배추 무사마귀병(Plasmodiophora brassicae)의 발병유인 및 약제방제 (Environmental Factors Favoring the Disease Development and Chemical Control of Clubroot(Plasmodiophora brassicae) in Chinese Cabbage)

  • 배추 무사마귀병의 발병에 관여하는 환경요인의 영향을 구명하고 약제처리 방법을 연구한 결과는 다음과 같다. 배추 무사마귀병의 발병율은 근침지접종이나 관주 접종 보다는 전염원 삽입접종에서 현저히 높았다. 발병 유인으로 pH와 온도를 조사한 결과 토양 pH는 5-6에서, 토양온도는 $20-25^{\circ}C$에서 발병율이 높았고 pH 7.0이상, 토양온도 $30^{\circ}C$ 이상에서는 발병율이 급격히 감소하였다. 배추 무사마귀병의 방제를 위해서는 fluazinam 0.5% 분제의 토양살포나 종자분의의 단용처리보다는 둘을 병용처리하는 것이 효과적이다.

Biological Control of Gray Mold Rot of Perilla Caused by Boftis cinerea 1. Resistance of Perilla Cultivars and Selection of Antagonistic Bacteria

  • Resistance of perilla varieties to Botrytis cinerea LVF12 was evaluated, while antagonistic bacteria were selected and tested for their efficacy towards biological control of gray mold rot caused by B. cinerea. Among 11 perilla varieties tested for disease resistance, Milyang variety showed some degree of resistance, while the rest of varieties showed no resistance. Among 250 bacterial isolates collected from perilla loaves and rhizosphere of perilla plants, six isolates showed high levels of inhibitory effect on mycelial growth and conidial germination of B. cinerea in in vitro test. Using the pot test in growth chambers these isolates showed high levels of disease suppression, with Nl isolate showing 95.3% of control value and N4 isolate showing 90.8% of control value. Further test was performed to evaluate the two isolates ability for disease prevention and/or disease therapy, and results showed almost 100% of control vague. Isolates Nl and N4 were identified as Bacillus licheniformis and 5. megatepium, respectively, according to Bergey's manual, API 20E and 50CHB test kit, and Transmission electron microscope.

몇 가지 식물추출물의 벼 병해충 방제 효과 (Controlling Effect of Some Plant Extracts on Pathogenic Fungi and Pest of Rice)

  • 본 연구는 고삼, 정향 및 neem 추출물에 대한 항균 항충력을 규명하여 벼 친환경 재배 포장에서 병해충 방제제로의 이용가능성을 검토하고자 수행하였다. 식물추출물에 대한 잿빛곰팡이균에 대한 항균력 실험결과 정향추출물이 가장 높은 활성을 나타내었으며, 벼멸구에 대한 살충력은 고삼추출물이 가장 높은 효과를 보였는데, 고삼추출물의 300배 희석용액에서도 100%의 살충 방제가를 나타내었다. 정향과 고삼추출물에 각각 목초액(10%)과 유화전착제(10%)를 혼합하여 친환경 벼 재배포장에 살포하고 병해충 발생률을 조사한 결과, 정향추출물 처리구의 병해 방제가는 무처리구(100%) 대비 50.8%, 해충 발생은 70.6%를 나타내었고, 고삼추출물 처리구는 무처리(100%) 대비 병해 방제가는 43.1%, 해충 방제가는 79.1%로 나타났다. 결론적으로, 고삼 및 정향 추출물은 벼 친환경 재배 시 병해는 약 50%, 충해는 약 70%를 감소시킬 수 있어서 친환경농자재로의 활용 가능성을 확인하였다.

Feasible Management of Southern Corn Leaf Blight via Induction of Systemic Resistance by Bacillus cereus C1L in Combination with Reduced Use of Dithiocarbamate Fungicides

  • Dithiocarbamate fungicides such as maneb and mancozeb are widely used nonsystemic protectant fungicides to control various plant fungal diseases. Dithiocarbamate fungicides should be frequently applied to achieve optimal efficacy of disease control and avoid either decline in effectiveness or wash-off from leaf surface. Dithiocarbamates are of low resistance risk but have the potential to cause human neurological diseases. The objective of this study was to develop a strategy to effectively control plant disease with reduced use of dithiocarbamtes. Southern corn leaf blight was the model pathosystem for the investigation. When corn plants were drench-treated with Bacillus cereus C1L, a rhizobacterium able to induce systemic resistance in corn plants against southern leaf blight, frequency of spraying dithiocarbamate fungicides could be decreased. The treatment of B. cereus C1L was able to protect maize from southern leaf blight while residues of dithiocarbamates on leaf surface were too low to provide sufficient protection. On the other hand, frequent sprays of mancozeb slightly but significantly reduced growth of corn plants under natural conditions. In contrast, application of B. cereus C1L can significantly promote growth of corn plants whether sprayed with mancozeb or not. Our results provide the information that plant disease can be well controlled by rhizobacteria-mediated induced systemic resistance in combination with reduced but appropriate application of dithiocarbamate fungicides just before a heavy infection period. An appropriate use of rhizobacteria can enhance plant growth and help plants overcome negative effects caused by dithiocarbamates.

정유를 이용한 환경친화적 수삼 저장병 방제 (Environment Friendly Control of Gray Mold, a Ginseng Storage Disease Using Essential Oils)

  • 본 연구는 정유를 이용하여 환경친화적으로 Botrytis cinerea에 의한 수삼 저장병 방제방법을 개발하기 위하여 수행되었다. 정유의 일종인 methyl eugenol은 200 ppm 농도의 배지상에서 80% 이상의 높은 균사 생육억제 효과를 나타냈다. 상온에서 수삼에 직접 처리 시 무처리구에서 80% 이상의 발병률을 보일 때 methyl eugenol이나 thymol 모두 분무와 침지처리구에서 10~20% 이하로 낮게 발병하여 높은 병 방제효과를 보였다. 저온($4^{\circ}C$)에서의 처리결과도 실온과 비슷하게 포장크기와 상관없이 무처리구에 비해 발병률이 매우 낮게 나타나 역시 실용적인 효과가 뛰어났다. 2 kg 이상을 분석한 대포장 실험에서도 무처리구는 발병지수가 1.65를 보인 반면 methyl eugenol은 0.60으로 개체별 발병면적이 상대적으로 매우 좁게 나타났다. 그러나 두 물질(methyl eugenol+thymol)의 혼합처리에 의한 상승효과는 확인할 수 없었다. Methyl eugenol은 잔류독성이 없고 수삼 저장병 발생 억제효과가 뚜렷해 수삼 저장 시 처리할 수 있는 환경친화적 제제로서 활용 가능하며 특히 신선한 상태로 섭취가 가능해야하는 수삼의 특성으로 볼 때 수삼 저장병 방제제로서의 활용가치는 더욱 클 것으로 판단된다.

Effect of Potassium Phosphonate on the Control of Phytophthora Root Rot of Lettuce in Hydroponics

  • The effect of potassium phosphonate ($KH_2PO_3 or $K_2 HPO_3$) on the control of Phytophthora root rot of lettuce was evaluated in a liquid hydroponic culture. Phosphonate 100 ppm strongly inhibited mycelial growth of Phytophthora species in vitro but did not affect normal growth of lettuce in a greenhouse test. Application of the chemical before infection showed over 94% control value, while it was less than 35% when applied after infection. In a field trial, phosphonate 100 ppm, which was directly supplemented into the nutrient solution, satisfactorily controlled the disease as it did not develop until 28 days after transplanting and remained at less than 2% infection rate at the end of cultivation. Meanwhile, in the control plot, the disease initiated at 7 days after transplanting and developed rapidly reaching over 70% infection rate at 28 days. Population density of the causal pathogen, R drechsleri, in a heavily infested farm was 22.0-25.0 cfu/100 ml of nutrient solution. However, it decreased to 1.3-2.0 cfu/100 ml at 7 days after treatment with phosphonate 200 ppm.

구제역바이러스 혈청형 A 검출을 위한 peptide nucleic acid (PNA)기반 multiplex real-time RT-PCR 개발 (Developing peptide nucleic acid based multiplex real time RT-PCR to detect Foot-and-Mouth-Disease virus Serotype A)

  • There have been a total tenth FMD outbreaks in Korea and for the first time, type O and A were detected simultaneously in 2017, which led to difficulties in FMD control. For the effective prevention of FMD, the importance of discrimination of serotypes became greater. Therefore, the most urgent requirement in case of FMD outbreak is differential diagnosis of serotypes. In this study, we developed a PNA probe-mediated multiplex real-time reverse transcription-polymerase chain reaction (rRT-PCR) assay using the peptide nucleic acid (PNA) probe, which is known to be stable to nucleotide mutation and that could specifically detect the all FMDV serotype A, FMDVA Yeoncheon strain which was occurred in Korea in 2017, and FMDV A viruses shown 96% similarity with FMDVA/Yeoncheon strain, at the same time. Therefore, It is believed that the newly introduced FMDVA will be effectively diagnosed using the PNA probe multiplex RT-PCR developed in this study, and ultimately contribute to the prevention of FMD.
