Protein phosphatase 2A (PP2A), a family of serine/threonine protein phosphatases, plays an important role in balancing the phosphorylation status of cellular proteins for regulating diverse biological functions in eukaryotic organisms. Despite intensive studies in mammals, limited information on its role is available in filamentous fungi. Here, we investigated the functional roles of genes for a putative B' delta regulatory subunit (FgPP2AR) and a catalytic subunit (FgPP2AC) of PP2A in a filamentous ascomycete, Fusarium graminearum. Molecular characterization of an insertional mutant of this plant pathogenic fungus allowed us to identify the roles of FgPP2AR. Targeted gene replacement and complementation analyses demonstrated that the deletion of FgPP2AR, which was constitutively expressed in all growth stages, caused drastic changes in hyphal growth, conidia morphology/germination, gene expression for mycotoxin production, sexual development and pathogenicity. In particular, overproduction of aberrant cylindrical-shaped conidia is suggestive of arthroconidial induction in the ${\Delta}FgPP2AR$ strain, which has never been described in F. graminearum. In contrast, the ${\Delta}FgPP2AC$ strain was not significantly different from its wild-type progenitor in conidiation, trichothecene gene expression, and pathogenicity; however, it showed reduced hyphal growth and no perithecial formation. The double-deletion ${\Delta}FgPP2AR;{\Delta}FgPP2AC$ strain had more severe defects than single-deletion strains in all examined phenotypes. Taken together, our results indicate that both the putative regulatory and catalytic subunits of PP2A are involved in various cellular processes for fungal development in F. graminearum.
Genetic transformation was carried out by using biolistic gun method. The hypocotyl derived embryogenic calli (explants) of cotton (Gossypium hirsutum L.) cv. Cocker-312 were transformed with a recombinant pGreen II plasmid, in which both, bar (selection marker) and GUS (${\beta}$-glucuronidase) reporter genes were incorporated. Explants were arranged on osmoticum-containing medium (0.5M mannitol) 4 hours prior to and 16 hours after bombardment that was resulted into an increase about >80% for GUS stable expression. 3 days after bombardment, GUS assay was performed, which exhibited, $18.36{\pm}1.00$ calli showed blue spots. The transformed embryogenic calli were cultured on selection medium (@ 6 mg/L basta) for 3 months. The putative transgenic plants were developed via selective somatic embryogenesis (@1.50 mg/L basta); maximum $27.58{\pm}1.25$ somatic embryos were obtained while $17.47{\pm}1.00$ embryos developed into plantlets (@ 0.75mg/L basta). In five independent experiments, up to 7.24% transformation efficiency was recorded. The presence of the transgenes was analyzed by using PCR and southern hybridization analysis. The transgenic plants were developed with in 6-7 months, but mostly transformants were abnormal in morphology.
Changes of in vitro true digestibility (IVTD) of Napiergrass (Pennisetum purpureum) were determined by a filter bag system, and their relationships to chemical composition, leaf to stem ratio, plant height, geographic location, climatic factors and harvest interval were studied and used to develop prediction models for the crude protein (CP), acid-detergent fiber (ADF), and neutral-detergent fiber (NDF) contents and IVTD. Partitioning the total variance of IVTD of Napiergrass showed that 80% was attributable to the effect of harvest interval. Days of growth, plant height, leaf/stem ratio, CP, ADF and NDF of Napiergrass had highly significant relationships (p<0.01) with IVTD. The highest coefficient of correlation between the ADF, NDF, and IVTD of Napiergrass and growth degree days was obtained when the base temperature was set at $0^{\circ}C$. Growth degree days could predict ADF, NDF, and IVTD of Napiergrass more accurately than plant height, and plant height is not suitable to predict IVTD.

The physico-chemical characteristics and nanostructure of the aerosol samples from a coal-fired power plant, a charcoal kiln and diesel vehicles were investigated with focusing on black carbon (BC). Aerosols from the coal-fired power plant were mostly comprised of mineral ash spheres which are heterogeneously mixed. The main components of the aerosols from coal-fired power plant were calcium compounds, iron oxide, alumino-silicate without BC. The typical combustion-generated BC which shows the shape of bunch of grapes with 20~50 nm particles which were detected in aerosol particles from diesel vehicles. The nanostructure of each BC particle shows the shape of concentric circles which is comprised of closely-packed graphene layers. Aerosols from charcoal kiln were likely condensed organic carbon generated from the low-temperature combustion process.

Opium poppy (Papaver somniferum L.) is one of the most important medicinal plants, which is used as a sole commercial source of narcotic analgesic, morphine. The transformant of opium poppy we have established by infection of Rhizobium rhizogenes (formerly Agrobacterium rhizogenes) strain MAFF03-01724 showed aberrant morphology and altered opium alkaloid composition. The major alkaloid produced by this transformant was thebaine (16.3%, opium dry weight) instead of morphine. It is likely that this 'thebaine poppy' phenotype was caused by the integration of T-DNA(s) into the poppy genome DNA, and their inserted loci are of great interest. To gain an insight into the mechanism of nonnarcotic thebaine accumulation for the further approach toward the creation of 'codeine poppy' which produces codeine as a major alkaloid, the genetical and morphological analyses on the transformant was carried out. Here we report the results of the detailed analysis on the T-DNA inserted loci of T0 transfromant and the correlation between opium alkaloid composition and segregated T-DNA integration pattern in the self-pollinated T1 transformants.
ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.

To develop transgenic cucumber tolerant to abiotic stress, a cotyledonary-node explants were co-cultivated with Agrobacterium tumefaciens (EHA101) carrying TPSP gene (pHC30-TPSP). After transfer to fresh medium every two week for eight weeks, putative transgenic plants were selected when shoots grown a length greater than 3 cm from the cotyledonary-node explants on selection medium supplemented with $5\;mgl^{-1}$ phospinotricin as selectable agent. The confirmation of transgenic cucumber was based on the Northern blot analysis. Thirty four shoots (5.2%) with resistance to phospinotricin were obtained from 660 explants inoculated. Of them, transformants were only confirmed from 11 plants (1.7%). Transgenic cucumber expressing TPSP gene was more synthesized at 3.8 times amounts of trehalose (0.014 mg g fresh $wt^{-1}$) than non-transformants (0.0037 mg g fresh $wt^{-1}$). However, all of transgenic plants showed abnormal morphology, including stunted growth (< height 15 cm), shrunken leaves, and sterility as compared with non-transgenic plants (> height 150 cm) under the same growth environment. These results lead us to speculate that the overproduction of trehalose was toxic for cucumber, even though that had known for rice as non-toxic.
In August 2015, we collected samples of gray mold from sweet basil growing in Sachunmeon, Gangneung, Gangwon Province, Korea. Symptoms included extensive growth of mycelia with gray conidia on young leaves, stems, and blossoms. The pathogen was isolated from infected leaves and blossoms and the fungus was cultured on potato dextrose agar. For identification of the fungus, morphology and rDNA sequencing analysis of the fungus were performed, which confirmed its pathogenicity according to Koch's postulates. The results of morphological examinations, pathogenicity tests, and the rDNA sequences of the internal transcribed spacer regions (ITS1 and ITS4) and the three nuclear protein-coding genes G3PDH, HSP60, and RPB2 showed that the causal agent was Botrytis cinerea. This is the first report of gray mold caused by Botrytis cinerea on sweet basil in Korea.
This study was carried out to investigate the morphological characteristics of the plants and seeds of Ainsliaea acerifolia and to determine the optimum condition for producing sprouts from the seeds. Plant height, flower stalk length, and pod number were higher in natural habitat than in campus farm. Average 1.2 seeds per pod was set but only 0 to 2 seeds per plant was set in plants with the enveloped flower stalks, indicating that this is an outcrossing species. Most of seeds were 9-11mm long and 1.1-1.4mm wide. Fresh weight of seeds was ranged from 10mg to 17mg. Seeds germinated well at 15$^{\circ}C$ and 2$0^{\circ}C$. Mean germination period was 11.5 day at 15 to $25^{\circ}C$. Sprouts grown at 15$^{\circ}C$ was longest(5.4cm) and heaviest(738mg/10 sprouts). Chlorophyll content was 333mg per fresh weight 110g. Protein, Fe, vitamin Bl, vitamin B2, and vitamin C were respectively 23.7mg, 6.4mg, 1.82mg, 0.49mg, and 10..7mg.

A new diatom species, Mayamaea vietnamica sp. nov., is described from Cát Tiên National Park in Vietnam. This species was discovered and described from soil samples. Algae from soil ecosystems in Vietnam are almost unknown. The new species is described on the basis of an integrated approach with molecular and morphological data, and comparison with similar species. In terms of molecular data, 18S rDNA (including V4 domain), and partial rbcL plastid genes show M. vietnamica sp. nov. is most closely related to M. terrestris N. Abarca and R. Jahn, and together they form a monophyletic group relative to other members of the genus. M. vietnamica sp. nov. differs from other species in the genus by the number of striae and areolae in 10 ㎛, number of areolae per stria, as well as shape and presence or absence of axial and central areas.
