다시마와 혈당강하제가 당뇨쥐의 지질대사에 미치는 영향 (Effect of Sea Tangle and Hypoglycemic Agent on Lipid Metabolism in Diabetic Rats)

  • The present study was conducted to investigate the effect of sea tangle and hypoglycemic agent on lipid metabolism in normal and dabetic rats. Male Sprague-Dawely rats were fed AIN-76 based experimental diets containing 5%(w/w) cellulose or 15%(w/w) sea tangle for 3 weeks, after which diabetic groups were made diabetic by intramuscular injection of streptozotocin(STZ, 45mg/kg BW). Metformin(350mg/kg BW) as a hypoglycemic agent was given once a day using a feeding tube for 5 days. Body weight grains were reduced significantly by STZ treatment, but not influenced by metformin feeding. Blood glucosel levels in sea tangle groups were reduced, compared with those in cellulose groups. Metformin feeding showed the lowering effect of blood glucose. Plasma levels of triglyceride were increased significantly in diabetic rats, but decreased in metformin group by sea tangel feeding. Total cholestero contents showed a similar tendency with triglyceride, but were reduced in diabetic groups without metformin by sea tangle feeding. Plasma levels of HDL-cholesterol were reduced in diabetic rats, compared with those in normal rats. There was a significant increase in fecal weights in diabetic rats fed sea tangle. Fecal contents of cholesterol were lower in diabetic rats than in normal rats. In normal rats, it tended to increase by sea tangle feeding, but not significantly. Fecal excretions of coprostanol and coprostanone were reduced significantly in diabetic rats, compared with those of normal rats. It tended to increase in diabetic rats by simultaneous feeding of sea tangle and metformin, but not significantly. Diabetes reduced fecal excretion of bile acid, but it was increased by sea tangle and metformin feeding.

레반 Diet 섭취에 의한 한국 여성의 체지방 축적 억제와 혈중 지질의 개선 효과 (The Effects of Fructose Polymer Levan on the Body Fat Accumulation and Serum Lipid Profiles of Korean Women)

  • This study was performed to investigate the effects of a levan diet on the body fat accumulation and serum lipid composition of 29 Korean women over a period of up to 12 weeks (n=13 for the control group, n=16 for the levan group). The subjects ate an uncooked diet (6 g) with 400$m\ell$ of tap water twice a day. The mean body weight and height measurements of the subjects (levan group) at the outset were 66.0$\pm$8.8kg and 156.7$\pm$5.3cm, respectively. The subjects showed a significant reduction in weight, body fat mass, anthropometric measurements and skinfold thickness during the experimental period. The waist hip ratio (WHR) was 0.88$\pm$0.03 at the outset, and fell to 0.82$\pm$0.05 after 4 weeks. The intake of levan was also influenced on the levels of serum Fe, leptin, lipoprotein lipase, HDL-cholesterol, LDL-cholesterol, and triglyceride. The serum glucose levels were within the normal range during the experimental period. The initial serum triglyceride level was 121mg/dl, but fell to 103mg/dl after 4 weeks of levan supplementation. The current study demonstrates that a levan diet is effective in controlling weight, body fat, HDL-cholesterol, LDL-cholesterol, and triglyceride levels.

Cholesterol-lowering Effect of Rice Protein by Enhancing Fecal Excretion of Lipids in Rats

  • The aim of this study was to investigate the effects of isolated protein from white rice on lipid metabolism in a hypercholesterolemic animal model. Male Sprague-Dawley rats were divided into three groups and fed either a normal diet or a high-cholesterol diet (HCD) containing either casein or isolated rice protein for 4 weeks. Compared with rats fed a HCD with casein, the total cholesterol (TC) level in the plasma was significantly reduced in the rats fed rice protein. However, no significant differences were observed in the triglycerides, high-density lipoprotein (HDL), and glucose levels among the experimental groups. Hepatic total lipids and TC levels were significantly decreased by supplementation with rice protein. In addition, rice protein significantly increased the levels of TC and bile acids in the feces. These results suggest that rice protein may improve HCD-induced hypercholesterolemia by enhancing fecal excretion of cholesterol.

The Hypoglycemic Effect of Saururus chinensis Baill in Animal Models of Diabetes Mellitus

  • The purpose of this study was to investigate the hypoglycemic effect of Saururus chinensis Baill in vitro and in vivo. Methanol extract of S. chinensis Baill inhibited yeast ${\alpha}$-glucosidase activity by 49.8%, which was twice as strong as that of acarbose at a concentration of 0.5 mg/mL in vitro. The effect of S. chinensis Baill methanol extract on the postprandial increase in blood glucose levels was studied in streptozotocin-induced diabetic rats using a carbohydrate load test. Oral administration of S. chinensis Baill extract (500 mg/kg) significantly decreased incremental blood glucose levels at 60 and 90 min (p<0.05) after oral ingestion of starch (1 g/kg). The area under the glucose response curve of the S. chinensis Baill group was significantly decreased compared to that of the control group (p<0.05). The effect of prolonged feeding of S. chinensis Baill was studied in an animal model of type 2 diabetes. Three-week-old db/db mice were fed an AIN-93G diet or a diet containing 0.5% S. chinensis Baill extract for 7 weeks after 1 week of adaptation. Plasma glucose, insulin, and blood glycated hemoglobin levels of the mice fed S. chinensis Baill extract were significantly lower than those of the control group (p<0.05). Therefore, we conclude that S. chinensis Baill is effective in controlling hyperglycemia in animal models of diabetes mellitus.

Effect of Chromium Dietary Supplementation on the Immune Response and Some Blood Biochemical Parameters of Transport-stressed Lambs

  • Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km from Al-Jouf to Riyadh, Saudi Arabia. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 and 0.9 ppm organic chromium (Cr). Each group consisted of four separately housed replicates of three lambs each. The animals were fed ad libitum on a grower diet for 84 days. Blood samples were obtained shortly before transportation, upon arrival and at weekly intervals thereafter from all lambs for analysis of plasma and serum. Plasma glucose and serum cortisol, total protein, albumin, urea-N and total cholesterol concentrations were determined. A cursory clinical examination of the lambs, along with rectal temperature, was undertaken at different intervals during the experiment. The lambs were inoculated each with 2 ml i.v. chicken red blood cells (CRBC) on days 0, 21, and 42. Serum total, IgG and IgM antibody titers were determined at weekly intervals post-immunization. An in vivo intradermal hypersensitivity test was carried out on 6 lambs from each group on days 10 and 70. Transportation of the lambs resulted in a significant (p<0.001) elevation of serum cortisol, total protein and albumin levels, as well as increased plasma glucose concentration, with corresponding decrease in total cholesterol, while blood urea-N remained largely unchanged. These constituents returned to normal levels during subsequent weeks, with no significant differences in their concentrations being observed between the Cr-supplemented groups and controls. Rise in rectal temperature after transportation was reduced to a greater extent (p<0.05) in Cr-supplemented versus control lambs. Total, IgG and IgM antibody titers against CRBC rose significantly (p<0.05) during immunizations in all groups, with significantly and linearly higher (p<0.05) total and IgG titers in Cr-supplemented versus control lambs. By contrast, no significant effect due to Cr supplementation was recorded in IgG titers, which increased equally in Cr-fed and control groups. Skin thickness in response to intradermal inoculation of phytohaemagglutinin (PHA) was also significantly (p<0.01) increased as a result of Cr supplementation. These results indicate that dietary Cr supplementation might be useful during stress especially for enhancing immune responses in transport-stressed lambs.

HD047703, a New Promising Anti-Diabetic Drug Candidate: In Vivo Preclinical Studies

  • G-protein coupled receptor 119 (GPR119) has emerged as a novel target for the treatment of type 2 diabetes mellitus. GPR119 is involved in glucose-stimulated insulin secretion (GSIS) from the pancreatic b-cells and intestinal cells. In this study, we identified a novel small-molecule GPR119 agonist, HD047703, which raises intracellular cAMP concentrations in pancreatic ${\beta}$-cells and can be expected to potentiate glucose-stimulated insulin secretion from human GPR119 receptor stably expressing cells (CHO cells). We evaluated the acute efficacy of HD047703 by the oral glucose tolerance test (OGTT) in normal C57BL/6J mice. Then, chronic administrations of HD047703 were performed to determine its efficacy in various diabetic rodent models. Single administration of HD047703 caused improved glycemic control during OGTT in a dose-dependent manner in normal mice, and the plasma GLP-1 level was also increased. With respect to chronic efficacy, we observed a decline in blood glucose levels in db/db, ob/ob and DIO mice. These results suggest that HD047703 may be a potentially promising anti-diabetic agent.

  • Rice(Oryza stiva L.) is a major cereal food providing nourishment to over half of the world's populations and was considered only as a source of energy. However, The recent many researches have been made to suggest that rice may relate to prevention chronic disease and health-promoting properties such as postprandial glucose response, hypocholesterolemic effect and blood pressure-lowering effect. There has been numerous observation supporting that rice has hypocholesterolemic effect. Rice, rice bran, rice bran oil and unsaponifiable matter of rice bran oil reduced plasma cholesterol in rat, hamster as well as human. Components of rice showing hypocholesterolemic effect include dietary fiber(hemicellulose, phytic acid). protein, ${\gamma}$-oryzanol, $\beta$-sitosterol, and tocotrienols. Crapo et al has been studied that the effect of various of starchy foods on the postprandial blood glucose and insulin responses in healthy and diabetic humans. The results showed that rice had lower blood glucose and insulin responses compared to potato, bread and dextrose. The different physical forms in the same starch also produce the different postprandial glucose and insulin responses. In recent years, several studies have shown that some components of rice have potent antioxidant activity against Fe$^{2+}$ -ascorbate induced lipid peroxidation in rat liver microsomal membranes. Cell culture and animal studies have shown that some components of rice have inhibitory effect on the growth and proliferation of several types of human cancer cell. It was also reported that the methanol extract of brown rice has antimutagenic activity against various mutagens. In addition, the pepsine hydrolysate from rice protein is reported to inhibit angiotensin converting enzyme activity. GABA (${\gamma}$ - aminobutyric acid) and GABA enriched rice germ is also effective for lowering blood pressure and triglyceride levels.s.

레이저 제2형 당뇨동물모델에서 macelignan과 한약제 열수 추출물의 병용효과 (The Hypoglycemic Effect of Complex of Chinese Traditional Herbs (CTH) and Macelignan in Type 2 Diabetic Animal Model)

  • 본 연구는 macelignan과 CTH를 제2형 당뇨병동물모델에 3주간 병용 투여하여 macelignan을 단독으로 투여했을 때보다 더 나은 효과가 있는지를 살펴 보았다. Macelignan과 CTH의 병용 투여는 공복혈당과 내당능을 비롯하여 혈중 free fatty acids, HTR, 그리고 AI에서는 macelignan 단독 투여군과 비교하여 유의적으로 나타나는 큰 차이를 발견할 수는 없었다. 그러나, HbA1c, insulin 민감도, 그리고 혈중 triglyceride 함량에서는 유의적인 변화를 보이며 효과가 있는 것으로 나타났다. 따라서, macelignan의 단독 투여보다 CTH를 병용 투여하는 것이 당뇨병 합병증을 예방하거나 개선시킬 수 있는 잠재력이 있을 것으로 판단된다. 이에 보다 명확하게 검증하기 위해 혈당 및 대사조절 기전연구를 위한 further study가 진행되어야 할 것으로 사료된다.

Psychosocial support interventions for women with gestational diabetes mellitus: a systematic review

  • Purpose: This study aimed to analyze the content and effectiveness of psychosocial support interventions for women with gestational diabetes mellitus (GDM). Methods: The following databases were searched with no limitation of the time period: Ovid-MEDLINE, Cochrane Library, Ovid-Embase, CINAHL, PsycINFO, NDSL, KoreaMed, RISS, and KISS. Two investigators independently reviewed and selected articles according to the predefined inclusion/exclusion criteria. ROB 2.0 and the RoBANS 2.0 checklist were used to evaluate study quality. Results: Based on the 14 selected studies, psychosocial support interventions were provided for the purpose of (1) informational support (including GDM and diabetes mellitus information; how to manage diet, exercise, stress, blood glucose, and weight; postpartum management; and prevention of type 2 diabetes mellitus); (2) self-management motivation (setting goals for diet and exercise management, glucose monitoring, and enhancing positive health behaviors); (3) relaxation (practicing breathing and/or meditation); and (4) emotional support (sharing opinions and support). Psychosocial supportive interventions to women with GDM lead to behavioral change, mostly in the form of self-care behavior; they also reduce depression, anxiety and stress, and have an impact on improving self-efficacy. These interventions contribute to lowering physiological parameters such as fasting plasma glucose, glycated hemoglobin, and 2-hour postprandial glucose levels. Conclusion: Psychosocial supportive interventions can indeed positively affect self-care behaviors, lifestyle changes, and physiological parameters in women with GDM. Nurses can play a pivotal role in integrative management and can streamline the care for women with GDM during pregnancy and following birth, especially through psychosocial support interventions.

Animal protein hydrolysate reduces visceral fat and inhibits insulin resistance and hepatic steatosis in aged mice

  • BACKGROUND/OBJECTIVES: An increasing life expectancy in society has burdened healthcare systems substantially because of the rising prevalence of age-related metabolic diseases. This study compared the effects of animal protein hydrolysate (APH) and casein on metabolic diseases using aged mice. MATERIALS/METHODS: Eight-week-old and 50-week-old C57BL/6J mice were used as the non-aged (YC group) and aged controls (NC group), respectively. The aged mice were divided randomly into 3 groups (NC, low-APH [LP], and high-APH [HP] and fed each experimental diet for 12 weeks. In the LP and HP groups, casein in the AIN-93G diet was substituted with 16 kcal% and 24 kcal% APH, respectively. The mice were sacrificed when they were 63-week-old, and plasma and hepatic lipid, white adipose tissue weight, hepatic glucose, lipid, and antioxidant enzyme activities, immunohistochemistry staining, and mRNA expression related to the glucose metabolism on liver and muscle were analyzed. RESULTS: Supplementation of APH in aging mice resulted in a significant decrease in visceral fat (epididymal, perirenal, retroperitoneal, and mesenteric fat) compared to the negative control (NC) group. The intraperitoneal glucose tolerance test and area under the curve analysis revealed insulin resistance in the NC group, which was alleviated by APH supplementation. APH supplementation reduced hepatic gluconeogenesis and increased glucose utilization in the liver and muscle. Furthermore, APH supplementation improved hepatic steatosis by reducing the hepatic fatty acid and phosphatidate phosphatase activity while increasing the hepatic carnitine palmitoyltransferase activity. Furthermore, in the APH supplementation groups, the red blood cell (RBC) thiobarbituric acid reactive substances and hepatic H2O2 levels decreased, and the RBC glutathione, hepatic catalase, and glutathione peroxidase activities increased. CONCLUSIONS: APH supplementation reduced visceral fat accumulation and alleviated obesity-related metabolic diseases, including insulin resistance and hepatic steatosis, in aged mice. Therefore, high-quality animal protein APH that reduces the molecular weight and enhances the protein digestibility-corrected amino acid score has potential as a dietary supplement for healthy aging.
