Influence of Chlordiazepoxide on the Changes of Blood Sugar and Plasma Corticosterone Level Induced by ACTH and Picrotoxin in Mice (ACTH와 Picrotoxin에 의(依)한 혈당(血糖)과 혈장(血漿) Corticosterone 치(値)의 변동(變動)에 미치는 Chlordiazepoxide의 영향(影響))

  • Marc et al. reported that diazepam increased plasma corticosterone level and Dasgupta et al. suggested that chlordiazepoxide(CDP) supressed the adrenal response to ACTH. In this paper, the influence of CDP on the changes of blood sugar and plasma corticosterone level induced by ACTH and picrotoxin were investigated in male mice. The results obtained were summarized as follows; 1) The blood sugar and plasma corticosterone level were increased by CDP, ACTH, and picrotoxin, respectively. 2) The hyperglycemia induced by ACTH and picrotoxin were not affected by the CDP pretreatment. 3) The increase of plasma corticosterone level induced by ACTH was inhibited by the CDP pretreatment. 4) The increase of plasma corticosterone level appeared 30 minutes after picrotoxin injection was slightly enhanced, but the level of 120 minutes after picrotoxin injection was significantly inhibited by the CDP pretreatment.

  • The influences of diazepam and naloxone on the increase of plasma corticosterone level induced by morphine, pentazocine, ACTH, or picrotoxin were investigated in male mice. The results obtained were summarized as follows: 1) The increase induced by morphine or pentazocine of plasma corticosterone level was not affected by naloxone pretreatment but markedly suppressed by diazepam pretreatment. 2) The increase induced by ACTH of plasma corticosterone level was not affected by diazepam or naloxone pretreatment. 3) The picrotoxin markedly increased plasma corticosterone level, and the inceease was not affected by diazepam or naloxone pretreatment. This above results suggest that the increase induced by opioids of plasma corticosterone level seems to be rather related with other than opiate- or GABArerecptor.

Effect of Each Constituent-Herb of Sagoonja-Tang on the Change of Corticosterone Level induced by Cold Stress in Mice (Cold Stress로 유발된 생쥐의 혈중 corticosterone 농도변화에 대한 사군자탕(四君子湯) 구성약물의 효능)

  • This study aimed to investigate the effects of each constituent-herb of Sagoonja- Tang on the plasma corticosterone level in mice exposed to cold stress. Sagoonja-Tang is composed of Ginseng Radix, Atractylodis Radix, Poria, and Glycyrrhizae Radix. Each constisuent-herb(0.5g/kg, 1.0g/kg) was administered intragastrically to mice 1hr prior to expose to forced cold water swimming($4{\circ}C$, 3min) before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. At 30 minutes after the cold stress for 3 minutes the most significant increase of corticosterone level was shown. 2. Ginseng group(PGR group) didn't show any significant changes. 3. Atractylodis group(AR group) showed significant decrease at 0.5g/kg dose (P<0.01). But it didn't show significant changes at 1.0g/kg dose. 4. Poria group(P group) didn't show significant changes. But it showed slightly increase at 1.0g/kg dose. 5. Glycyrrhizae group(GR group) didn't show significant changes. These data revealed that Atractylodis macrocephalae Rhizoma might have the anti-stress effect through the control of plasma corticosterone level and the dose of herbs had a different effect on the plasma corticosterone level in mice exposed to cold stress.

  • This study aimed to investigate the effect of each constituent-herb of Sagoonja-Tang on the plasma corticosterone level in mice exposed to heat stress. Each formula(0.5g/kg, 1.0g/kg) was injected intragastrically to mice exposed to forced hot water swimming(42$^{\circ}C$, 3min) before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. At 15 minutes after the heat stress for 3 minutes the most significant increase of corticosterone level was shown. 2. Ginseng group didn't show significant changes. 3. Atractylodis group showed significant decrease at 0.5g/kg dose(P<0.05). But it showed slightly increase at 1.0g/kg dose. 4. Poria group didn't show significant changes. 5. Glycyrrhizae group showed slightly increase, but insignificant. These data revealed that Atractylodis macrocephalae Rhizoma might have the anti-stress effect through the control of plasma corticosterone level in mice exposed to heat stress.

Yookmijihwang-Tang on the Plasma Corticosterone level in Mice exposed to Heat, Cold and Immobilization Stress (한(寒), 열(熱), 구속(拘束) 스트레스에 대한 사군자탕(四君子湯), 사물탕(四物湯), 육미지황탕(六味地黃湯)의 효능(效能))

  • In order to investigate the effect of Sagoonja-Tang, Samool-Tang and Yookmijihwang-Tang against the cold, heat and immobilization stress, each formula was injected intragastically to mice exposed to forced cold water swimming($4^{\circ}C$), forced hot water swimming($42^{\circ}C$) and immobilization stress before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. One hour after each Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administered to mice, we put cold stress($4^{\circ}C$) on mice for 3min. Then those prescriptions didn't show any significant effect on plasma corticosterone level. however, Yookmijihwang-Tang(1g/kg) showed a significant increase(p〈0.05), and Samool-Tang(3g/kg) showed a small increase. 2. One hour after Samool-Tang, Sagoonja-Tang and Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administerd, mice exposed to heat stress($42^{\circ}C$) for 3 min. Samool-Tang(1g/kg) and Yookmijihwang-Tang(lg/kg) showed significant decrease of corticosterone level(p〈0.01, p〈0.05) but Sagoonja-Tang didn't change it. In the case of the dose of 3g/kg, only Samool-Tang decreased corticosterone level(P〈0.01), but Sagoonja-Tang, Yookmijihwang-Tang didn't make significant effect on plasma corticosterone level of the mice exposed to heat stress($42^{\circ}C$ ). 3. Those mice which exposed to immobilization stress one hour after Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg and 3g/kg didn't show effective decrease of corticosterone level. However, Sagoonja-Tang(3g/kg), Samool-Tang(lg/kg, 3g/kg) showed the effect of mild increase of corticosterone level rather than decrease. These data revealed that the effect-variation of prescriptions depended on the kinds of stress and the dose of prescriptions had a different effect on therapy.

  • In this paper, the influence of phenytoin and phenobarbical on the changes of brain norepinephrine(NE) content, plasma corticosterone and blood sugar level in mice were studied. The results obtained were summarized as follows: 1) Phenytoin(50 mg/kg) increased the brain NE content but phenobarbital(50 mg/kg) did not affect. The increase of the brain NE content induced phenytoin was potentiated by phenobarbital pretreatment. 2) Phenytoin(25 mg/kg, 50 mg/kg) markedly increased the level of plasma corticosterone but phenobarbital did not affect. The increase of the plasma corticosterone induced by phenytoin was inhibited by phenobarbital pretreatment. 3) Phenytoin(50 mg/kg) markedly increased the blood sugar level but phenobarbital did not affect. The increase of the blood sugar induced by phenytoin was not affected by phenobarbital pretreatment.

  • This study was performed to investigate what effect the taste and quality of drugs would have on the cold and heat mechanisms of human body. We administered Hwangryeunhaedok-tang composed of bitter-tasted, cold-qualified drugs and Gungangbuza-tang composed of spice-tasted. hot-qualified drugs, respectively to the mice 1 hr before exposure to heat stress or cold stress. Plasma corticosterone level of mice was measured. The results were as follows: 1. The elevated corticosterone level in the mice exposed to heat stress was significantly decreased after administration of Gungangbuza-tang but there was no decrease after administration of Hwangryeunhaedok-tang. 2. The elevated corticosterone level in the mice exposed to cold stress was significantly decreased after administration of Hwangryeunhaedok-tang but there was mild decrease after administration of Gungangbuza-tang. 3. When the doses, 3g/kg and 1g/kg were administerd to mice exposed to heat stress. both dose showed significant decrease of corticosterone level and the dose. 3g/kg was more effective. However, in the mice exposed to cold stress, the dose, 3g/kg showed mild decrease and 1g/kg showed significant decrease. These data suggested that HW decreased the plasma corticosterone level in the mice exposed to cold stress and GB also decreased the plasma corticosterone level in the mice exposed to heat stress. In conclusion, our study revealed that the taste and quality of drugs controled the cold and heat mechanism of human body.

Effects of Coptidis Rhizoma and Aconiti Lateralis Preparata Radix on the Change of Plasma Corticosterone Level and Rectal Temperature Induced by LPS (LPS에 의해 유발된 염증(炎症) 스트레스에 대한 황련(黃蓮)과 부자(附子)의 효과)

  • Objectives : We investigated the effects of Coptidis Rhizoma and Aconiti Lateralis Preparata Radix on the LPS(Lipopolysaccharide) ICV(intracerebroventricular) injection. Methods : We measured plasma corticosterone level and rectal temperature in mice induced by I.C.V. injection of LPS (100ng/mouse). Results : The results were as follows.. 1. The plasma corticosterone levels in CR-1(0.5g/kg), CR-2(1.0g/kg), CR-3(3.0g/kg) were not decreased significant comparing with the control group.(P<0.05) 2. The plasma corticosterone level in AR-1(0.5g/kg) was decreased significant comparing with the control group.(P<0.05), but AR-2(1.0g/Kg) and AR-3(3.0g/kg) were not decreased significant comparing with the control group.(P<0.05). 3. The rectal temperature in CR-1(0.5g/kg), CR-2(1.0g/kg), CR-3(3.0g/kg) was decreased significant comparing with the control group.(P<0.05) 4. The rectal temperature in AR-1(0.5g/kg), AR-2(1.0g/kg), AR-3(3.0g/kg) was not decreased significant comparing with the control group.(P<0.05) Conclusion : These data revealed that Rhizoma Coptidis might have no significant effect on inflammation stress and Aconiti Lateralis Preparata Radix(0.5g/kg/mouse) might have significant effect on inflammation stress.

  • Footpad swelling having the relationship with arthus-reaction of antibody-mediated hypersensitivity and delayed type hypersensitivity was recovered to the almost normal level. The nethemoglobin induced by aniline showed no significant deviation. PFC/spleen cell and PFC/106 spleen cell were increased slightly, but not in case of RFC. Hemagglutination value was increased slightly, but hemolytic value was not changed significantly. Mice in the administration of the polysaccharide does not show any significant stress factor in the cage for mice admlrustered plasma corticosterone. Key words: antibody-mediated hypersensitivity, delayed type hypersensitivity, methemoglobin, Hemagglutination value, hemolytic value, plasma corticosterone.

  • Objective : This study was aimed to evaluate the anti-starvation stress effect of Liriopis Tuber on mice. Method : The first experiment was done to mice which have a high corticosterone level at a short term starvation. The plasma corticosterone level of each mouse was measured over time at 12, 24, 36, 48, 72 hours of starvation respectively. The second experiment was done in the two groups(LT A and LT B) of mice which were in famine for 36.5 hours after being administered Liriopis Tuber three times with different doses(LT A: 1.0 g/.kg and LT B: 3.0g/kg) for three days. The plasma corticosterone levels and rectal temperature were measured when a 36.5-hour of starvation period was finished. Results : 1. The corticosterone level in the mice was significantly increased after a 36.5-hour of starvation. (P<0.001). 2. The plasma corticosterone level was decreased significantly in the group of 3.0g/kg, compared to the control group.(P<0.05) 3. The rectal temperature in the groups of 1.0g/kg and 3.0g/kg were increased slightly in both group. But the two groups didn't show significant change. Conclusion: Based on the above results, Liriopis Tuber might have anti-starvation stress effect and it may be used as anti-starvation stress medicine and anti-stress medicine.
