Variovorax sp. PMC12 균주에 의한 토마토의 생물학 및 비생물학적 스트레스 저항성 증진 (Enhancement of Tomato Tolerance to Biotic and Abiotic Stresses by Variovorax sp. PMC12)

  • 근권세균은 식물 생육과 건강 증진에 중요한 역할을 하며 생물학적 스트레스뿐만 아니라 저온, 고온, 건조 및 염과 같은 비생물적 스트레스에도 내성을 부여한다. 본 연구는 토마토에 생물적 및 비생물적 스트레스를 완화시키는 기능을 가진 식물생장촉진 근권세균(plant growth promoting rhizobacteria, PGPR)을 선발하는 것을 목표로 하였으며 토마토 근권에서 Variovorax sp. PMC12균주를 분리하였다. PMC12균주는 in vitro에서 PGPR의 특성으로 알려진 암모니아, IAA, 시드로포아 및 ACC 탈아민효소를 생성하였다. PMC12 균주를 처리한 토마토는 대조구에 비해 염, 저온 및 건조 스트레스 조건에서 지상부 생체중이 유의적으로 높았다. 또한 PMC12 균주를 처리한 토마토는 Ralstonia solanacearum에 의한 세균성 시들음병에 대한 저항성이 증가되었다. 결과적으로 PMC12 균주는 식물의 비생물적 스트레스 및 생물적 스트레스에 대한 감수성을 감소시키는 유망한 생물학적 방제제 및 생물활성제로 사용될 수 있을 것으로 전망된다.

An enhanced root system developmental responses under drought by inoculation of rhizobacteria (Streptomyces mutabilis) contributed to the improvement of dry matter production in rice

  • Drought limits rice production under upland condition. This study quantified the effect of rhizobacteria inoculation on rice root system developmental response to drought and its role in maintaining high soil water use, and dry matter production under drought using NSIC Rc192 (rainfed lowland rice variety). The source of inoculant was Streptomyces mutabilis, a recently isolated rhizobacteria containing plant growth promoting compounds such as ACC deaminase, indole-3-acetic acid and phosphatase (Cruz et al., 2014, 2015). In the first experiment, pre-germination inoculation of seeds with S. mutabilis significantly increased the shoot and root (radicle) length as well as root hair lengths, relative to the non-inoculated control. In the second experiment, rice plants inoculated with S. mutabilis and grown in rootbox with soil generally had greater total root length under drought regardless of the timing of inoculations, relative to the non-inoculated control. Consequently, improved root system development contributed to the increase in soil water uptake under drought and thus, dry matter production. Among inoculation treatments, one-time inoculation of S. mutabilis either at pre-germination or pre-drought stress at 14 days after sowing (DAS), had significantly greater shoot dry matter production than three-time inoculation at pre-germination, at thinning (3 DAS) and at pre-drought (14 DAS). This study demonstrated the effectiveness of rhizobacteria (S. mutabilis) containing growth promoting compounds for enhancing drought dehydration avoidance root traits and improving the growth of rice plants under drought condition.

고추 식물의 건조 스트레스 완화를 위한 미생물 선발 (Screening of Bacterial Strains for Alleviating Drought Stress in Chili Pepper Plants)

  • 식물 근권과 내생에서 분리한 447 균주 중 식물 생장 촉진특성과 건조 내성이 있는 28 균주를 일차적으로 스크리닝 하였으며, PEG에 의한 인위적 건조 스트레스 조건에서 잎의 상대수분함량과 MDA를 기반으로 GLC02와 KJ40을 선발하였다. 이 두 균주의 효과를 검증하기 위해 밭흙을 사용한 자연 건조에서 식물 포트 검정을 하였으며, 기공전도도와 지상부(줄기와 잎) 무게가 유의하게 증가한 반면 MDA가 감소하였다. 병 억제 효과에서는 GLC02를 처리할 경우 역병의 병진전도가 감소하였으며, KJ40을 처리할 경우 세균성 점무늬병에 대한 억제효과가 있었다. 이를 토대로, GLC02와 KJ40을 처리할 경우 건조 스트레스를 경감시켜주며 식물생장의 증진, 병 억제효과를 유도하여 생물비료의 소재로 사용할 수 있을 것으로 생각된다.

Identification and Characterization of Microbial Community in the Coelomic Fluid of Earthworm (Aporrectodea molleri)

  • Earthworms play an important role in soil fertilization, interacting continually with microorganisms. This study aims to demonstrate the existence of beneficial microorganisms living in the earthworm's immune system, the coelomic fluid. To achieve this goal, a molecular identification technique was performed, using cytochrome c oxidase I (COI) barcoding to identify abundant endogenic earthworms inhabiting the temperate zone of Rabat, Morocco. Then, 16S rDNA and ITS sequencing techniques were adopted for bacteria and fungi, respectively. Biochemical analysis, showed the ability of bacteria to produce characteristic enzymes and utilize substrates. Qualitative screening of plant growth-promoting traits, including nitrogen fixation, phosphate and potassium solubilization, and indole acetic acid (IAA) production, was also performed. The result of mitochondrial COI barcoding allowed the identification of the earthworm species Aporrectodea molleri. Phenotypic and genotypic studies of the sixteen isolated bacteria and the two isolated fungi showed that they belong to the Pseudomonas, Aeromonas, Bacillus, Buttiauxella, Enterobacter, Pantoea, and Raoultella, and the Penicillium genera, respectively. Most of the isolated bacteria in the coelomic fluid showed the ability to produce β-glucosidase, β-glucosaminidase, Glutamyl-β-naphthylamidase, and aminopeptidase enzymes, utilizing substrates like aliphatic thiol, sorbitol, and fatty acid ester. Furthermore, three bacteria were able to fix nitrogen, solubilize phosphate and potassium, and produce IAA. This initial study demonstrated that despite the immune property of earthworms' coelomic fluid, it harbors beneficial microorganisms. Thus, the presence of resistant microorganisms in the earthworm's immune system highlights a possible selection process at the coelomic fluid level.

Siderophore-producing rhizobacteria reduce heavy metal-induced oxidative stress in Panax ginseng Meyer

  • Background: Panax ginseng is one of the most important medicinal plants and is usually harvested after 5 to 6 years of cultivation in Korea. Heavy metal (HM) exposure is a type of abiotic stress that can induce oxidative stress and decrease the quality of the ginseng crop. Siderophore-producing rhizobacteria (SPR) may be capable of bioremediating HM contamination. Methods: Several isolates from ginseng rhizosphere were evaluated by in vitro screening of their plant growth-promoting traits and HM resistance. Subsequently, in planta (pot tests) and in vitro (medium tests) were designed to investigate the SPR ability to reduce oxidative stress and enhance HM resistance in P. ginseng inoculated with the SPR candidate. Results: In vitro tests revealed that the siderophore-producing Mesorhizobium panacihumi DCY119T had higher HM resistance than the other tested isolates and was selected as the SPR candidate. In the planta experiments, 2-year-old ginseng seedlings exposed to 25 mL (500 mM) Fe solution had lower biomass and higher reactive oxygen species level than control seedlings. In contrast, seedlings treated with 108 CFU/mL DCY119T for 10 minutes had higher biomass and higher levels of antioxidant genes and nonenzymatic antioxidant chemicals than untreated seedlings. When Fe concentration in the medium was increased, DCY119T can produce siderophores and scavenge reactive oxygen species to reduce Fe toxicity in addition to providing indole-3-acetic acid to promote seedling growth, thereby conferring inoculated ginseng with HM resistance. Conclusions: It was confirmed that SPR DCY119T can potentially be used for bioremediation of HM contamination.
