Hypogonadism is a clinical syndrome that results in hormone deficiency and can be classified as 1) primary caused by the gonadal failure and 2) secondary by the hypothalamus-pituitary gland dysfunction and/or cardiometabolic complications. Recently the presence of thyroid hormone receptors in different testicular cell types was demonstrated, and thus thyroid dysfunctions would be another cause of secondary hypogonadism. Thus, we investigated the effects of perinatal hypothyroidism on hypogonadism in male Sprague-Dawley rats. Perinatal hypothyroidism was induced by daily administration of 0.05% 6-propyl-2-thiouracil (PTU) by tap water from gestation day 15, which were compared with negative control (PTU (-)) group. At postnatal day 28, hypothyroid pups were divided into 2 groups: PTU (+) group - continued PTU treatment and PTU (+/-) group - stopped PTU until postnatal day 49. Body weights, dehydrotesosterone (DHT), and testosterone levels were checked 2 and 3 weeks after grouping. Body weights were significantly decreased in PTU(+) and PTU(+/-) groups compared with PTU (-) group at postnatal day 28. 3 weeks later, PTU (+/-) group significantly gained weight compared with PTU (+) group. DHT and testosterone levels significantly decreased with PTU treatment, but increased 3 weeks after stopping PTU administration. Perinatal PTU-induced hypothyroid hypogonadism was sustained for 2 weeks after stopping PTU administration, but restored gonadal hormone levels 3 weeks after stopping PTU. These results suggest that researchers should design an experiment on hypothyroid hypogonadism based on the estimated period.
Objective: Elevated serum progesterone (P) levels on triggering day have been known to affect the pregnancy rate of in vitro fertilization (IVF). This study aimed to identify the possible factors influencing serum P levels on triggering day in stimulated IVF cycles. Methods: Three hundred and thirty consecutive fresh IVF cycles were included in the study. All cycles were first attempts and were performed in a single infertility center. The indications for IVF were male factor infertility (n=114), ovulatory infertility (n=84), endometriosis (n=61), tubal infertility (n=59), unexplained infertility (n=41), and uterine factor infertility (n=39). A luteal long protocol of a gonadotropin-releasing hormone (GnRH) agonist (n=184) or a GnRH antagonist protocol (n=146) was used for pituitary suppression. Ovarian sensitivity was defined as the serum estradiol level on triggering day per 500 IU of administered gonadotropins (OS[a]) or the retrieved oocyte number per 500 IU of administered gonadotropins (OS[b]). Results: Univariate analysis revealed that the serum P level on triggering day was associated with the serum estradiol level on triggering day (r=0.379, p<0.001), the number of follicles ${\geq}14mm$ (r=0.247, p<0.001), the number of retrieved oocytes (r=0.384, p<0.001), and ovarian sensitivity (OS[a]: r=0.245, p<0.001; OS[b]: r=0.170, p=0.002). The woman's age, body mass index, antral follicle count, and basal serum follicle stimulating hormone and estradiol levels were not associated with serum P level on triggering day. The serum P level on triggering day did not show significant variation depending on the type or cause of infertility, pituitary suppression protocol, or the type of gonadotropins used. Conclusion: The serum P level on triggering day was closely related to the response to ovarian stimulation.
In 27 patients with the past history of poor response to the gonadotropin superovulation induction due to poor follicular growth or permature surge of endogenous luteinizing hormone, the effectiveness of pituitary supperssion with the gonadotropin releasing hormone agonist(GnRH-a) in in vitro fertilization(IVF) program was evaluated in 43 cycles using a combination regimen of D-Trp-6 LHRH(Decapeptyl, Ferring)and FSH/hMG from June, 1989 to August, 1990 at Korea University Hospital IVF Clinic. At midluteal phase of menstrual cycle, Decapeptyl-CR was administered by long-term protocol to minimize initial agonistic effect of endogenous gonadotropins. After the confirmation of pituitary suppression, about 2-3 weeks after GNRH-a administration, ovarian follicle growth was stimulated with FSH/hMG and followed by transvaginal ultrasonic measurement of follicle size and by monitoring of serm E2 and LH if necessary. When compared with the control group stimulated with gonadotropin regimen only, the cancellation rate and occurrence rate of premature LH surge during gonadotropin treatment were significantly lower in study group(11.6% and 2.4%, respectively). There is no significant differences in the mean number of aspirated oocytes, fertilization/cleavage rate, embryo transfer(ET) rate, and mean number of embryos transferred between the two groups. The pregnancy rate per treatment cycle, 16.3%, and per ET cycle, 23.3%, were significantly higher in the study group compared with those of control group. These data suggest that GnRH-a therapy is effective for previous poor responder In gonadotropin superovulation induction for IVF.
Treatment of newborn female rats with gonadal steroids induces permanent sterility in adulthood. We investigated the alteration in expression patterns of gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) in neonatally estrogenized sterile rats (ESR). Newborn female rats received daily injections of 17${\beta}$-estradiol (E, 10 ${\mu}$g) from the day of birth (day 1) to postnatal day 5. Controls were subjected to vehicles over the same period. All animals were sacrificed on week 7 after birth. Hypothalamic GnRH mANA levels were markedly higher in all ESR than in controls, while hypothalamic GnRH contents in ESR increased in proportion to the frequency of daily administration of E. However, both pituitary LH6 mRNA and serum LH levels were inversely decreased by the same treatment. The data indicate that neonatal exposure of E equally elevates the expression of GnRH gene, but reduces the secretion of GnRH, accordingly leading to attenuation of LH6 gene expression and circulating LH levels. The temporal effect of E and/or progesterone (P) on GnRH and LH6 mRNA levels was also examined in ESR. Newborn female rats were daily injected with E (10 ${\mu}$g) or vehicle for five successive days from day 1 and ovariectomized at week 5. They were implanted with E (235 ${\mu}$g/ml) two days prior to week 7, injected with P (1 mg) 42 h later, and sacrificed 7 h after P administration. In ovariectomized controls, hypothalamic GnRH mRNA levels were dropped to half by treatment of E and restored by subsequent treatment of P. The negative feedback action of E on GnRH mRNA levels observed in ovariectomized rats was completely blocked by neonatal exposure of E. The change in pituitary LH mRNA levels was similar to that in hypothalamic GnRH mRNA levels. Taken together, the results suggest that neonatal treatment of E alters the synthesis and release of GnRH in adulthood and furthermore blocks the negative feedback regulation of E which occurs normally after ovariectomy.
목 적 : 특별한 원인 없이 발생하는 특발성 중추성 요붕증 중에서 일부는 후에 뇌종양이 발견된다. 이에 저자들은 요붕증 진단 시 특발성 중추성 요붕증으로 진단 받은 20명의 환자들에서 임상적, 방사선학적, 그리고 내분비학적 경과를 추적 관찰하여 기질성 원인으로의 진행 유무를 예측할 수 있는 지표가 있는지 연구하였다. 방 법 : 1984년 1월부터 2006년 12월까지 서울대학교병원에서 중추성 요붕증으로 진단 받았던 환자들 중 뇌 자기공명영상에서 뚜렷한 기질적 원인의 증거가 없었던 20명의 환자들에서 후향적으로 임상소견과 검사소견을 검토하였다. 중추성 요붕증 진단 시 뇌종양, 선천성 중추신경계 기형이 있는 경우는 제외시켰다. 결 과 : 남아 15명, 여아 5명이었고, 진단 시 평균 연령은 $7.8{\pm}3.6$(2.1-14.7)세로, 야뇨증이 11명, 두통이 4명, 구토가 2명에서 있었다. 평균 추적관찰 기간은 $8.6{\pm}5.1$(1.5-18)년 이었다. 추적관찰 도중 뇌종양으로 진단 받은 환자는 6명(30%)이었다(생식세포종 5명, 랑게르한스 조직구증 1명; 추적기간 $2.6{\pm}2.1$년). 성장호르몬결핍증은 10명(50%)에서 관찰되었다. 복합 뇌하수체전엽호르몬결핍은 뇌종양 환자군에서 비뇌종양 환자군에서보다 더 높은 빈도로 관찰되었다(60% vs. 7%, P=0.037). 진단 시 뇌하수체 줄기 비후는 9명(47%), 뇌하수체 후엽 신호 소실은 9명(47%)에서 관찰되었으며, 추적 중 새롭게 뇌하수체 줄기 비후가 생긴 3명의 환자 모두에서 뇌종양이 발생하였다. 결 론 : 특발성 중추성 요붕증 환자의 일부는 추적관찰 중 뇌종양을 발견할 수 있는데, 특히 복합 뇌하수체전엽호르몬결핍증이 동반되거나, 뇌하수체 줄기 비후가 새롭게 발생하는 경우에는 더욱 세심한 뇌종양에 대한 추적관찰이 필요하다.
Growth hormone-releasing hormone (GHRH), a hypothalamic neuropeptide can stimulate the growth hormone secretion from the anterior pituitary. In this study, a porcine GHRH expression plasmid pHC-GHRH was used to enhance growth performance through ectopic expressions in muscle tissues of rats. Rats injected with the plasmid of pHC-GHRH and pCMV-GHRH exhibited cumulative weight gains 6.4% and 1% greater than controls. During a 5-day period, significant weight gain differences were observed as follows compared with that of control: during 5-10 days post-injection (DPI) period, the group pHC-GHRH on average 14.5% heavier than controls, $40.73{\pm}0.88$ g vs. $35.57{\pm}1.23$ g (p = 0.0023); during 10-15 DPI period, the group pHC-GHRH on average 13.6% heavier than controls, $37.49{\pm}2.85$ g vs. $33.00{\pm}1.56$ g (p = 0.0146); during 15-20 DPI period, the group pHC-GHRH on average 17.8% heavier than controls, $25.64{\pm}1.39$ g vs. $21.77{\pm}1.27$ g (p<0.05). In addition, plasmids-treated rats maintained higher serum IGF-I than controls. Significant differences of IGF-I were observed on 13 DPI and on 40 DPI in pHC-GHRH group compared with that of controls. This was accomplished through the use of an improved expression cassette that included the cytomegalovirus (CMV) immediate early enhancer/promoter in combination with a 1.5-kilobase portion of porcine ${\alpha}$-skeletal muscle actin promoter.
미꾸라지의 성장호르몬 유전자를 분리하기 위하여 미꾸라지의 cDNA library를 준비하였다. total RNA는 미꾸라지의 뇌하수체로부터 얻었으며 oligo (dT)-coupled magnetic bead를 이용하여 total RNA로부터 mRNA를 순수분리하였다. 정제된 mRNA는 cDNA를 합성하기 위한 기질로 사용하였으며, 합성된 cDNA는 EcoRV/Smal으로 절단된 pBlueKS+ plasmid vector에 삽입하였다. 모든 ligation 반응용액을 E. coli, JM109 균주에 형질전환을 유도하였으며 형질전환 효율을 최대화시키기 위하여 전기천공법을 이용하였다. 얻어진 모든 형질전환주들을 DIG로 표지된 Tilapia의 성장호르몬 유전자를 이용하여 고밀도 colony hybridization 에 의하여 검색하였다. 양성반응을 나타내는 10개의 형질전환주를 분리하여 2차 colony hybridization 및 southern hybridization에 의하여 성장호르몬 유전자가 cloning 되었음을 확인하였다. 10 개의 형질전환주 중 하나인 pCGH1을 probe로 사용한 Tilapia 성장 호르몬 유전자의 염기서열과 비교분석하였으며 53.2%의 유사성을 나타냄을 확인하였다.

Human follicle-stimulating hormone (hFSH) is a pituitary glycoprotein that regulates follicular development and ovulation. Clinically, hFSH has been used to induce follicular growth in infertile women. The hormone is composed of heterodimers, including a common ${\alpha}$ subunit among the gonadotropin family and a hormone-specific ${\beta}$ subunit. Since assembly of the heterodimer is a rate-limiting step in the production of functional hFSH, transgenic clone cows carrying a single-chain hFSH transgene may efficiently produce functional hormone. Genes encoding the ${\alpha}$ and ${\beta}$ subunits of hFSH were linked using the C-terminal peptide sequence from the ${\beta}$ subunit of human chorionic gonadotropin. Bovine fetal fibroblasts were transfected with the gene construct, including the goat ${\beta}$-casein promoter and a single-chain hFSH coding sequence. Transfected fibroblasts were transferred into enucleated oocytes, and individual nuclear transfer (NT) embryos developed to the blastocyst stage were analyzed for the transgene by polymerase chain reaction. Seventy eight blastocysts (30.8%) were developed from 259 reconstructed embryos. Among these blastocysts, the hFSH gene was detected in 70.8% (34/48) of the embryos. Subsequent transfer of hFSH-transgenic clone embryos to 31 recipients results in 11 (35.5%) early pregnancies. However, all fetuses were lost before reaching day 180 of gestation. The results from this study demonstrated that bovine NT embryos carrying single-chain hFSH could be produced, and further extensive studies in which NT embryos are transferred to more recipients may give rise to single chain hFSH-transgenic cows for biomedical applications.

In the present study, we investigated the effects of recombinant eel gonadotropins (rec-GTHs) on maturation induction in immature ovarian culture in vitro and sex steroid hormones in vivo in the Japanese eel Anguilla japonica. To study the in vitro effects of rec-GTHs on estradiol-17β (E2) production in immature ovarian tissues, ovarian tissues were incubated with different doses of rec-follicle-stimulating hormone (rec-FSH) or rec-luteinizing hormone (rec-LH). The results revealed that the E2 levels in the rec-FSH (0.1, 0.5, or 1 ㎍/mL)- and rec-LH (0.1 or 0.5 ㎍/mL)-treated groups were significantly higher than those in the female eels from the control group. Furthermore, to investigate the in vivo effects of rec-GTHs on the gonadosomatic index (GSI) and plasma sex steroid hormone levels, the eels were injected intraperitoneally with eel's ringer (control), salmon pituitary extract (SPE; for female eels), human chorionic gonadotropin (hCG; for male eels), rec-FSH, rec-LH, and rec-FSH + rec-LH once a week. The results revealed that except for the SPE and the hCG groups, none of the groups exhibited a significant difference in GSI values. However, in vivo plasma E2 levels increased at the end of 4 weeks after rec-FSH treatment in female eels. Based on these results, it is suggested that rec-GTHs may have a positive effect on sexual maturation in female eels; however, further studies on complementary rec-protein production systems and additional glycosylation of rec-hormones are needed to elucidate hormone bioactivity in vivo and in vitro.
Objectives: The objective of this review was to summarize the primary role of three representative endocrine axes in aquatic vertebrates and discuss the effects on endocrine systems and their interactions in teleost fish after exposure to environmental contaminants. Methods: We summarized individual traits and mechanisms for hormonal and transcriptional interactions between the hypothalamic-pituitary-gonad (HPG), hypothalamic-pituitary-thyroid (HPT), and hypothalamic-pituitary-adrenal (HPA) axes in fish. We also provided a brief discussion on the effects of nonylphenol-induced toxicity on endocrine systems and their interactions in fish as a demonstration of holistic explanation. Results: Currently-available data showed that thyroid dysfunction is associated with reproductive toxicity due to changes in steroidogenic gene expressions and sex hormone levels as well as gonad glands in fish. As an example, we demonstrated that exposure to nonylphenol could induce estrogenicity in male fish by decreasing thyroid hormones, which contributes to increased aromatase expression. Although the mechanisms are complicated and involved in multiple ways, a number of studies have shown that sex steroids influence the HPT axis or the HPA axis in fish, indicating bi-directional crosstalk. Critically missing is information on the primary target or toxicity mechanisms of environmental contaminants among the three endocrine axes, so further studies are needed to explore those possibilities. Conclusions: This review highlights the interactions between the HPG, HPT, and HPA axes in fish in order to better understand how these endocrine systems could interact with each other in situations of exposure to endocrine disrupting chemicals.

제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
