To determine whether the reproductive processes of sea bass, Lateolabrax japonicus, proceed normally after transportation from an outdoor net-cage into indoor tanks, we examined changes in the gonadosomatic index (GSI), histological gonadal tissue, and plasma levels of sex hormones (testosterone and estradiol-17ß) during their annual reproductive cycle. We also measured maturation and spawning across two sea water salinity levels (full and low salinity). Fecundity was estimated by the relationship between egg number and body size in female sea bass. Monthly changes in the GSI, histological gonadal tissues, and oocyte size showed both male and female sea bass reach final maturation in January and February, respectively, indicating that the spermiation of males occurs earlier than the spawning of females. The histological results indicated that the sea bass is a multiple spawner, similar to many marine teleosts, exhibiting group-synchronous oocyte development. Female maturation and spawning were enhanced in lower salinity seawater (29.6-31.0 psu) compared to that of normal salinity (34.5-35.1 psu). These results confirm that sea bass reproduction can occur successfully in captivity and imply that fertilized eggs can be collected from February to March. Additionally, our results show that lower salinity enhances oocyte maturation and spawning of female sea bass.
Twenty crossbred (Yorkshire ${\times}$ Landrace) gestating sows were used to determine the effects of Moringa Oleifera leaf (MOL) supplementation in lactating sow diets with or without banana peel powder (BPP) on reproductive performance, fecal moisture content, rectal temperature and hormone profile. The treatments were as follows: 1) CON (control diet), 2) M1 (control diet + 0.5% MOL), 3) M2 (control diet + 1.0% MOL), 4) M3 (control diet + 0.75% MOL and 0.75% BPP), and 5) M4 (control diet + 1.5% MOL and 1.5% BPP). The diets was fed from da 107 of gestation to weaning. MOL and BPP supplementation decreased (p < 0.05) the fecal moisture content compared with that of the CON at day 0 and 7 after farrowing, and 1.0% supplementation of MOL also decreased (p < 0.05) the fecal moisture content at day 14 after farrowing. On farrowing day, MOL supplementation decreased (p < 0.05) the blood cortisol and norepinephrine concentrations compared with the CON. In conclusion, the results of this study show that MOL supplementation decreased the fecal moisture content and plasma concentrations of cortisol and norepinephrine, and MOL and BPP supplementation also decreased the fecal moisture content in lactating sows.
Objectives : This study investigated the effects of electroacupuncture on memory, depression, and plasma stress hormone levels in rats that were under immobilization stress. Materials and Methods : The immobilization-only group was given two hours of immobilization stress for 10 consecutive days. The immobilization and high frequency EA group was given two hours of immobilization stress simultaneously with high frequency (100Hz) electroacupuncture stimulation on the right ST-36 (Zusanli) for 10 consecutive days. We conducted the inhibitory avoidance and forced swimming tests to recognize whether immobilization stress and electroacupuncture have effects on memory and depression. We collected blood samples from the tail of each rat at 30, 60, 90, and 120-minute intervals during the immobilization stress and EA stimulation to measure plasma concentrations of ACTH, corticosterone, melatonin, and norepinephrine induced by immobilization stress and electroacupuncture. Results : There was a significant effect of high frequency on the increase in anamnesis based on the result of the inhibitory avoidance test, but there was no significant effect of decreasing depression based on the result of the forced swim test. Also, there was no significant effect on the response indicated by stress hormones. Conclusions : 1. High frequency electroacupuncture (100Hz) improved anamnesis in immobilization stress states under the inhibitory avoidance task. 2. High frequency electroacupuncture (100Hz) did not reduce depression induced by immobilization stress under the forced swimming test. 3. High frequency electroacupuncture (100Hz) did not decrease stress hormones through blood sampling.

Cholesterol $7\alpha-hydroxylase$ (CYP7A1) is the rate-limiting enzyme in the conversion of cholesterol to bile acids and plays a central role in regulating cholesterol homeostasis. We previously showed that a fermentable $\beta-glucan$ ingestion decreased plasma cholesterol levels due to fecal bile acid excretion elevation involved inincrease of cholesterol $7\alpha-hydroxylase$ mRNA expression and activity. It is proposed that short chain fatty acids (SCFA) produced by cecal and colonic fermentation of soluble fiber are associated with cholesterol-lowering effect of fiber. In the present study, we investigated whether CYP7A1 expression is up-regulated by short chain fatty acids or by hormones in cultured human hepatoma (HepG2) cells. Confluent HepG2 cell were incubated with acetate, propionate, or butyrate at 1 mM concentration for 24 hrs. Acetate as well as propionate increased to 1.8-fold expression of CYP7A1 mRNA than the control. Butyrate also increased 1.5-fold expression of CYP7A1 mRNA. Our data show for the first time that SCFA increase expression of CYP7A1 mRNA. Adding insulin, dexamethasone and triiodothyronine $(1\;{\mu}M)$ to HepG2 cell increased the expression of CYP7A1 mRNA to $150\%,\;173\%,\;141\%$, respectively. These results suggest that SCFA produced by cecal fermentation stimulate enteric nervous system, in which secreted some neuropeptides may be responsible for change in cholesterol and bile acid metabolism. These findings suggest that SCFA are involved in lowering plasma cholesterol levels due to the up-regulation of CYP7A1 and bile acid synthesis.

To understand the steroidogenic activities and plasma vitellogenin (VTG) profiles according to the reproductive phases in the oblong rockfish Sebastes oblongus, we examined changes in sex steroid hormones and plasma vitellogenin. Plasma levels of testosterone (T) was significantly higher value in only ovulation stage (P<0.05). In vitellogenesis, plasma estradiol-$17{\beta}$ ($E_2$) had a high level in August which was a similar higher level until ovulation than other ovarian development stages (P<0.05). However, $E_2$ was significantly decreased after embryo stage (P<0.05). This indicates that variability in $E_2$ at different stage is associated with the development of the oocytes. Plasma levels of $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) were significantly high at the stages of vitellogenesis and ovulation (P<0.001). It is assumed that DHP plays an important role in vitellogenesis. Also, We determined the plasma levels of vitellogenin (VTG) divided the development stage into four steps: immaturation, vitellogenesis, and ovulation and parturition. A significant lower levels of VTG were shown in immaturation and parturition (P<0.05), which did not discriminate between them. However, in vitellogenesis and ovulation were shown in a remarkable higher levels of VTG(P<0.05), but not significantly different between them. Consequently, plasma VTG levels were considerably increased after October and maintained a higher concentration until ovulation, but significantly decreased after ovulation. It is suggested that VTG plays also an important role in the development of vitellogenesis and oogenesis.
Gonad and blood samples were taken from the cultured female Korean sea bass, Lateolabrax japonicus from October to February between 1997 and 1999. Gonadosomatic index began to increase in November and reached the highest value in December (12.8$\pm$1.5) and January (14.8$\pm$3.5), and then decreased sharply in February (2.6$\pm$1.5, p<0.05). The ovarian oocytes developed to tertiary yolk stage and reached fully-Brown stage in December and January, and then underwent atresia without maturation and ovulation in February. The plasma estradio3-17 $\beta$ level increased from November, and reached the highest value in December (1,152.3$\pm$107.2 pg/ml) and January (1,315.4$\pm$99.5 pg/ml), after then decreased in February (P<0.05). The concentration of plasma 17 $\alpha$ ,20 $\beta$-dihydroxy-4-pregnen-3-one was not significantly changed at low levels (86.6$\pm$6.5∼93.8$\pm$2.8 pg/ml) during the experimental period (P<0.05). All the fish with fully-grown oocytes in the ovary were matured and ovulated by HCG injection. The number of floating eggs were 325,000$\pm$26,000 at HCG 1,000 luhg and 195,000$\pm$35,000 at 2,000 lUikg. There was no difference in fertilization rate and hatching rate of the eggs (P<0.05). Considering these results, we could infer that the ovarian oocyte of the cultured Korean sea bass were not matured and ovulated because of the lack of gonadotropin surge. Moreover, HCG injection could induce oocyte maturation and ovulation in the cultured fish, and the effective dose was 1,000 IU/kg.
This study was conducted to investigate the effects of castration and ovariectomy on growth performance and plasma hormone concentration in pigs. A total of 48 pigs of 35 days of age were used. The results obtained in the present study are summarized as follows: 1. No significant difference was found in average daily gain between ovariectomy group (898.6g) and control gilt group (862.7g), and between castration group (926.0g) and control boar group (945.5g), respectively. Average daily gain of control boar group, however, was significantly higher than that of control gilt group (p<0.05). There was no significant difference in feed/gain between ovariectomy and control gilt group and between castration and control boar group, respectively. Backfat thickness was significantly (p<0.05) higher in ovariectomy or castration group than in control gilt or boar group, respectively. 2. Plasma concentration of IGF-I was significantly (p<0.05) increased during the period of 5 weeks of age (45.1 $\pm$0.72 ng/ml) to 15 weeks of age (356.3$\pm$3.05 ng/ml), and maintained constantly afterwards in control gilt group, as was in control boar group. That of ICF-I tended to be lower in ovariectomy or castration group than in control gilt or boar group, respectively. Regarding steroid hormones of estradiol-17$\beta$, progesterone, and testosterone, the concentration was extremely low at 5 weeks of age, however, increased from 11 weeks to 23 weeks of age in control gilt or boar group, while it was nearly under detection limit in ovariectomy or castration group. 3. Chemical compositions of pork loins were not affected by ovariectomy or castration, except that crude ash content was significantly (p<0.05) higher in castration group than in control boar group. These results indicated that ovariectomy or castration had no effects on growth performance and feed utilization. However, the concentration of sex steroid hormones was under detection limit in ovariectomy and castration group. Further studies, however, are needed to develope the techniques which minimize the stress related with castration or ovariectomy for the production of high quality pork.
The present studies were carried out to examine the effects of exogenous oxytocin(OT) on plasma, uterine and placenta of estradiol-17$\beta$, progesterone, prostaglandin F$_2$$_{\alpha}$ (PGF$_2$$_{\alpha}$), Prostaglandin E$_2$(PGE$_2$) and OT receptor concentrations in pregnant rats. Pregnant rats received an injection of exogenous OT on days 14, 16, 18, 20, 22 of pregnancy and day 1 of postpartum. Concentrations of plasma estradiol-17 $\beta$ after OT injection started to increase after day 18 and peaked on day 22 of pregnancy but decreased on day 1 of postpartum. Plasma progesterone concentrations declined gradually from day 18 of pregnancy and decreased more rapidly until postpartum 1 day. Concentrations of estradiol-17$\beta$in uterine tissues after OT injection were sharply increased from day 20 to 22 of pregnancy and progestrone concentrations were peaked on day 16 and decreased rapidly from day 16 to 20 and maintained the same level until day 1 of postpartum. Uterine concentrations of PGF$_2$$_{\alpha}$ and PGE$_2$increased gradually until day 20 and peaked on day 22 of pregnancy but showed a marked decrease on day 1 of postpartum. Concentrations of PGF$_2$$_{\alpha}$ in placental tissues increased rapidly from day 14 of pregnancy and decreased sharply on day 1 of postpartum. Concentrations of PGE$_2$increased gradually after day 14 and peaked on day 20 of pregnancy. The concentration of OT receptor in uterus was significantly elevated from day 20 and rose to maximum on day 22 of pregnancy. These findings show that OT suppress the concentration of progestrone and stimulate productions of estradiol-17 $\beta$, PGF$_2$$_{\alpha}$, PGE$_2$ and oxytocin receptor concentrations in pregnant rats.

A study was conducted to investigate the effect of induced molting on the relative weight and hormone levels of thyroid, ovary, and adrenal glands in spent laying hens. Three hundred sixty 77-wk-old, Babcock White hens were divided into 36 experimental units of 10 hens each and induced to molt for seven weeks. A diet containing 16% CP and 2,800 kcal ME/kg was fed ad libitum from 84 to 126 weeks of age. Thirty-six birds were randomly selected for blood collection and slaughtered at Pre-molt, 5% egg Production, Peak, and end Phase of the egg Production. Weights of the thyroid, ovary, adrenal glands, and plasma T3, T4, cortisol, and estradiol contents were measured at each stage. Weight of the thyroid reached its highest level at 5% e99 Production stage. The ovary weight was greatly reduced during molting, but started to increase after induced molting until the end of the egg Production Phase (P<0.05). Plasma T3,T4 and cortisol were found to be the highest at the start of the Post-molt Production stage, but reached their minimum at the end Phase of e99 Production. Plasma estradiol was the lowest at the 5% egg Production stage. The Present study demonstrated that molting is a complex Process that require the involvement of endocrine glands to trigger their specific hormones, which play a key role in molt induction.
A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.
