Seed Germination and Dormancy Breaking of Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai

  • This study was carried out to develop an effective seed propagation method for Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai by analyzing seed dormancy types and germination characteristics. Seeds were collected between September to October at Gangwon province, and well-selected seeds were used while being dry-stored at 4±1℃. The seed size ranged 4.52 × 1.58 ㎜ and the weight of thousand seeds were 1,603.5 ± 0.02 ㎎. The moisture content was 7.2%. Seeds were achene type, and morphology characters showed an elliptical shape and rough texture, and light brown in color. Moist-chilling treatment was conducted for dormancy breaking because the seeds had an undeveloped embryo of liner type. The embryo had developed during a moist-chilling period, constantly, and fully developed in 10 weeks. Consequently, it seemed to be non-deep complex or intermediate complex type of morphophysiological dormancy, and embryo dormancy was broken by wet-chilling for 10 weeks. After 10 weeks of wet-chilling treatment, seed germination began. Germination percentage was higher in dark condition raher than light condition and recorded the maximum at 25℃ in the dark (16.3%). A pre-soaking treatment with a combined plant growth hormones promoted germination and shortened T50. Specifically, seed germination of 84.5% was achieved by pre-soaking of seeds with a combined solution of 500 ㎎/L GA3 and 10 ㎎/L kinetin for 24 h after a wet-chilling treatment for 10 weeks. Thus the effect of plant growth hormones coupled with chilling temperature on seed breaking dormancy provide asubsequent growth of seedlings for successful plantation.

  • This study was carried out to develop a vacuum nozzle seeder for large seeds and performance was tested on seed feeding, arranging. and sowing peformance. The results of this study were as follows: The operation of feeding device of the seeder was programmed to operate a period of setting time after sowing 6 rows. The setting time was decided based on a discharged seed by the angular speed of feeding roller. The arranging accuracy of 'tuktozwa', 'hukjong' and 'chambak' was 96.4%, 95.2% and 89.4% respectively. The working performance was 75.6sheet/hr which was 3.8 times higher than that of manual work. An average seeding rate of 1 grain was 97.8%.

Germination Characteristics and Early Growth of Papaver radicatum var. pseudoradicatum Seeds (두메양귀비(Papaver radicatum var. pseudoradicatum) 종자의 발아특성과 초기생장에 관한 연구)

  • This experiment was held to identify the germination feature of Papaver radicatum var. pseudoradicatum which has high economical value. Seeds were collected in the Mt. Changbai in China. To eliminate dormancy of the seeds, chilling treatment were carried out in the $5^{\circ}C$ refrigerator. 30 days, 60 days, 90 days, 120 days of chilling treatments were taken. After being Treated, seeds were under 15, 20, $25^{\circ}C$ of temperature and light, dark conditions respectively to see the differences in germination. Within many features, germination ratio of seeds and germination period were investigated. Also, features related to early growth were checked through the closer look into acrospire, growth of cotyledon of young plant. Over 30 days of chilling treatment was all effective. Especially, 120 days of chilling treatment was most effective. The seeds germinated well in the light conditions. 120 days of chilling treatment, $15^{\circ}C$, light conditions were found to be most effective conditions as 54.5% of the seeds germinated. The germination period was 4.29 days. 30 days after germinating, length of the aerial part had been 1.90 cm. But after 45 days, the length grew rapidly to 4.58 cm. Fresh weight of scales plant was also increased dramatically after 45 days. Therefore, transporting the plant 45 days after seeding judged to be good.

Effect of Gibberellin Acid on Embryo Development and Germination of Dicentra spectabilis (L.) Lem. Seeds

  • This study was carried out to develop the seed propagation method of Dicenta spectabilis (L.) Lem. which is an ornamental plant native to Korea. In the previous studies, it was found that the seeds of D. spectabilis were morphophysiologically dormant (MPD), and high and low temperature of stratification were continuously required for the embryo growth and germination of the seeds. Especially, it was most effective to store for 1 month at $20^{\circ}C$ and then to transfer to $4^{\circ}C$. The treatment of $GA_3$ was carried out to promote embryo development and germination. The seeds were submerged in 100, 200 or $500mg{\cdot}L^{-1}$ $GA_3$ for 72 hours and then stored at various conditions as follow. The temperature conditions disposed of this experiment were 1 month at 10, 15, 20, and $25^{\circ}C$ or 2, 4, 8, and 12 months at $4^{\circ}C$, respectively. As a result, the length of embryo and germination rate of the seeds were the best when stored at $4^{\circ}C$ for 8 months after $500mg{\cdot}L^{-1}$ $GA_3$ treatment. Besides, when the seeds stored at $4^{\circ}C$, significant differences in embryo length and germination rate were shown with $GA_3$ concentration and storage period. It was also proved that high-concentration of $GA_3$ could replace the high temperature and could promote germination. Consequentially, the D. spectabilis seeds were classified into intermediate simple levels among MPD types.

Effect of Tall fescue (Schedonorus phoenix Scop.) Genotype on Endophyte (Neotyphodium coenophialum) Transmission under Water stress

  • It has been known that endophyte (Neotyphodium coenophialum) is beneficial to tall fescue (Schedonorus phoenix Scop.) because the mutualistic endophyte is able to confers tolerance against abiotic and biotic stresses to tall fescue. However, this fungal endophyte produces toxic alkaloid resulting in negative effects on animal performance. Recently, Non-toxic endophyte have been developed and inserted into tall fescue to avoid detrimental effect on animal but remaining positive influence on tall fescue. In order to keep this beneficial impact, it is essential to have endophyte infected tall fescue through vertical transmission from maternal plants to seeds. Little research has been carried out on endophyte transmission. To get basic information related to endophyte transmission, experiment was conducted to examine the effect of plant genotype on endophyte transmission under water stresses. Overall endophyte concentration in seeds was higher than that in panicles and endophyte concentration in seeds and panicles relied on plant. This study revealed that drought is not a critical component to control the endophyte transmission from maternal plants to seeds. Plant genotype is an important factor controlling the endophyte transmission from plant to seed.

Effect of Cold Stratification and Gibberellin Treatment on Androsace septentrionalis L. Seed Germination

  • Androsace septentrionalis is a grass species restricted to North Korea; however, it is at the brink of extinction due to habitat loss and environmental changes caused by natural disasters and anthropogenic activities. This study was conducted to characterize the dormancy conditions of A. septentrionalis in an effort to conserve this North Korean plant resource. For this purpose, the morphological characteristics and vigor of A. septentrionalis seeds were examined, and its germination characteristics under different temperature conditions (15/6℃ and 25/15℃ ), low-temperature stratification, and gibberellin (GA3) treatment were determined. The results revealed that A. septentrionalis exhibits non-deep type morpho-physiological dormancy, and low-temperature stratification treatment was not effective in breaking the dormancy of A. septentrionalis seeds. Meanwhile, GA3 treatment significantly increased the mean germination time, rate, and speed of the seeds. The optimal conditions for the germination of A. septentrionalis seeds were 25/15℃ fluctuating temperature and 500 mg·L1 GA3 treatment. The results of this study are useful for the mass propagation of A. septentrionalis.

Propagation of asymbiotically germinated seedlings with liquid culture for endangered lady's slipper orchid (Cypripedium macranthos Sw.) (액체배양 방법을 이용한 멸종위기종 복주머니란 종자 무균발아 및 증식)

  • We had already reported the successful germination for green pods of purple lady's slipper orchid (Cypripedium macranthos Sw.). The green pod methods is to take immature seeds in green capsules, sterilize the capsule, and take out the sterile seeds. This method, however, needs very critical time of harvest. The critical time of seed harvest changes depending upon the species, condition of the specimen, and climatic influence, and the right time lies between 5 and 12 weeks after fertilization. In this study, the mature seeds were collected after 120-130 days with hand-polination of lady's slipper orchids. Mature seeds are usually dormant and it has to be overcome, either with hormone or storing the seeds near freezing for two or three months to break dormancy. The seeds were first surface sterilized with 70% ethanol and then transferred 1% NaOCl for 10-15 minutes, followed by rinses 3 times with sterilized distilled water. The cypripedium seeds consists of an embryo within a seed coat known as a testa. The testa is water repellent and the seed has a large air space between the embryo and testa so the seed tends to float on water. We had resolved the problems with vacuum pump to soak water into the testa before sterilization. The seeds were placed on liquid or agar solidified germination media. Cultures were incubated at $24{\pm}1^{\circ}C$ in dark. The seeds were germinated in 6-8 weeks in liquid suspension culture (germination percentage over 18%); however, the seeds on agar solidified media took more than 5 months to germinate and the germination percentage less than 5%. The most effective media for liquid culture was 1/4 strength Murashige and Skoog (MS) medium with 50 ml/l coconut water ($4brix^{\circ}$) at pH 5.8.

Effects of Priming and Growth Regulator Treatment of Seed on Emergence and Seedling Growth of Rice

  • An experiment was carried out to determine the effects of priming and growth regulator treatment of seeds on the emergence and seedling growth of rice, Oryza sativa L. (cv. 'Ilpumbyeo'). Normal seeds were primed in a -0.6 MPa polyethylene glycol solution at 15$^{\circ}C$ for four days with air-bubbling. Then both primed and non-primed seeds were soaked in water, 100 ppm GA$_3$, 2 ppm ABA, and 10 ppm kinetin solutions for 24 hours. The seeds were planted in soil at 3 and 5 cm depths and allowed to germinate in a growth cabinet at 2$0^{\circ}C$. Generally, the emergence rate at the 3 cm seeding depth was higher and emerged faster compared with the 5 cm seeding depth. The emergence rate of primed seeds was higher and emerged faster compared to non-primed seeds. GA$_3$ and kinetin treatments were the most effective to improve the emergence rate of non-primed seeds. Coleoptile length at the 5 cm seeding depth was longer than that at the 3 cm seeding depth. Leaf number, plant height, and root length of primed seedlings were higher compared with non-primed seeds. GA$_3$ promoted the elongation of plant height and mesocotyle length.

Change in Germination and Physiological Properties of Hippophae rhamnoides Seeds by Different Storage Period (갈매보리수나무 종자의 저장기간에 따른 발아 및 생리적 특성)

  • Germination properties, leachate electrical conductivity (EC), and inorganic compound leaching were analyzed to ascertain the storage ability and change of physiological characteristics during storage of Hippophae rhamnoides seeds. Seeds were placed in an incubator at 25℃ and sown in different soil media (sand, vermiculite and horticultural substrate) after being stored for 6, 18 and 30 months at 2℃. All germination properties decreased in accordance to an increase of the seed storage period. Compared with the seed storage for 18 months, germination percentage (GP), germination performance index (GPI), and germination value (GV) of seeds stored for 30 months decreased by more than 50%. When the seeds were sown in different soil media in a greenhouse, those germination properties were similar to the seeds germinated in an incubator, and mean germination time, GPI and GV had a significant difference except GP among soil media. EC and inorganic ion concentration had a strong positive correlation with the seed storage period, but the ratios of inorganic ions from stored seeds revealed that K+/Mg2+ and Na+/Mg2+ were inversely correlated with the storage period.

Study on the Seed Germination and Salt Tolerance of Plants in Reclaimed Salt Area (간척지내 식물종자의 내염성과 발아에 관한 연구)

  • The germination character and the salt tolerance of seeds collected in a salt reclaimed area were studied. Twenty species of seeds out of 30 species collected were germinated under natural or continuous light conditions but only 16 species under dark condition. Germination percentage of seeds under dark was more decreased than those under other two conditions and speed of germination was accelerated at high temperature. It is clear that almost all the wild weeds were the light germinated seeds. The higher concentration of NaCl for germination inhibited to make the lower percentage and rate of germination. The germination curves of seeds treated with NaCl solution were classified into 3 different curves; a steeper, a parabola and intermediate. The critical concentrations of NaCl for germination were 2.3% for Brassica napus, 1.8% for Echinochloa hispidula, 1.5% for Setaria lutescens, 1.3% for Aster koraiensis, 0.7% for Bromus japonicus and 0.6% for Glyceria acutiflora.
