Molecular Cloning and Characterization of a Novel Stem-specific Gene from Camptotheca acuminata

  • In higher plants, P450s participate in the biosynthesis of many important secondary metabolites. Here we reported for the first time the isolation of a new cytochrome P450 cDNA that expressed in a stem-specific manner from Camptotheca acuminata (designated as CaSS), a native medicinal plant species in China, using RACE-PCR. The full-length cDNA of CaSS was 1735 bp long containing a 1530 bp open reading frame (ORF) encoding a polypeptide of 509 amino acids. Bioinformatic analysis revealed that CASS contained a heme-binding domain PFGXGRRXCX and showed homology to other plant cytochrome P450 monooxygenases and hydroxylases. Southern blotting analysis revealed that there was only one copy of the CaSS present in the genome of Camptotheca acuminata. Northern blotting analysis revealed that CaSS expressed, in a tissue-specific manner, highly in stem and lowly in root, leaf and flower. Our study suggests that CaSS is likely to be involved in the phenylpropanoid pathway.

LED array design for optimal combination of plant grown (식물재배를 위한 최적LED 배열조합설계)

  • This paper is suitable for household plant factory by design and using both energy-saving LED and solar technology. Conventional household plant factory only depending on natural sunlight is sensitive for the change of external environment. Another a big problem of conventional common household plant factory is large power consumption. Recently interest in wellbeing food such as chemical-free is increased abruptly. To solve these two problems, this paper describes hybrid type of household plant. In particular, reducing the power photosynthesis photon flux density (PPFD) is kept uniform to enhance the growth of the plant. Ambient light sensor is adopted for the control of proper combination of sunlight and LED to keep PPFD constant.

  • Plant tissue culture and molecular biology techniques are powerful tools of biotechnology that can complement conventional breeding, expedite crop improvement and meet the demand for availability of uniform clones in large numbers. Jatropha curcas Linn., a non-edible, eco-friendly, non-toxic, biodegradable fuel-producing plant has attracted worldwide attention as an alternate sustainable energy source for the future. This review presents a consolidated account of biotechnological interventions made in J. curcas over the decades and focuses on contemporary information and trends of future research.

  • Many therapeutic glycoproteins have been successfully generated in plants. Plants have advantages regarding practical and economic concerns, and safety of protein production over other existing systems. However, plants are not ideal expression systems for the production of biopharmaceutical proteins, due to the fact that they are incapable of the authentic human N-glycosylation process. The majority of therapeutic proteins are glycoproteins which harbor N-glycans, which are often essential for their stability, folding, and biological activity. Thus, several glyco-engineering strategies have emerged for the tailor-making of N-glycosylation in plants, including glycoprotein subcellular targeting, the inhibition of plant specific glycosyltranferases, or the addition of human specific glycosyltransferases. This article focuses on plant N-glycosylation structure, glycosylation variation in plant cell, plant expression system of glycoproteins, and impact of glycosylation on immunological function. Furthermore, plant glyco-engineering techniques currently being developed to overcome the limitations of plant expression systems in the production of therapeutic glycoproteins will be discussed in this review.

  • The hazard conditions on plant works of the steel industry have been investigated to efficiently prevent from industrial accidents. The plant works are eventually carried out by workers of smaller service companies where the safety management systems are not well established, on behalf of the main steel company. Based on the preliminary risk assessment on the plant working and open literature reviews, the safety management systems, which can be applied to the plant workplace of each process, are studied and then the step-by-step 4M check-list method is established. Accordingly, the risk assessment technique using 4M checklist for the plant workplace is proposed. In addition, a standardized flow-chart for safety management of plant work is introduced. These risk assessment technique and flow-chart can definitely contribute to the reduction of industrial accidents, considering the risk characterization and the unsafe conditions in small and medium-sized plant workplace, as effective safety management tools and safety guide line.

  • The power plant companies reached the culmination for growth in the domestic market. Besides, we have faced many challenges such as an electricity opening market trend for overseas, banning the use of coal due to the United nations framework convention on climate change, and the meticulous attention regarding the government's electricity charge plan. On the other hand, the overseas business has been a critical issue since China and many other developing countries expanded their projects abroad. Another factor is that power plant industry is being privatized, and it made the market regulation a bit loose. Thus, the market environment became favorable toward those companies which planned many overseas businesses. In this research, the power plant company's current condition for construction and operation as well as its technical competitiveness were analyzed, and an alternative plan using SWOT analysis for entering an oversea market was made. It dealt with both internal and external factors. Also, examined was the current situation under the power plant industry dealing with restructure for electric industry, lack of fossil fuel, and the United nations framework convention on climate change. From the research, it was suggested that many successful strategies to enter the overseas business by using the market trend I researched.

Development and Working Efficiency of Supporting Program for the Parametric Electrical Outfit Production Design of Offshore Plant Based on PML (PML 기반 파라메트릭 해양플랜트 전장생산설계 지원 프로그램 개발 및 업무 효율성 연구)

  • Recently, because of the global recession of the offshore plant industry and low-cost orders, there has been increasing interest in strengthening the competitiveness of domestic companies for the design and production technologies of offshore plants. However, in the offshore plant design field, the Plant Design Management System (PDMS), which is a 3D CAD program for plant layout developed by AVEVA Marine, is already commonly used as offshore plant design software and widely used in large domestic shipyards and cooperative design companies. Under this background, we have been thinking about ways to design better with the existing software. In this study, we developed a parametric design program to maximize the efficiency and reduce the working time for offshore plant electrical outfit production design based on the Programmable Macro Language (PML) of PDMS. We also examined its performance. By applying the developed program to the offshore plant module selected as an application example, it was confirmed that a 50% improvement in the work efficiency of cable tray design could be obtained compared with the existing method, with work efficiency improvements of 80% or more in other field design work.

  • The purpose of this study is to present a cost classification system that can be used in gas plant construction projects. Towards this end, it examined the detailed statements of the construction companies which had experience in carrying out plant construction projects. Based on the above, it also presented a life-cycle (i.e., EPCC) cost classification system for the gas plant construction projects by conducting the Delphi analysis through the expert opinions.

A Real-Time PCR Assay for the Quantitative Detection of Ralstonia solanacearum in Horticultural Soil and Plant Tissues

  • A specific and rapid real-time PCR assay for detecting Ralstonia solanacearum in horticultural soil and plant tissues was developed in this study. The specific primers RSF/RSR were designed based on the upstream region of the UDP-3-O-acyl-GlcNAc deacetylase gene from R. solanacearum, and a PCR product of 159 bp was amplified specifically from 28 strains of R. solanacearum, which represent all genetically diverse AluI types and all 6 biovars, but not from any other nontarget species. The detection limit of $10^2\;CFU/g$ tomato stem and horticultural soil was achieved in this real-time PCR assay. The high sensitivity and specificity observed with field samples as well as with artificially infected samples suggested that this method might be a useful tool for detection and quantification of R. solanacearum in precise forecast and diagnosis.

Modeling of a Small Group Scale TMR Plant for Beef Cattle and Dairy Farm in Korea(II) - Performance Test and Cost Analysis of the Model Plant - (한우 및 낙농 단지용 소형 TMR 플랜트 모델 개발(II) - 모델의 성능시험 및 경제성분석 -)

  • A Model of small scale total mixed rations(TMR) plant which can be utilized round bales was developed, tested and analyzed in this study. This study consist of two parts. One is development of a small scale TMR plant model which was already reported at the previous paper. This is the second part of the study. For the study, a series of tests of the model plant were performed and its costs was analyzed. Also, the break-even point of the model plant by comparing with market price of commercial TMR feed was determined. Results of the research are summarized as follows ; As the results of mixing test, the average coefficient of variation(CV) value for mixing of the feed was 13.0 % at the gate of the mixer. The production cost was estimated as 8,298 won/head for dairy cattle farm and 2,495 won/head for beef cattle farm, when producing 8 batch a day. Also, it is recommended to utilize the model plant when farm size is over 79 heads for dairy cattle farm and 113 heads for beef cattle farm. As an overall conclusion, the model plant designed for farm size TMR feed mill will be very useful model for both beef cattle and dairy farms in Korea. Also it is expected that the capital investment for the model plant can be recovered with 8 months compare with purchasing commercial TMR feed if the model plant feeds 1,000 beef cattle approximately.
