Foliar sprays of three plant resistance inducers, including chitosan (CH), potassium sorbate (PS) ($C_6H_7kO_2$), and potassium bicarbonates (PB) ($KHCO_3$), were used for resistance inducing against Erysiphe cichoracearum DC (powdery mildew) infecting okra plants. Experiments under green house and field conditions showed that, the powdery mildew disease severity was significantly reduced with all tested treatments of CH, PS, and PB in comparison with untreated control. CH at 0.5% and 0.75% (w/v) plus PS at 1.0% and 2.0% and/or PB at 2.0% or 3.0% recorded as the most effective treatments. Moreover, the highest values of vegetative studies and yield were observed with such treatments. CH and potassium salts treatments reflected many compounds of defense singles which leading to the activation power defense system in okra plant. The highest records of reduction in powdery mildew were accompanied with increasing in total phenolic, protein content and increased the activity of polyphenol oxidase, peroxidase, chitinase, and ${\beta}$-1,3-glucanase in okra plants. Meanwhile, single treatments of CH, PS, and PB at high concentration (0.75%, 2.0%, and/or 3.0%) caused considerable effects. Therefore, application of CH and potassium salts as natural and chemical inducers by foliar methods can be used to control of powdery mildew disease at early stages of growth and led to a maximum fruit yield in okra plants.

Plant growth promoting fungi(PGPF) obtained from zoysiagrass rhizosphere offer dual advantages - induse systemic disease resistance response in cucumber to C. orbiculare infection and cause enhancement of plant growth and increase yield. PGPF protected plants either by colonizing roots or by their metabolites. PGPF offer an advantage by protecting plants for more than 9 weeks and 6 week in the greenhouse and field. PGPF-induced plants limited pathogen spore germination and decreased the number of infection hyphae on the leaf, and increased lignification at places of attempted pathogen infection, thus reducing the pathogen spread. PGPF elicited increased activities of chitinascs, glucanases, peroxidase, polyphenol oxidase, and phenylalanine ammonia lyase to C. orbiculare infection in cucumber plants. The role of PGPF in elevating cucumber defense response to pathogen infection suggests potential application of PGPF as biological control agents.
Phytophthora infestans is the causal agent of potato and tomato late blight, one of the most devastating plant diseases. P. infestans secretes effector proteins that are both modulators and targets of host plant immunity. Among these are the so-called RXLR effectors that function inside plant cells and are characterized by a conserved motif following the N-terminal signal peptide. In contrast, the effector activity is encoded by the C terminal region that follows the RXLR domain. Recently, I performed in planta functional profiling of different RXLR effector alleles. These genes were amplified from a variety of P. infestans isolates and cloned into a Potato virus X (PVX) vector for transient in planta expression. I assayed for R-gene specific induction of hypersensitive cell death. The findings included the discovery of new effector with avirulence activity towards the Solanum bulbocastanum Rpi-blb2 resistance gene. The Rpi-blb2 encodes a protein with a putative CC-NBS-LRR (a coiled-coil-nucleotide binding site and leucine-rich repeat) motif that confers Phytophthora late blight disease resistance. We examined the components required for Rpi-blb2-mediated resistance to P. infestans in Nicotiana benthamiana. Virus-induced gene silencing was used to repress candidate genes in N. benthamiana and to assay against P. infestans infections. NbSGT1 was required for disease resistance to P. infestans and hypersensitive responses (HRs) triggered by co-expression of AVRblb2 and Rpi-blb2 in N. benthamiana. RAR1 and HSP90 did not affect disease resistance or HRs in Rpi-blb2-transgenic plants. To elucidate the role of salicylic acid (SA) in Rpi-blb2-mediated resistance, we analyzed the response of NahG-transgenic plants following P. infestans infection. The increased susceptibility of Rpi-blb2-transgenic plants in the NahG background correlated with reduced SA and SA glucoside levels. Furthermore, Rpi-blb2-mediated HR cell death was associated with $H_2O_2$, but not SA, accumulation. SA affects basal defense and Rpi-blb2-mediated resistance against P. infestans. These findings provide evidence about the roles of SGT1 and SA signaling in Rpi-blb2-mediated resistance against P. infestans.

In this research, the relationships among the 31 microsatellite markers with charcoal rot disease resistance related indices in 130 different soybean cultivars and lines were evaluated using association analysis based on the general linear model (GLM) and the mixed linear model (MLM) by the Structure and Tassel software. The results of microsatellite markers showed that the genetic structure of the studied population has three subpopulations (K=3) which the results of bar plat also confirmed it. In association analysis based on GLM and MLM models, 31 and 35 loci showed significant relationships with the evaluated traits, respectively, and confirmed considerable variation of the studied traits. The identified markers related to some of the studied traits were the same which can probably be due to pleiotropic effects or tight linkage among the genomic regions controlling these traits. Some of these relationships were including, the relationship between Sat_252 marker with amount of charcoal rot disease, Satt359, Satt190 and Sat_169 markers with number of microsclerota in stem, amount of charcoal rot disease and severity of charcoal rot disease, Sat_416 marker with number of microsclerota in stem and amount of charcoal rot disease and the Satt460 marker with number of microsclerota in stem and severity of charcoal rot disease. The results of this research and the linked microsatellite markers with the charcoal rot disease-related characteristics can be used to identify the suitable parents and to improve the soybean population in future breeding programs.

TMV resistant lines (TRLs) originated from the Blo plant of Nicotiana tabacum cv. NC82 transformed with TMV coat protein cDNA which initially showed delayed disease symptom were selected for increased resistance in each subsequent generation. The result of field experiment of the transgenic tobacco lines in the fifth generation for TMV resistance and their response to other tobacco diseases (black shank, bacterial wilt, and powdery mildew) is described in this report. When fifteen TRLs of the fifth generation were tested for TMV resistance by mechanically inoculating the individual plants, over 95 percent of the plants of 6 lines showed complete resistance even 8 weeks after the inoculation. Average frequency of the resistant plants in TRLs of the fifth generation 8 weeks after the inoculation was 87%. Stable insertion and expression of TMV coat protein cDNA in the fifth generation of the transgenic tobacco plant were confirmed by PCR and immunoblot hybridization, respectively. All TRLs were resistant to the black shank but were susceptible to the bacterial wilt disease and the powdery mildew to the same degree as non-transgenic NC82 was. Therefore, it was indicated that the phenotypes related at least to disease resistance were not changed in the transgenic tobacco. We investigated the virulence genes, O-serotypes, antimicrobial resistance of pathogenic E. coli isolated from carcasses (n=455) and environmental (n=372) samples of 11 cattle and 12 pig slaughterhouses from December 2020 to December 2021. E. coli were isolated from nine carcasses (2.0%), three slaughter facilities (1.4%), two utensils (2.7%) and three abattoir workers (3.5%) from four cattle and four pig slaughterhouses. Among all isolates, 13 STEC (76.5%) were identified, followed by four EPEC (23.5%). As a result of the antibiotic susceptibility test, all isolates were resistant to at least one antimicrobial, of which 70.6% isolates showed multidrug resistance patterns. The serotypes were diverse in pigs compared to cattle, with serotypes O18, O66, O109 in cattle and O9, O76, O85, O100, O153, and O159 in pigs. In a single cattle slaughterhouse, eight STEC O66 were isolated from various types of sample (4 slaughter animal surfaces, 3 gloves, and 1 knife) with two antimicrobial resistance patterns (CHL-FIS-STR and CHL-FIS). Those two types of strain were suspected cross-contamination from utensils to slaughter animal surfaces. These results showed that pathogenic E. coli were detected in carcasses and various environmental samples in cattle and pig slaughterhouses. Nationwide monitoring and hygiene management are required to prevent cross-contamination of STEC isolate slaughterhouses.
