Rice functional genomics using T-DNA mutants (T-DNA 돌연변이를 이용한 벼 기능 유전체 연구)

  • Rice (Oryza sativa) is a major cereal crop that has been developed as a monocot model species. In past decades rice researchers have established valuable resources for functional genomics in rice, such as complete genome sequencing, high-density genetic maps, a full length cDNA database, genome-wide transcriptome data, and a large number of mutants. Of these, rice mutant lines are very important to definitively determine functions of genes associated with valuable agronomic traits. In this review we summarize the progress of functional genomics approaches in rice using T-DNA mutants.

  • As the completion of genome sequencing, large collection of expression data and the great efforts in annotating plant genomes, the next challenge is to systematically assign functions to all predicted genes in the genome. Functional genome analysis of plants has entered the high-throughput stage. The generations and collections of mutants at the genome-wide level form technological platform of functional genomics. However, to identify the exact function of unknown genes it is necessary to understand each gene's role in the complex orchestration of all gene activities in the plant cell. Gene function analysis therefore necessitates the analysis of temporal and spatial gene expression patterns. The most conclusive information about changes in gene expression levels can be gained from analysis of the varying qualitative and quantitative changes of messenger RNAs, proteins and metabolites. New technologies have been developed to allow fast and highly parallel measurements of these constituents of the cell that make up gene activity. We have reviewed currently employed technologies to identify unknown functions of predicted genes including map-based cloning, insertional mutagenesis, reverse genetics, chemical mutagenesis, microarray analysis, FOX-hunting system, gene silencing mutagenesis, proteomics and chemical genomics. Recent improvements in technologies for functional genomics enable whole-genome functional analysis, and thus open new avenues for studies of the regulations and functions of unknown genes in plants.

S-Nitrosoglutathione (GSNO) Alleviates Lead Toxicity in Soybean by Modulating ROS, Antioxidants and Metal Related Transcripts

  • Heavy metals, including lead (Pb) toxicity, are increasing in soil and are considered toxic in small amounts. Pb contamination is mainly caused by industrialization - smelting, mining. Agricultural practices - sewage sludge, pests and urban practices - lead paint. It can seriously damage and threaten crop growth. Pb can adversely affect plant growth and development by affecting the photosystem, cell membrane integrity, and excessive production of reactive oxygen species (ROS) such as hydrogen peroxide (H2O2)andsuperoxide(O2.-). NO is produced via enzymatic and non-enzymatic antioxidants to scavenge ROS and lipid peroxidation substrates in terms of protecting cells from oxidative damage. Thus, NO improves ion homeostasis and confers resistance to metal stress. Our results here suggest that exogenous NO may aid in better growth under lead stress. These enhancements may be aided by NO's ability in sensing, signaling and stress tolerance in plants under heavy metal stress in combination with lead stress. Our results show that GSNO has a positive effect on soybean seedling growth in response to axillary pressure and that NO supplementation helps to reduce chlorophyll maturation and relative water content in leaves and roots following strong burst under lead stress. GSNO supplementation (200 µM and 100 µM) reduced compaction and approximated oxidative damage of MDA, proline and H2O2. Under plant tension, a distorted appearance was found in the relief of oxidative damage by ROS scavenging by GSNO application. In summary, modulation of these NO, PCS and prolongation of metal past reversing GSNO application confirms the detoxification of ROS induced by toxic metal rates in soybean. Molecular approaches for improvement of medicinal and aromatic plants

  • Medicinal and aromatic plants (MAPs) are important sources for plant secondary metabolites, which are important for human healthcare. Improvement of the yield and quality of these natural plant products through conventional breeding is still a challenge. However, recent advances in plant genomics research has generated knowledge leading to a better understanding of the complex genetics and biochemistry involved in biosynthesis of these plant secondary metabolites. This genomics research also concerned identification and isolation of genes involved in different steps of a number of metabolic pathways. Progress has also been made in the development of functional genomics resources (EST databases and micro-arrays) in several medicinal plant species, which offer new opportunities for improvement of genotypes using perfect markers or genetic transformation. This review article presents an overview of the recent developments and future possibilities in genetics and genomics of MAP species including use of transgenic approach for their improvement.
