The purpose of this study was to develop a system dynamics model for management of glucose metabolism disorders that demonstrated dynamic relationships between insulin and plasma glucose levels over the time. The model was developed to 1) represent the physiology of glucose metabolism for an normal adult subject, 2) to draw causal loop diagram that demonstrate feedback systems of glucose regulation in normal condition and pathologic condition of the type 2 diabetes, 3) to develop an interactive computer simulation model for management of glucose metabolism disorders. The simulation results showed the plasma glucose level for normal persons varied from 75 to 140 which was consistent with clinical findings. As an example for patients we selected a case which varied from 110 to 310. Two types of interventions were chosen to review the model; meal control and insulin administration. The simulation results for those cases also matched well with clinical findings. The developed model can be used as an effective educational tool for patients to develop healthy lifestyle choices. The results also provide a blueprint for health providers to maintain normal blood glucose levels in diabetes patients.
Background: Chronic inflammation could affect the occurrence and development of malignant tumors. To explore the levels of tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$) and C-reactive protein (CRP) in patients accompanied by impaired glucose tolerance (IGT) and their clinical significance. Materials and Methods: A total of 210 patients hospitalized in Affiliated Hospital of Taishan Medical University from Jun., 2013 to Dec., 2014 were selected, in which 92 cases were accompanied by IGT. Meanwhile, 80 randomly-selected healthy people by physical examination were as the control. The levels of routine biochemical indexes, plasma TNF-${\alpha}$ and CRP in all subjects were measured. Results: Both systolic and diastolic pressures in hypertension group and hypertension plus IGT group were significantly higher than in control group (p<0.01), but there was no statistical significance between these two groups (p>0.05). The levels of fasting plasma glucose (FPG) and blood glucose 2 h after taking glucose in hypertension plus IGT group were markedly higher than other groups (p<0.01). Homeostasis model assessment-insulin resistance (HOMA-IR), TNF-${\alpha}$ and CRP contents were on the progressive increase in control, hypertension and hypertension plus IGT groups, but significant differences were presented among each group (P<0.01). Hypertension accompanied by IGT had a significantly-positive association with CRP, TNF-${\alpha}$, FPG and blood glucose 2h after taking glucose. Conclusions: The levels of plasma TNF-${\alpha}$ and CPR in patients with hypertension accompanied by IGT increase significantly, indicating that inflammatory reaction in these patient increases, thus suggesting that these patients should be focused regarding cancer prevention.
The ability of exercise and buckwheat diet to modify plasma glucose and cholesterol levels in streptozotocin-induced diabetic rats has been studied. Diabetic rats were fed corn starch as a control diet or buckwheat as an experimental diet for 4 weeks. One group of rats were exercise-trained to swim for 60 min/day, 6 days a week. Plasma glucose levels of sedentary rats both on the control diet and on the buckwheat diet were significantly increased from 367.0$\pm$33.6 mg/dl to 545.0$\pm$38.7 mg/dl and from 380.3$\pm$18.9 mg/dl to 540.5$\pm$36.6 mg/dl, respectively. However, this large increase of plasma glucose was not seen in exercised rats on the buckwheat diet (from 345.0$\pm$25.6 mg/dl to 391.4$\pm$34.7 mg/dl). The total plasma cholesterol level was not affected by either diet or exercise. The HDL-cholesterol level tends to increase due to the buckwheat diet or an exercise, but not with a significant increase. Our results suggest that the buckwheat diet is beneficial in lowering the plasma glucose level only when diabetic rats ar exercised.
The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.
Background: Preoperative blocking of surgical nociceptive inputs may prevent sensitization of CNS and reduce postoperative pain. The stress responses to surgical trauma consist of increase in catabolic hormones and decrease in anabolic hormones. We studied whether preoperative intravenous morphine could affect postoperative pain and change plasma cortisol and serum glucose levels. Methods: Thirty eight patients undergoing total abdominal hysterectomy were randomly assigned to one of three groups. Control group (n=11) did not received intravenous morphine, preoperative group (n=13) received intravenous morphine (0.1 mg/kg as a bolus 10 min before operation and followed by 1.5 mg/hr for 10 hours), postoperative group (n=14) received the same doses and method of intravenous morphine of preoperative group postoperatively. Postoperative pain relief was provided with i.v. fentanyl through Patient-Controlled-Analgesia Pump. Postoperative visual analogue scores (VAS), analgesic requirement (first request time, total amounts used), side effects, plasma cortisol and serum glucose levels were compared. Results: VAS were different between control group and the other two goups, but were not different between preoperative and postoperative group. Total amounts of used fentanyl were not different among groups, but first request time were significantly delayed in the preoperative group compared with the other two groups ($66.2{\pm}33.9$ vs $39.0{\pm}15.4$ and $45.0{\pm}14.9$ min respectively, p<0.05). Plasma cortisol and serum glucose levels were not different among groups. Conclusions: Above dosage of preoperative and postoperative morphine has analgesic effect, but could not block surgical stress induced plasma cortisol and serum glucose increase.
In this study, the two sampling methods for blood glucose measurement were evaluated in order to reduce pre-analytical error. For this purpose, glucose levels of Li-heparinized plasma (LHP) and Na-fluorinated plasma (NFP) in blood collected from healthy volunteers were determined by using routine hexokinase procedures. The percentage range of pre-analytical error was quantitated by comparing LHP glucose values with NFP glucose values according to delaying analytical time (0, 60, and 120 min). LHP glucose values were decreased by 6.6% in 60 min., 17.7% in 120 min, whereas NFP glucose values decreased by 1.1% in 60 min, 2.0% in 120 min. Therefore it may be recommended that the NFP sampling method should be used for routine blood glucose determination in diabetes mellitus diagnosis.
Anti-diabetic effect of the exopolysaccharides (EPS) produced from submerged mycelial culture of Cordyceps sinensis (Cs) was studiedin a type II diabetic animal model (C57BL/6J ob/ob). This study was designed to determine whether Cs-EPS improves clinical symptoms of type II diabetes in ob/ob mice. After Cs-EPS treatment at doses of 200 mg/kg body weight, the fasting blood glucose levels decreased by 47% after 7 weeks compared with those of the control mice. According to the oral glucose tolerance test, the glucose levels recovered its baseline after 120 min in Cs-EPS-treated mice, although the blood glucose levels increased significantly after 30 min. On the other hand, the control group (not-treated) did not recovered its initial level of glucose after 120 min. Furthermore, food intake, body weight, total plasma cholesterol and triglyceride concentrations in ob/ob mice treated with Cs-EPS were significantly decreased, compared with those in control ob/ob mice. Cs-EPS treatment increased significantly the plasma insulin level and the expression of leptin mRNA in adipose tissue of Cs-EPS-treated ob/ob mice. From these results, it is demonstrated that Cs-EPS could be effective for regulating normal blood glucose levels by increasing the amounts of plasma insulin and leptin expression in ob/ob mice, indicating that this compound could be a candidate material as a dietary supplement to control hyperglycemia in patients suffering from type II diabetes.

The black seabream Acanthopagrus schlegelii is an important species for aquaculture in Korea. There are, however, no reports on the physiological responses to hypoxia in this species. The objective of this study was to evaluate the effects of acute hypoxia on the physiological stress responses (plasma cortisol as the primary response, and plasma glucose, hematocrit (Ht), hemoglobin (Hb), sodium, chloride, osmolality, AST, ALT and total protein as the secondary responses) of the fish. The experimental fish were exposed to 0.5 ppm dissolved oxygen (DO) in the seawater via two methods (progressive stepwise decline (Exp. I), and direct decline (Exp. II)). A highly significant increase was detected in plasma cortisol levels due to the hypoxia treatments, and fish in Exp. I had hormone levels that were significantly higher than the fish in Exp. II. For plasma glucose, there was no significant difference between the Exp. I fish and the control fish, whereas Exp. II fish showed significantly higher plasma glucose levels than Exp. I fish and control fish. Ht values increased in both hypoxia treatments; however, Hb concentrations increased only in Exp. I. Although plasma chloride levels were unaffected by acute hypoxia, plasma sodium and osmolality levels increased in Exp. I. Progressive hypoxia (Exp. I fish) increased plasma AST, ALT and total protein. These results suggest that the value and direction of changes to the investigated parameters can be used to determine the resistance of black seabream to acute hypoxia.
Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-245g by injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg and were divided into seven groups ; normal, diabetic control, and five experimental groups(fractions of hexane, chloroform(CHCl$_3$), ethylacetate(EtOAc), butanol(BuOH) and water($H_2O$)). The rats of all groups were fed on AIN-93 diet and the five experimental groups were orally administered each fraction for 14 days. The body weight and diet intake were monitored daily. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol, free fatty acid and aspartate and alanine aminotransferase activities were analyzed. Diabetic rats showed the lower weight gain compared to the normal rats. The plasma glucose levels of the CHCl$_3$, and $H_2O$ fraction groups were significantly lower than the other experimental groups. The plasma insulin level of the CHCl$_3$ fraction group was much higher than that of diabetic control group. The plasma cholesterol levels were increased in all the experimental groups. The groups of hexane, BuOH and H2() fractions showed the lower plasma triglyceride levels compared to diabetic control group. The plasma free fatty acid levels were not significantly different in all groups. HDL-cholesterol levels were definitely higher in hexane, CHCl$_3$ and EtOAc fraction groups than that of diabetic control group. In conclusion, administration of CHCl$_3$ and $H_2O$ fractions of methanol extract of Alisma canaliculatum exhibited hypoglycemic effects in STZ induced diabetic rats, showing the possibility of therapeutic use of Alisma canaliculatum to the diabetes mellitus.
This study investigates the effects of a raw diet (RD) on blood glucose and lipid metabolism in non-diabetic (normal) and streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley vats were assigned to four groups (normal control, normal RD, diabetic control, and diabetic RD), for the four-week experimental period. The control groups were fed the AIN diet and the RD groups were fed a diet consisting only of raw materials. Weight gain was statistically lower in the RD group than the control. fasting plasma glucose was significantly lower in the diabetic RD group compared to the diabetic control group. The levels of triglycerides (TG), and of total cholesterol (TC) and LDL-cholesterol in the plasma, were lower in the RD groups than the control groups, but not significantly. There was a statistically significant decrease in the levels of TG and TC in the livers of the diabetic RD group, compared to the diabetic control group. The fecal levels of total lipids, TG, and TC were significantly higher in the RD groups, compared to the non-RD groups. It can be postulated that this raw diet may possess substantial hypoglycemic/hypolipidemic properties in diabetic rats.
