Plasma Protein Profile of Neonatal Buffalo Calves in Relation to the Protein Profile of Colostrum/Milk during First Week Following Parturition

  • An investigation was made into the protein profile of colostrum/milk of ten Murrah buffaloes and of their ten buffalo calves during their first week of neonatal life to study the materno-neonatal transfer of immunoglobulins (Ig). Calves were pail fed 3.5 liter of colostrum and/or milk per calf/day exclusively from their dam. First blood sample from newborn calves was collected before colostrum feeding on the day of birth (day zero) and the sampling continued daily for seven days after colostrum/milk feeding. Colostrum/milk Ig and IgG values were $4.82{\pm}2.60$, $2.19{\pm}1.90$, $1.12{\pm}0.82$, $0.69{\pm}0.44$, $0.59{\pm}0.31$, $0.47{\pm}0.20$, $0.40{\pm}0.22$, $0.40{\pm}0.25$ and $3.58{\pm}1.90$, $1.08{\pm}0.92$, $0.52{\pm}0.40$, $0.31{\pm}0.20$, $0.27{\pm}0.14$, $0.22{\pm}0.08$, $0.18{\pm}0.09$, $0.14{\pm}0.08$ respectively during 0-7 days post partum. The concentration of total colostrum/milk proteins, Ig, IgG and albumin were highest within 12 h post-partum. Thereafter, the concentrations followed a declining trend which may be attributed to the reduced transfer of proteins from the maternal blood, declining synthesis by the mammary glands and/or depletion of stored proteins. The concentrations of plasma Ig and IgG before colostrum feeding on day zero were $0.42{\pm}0.09$ and $0.08{\pm}0.03$ respectively. The levels of plasma Ig were $1.90{\pm}0.37$, $1.80{\pm}0.31$, $1.80{\pm}0.26$, $1.81{\pm}0.28$, $1.78{\pm}0.31$, $1.79{\pm}0.21$, $1.80{\pm}0.32$ and of IgG were $1.57{\pm}0.41$, $1.30{\pm}0.29$, $1.31{\pm}0.21$, $1.27{\pm}0.18$, $1.23{\pm}0.21$, $1.23{\pm}0.16$, $1.26{\pm}0.21$ on days 1-7 after birth after colostrum/milk feeding. The concentrations of total plasma proteins, Ig, IgG were lowest before colostrum feeding and increased significantly (p<0.05) after colostrum feeding in buffalo neonates. The results suggest that the highest amounts of colostral Ig and IgG were available on the day of parturition and thus the calves should receive colostrum as early after birth as possible. Colostrum Ig and IgG concentrations were not correlated to plasma Ig and IgG concentrations in the post-suckle buffalo calves and therefore, colostrum Ig and IgG concentrations were probably not the principle determinants of calf post-suckle plasma Ig and IgG concentrations.

  • Immunoglobulin G4 (IgG4)-associated autoimmune hepatitis (AIH) is a very rare subtype of autoimmune hepatitis and characterized by marked elevated serum IgG and hepatic infiltration of IgG4-expressing plasma cells. Pathologic confirmation of hepatic IgG4-expressing plasma cells is usually required for the final diagnosis of IgG4-associated AIH. Herein, we report the case of a 47-year-old female diagnosed with autoantibody-negative IgG4-associated AIH mimicking lymphoproliferative disorders.

IgG4-related Ophthalmic Disease Associated with Adult Xanthogranulomatous Disease (황색육아종과 동반된 면역글로불린G4관련안질환 1예)

  • Purpose: To report a case of immunoglobulin G4 (IgG4)-related ophthalmic disease associated with adult xanthogranulomatous disease. Case summary: A 38-year-old male with a history of cholecystectomy visited our clinic for bilateral periorbital swelling. Histopathology of the orbital biopsy showed diffuse infiltration of foamy histiocytes with Touton giant cells and lymphoid follicles, with a diagnosis of adult-onset xanthogranuloma. After excisional biopsy, he was treated with azathioprine and prednisolone. Four years after treatment, he again visited the clinic due to bilateral, yellowish eyelid masses. Serological examinations were all nonspecific findings, except for elevation of IgG and IgG4 levels. Magnetic resonance imaging showed bilateral symmetric soft tissue enlargement with slightly heterogeneous T1/T2 isosignal intensity, with contrast enhancement at the superolateral aspect of extraconal spaces. Excisional biopsy and blepharoplasty were performed. Immunohistochemical sections showed that the IgG4+/IgG plasma cell ratio was 10-20% and the IgG4 plasma cell count was 22/high power field (HPF). His past sections of 2013 from the pathology department were again stained and showed that the IgG4+/IgG plasma cell ratio was 40-50% and the IgG4 plasma cell count was 59/HPF. Thus, he was definitely diagnosed with IgG4-related ophthalmic disease. Conclusions: If there is recurrent eyelid swelling, IgG4-related ophthalmic disease should be considered as a differential diagnosis. And the patient with adult xanthogranulomatous disease can be diagnosed with IgG4-related ophthalmic disease.

IgG4-Related Intracranial Hypertrophic Pachymeningitis : A Case Report and Review of the Literature

  • Hypertrophic pachymeningitis is an uncommon disorder that causes a localized or diffuse thickening of the dura mater. Recently, the possibility that IgG4-related sclerosing disease may underlie some cases of intracranial hypertrophic pachymeningitis has been suggested. We herein report the tenth case of IgG4-related intracranial hypertrophic pachymeningitis and review the previous literature. A 45-year-old male presented with left-sided focal seizures with generalization. Magnetic resonance imaging (MRI) revealed a diffuse thickening and enhancement of the right convexity dura matter and falx with focal nodularity. The surgically resected specimens exhibited the proliferation of fibroblast-like spindle cells and an infiltration of mononuclear cells, including predominantly plasma cells. The ratio of IgG4-positive plasma cells to the overall IgG-positive cells was 45% in the area containing the highest infiltration of plasma cells. On the basis of the above findings, IgG4-related sclerosing disease arising from the dura mater was suspected. IgG4-related sclerosing disease should be added to the pachymeningitis spectrum.

The effects of high-dose intravenous immunoglobulin on plasma protein and lipid levels in the patients with Kawasaki disease (가와사끼병에서 고용량 정맥용 면역글로불린이 혈장 단백 및 지질에 미치는 영향(면역글로불린 투여 후 단백질 농도 변화))

  • Purpose : The reticuloendothelial system is composed of sinusoidal capillaries, through which even large protein molecules are freely movable between plasma and interstitial space, including the lymphatic system. Therefore, high-dose intravenous immunoglobulin (IVIG) would cause a redistribution of proteins between two compartments. To investigate this hypothesis, we measured plasma protein and lipid levels in patients with Kawasaki disease before and after high-dose IVIG treatment. Methods : Thirty four children with Kawasaki disease who had complete responses to high-dose IVIG treatment (1 g/kg/day for two consecutive days), were analyzed. Before and after the administration of IVIG, serum analyses were performed for such parameters as total protein, albumin, ${\gamma}$-globulins (IgG, IgM, IgA), ${\alpha}1-$, ${\alpha}2-$, and ${\beta}-$ globulin fractions, and lipid profiles (total cholesterol, HDL-cholesterol, LDL-cholesterol and triglyceride). Results : The levels of ${\gamma}$-globulins including IgG, IgM, IgA were significantly increased, and IgG was increased by $1,779{\pm}304mg/dL$ after two-dose of IVIG infusion. The levels of albumin, ${\alpha}1-$, ${\alpha}2-$, and ${\beta}$ globulin fractions were significantly decreased by 18 percent, 24 percent, 19 percent and 12 percent, respectively. HDL-cholesterol level was significantly decreased by 20 percent, while LDL-cholesterol and triglyceride levels were significantly increased by 21 percent and 50 percent, respectively. The total cholesterol level was not changed. Conclusion : High-dose IVIG treatment decreased the levels of a variety of proteins except immunoglobulins, and the increase of IgG after IVIG treatment was lower than expected. Our results suggest that a part of infused IVIG and plasma proteins, including etiologic proteins for Kawasaki disease, may be distributed to the extravascular compartments. The rapid improvement of symptoms induced by IVIG in Kawasaki disease might be explained by this mode of action of IVIG.

Effects of Dietary Ginger and Scutellaria Dry Powder on Antioxidant Capacity and IgG Concentration in Colostrum and Plasma of Sows (모돈에 대한 생강 및 황금 분말 급여가 초유와 혈청의 항산화력과 IgG 농도에 미치는 영향)

  • The objective of this study was to investigate the effects of dietary ginger and Scutellaria on antioxidant capacity and IgG concentration in plasma and colostrum of sows. Twenty-four sows (Landrace $\times$ Yorkshire) in parity 2 were assigned to 3 treatment groups (8 sows per treatment) from 30 day before farrowing up to weaning on day 21 after farrowing. The treatment diets included 1) CON (Basal diet), 2) GDP (0.3% ginger dry powder was used as the replacement of wheat in basal diet) and 3) SDP (0.3% Scutellaria dry powder was used as the replacement of wheat in basal diet). No differences were found in antioxidant capacity and IgG concentration in plasma of sows among all the three treatments. Antioxidant capacity in colostrum was not affected by replacement of ginger and Scutellaria dry powder. IgG and protein concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. However, fat and lactose concentration were not affected by treatments. The total amino acid concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. Plasma antioxidant capacity was not affected by treatments in piglet on 0 day and 21 day, but tended to increase by supplementation of ginger. On 0 day, plasma IgG in piglet was not detected by replacement of ginger and Scutellaria dry powder. However, plasma IgG concentration in piglets on 21 day was significantly higher (P<0.05) in GDP than that in CON and SDP. These results indicate that colostrum IgG concentration in sows and plasma IgG concentration in piglets were improved by feeding ginger dry powder 30 days before farrowing in comparison to basal diet.

The Effects of Tofu (Soybean curd) Containing Mushrooms on the Immune Activities (버섯첨가 두부의 면역활성에 미치는 영향)

  • This research was carried out to evaluate the value of tofu containing mushroom as a immunomodulator. Tofu was made using $CaSO_4{\cdot}2H_2O$ or Lactobacillus extract as a coagulant after adding powder of fruit bodies or mycelia of Letino edodes and Lepista nuda to soybean milk. Proximate compositions of tofu and tofu containing mushroom were analyzed. Levels of interferon ${\gamma}$ (IFN-${\gamma}$), interleukin 4 (IL-4) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in culture media of lymphocytes collected from mouse spleens after being injected with mushroom, regular tofu, or tofu made with mushroom were measured by sandwich ELISA. In addition, concentrations of IgG1, IgG2a and IgE in plasma or lymphocyte culture media were analyzed. Crude protein, crude lipid and crude ash were decreased in tofu containing mushroom but phosphorus was increased significantly. IFN-${\gamma}$ concentration was significantly decreased in mice injected with fruit body or tofu alone. IL-4 level was decreased significantly in mice injected with tofu containing fruit body of L. edodes. However, TNF-${\alpha}$ was increased in mice injected with tofu containing fruit body of L. edodes. Plasma levels of IgG1 were increased in almost all groups, while there was no significant change in IgG2a levels among treated mice groups. Concentrations of IgG1 and IgG2a were increased significantly in lymphocyte culture media of mice injected with tofu containing mushroom. Plasma levels of IgE level was significantly increased in mice injected with tofu or fruit body of L. edodes, but not in mice treated with tofu containing mushroom. These results showed that tofu with mushroom affected immune activities, and it seems valuable to consider developing the mixture of tofu and L. edodes as an immunomodulator.

  • This stduy was carried out to investigate the effects of Isoimmunization by sperm and seminal plasma on density of immunoglobulins in reproductive tract fluids and serum of immunized rabbits. The results obtained were summarized as follows: 1. Antibody titers against sperm and seminal plasma antigen ranged from 8 to 64 to 512, respectively. 2. All immunoglobulins; IgG, IgA and IgM were detected with Indirect ELISA method in the uterine and oviductal fluids as well as the sera of immune rabbits. 3. Concentrations of IgGs in the uterine and oviductal fluids of rabbits immunized with sperm and seminal plasma were higher than those of the control rabbits, but not showed any differences in sera. 4. Amount of IgA in the sera and oviductal fluids of control animals was more than that of the immune animals, while that of IgA in the uterine fluids of control and seminal plasma-immunized animals was higher as compared to sperm immune animals. 5. Average concentration of IgM in the uterine fluids of control and seminal plasma-immunized rabbits was higher than that of sperm-immunized ones. In the oviductal fluids, average concentrations of IgM of immune rabbits was higher than that of immune rabbits.

Adsorptive Binding of Plasma Proteins to Red Blood Cell Ghosts (플라즈마 단백질의 고스트 적혈구 흡착 특성)

  • Blood plasma proteins dissolved in isotonic phosphate-buffered saline were found to be adsorbed to red blood cell ghosts suspended in the solution. This was evidenced by concentration reduction of the plasma proteins in the bulk solution. For initial concentration of 0.1mg/mL immuno globulin, concentration reduction increased from 14% to 45% as the volume fraction of red blood cell ghosts in the solution increased from 5% to 45%. For initial concentration of 0.075mg/mL albumin, the concentration reduction increased from 12% to 47% as the volume fraction of red blood cell ghosts in the solution increased from 5% to 70%. The concentration reduction of plasma proteins in hardened red blood cell ghosts was higher than that in red blood cell ghosts. The number of adsorbed protein molecules per a red blood cell ghost were reduced as volume fraction of the ghosts in the solution increased.

An Effective Method of Isolating Immunoglobulins from Bovine Plasma Proteins (도축혈액으로부터 면역단백질의 효과적인 분리법)

  • Imunoglobulins from bovine plasma proteins were isolated by IMAC which $Cu^{2+}$ was chelated on a chelating sepharose fast flow gel. Most plasma proteins were eluted by 1st (0.01 M $Na_2HPO_4$, 0.5 M NaCl, pH 4.0) and 2nd elution buffers (0.01 M imidazol). According to the reverse phase HPLC analysis, it was found that proteins which were eluted by 1st elution buffer were mainly composed of serum albumin, while most IgG and transferrin were eluted by 2nd elution buffer. When protein fractions obtained by 2nd elution buffer was applied to ultra filtration system (molecular weight cut off: 100 kD), IgG was further purified. These results indicate that IMAC is an excellent tool for isolating imunoglobulins from plasma proteins.
