4. 290 Kd, 200 Kd, 28 Kd의 항원성분은 sparganosis환자의 196항체에서만 인지되었고 228 Kd, 152 Kd, 66 Kd항원성분은 hydatidosis환자의 19G항체에서만 인지되었으며 66 Kd항원성분은 sparganosis, cysticercosis, hydatidosis, 정상인의 혈청에서 모두 인지되었다. We studied the serological reaction between the specific and nonspecific antigenic components from metacestode (plerocercoid) of spiromeko erimacei and IgG antibodies in sparganosis, cysticercosis. hydatidosis patients and normal human sera by ELISA and EITB. We prepared the crude extracts of sparganlim from snake, Matrix tigrina laterolis and used as antigenic components. By SDS-PAGE, we detected a total 25 peptide bands (fractions) with 290 Kd to 23 Kd molecular weight, and 8 bands of these detected bands developed strongly by silver stain. In serological test, ELISA, we recognized the cross-reaction of antigenic components reacting with IgG antibodies in heterogenous sera, cysticercosis and hydatidosis patients sera. The crude antigenic components of sparganum showed the high sensitivity in sparganosis, hydatidosis patients sera, but showed lower sensitivity in cysticercosis patients sera than the sparasanosis, hydatidosis patients sera. Sixteen antigenic components of these 25 separated bands were recognized by antibodies In sparsanosis patients sera,8 antigenic components in normal human sera, 4 antigenic components in cysticercosis patients sera and 19 antigenic compoenents in hydatidosis patients sera. The crude antigenic compnenets with 290 Kd, 200 Kd, 125 Kd and 28 Kd molecular weight was only recognized in sparganosis patients sera, but 64 Kd antigenic component was nonspecific antigenic components which were also cross-reacted with sparganosis, hydatidosis, cysticercosis patients sera and normal human sera.

The changes of histopathology and serum anti-sparganum IgG in experimental sparganosis of mice (마우스 피하 스파르가눔증에 있어서 감염 경과에 따른 조직병리학적 병변 및 혈청 항체가의 변화 양상)

  • The present study is intended to observe the chronologic changes of experimental sparganosis by histopathological observation and detection of circulating anti-sparganum IgG antibody using ELISA. Each of 25 mice was infected with aye spargana, and they were examined after 1, 2, 4, 10 weeks or 6 months from infection. The followings are summarized results. - 1. The plerocercoids were detected in the subcutaneous tissue of the trunk, neck or axilla, but a few often extended into the skeletal muscle. The recovery rates were 72% at the first week, 80% at the second week, 95% at the fourth week, 92% at the tenth week and 100% at .the sixth month. The larvae grew slowly in both length and weight until 6 months. 2. Histopathologically, most of the larvae were observed alive in the soft tissue or skeletal muscle. Numerous eosinophils, neutrophils, Iymphocytes and plasma cells were infiltrated focally around the worms by the second week, but they surrounded the worms to form a layer of inflammatory reaction after 4 weeks of infection. Also histiocytes and fibroblasts began to appear around the inflammatory cells at 4 weeks. After 10 weeks, the worms encircled by a thin fibrous layer were found. After 6 months, the worms were surrounded by either fibrous tissue or active inflammatory cells. The inflammation looked more severe in the tracks left by the worms, rather than around the worms. 3, The level of anti-sparganum IgG antibody in the serum showed an increase by the fourth week, and a rapid and continuous increase was observed thereafter by the tenth week after infection. The high level of the IgG antibody was maintained up to 6 months forming a plateau curve. The present results suggest that the tissue reaction and antibody production in subcutaneous sparganosis become distinctive by the fourth week after infection.
