The Various Suspension Culture Methods on the Growth of Culture Cells of Wild Viola (Viola patrinii Dc.) (야생 흰 제비꽃 (Viola patrinii DC) 배양세포의 활성에 미치는 다양한 현탁배양 방법)

  • This experiment was carried out to examine the effects of culture medium on cell growth of the viola (Viola patrinii DC.) suspension culture. The results are as follows: The greatest cell growth rates were found with MS medium suggesting that this medium could be recommendable for the viola suspension cell culture. When the nitrogen sources (NH$_4$NO$_3$ and KNO$_3$) of MS salts were diluted at half concentrations, the cell growth rates were slightly increased, but when the combined concentration rations of NH$_4$+ and NO$_3$ions were 25 to 75 the greatest cell growth rates were obtained. This result imply that the nitrogen ion sources had slight influence on the rates. Another feature was obtained. This result implys that the nitrogen ion sources had slight influence on the rates. Another feature was that as the concentration of NH$_4$+ ion lowered, the callus color changed to pale yellow with some red spots. The addition of casein hydrolysate (5 g/L) was more effective for the cell growth. On the basis of microscopic observation, the highest cell growth rates were detected during 2-4 weeks culture and after 6 weeks of the culture, some elongated and vacuolated cells were determined.

Formation of Genetic Tumor and Characteristics of Teratoma Shoot from Tobacco Interspecific Reciprocal Hybrids (연초종간 상호교잡에 의한 Genetic Tumor의 유도 및 Teratoma Shoot의 특성)

  • Reciprocal interspecific hybrids between N. glauca(2n=24) and N. langsdorffii(2n=18) were obtained by intercrossing. One hundred percent of F$_1$ seeds was produced from intercrossing of N. glauca $\times$ N. langsdorffii, whereas the frequency of F$_1$ hybrid seed formation from N. langsdorffii $\times$ N. glauca was very low. However, all the hybrid seeds were germinated well and then grown to normal plantlets. All the plants of F$_1$ hybrids have chromosome number of interspecific hybrids (2n=21). From observation of morphological characteristic, the structure of petrol, leaf, flower, and the morphology of pollen have characteristics of F1 hybrid. Spontaneous tumors (genetic tumor) were formed from each F$_1$ hybrid; the genetic tumor arose at the reproductive phase when the maternal type of F$_1$ hybrid came from N. glauca, while the genetic tumor arose only after reproductive phase when the maternal type of F$_1$ hybrid came from N. langsdorffii. The genetic tumor actively proliferated on hormone-free medium and produced numerous teratoma shoots. In addition, normal leaf or stem explants of F$_1$ hybrid produced calli on hormone-free medium after 15 days of culture, the calli produced new numerous teratoma shoots after 30 days. The frequency of teratoma shoot formation from rnterspecific hybrid was higher in the N. glauca $\times$ N. langsdorffii than in the N. langsdorffii $\times$ N. glauca. Root development from the teratoma shoots was hardly obtained. Teratoma shoots without roots in vitro can form genetic tumor at the vegetative growth phase after tissue culture.

Several Factors on Bulblets Regeneration from Callus Culture in Lilium longiflorum 'Celia' (백합 'Gelia' 캘러스로부터 자구 재분화에 미치는 제요인)

  • Callus from scale segments of Lilium longiflorum 'Gelia' was effectively induced and maintained from unorganized tissue on the semi-solid medium by 0.42% Bacto agar with MS basal salts and vitamins of SH medium supplemented with 0.5 mg/L 2, 4-D, 1.0 mg/L NAA, 0.3 mg/L BA, and 3% sucrose. More than 5% of high sucrose level had inhibiting effect on regeneration capacity of formed callus and decreased callus growth. Various combinations of nitrogen did not effective to proliferate the ELC (Embryogenic-like callus), but friability of callus was increased in the medium containing only nitrate as nitrogen source. 5 mL conditioned medium into 30 mL fresh medium was good for cell growth. However friable cell aggregates during suspension culture had to form hard callus which hindered to establish suspention culture system. Addition of 2 g/L casein hydrolysate increased callus growth and friability of the hard callus. As a result of anatomical observation of callus, organogenesis such as shoots, roots and bulblets was independently induced from callus tissue. Somatic embryogenesis from callus tissue could be observed with low frequency.

Application of a Climate Suitability Model to Assess Spatial Variability in Acreage and Yield of Wheat in Ukraine (우크라이나 밀 재배 면적 및 수량의 공간적 변이 평가를 위한 기후적합도 모델의 활용)

  • It would be advantageous to predict acreage and yield of crops in major grain-exporting countries, which would improve decisions on policy making and grain trade in Korea. A climate suitability model can be used to assess crop acreage and yield in a region where the availability of observation data is limited for the use of process-based crop models. The objective of this study was to determine the climate suitability index of wheat by province in Ukraine, which would allow for the spatial assessment of acreage and yield for the given crop. In the present study, the official data of wheat acreage and yield were collected from the State Statistics Service of Ukraine. The EarthStat data, which is a data product derived from satellite data and official crop reports, were also gathered for the comparison with the map of climate suitability index. The Fuzzy Union model was used to create the climate suitability maps under the historical climate conditions for the period from 1970 to 2000. These maps were compared against actual acreage and yield by province. It was found that the EarthStat data for acreage and yield of wheat differed from the corresponding official data in several provinces. On the other hand, the climate suitability index obtained using the Fuzzy Union model explained the variation in acreage and yield at a reasonable degree. For example, the correlation coefficient between the climate suitability index and yield was 0.647. Our results suggested that the climate suitability index could be used to indicate the spatial distribution of acreage and yield within a region of interest.

Comparison of Soil Moisture Changes Based on the Installation Position of Soil Moisture Sensors in the Korean Orchard Field Soils (노지 과수원에서 토양수분센서 설치 위치에 따른 토양수분 변화 비교)

  • For efficient soil water management in open fields, the proper use of soil moisture sensors is a prerequisite. Particularly in open-field environments like orchards with extensive root systems, the appropriate positioning of sensors is very important. The present study was conducted to identify the optimal placement of soil moisture sensors by assessing changes in soil water potential across various positions within orchard field soils after installing tensiometers. In apple and Asian pear orchards located in two regions of Korea, nine soil water potential sensors (TEROS 21, METER Group) were installed at distances of 20, 40, and 60 cm from the tree trunk and depths of 10, 20, and 30 cm from the soil surface, and monitored the soil water potential changes over two years. Results indicated that the positions closer to the tree trunk and the soil surface exhibited more pronounced changes in soil water potential. The greatest magnitude of change in soil water potential was observed at a distance of 20 cm and a depth of 10 cm, suggesting this position as the most suitable for soil moisture sensor installation. However, variations in the degree and pattern of changes in soil water potential were noted across sensor positions due to root system growth over time. Therefore, periodic observation and adjustments in sensor placement would be advisable to accurately monitor the soil moisture condition in long-term crops such as fruit trees in open fields.

  • It is a recent observation that about 80 per cent of the body's immune system is localized in the gastrointestinal tract. This explains to a large extent why eating right is important for the modulation the immune response and prevention of disease. In addition it is increasingly recognized that the body has an important digestive system also in the lower gastrointestinal tract where numerous important substances are released by microbial enzymes and absorbed. Among these substances are short chain fatty acids, amino acids, various carbohydrates, poly-amines, growth factors, coagulation factors, and many thousands of antioxidants, not only traditional vitamins but numerous flavonoids, carotenoids and similar plant- and vegetable produced antioxidants. Also consumption of health-promoting bacteria (probiotics) and vegetable fibres (prebiotics) from numerous sources are known to have strong health-promoting influence. It has been calculated that the intestine harbours about 300,000 genes, which is much more than the calculated about 60,000 for the rest of the human body, indicating a till today totally unexpected metabolic activity in this part of the GI tract. There are seemingly several times more active enzymes in the intestine than in the rest of the body, ready to release hundred thousand or more of substances important for our health and well-being. In addition do the microbial cells produce signal molecules similar to cytokines but called bacteriokines and nitric oxide, with provide modulatory effects both on the mucosal cells, the mucosa- associated lymphoid system (MALT) and the rest of the immune system. Identification of various fermentation products, and often referred to as synbiotics, studies of their role in maintaining health and well-being should be a priority issue during the years to come.

The Study of Free Radical Scavenging Effect of Lycii Fructus by Liver Injury of Rats (백서 간손상에 의한 구기자의 유리자유기 소거능에 관한 연구)

  • In the present study, we investigated the protective effect of the Lycii Fructus water extracts (LFE) against CCl4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of LFE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation, In addition, the pretreatment of LFE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CC1₄-injected rats. The LFE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 μg/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of LFE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the LFE may be related to antioxidant effects and regulation of CYP2E1 gene expression.

A Study on a Morphological Identification of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma (반하(半夏), 수반하(水半夏) 및 천남성(天南星)의 형태(形態) 감별에 관한 연구)

  • Objectives : A morphological classification among Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Pinelliae Tuber is like a ball with a $1{\sim}1.5cm$ diameter, with a concave apex and pockmarked root trace around. 2. Typhonii Flagelliformis Rhizoma is somewhat oval or like a cone, with a $4{\sim}8mm$ diameter, a $7{\sim}15mm$ height, a projected round apex, and a pin-pointed back. 3. Arisaematis Rhizoma is somewhat flat round with a $2{\sim}2.5cm$ diameter, a little than Pinelliae Tuber, and its apex has incomplete lobopodium and root trace. 4. Fundamental tissues of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma have starch grains and mucilage cells, and their mucilage cells include the raphides of calcium oxalate. Conclusion : Internal forms of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma are difficult to distinguish from one another, but Pinelliae Tuber and Arisaematis Rhizoma can be distinguished by their external forms and sizes, and Pinelliae Tuber and Typhonii Flagelliformis Rhizoma were distinguished by the forms of apex.

Resistance to Root Penetration of Root Barrier for Green Roof System (옥상녹화용 방근층 구성재료의 방근성능에 관한 실험적 연구)

  • The purpose of this study is to test performances of 14 types of root barrier materials by applying testing plant: and soils suitable for weather and natural features of Korea. For testing Plants, Plioblastus pygmaed Mitford A and Pyracantha angustifolia have been selected. For testing soil, mixture of pearlite and peat moss in 3:1 ratio Testing container has been fabricated with duplicated structure having inner and outer containers. And the outer container has 2 hinges on its side wall to allow opening and closing. Wet rock wool with 50mm in thickness has been inserted between inner and outer containers to allow root to penetrate through root barrier material and continue to grow. We planted 12 Plioblastus pygmaed Mitford A. and 4 Pyracantha angustifolia per one testing container. Three testing samples have been made for 1 type of root barrier material, which become a total 42 specimens. Planted testing samples have been installed within the greenhouse, which will be observed regularly for 2 years from now on. We started test from July 11, 2008 and had performed intermediate observations every month for initial 3 months. From the 3rd intermediate observation on Sept. 18, we confirmed that 6 types of roe barrier materials have penetrated roots. Even though two types of them have been generally used as root barrier materials for roof planting system, all of three testing samples have a lot of penetrated roots. This result proves that it is not reasonable to introduce testing methods of root barrier from Europe. USA or Japan.

Factors Involved in Promoting Seed Germination of Foeniculum vulgare (회향종자의 발아촉진에 관여하는 요인)

  • The effects of temperature, prechilling, chemicals such as $GA_3$, IAA, kinetin and $KNO_3$ on the germinability of Foeniculum vulgare seed of medicinal plant were examined. In Foeniculum vulgare, the germination rate appeared to be around 50~60% or more in general, showing no differences in germination rate with different temperatures, promoting substances, physical or chemical treatments, and prechilling treatments. The observation of embryo under stereoscopic microscope for Foeniculum vulgare in Umbelliferae showed that seeds with or without embryo was almost the same in number. This result suggests that the lower rate of germination in this species is caused by embryolessness of seeds. The straight-shaped embryos as well as Y-shaped embryos were also observed. Foeniculum vulgare of medicinal plants in Umbelliferae were observed under scanning electron microscope, and did not show any opening problem near micropyle area. Final count should be made on 7th day of germination test.
