Chemical Constituents from the Aerial Parts of Artemisia capillaris Thunb. and Their Anti-allergic and Anti-inflammatory Effects

  • Artemisia capillaris Thunb. (Compositae) is a traditional medicinal plant with various pharmacological activities. To elucidate new anti-allergic and anti-inflammatory constituents, the aerial parts of A. capillaries were investigated to afford a new compound, (6E,8E)-6-methylundeca-6,8-diene-2,5,10-trione (17) together with 19 known compounds (1 - 16, 18 - 20). The structures of these compounds were determined by extensive spectroscopic analyses including 1D, 2D NMR, HREIMS, and optical rotation [α]D. The absolute configuration of compound 2 was determined to be S form for the first time. All isolates (1 - 20) were tested their inhibitory effects on interleukin 2 (IL-2) expression in T cells and NO production in lipopolysaccharide (LPS)-stimulated RAW246.7. Among them, compounds 10, 11, 19, and 20 reduced IL-2 expression in a dose-dependent manner. In addition, compound 10 also inhibited NO production with an IC50 value of 37.3 ± 0.4 μM.

Quantitative Determination of Bakkenolide D in Petasites japonicus and Farfugium japonicum by HPLC/UV

  • A quantitative analysis of bakkenolide D in the different parts of Petasites japonicus and Farfugium japonicum was performed by HPLC. A gradient HPLC elution system with a mobile phase consisting of water: acetonitrile solution (20:80 to 0:100 for 45 min) was followed and an INNO $C_{18}$ column was used for the chromatographic separation. The injection volume, flow rate, and UV detection were $10{\mu}L$, 1 mL/min, and 290 nm, respectively. Results show that both species showed the highest amount of bakkenolide D in the roots being 107.203 and 166.103 mg/g for P. japonicas and F. japonicum, respectively. Content analysis on the different parts of both plants displayed remarkably lower values which ranged from 0.403 - 4.419 and 7.252 - 32.614 mg/g for P. japonicas and F. japonicum, respectively. The results show that the roots of both plants are rich in bakkenolide D showing a promising use in the development of nutraceuticals and industrial application of the compound.

Tyrosinase Inhibitory Activity of Plant Extracts (III): Fifty Korean Indigenous Plants

  • The purpose of this study was to evaluate tyrosinase inhibitory activity of plant extracts, especially Korean indigenous plants, for the cosmetic use of skin whitening. When 50 plant extracts were tested, the methanol extracts of Agrimonia pilosa, Aster scaber; Dianthus sinensis, Fatsia japonica, Hemistepta lyrata, Lespedeza cuneata, Osmunda japonicum, Pyrvla japvnica, Rodgersia phodophylla and Veratrum grandiforum possessed the considerable tyrosinase inhibitory activity at 3-300 $\mu\textrm{g}$/mL. Especially, L. cuneata, aerial part of O. japonicum and V. gandiforum exhibited the strong inhibition (>50% inhibition at 300 $\mu\textrm{g}$/mL). In particular, the methanol extract of V. grandiforum and its ethylacetate fraction showed potent inhibition ($IC_{50}$/=30 and 13$\mu\textrm{g}$/mL, respectively), while the reference compound, kojic acid, showed $IC_{50}$/ value of 26$\mu\textrm{g}$/mL. These plant extracts may be used as tyrosinase inhibitors in cosmetics.

Isolation and Characterization of Plant Growth Promoting Rhizobacterium Bacillus subtilis YK-5 from Soil (토양에서 식물생육촉진 활성을 가진 균주 Bacillus subtilis YK-5의 분리 및 특성)

  • For the isolation of a plant growth-promoting rhizobacteria, strain YK-5 was selected from approximately 400 thermostable strains isolated from special soil samples. Strain YK-5 produced an antifungal compound, and optimum carbon and nitrogen sources for the production of the antifungal compound were investigated against Aspergillus flavus as a test strain. Modified LB medium containing 1% peptone, 1% yeast extract and 5% black sugar was determined to be the optimal medium for growth and antifungal compound production. Culture broth of strain YK-5 potently inhibited growth of the phytopathogenic fungus Fusarium oxysporum KACC 40052 for 7 days. The plant growth-promotion function of strain YK-5 was tested against radish and rice in pot trials. Leaf number, plant height and root length in YK-5-treated radish markedly exceeded (> 60%) those of untreated radish. Leaf length and white rootlet development were markedly more prominent than in commercially-treated rice plants. Strain YK-5 was determined to be Bacillus subtilis YK-5 by physiological, chemotaxonomical, and phylogenetical analyses.

Radical Scavenging Effects of 10 Plant Essential Oils and Active Compound Screening Analysis (식물정유 10 종의 라디칼 소거 활성과 주요 활성 성분의 탐색)

  • Background: A growing interest in health has increased the need for the development of potent antioxidant materials known to play a role in various physiological activities. Currently research and development of non-toxic natural antioxidants with high activity is ongoing. Methods and Results: In this study, we measured 2,2'-azinobis- (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability of 10 plant essential oils, selecting samples of Dendranthema indicum, Dendranthema zawadskii, and Citrus sunki essential oils. The samples were analyzed using liquid chromatography (LC) and the radical scavenging activity on LC-based systems with the same conditions. In the LC-mass spectroscopy (MS)/MS analysis of the active compound peak, 2-methoxy-4-vinylphenol with a molecular weight of 150.1 g/mol was identified in C. sunki essential oils. Eugenol or isoeugenol with a molecular weight of 164.1 g/mol as identified in D. indicum and D. zawadskii essential oils as radical scavenging active compounds. Conclusions: In the LC-based measurement system, the active ingredient can be identified by simultaneously conducting profile analysis and the radical scavenging activity of essential oil samples. In addition, LC-MS/MS analysis of the active compound peaks can be performed under the same separation conditions to obtain data that can identify the active compounds in the sample.

Identification of Novel Saringosteryl Glucoside in Phaseolus vulgaris Seed (강낭콩 미숙종자내 신규 Saringosteryl Glucoside의 동정)

  • From immature seed of Phaseolus vulgaris L., a novel phytosteryl glucoside was isolated. Strong ion peaks at m/z 613 $[M+Na}^{+},\;696\;[M+Matrix]^{+}$ in positive F AB- MS and at m/z 589 $[M-1]^{-}$ in negative F AB- MS indicated the molecular weight of the compound is 590. Four hundred MHz $^IH-NMR$ analysis revealed that the compound canys a 24-hydroxy-24-vinyl-cholesterol (saringosterol) as an aglycone and a ${\beta}-D-glucopyranose$. Four hundred MHz $^IH-NMR$ analysis of the acetate derivate of the compound revealed that hydroxyls at C-1' in glucose moeity and at C-3 in aglycone have been condensed. Therefore, the phytosteryl glucoside was characterized to be $3-0-{\beta}-D-glucopyranosyl-24-hydroxy-24-vinyl-cholesterol$ (saringosteryl glucoside). This is the first demonstration for the presence of saringosterol in higher plants. Also this is the first identification of saringosteryl glucoside in natural materials.erials.

Cytotoxic and Antioxidant Compounds Isolated from the Cork of Euonymus alatus Sieb.

  • Seventeen compounds (1 - 17), ${\beta}$-sitosterone (1), lupenone (2), arborinone (3), ${\beta}$-sitosterol (4), lupeol (5), epi-lupeol (6), taraxerol (7), betulinic acid (8), 24R-methyllophenol (9), germanicol (10), hexatriacontane (11), nonacosan-1-ol (12), benzoic acid (13), tetradecyl(E)-ferulate (14), di(2-ethylhexyl) phthalate (15), trilinolein (16) and monopalmitin (17), were isolated from the methylene chloride-soluble fraction of the cork of Euonymus alatus Sieb. The structures of these compounds were elucidated on the basis of spectroscopic evidence. Compounds 6, 11, 13 and 14 were isolated for the first time from this plant. Compound 4 showed moderate cytotoxic activity with an $IC_{50}$ value of 6.22 ${\mu}M$ in HL-60 cell line. Compound 9 exhibited moderate cytotoxic activity with $IC_{50}$ values of 63.31, 15.45, 15.14 and 21.72 ${\mu}M$ in four kinds of human cancer cell lines, Jurkat T, HeLa, HL-60 and MCF-7, respectively. Compound 17 showed moderate cytotoxic activity with an $IC_{50}$ value of 70.71 ${\mu}M$ in Jurkat T cell line. In addition, compounds 2, 3, 14 and 16 exhibited weak antioxidant activity with $IC_{50}$ values of 151.76, 170.79, 137.46 and 139.37 ${\mu}M$, respectively.

Biological Control Activity of Two Isolates of Pseudomonas fluorescens against Rice Sheath Blight

  • Two isolates of mucous bacteria, mc75 and pc78, were isolated from fungal culture plate as culture contaminants with an interesting swarming motility. Both isolates were identified as Pseudomonas fluorescens based on microscopy, biochemical analysis, Biolog test and DNA sequence analysis of the 16S rRNA gene. Both strains have the exactly the same 16S rRNA gene sequences, and yet their biological control activity were not identical each other. In vitro analysis of antagonistic activity of two isolates against several plant pathogenic fungi indicated that both produced diffusible and volatile antifungal compounds of unknown identities. Treatment of the bacterial culture of P. fluorescens pc78 and its culture filtrate exhibited a strong biological control activity against rice sheath blight in vivo among six plant diseases tested. More effective disease control activity was obtained from treatment of bacterial culture than that of culture filtrate. Therefore, in addition to antifungal compound and siderophore production, other traits such as biofilm formation and swarming motility on plant surface may contribute to the biological control activity of P.fluorescens pc78 and mc75.

Synthesis, Antioxidant and Molecular Docking Studies of (-)-Catechin Derivatives

  • 12 kinds of (-)-catechin derivatives were designed and synthesized. The catechin derivatives were evaluated their antioxidant activities using DPPH method. Most of them showed good antioxidant activity, particularly compounds 1d, 1e and 1j exhibited more activity than butylated hydroxytoluene (BHT). Molecular docking studies for compounds 1d, 1e and 1j with STAT1 showed not only sufficent characteristics binding cavity but also agreement with the observed biological activity. Acording to docking results, the compounds showed greater than hydrogen bonding, hydrophobic interactions, electrostatic interactions, and Van der Waals interactions as compared to the reference compound. They formed hydrogen bonds with important residues such as Lys566, His568, Leu570, and Phe644. The compounds showed a novel hydrogen bonding interaction with Arg649, which was not reported previously. Our results might suggest the compounds could serve as a novel anti-oxidant agent.

Biological Activity Analysis of Potato-derived Polar Compounds (감자 유래 극성화합물의 생리활성 분석)

  • Natural substances have various physiological activities. Substances isolated from natural substances are known to be safer and more potent than pharmaceuticals. Potatoes not only act as energy sources but also contain active ingredients such as vitamins and minerals. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by chemical base anti-oxidation assay and cell based anti-oxidation assay. In the chemical base anti-oxidation assay, DPPH experiment showed activity in the order of Hongyoung, Haryung, Seohong, Sumi, and Jayoung. In the LPA experiment, IC50 was lower in the order of Jayoung, Seohong, Sumi, Hongyoung, and Haryung. In the cell based anti-oxidation assay, the smallest amount of ROS was generated when the compound was derived from Haryung and hongyoung, and strong SOD activity was observed in Sumi and Jayoung. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers.
