The Effects of Different Wavelengths of Light-Emitting Diodes on the Expression of Reproduction-Related Genes in Goldfish Carassius auratus

  • We investigated the differences in the expression of the neurohormones kisspeptin (Kiss) and gonadotropin-inhibitory hormone (GnIH) and cytochrome P450 aromatase (P450arom), gonadotropin hormones (GTHs), and sex steroids in the goldfish Carassius auratus exposed to light-emitting diodes (LEDs). The expression levels of Kiss1, Kiss2, G-protein-coupled receptor 54 (GPR54), GTHs, GnIH, and P450arom were compared between the control (white light) and LED-treated goldfish. Furthermore, we measured the plasma levels of follicle-stimulating hormone (FSH) and luteinizing hormone (LH). The levels of Kiss1 mRNA and protein; Kiss2, GPR54, and $GTH{\alpha}$ protein; GTH mRNA; and plasma FSH and LH in the hypothalamus and cultured hypothalamus cells were significantly higher in the green and purple LED treatment groups than in the other groups. These results suggested that red LEDs inhibit the sex maturation hormones, Kiss, GPR54, GTHs, and P450arom, and that GnIH plays a role in the negative regulation of reproductive function in goldfish.

시험 스트레스와 Vitamin B 복합제가 혈장 ACTH, Cortisol 및 Prolactin치에 미치는 영향 (The Effect of Exam Stress and Vitamin B Complex on Plasma ACTH, Cortisol and Prolactin Level)

  • 본 연구의 목적은 시험스트레스 및 vitamin B 복합제가 혈장 ACTH, cortisol 및 prolactin치에 미치는 영향을 알아보고자 하는 데 있다. 본 연구는 의과대학 2학년 학생들 중 시험 4주전, 시험 2주전 및 시험기간 세차례를 모두 완료한 21명을 대상으로 하였다. 상기 혈장 hormone치는 방사성 면역분석법에 의해 측정되었다. 그리고 스트레스지각 및 정신병리는 global assessment of recent stress(GARS)척도와 SCL-90R척도를 사용하여 평가되었다. 혈장 ACTH치는 시험 4주전에 비해 시험 2주전과 시험기간 중에 각각 유의하게 더 높게 나타났다. 그러나 세기간 중 혈장 cortisol과 prolactin치는 각각 유의한 차이를 보이지 않았다. 한편 vitamin B투여 유무 간에는 혈장 ACTH, cortisol 및 prolactin치는 각각 유의한 차이를 보이지 않았다. 또한 시험 2주전에 경제적 문제와 관련된 스트레스지각 점수는 혈장 ACTH 및 prolactin치와 유의한 양상관성을 보였다. 시험 2주전에 적대감척도 점수는 혈장 ACTH치와 유의한 양상관성을, 편집증척도 점수는 혈장 ACTH치와 양상관성의 경향을, 시험기간 중 불안척도 점수는 혈장 cortisol치와 양상관성의 경향을 보였다. 시간경과는 물론 vitamin투여 유무에 따른 혈장 ACTH, cortisol 및 prolactin치의 변화량에 있어서도 유의한 차이를 보이지 않았다. 결론적으로 시험스트레스에 대해서 cortisol이나 prolactin보다는 ACTH가 더 예민한 반응을 나타내고, 주관적 스트레스지각이나 시험전 적대감 및 편집증, 그리고 시험기간 중 불안과 같은 정신병리가 상기 hormone과 관련될 가능성이 높다. 그러나 vitamin B복합제는 스트레스에 의한 상기 hormone의 변화에 영향을 미치지 않음을 시사한다.

EFFECTS OF XYLAZINE (ALPHA 2-ADRENERGIC AGONIST) ON THE STRESS RESPONSE TO IMMOBILIZATION AND HEAT IN RATS

  • The effect of xylazine administration on plasma cortisol, prolactin, glucose and packed cell volume (PCV) responses to immobilization and heat stress was investigated. Immobilization of rats for 2 hours by ligation of the fore and hind legs strongly caused approximately two-fold increase in plasma cortisol and prolactin levels. Plasma glucose and PCV were not significantly changed. Pretreatment of immobilized rats with xylazine (20 mg/kg body weight i.m.) resulted in approximately 20% reduction in both plasma cortisol and prolactin concentrations. A marked hyperglycemia and increase in the PCV value was observed. On the other hand, rats exposed to acute heat stress ($40^{\circ}C$, and 60% relative humidity) for 2 hours, also developed two fold increase in both plasma cortisol and prolactin concentrations and the pretreatment with xylazine caused a 20% reduction in the levels of both hormones. Plasma glucose level was not significantly changed in heat stressed rats but it was markedly increased after pretreatment with xylazine. PCV was significantly incrcased under heat stress and pretreatment with xylazine induced a pronounced elevation in this value. It was suggested that stimulation of cortisol and prolactin secretion in response to immobilization or heat stress can be partially reduced by an alpha 2-adrenergic agonist.

Effect of the Dietary Protein Level on Plasma Glucose, Lipids and Hormones in Streptozotocin-Diabetic Rats

  • Atherosclerotic vascular disease is a major cause of the increased morbidity and mortality assciated with diabetes mellitus. The prominent role of nutrition in hypercholesteolemia and atherosclerosis is generally accepted. Diet is a key element in the management of diabetes (type I-IDDM), yet the appropriate diet for patient with diabetes mellitus is not well known. Dietary protein has been shown to have a significant effect on plasma cholesterol levels in both experimental animals and humans. The present experiment was designed to determine the effect of the dietary protein level(20% vs 60%) on plasma glucose concentration, lipids profile, insulin and glucagon levels from non-diabetic and streptozotocin-induced diabetic rats. Results showed that a high protein diet decreased triglyceride concentration in diabetic rats. Also diabetic rats fed a high protein diet were hypocholesterolemic than rats fed a control diet. There were no effects by level of protein on fasting blood glucose concentration and insulin/glucagon ratio. Results from the present study suggest that a high protein diet may be beneficial to control pasma lipids in chemically-induced diabetic rats.

정상생활을 하는 고혈압 여성에 있어서 일상적인 나트륨, 칼슘 섭취습관이 혈압조절 관련 호르몬에 미치는 영향 (The Effect of Habitual Calcium and Sodium Intakes on Blood Pressure Regulating Hormone in Free-Liveing Hypertensive Women)

  • In order to evaluate the effect of habitual Na and Ca intake on blood pressure regulation, we measured the habitual dietary intakes of Na and Ca, urinary excretion of Ca, Na and K, and plasma level of renin activity, aldosterone, and indices of Ca metabolism in 27 untreated hypertensive women and 30 age-matched normal women on a free diet. Hypertensive and total subjects were divided into four groups according to habitual dietary intakes of Na and Ca as low Na-low Ca(LNLC), low Na-high Ca(LNHC), high Na-low Ca(HNLC), and high Na-high Ca(HNHC). HNLC hypertensive group showed the lowest level of plasma renin activity, 25-(OH) Vit D$_3$, calcitonin and serum total Ca, and presented the highest level of PTH and urinary excretions of Na/K and Ca/Cr. There were no significant difference in plasma level of aldosterone and urinary excretion of Na and K among four hypertensive groups. When all subjects were divided into four groups according to the same method, HNLC group showed the highest level of blood pressure with no statistical significance and the lowest level of calcitonin and total serum Ca. The above results indicated that renin-aldosterone system and Ca regulating hormone has a mutual relationship in hypertension. Na and Ca may interact each other, rather than affecting independently blood pressure control. As a result, considering the fact that daily balance of Na and Ca intakes affects Na and Ca regulating hormones and urinary excretion of Na and Ca, it may be involved in blood pressure control. These results suggest that maintaining an adequate intake of Ca with less intake of Na may prevent from the risk of hypertension. (Korean J Nutrition 34(4) : 409~416, 2001)

Effects of The Soy Protein Level on Plasma Glucose, Lipids, and Hormones in Streptozotocin-Diabetic Rats

  • The number of diabetics in Korea is about 3 to 5 percent of the population, and the incidence is increasing yearly due to changes of life style and food intake. Diet is a key element in the management of diabetes, yet the appropriate diet for diabetes remains controversial. We have recently shown that a diet rich in protein of animal origin(casein) seems beneficial to controling plasma glucose and lipids in streptozotocin-induced diabetic rats. It therefore seemed desirable to find out whether the beneficial effect of high casein diet in experimental diabetes could also be reproduced with a vegetable source of protein(soy). The purpose of this study is to compare these results with the results of our previous study. In the present study, non-diabetic and streptozotocin-induced diabetic rats were studied in order to examine the effects of altering the level(20% vs 60%) of dietary soy protein on blood glucose, lipids, and hormones. Results of the present study showed that a high soy protein diet decreased triglyceride concentration in diabetic rats. However, diabetic rats fed a high soy protein diet were not hypocholesterolemic compared to rats fed a control diet. Moreover, diabetic rats fed a high soy protein diet had significantly increased plasma glucose concentration compared to rats fed a control diet. This study was not able to discern a specific effect of dietary protein level on insulin, glucagon, or insulin/glucagon ratio. Except for the hypotriglyceridemic effect, the results were not similar to the findings of our previous study which showed a beneficial effect on streptozotocin-induced diabetic rats fed a high casein diet.

Effects of Ensiled Cassava Tops on Rumen Environment Parameters, Thyroid Gland Hormones and Liver Enzymes of Cows Fed Urea-treated Fresh Rice Straw

  • Four rumen-cannulated cows (330 kg average weight at 4 years) were used to evaluate the supplement of ensiled cassava tops (ECT) (variety KM 94, 39% DM) on rumen functions, thyroid hormones and liver enzymes. The treatments, arranged in a 4 $\times$4 Latin square design, were ECT at 0, 50, 100 and 150 g CP 100 kg$^{-1}$ body weight (BW), and a basal diet of urea-treated fresh rice straw (UFRS) ad libitum and 1.1 kg dry matter (DM) cassava root meal (CRM) in each 30 day study period. The results showed a continuous decrease in dry matter intake (DMI) of UFRS with increasing level of ECT supplement (p<0.001). The highest total DMI was observed for treatment ECT$_{150}$ (2.68 kg DM 100 kg$^{-1}$ BW day$^{-1}$) followed by treatments ECT$_{100}$, ECT$_{50}$ and ECT$_{0}$, with 2.47, 2.24 and 2.06 kg DM 100 kg$^{-1}$ BW⋅day$^{-1}$, respectively. Increasing levels of ECT supplement increased the concentration of total volatile fatty acids (p<0.05) and ammonia nitrogen (p<0.05) and resulted in a decrease in pH (p<0.05). Overall average plasma triiodothyronine and thyroxine concentrations were 0.80, 0.82, 0.85 and 0.69 ng ml$^{-1}$ (p>0.05), and 50.9, 49.5, 50.7 and 42.4 ng ml-1 (p>0.05) for treatments ECT$_{0}$, ECT$_{50}$, ECT$_{100}$ and ECT$_{150}$, respectively. There were non-significant differences in alanine aminotransferase and aspartate aminotransferase among treatments. It is concluded that ECT is a valuable protein-rich feed supplement to cattle, and the highest level of on average 2.48 kg DM ECT per cow and day (28% of total DMI) did not significantly affect thyroid gland hormones and liver enzymes in cows.

A systematic review of the biological mechanisms linking physical activity and breast cancer

  • [Purpose] Epidemiological evidence has shown that leisure-time physical activity and structured exercise before and after breast cancer diagnosis contribute to reducing the risk of breast cancer recurrence and mortality. Thus, in this review, we aimed to summarize the physical activity-dependent regulation of systemic factors to understand the biological and molecular mechanisms involved in the initiation, progression, and survival of breast cancer. [Methods] We systematically reviewed the studies on 1) the relationship between physical activity and the risk of breast cancer, and 2) various systemic factors induced by physical activity and exercise that are potentially linked to breast cancer outcomes. To perform this literature review, PubMed database was searched using the terms "Physical activity OR exercise" and "breast cancer", until August 5th, 2020; then, we reviewed those articles related to biological mechanisms after examining the resulting search list. [Results] There is strong evidence that physical activity reduces the risk of breast cancer, and the protective effect of physical activity on breast cancer has been achieved by long-term regulation of various circulatory factors, such as sex hormones, metabolic hormones, inflammatory factors, adipokines, and myokines. In addition, physical activity substantially alters wholebody homeostasis by affecting numerous other factors, including plasma metabolites, reactive oxygen species, and microRNAs as well as exosomes and gut microbiota profile, and thereby every cell and organ in the whole body might be ultimately affected by the biological perturbation induced by physical activity and exercise. [Conclusion] The understanding of integrative mechanisms will enhance how physical activity can ultimately influence the risk and prognosis of various cancers, including breast cancer. Furthermore, physical activity could be considered an efficacious non-pharmacological therapy, and the promotion of physical activity is probably an effective strategy in primary cancer prevention.

Modulation of the Somatotropic Axis in Periparturient Dairy Cows

  • This review focuses on modulation of growth hormone (GH) and its downstream actions on periparturient dairy cows undergoing physiological and metabolic adaptations. During the periparturient period, cows experience a negative energy balance implicating that the feed intake does not meet the total energy demand for the onset of lactation. To regulate this metabolic condition, key hormones of somatotropic axis such as GH, IGF-I and insulin must coordinate adaptations required for the preservation of metabolic homeostasis. The hepatic GHR1A transcript and GHR protein are reduced at parturition, but recovers on postpartum. However, plasma IGF-I concentration remains low even though hepatic abundance of the GHR and IGF-I mRNA return to pre-calving value. This might be caused by alternation in IGFBPs and ALS genes, which consequently affect the plasma IGF-I stability. Plasma insulin level declines in a parallel manner with the decrease in plasma IGF-I after parturition. Increased GH stimulates the lipolytic effects and hepatic glucose synthesis to meet the energy requirement for mammary lactose synthesis, suggesting that GH antagonizes insulin-dependent glucose uptake and attenuates insulin action to decrease gluconeogenesis.

Gonadal Development, Spawning and Plasma Sex Steroid Levels of the Indoor Cultured Grunt, Hapalogenys nitens

  • The gonadosomatic index (GSI), gonadal development and changes in hormones in plasma level of the indoor cultured grunt (Hapalogenys nitens) were investigated by histological study from August 2011 to October 2012. The GSI showed similar trends with gonad developmental stages during the culture periods. Changes in plasma level of estradiol-$17{\beta}$ of female H. nitens reached the highest value before the spawning period, and seasonal changes in plasma level of estradiol-$17{\beta}$ were similar in trends of oocyte developments and GSI changes. Testosterone levels of male H. nitens reached the highest value before and after the spent stage. Ovarian developmental stages of H. nitens could be classified into early growing stage, late growing stage, mature stage, ripe and spawning stage, recovery and resting stage. The testicular developmental stages could be divided into growing stage, mature stage, ripe and spent stage, and recovery and resting stage.
