Monascus sp. 의 적색색소생성에 대한 용존산소량의 영향

  • 이차대사산물의 생산에 영향을 미치는 인자 중 통기성의 조절을 통한 Monascus 적색색소의 생산을 증가 시켰다. 높은 산소공급조절은 균체량을 증가 시켰으며, 낮은 산소공급조절은 고농도의 적색색소를 생산하였다. 따라서, 산소 공급량의 조절을 통하여 적색색소 생합성 과정을 활성화 시킬 수 있다.

Monascus ruber의 액체배양을 통한 색소 생산 및 고체발효를 통한 홍국쌀의 monacolin K 생산 특성 (Production of Pigment by Liquid Culture and Monacolin K in Red Mold Rice by Solid State Fermentation of Monascus ruber Strains)

  • The growth characteristics and production of color pigments by Monascus strains were investigated during liquid culture, and production of monacolin K in red mold rice was carried out by solid state fermentation. Four different Monascus ruber strains were cultured in potato dextrose yeast extract broth (PDYB) media at $25^{\circ}C$ for 15 days. The high producing strain for red pigment was not corresponded to the strain for yellow pigment. Production of red pigment was high in the strain causing the fast pH change in culture broth. Production of monacolin K in red mold rice by solid state fermentation was influenced by a combination of wet cell weight and spore density in inoculum by liquid culture. Most strains showed the high production of monacolin K in red mold rice, when submerged fermentation was carried out for 5 days as inoculum for solid state fermentation. These results suggest that submerged fermentation period of inoculum have an effect on the production of monacolin K in red mold rice by solid state fermentation, and monacolin K in red mold rice could be increased by controlling the condition of submerged fermentation for inoculum.

Production of Antibacterial Violet Pigment by Psychrotropic Bacterium RT102 Strain

  • The antibacterial action of violet pigment, a mixture of violacein and deoxyviolacein, isolated from phychrotrophic bacterium RT102 strain was examined, and the operational conditions for the effective production of violet pigment were studied. The antibacterial activity of the violet pigment was confirmed for several bacteria such as Bacillus licheniformis, Bacillus subtilis, Bacillus megaterium, Staphylococcus aureus, and Pseudomonas aeruginosa, and the high concentration of violet pigment, above about 15mg/L, caused not only growth inhibition but also death of cells. The growth properties of RT102 strain were clarified under various incubation conditions such as pH, temperature, and dissolved oxygen concentration. The maximum violet pigment concentration, i.e. 3.7 g/L, and the maximum productivity of violet pigment, i.e. 0.12 g .L$\^$-1/H$\^$-1/, were obtained in a batch culture of pH 6, 20$^{\circ}C$, and 1 mg/L of dissolved oxygen concentration.

Phellinus SP.의 액체배양에 의한 세포외 색소물질의 생산 및 특성 (Production and Characteristics of an Extracellular Pigment through the Submerged Cultivation of Phellinus Sp.)

  • An extracellular pigment production of three Phellinus sp. (Phellinus 421, P. linteus and P. hartigil) through submerged cultivation was investigated. The maximum brown pigment from culture broth was obtained from the precipitate by addition of 10% 1M HCI solution. This precipitate showed absorption characteristics with ${\lambda}_{max}$ of 360nm. The maximum production of extracellular pigment obtained at optimum medium and culture condition was 3.54 ($A_{360}$). The precipitate was fractionated by Sephadex G-75 gel chromatography, and the isolated brown pigment contained a large amount of polyphenol and the small amounts of sugar and protein. The brown pigment fraction was stable in temperature range of $30{\sim}60^{\circ}C$, pH range of $4{\sim}6$, sugar addition ranges of $1{\sim}5%$ and salt addition concentration of 3 molarity. Antioxidative activity of the brown pigment by TBA method was better than that of vitamin E (${\alpha}$-tocopherol).

Monascus purpureus 에 의한 세포외 적색색소 생산성 증가에 대한 배지내 pH 조절의 영향 (Effect of medium pH on the extracellular production of red pigments using Monascus purpureus)

  • Monascus purpureus ATCC 16365에 의한 5-liter 배양기에서 pH 조절을 통하여 세포외로 이차대사물인 수용성 적색소의 생산 증가 현상 와 황색소 보다 적색소로의 대사과정을 촉진시켜 적색소의 생산성을 향상시킬수 있는지의 가능성에 대하여 실험을 수행하였다. 황색소와 적색소의 분석은 UV-Vis spectrophotometer를 이용하여 각각 385nm 와 495nm에서 측정하였다. 실험결과 곰팡이의 최대균체량과 황${\cdot}$적색소를 포함한 총 색소 생산양은 pH 8보다 pH 4.0-5.5인 산성조건에서 2배 및 4배정도 증가하였다. 적색소 및 황색소의 전구물질인 홍색소는 pH 4.0에서 많이 생성되며 또한 홍색소에서 적색소로의 대사과정도 산성 조건 하에서 촉진된다고 보고되었다. 본 실험에서도 같은 경향을 보였으며 또한, 산성 조건 하에서 특히 수용성 적색소가 곰팡이 세포 외로 유출되는 양이 증가하는 경향을 나타내었다.

Serratia sp. KH-95가 생산하는 Prodigiosin계 적색 색소의 분리 및 특성 (Isolation and Characteristics of Prodigiosin-like Red Pigment Produced by Serratia sp. KH-95.)

  • 토양으로부터 다량의 적색 색소를 생산하는 KH-95 균주를 분리하였다. 형태학적 특성 및 생리학적 특성을 조사한 결과 KH-95는 Serratia sp.로 동정되었다. 본 균주가 생산하는 적색색소를 용매 추출 및 실리카겔 칼럼 크로마토그라피를 이용하여 분리, 정제 후 기기분석을 수행한 결과 prodigiosin의 일종으로 밝혀졌다. Serratia sp. KH-95 균주가 prodigiosin계 적색색소를 생산하기 위한 최적 조건은 배양온도 28$^{\circ}C$, 초기 pH는 7.0였으며 배지 조성을 검토 결과 카제인이 색소의 생산에 가장 적합하였다. 대두유, 미강유와 같은 oil을 제외한 대부분의 탄소원과 이용하기 쉬운 질소원인 효모즙, 육즙, 펩톤 등은 균체의 생장은 촉진 하였으나 색소 생산을 현저하게 저해하였다. 최적화된 배지 및 배양조건에서 대두유를 탄소원으로 첨가할 경우 3,380 mg/l의 많은 적색색소가 생산되었다.

  • 식품용 색소로 사용되고 있는 Monascus-pigment(홍국색소) 를 산업적으로 생산하기 위하여 공기, 토양등으로부터 균주를 순수분리하여 색소생산용 배지에서 $30^{\circ}C$로 5일 동안 액체배양한 결과 색소생산이 가장 높은 Monascus sp. KS 2 를 분리하였다. 이균주를 모균주로 하여 NTG로 돌연변이를 유발한 결과 6개의 돌연변이주를 얻었으며, 색소생산성을 비교한 결과 가장 강력한 색소생산성 돌연변이주인 Monascus sp. SUR 37 을 선별하였으며, 모균주에 비해 적색색소와 황색색소가 각각 2.4배, 1.6배 높았다. 색소생산의 최적배양 조건은 온도 $30^{\circ}C$, pH6.0, 탄소원으로는 rice powder 5%, 질소원으로는 monosodium glutamate 0.15%, 그리고 아미노산으로는 DL-threonine, 배양기간은 4일이 가장 효과적이었다.

Isolation and Characterization of Pigment-deficient Mutants from Azomonas agilis PY101

  • To investigate the mechanism of cadmium tolerance in a cadmium-resistant Azomonas agilis PY101 that produces a specific fluorescent pigment promoted by cadmium, we carried out Tn5 mutagenesis and isolated four pigment-deficient mutants. In these mutants, Ppg1, Ppg2, and Ppg3 remarkably reduced the pigment production to 15.3%, 11.2%, and 13.9%, respectively. Especially, Ppg4 mutant did not produce the pigment at all. None of the mutants grew in the presence of 1500 ppm of CdCl2 in growth medium, and they exhibited differential sensitivities to cadmium. Ppg1, Ppg2, Ppg3, and Ppg4 mutants were sensitive to 900 ppm, 1100 ppm, 1000 ppm, and 800 ppm of CdCl2, respectively. These mutants also showed noticeable increase, from 8.8-fold to 13.2-fold, in the size of growth inhibition zone compared with that of the will type after treatment with cadmium. Therefore, the pigment production of A. agilis PY101 was found to decrease the toxic effects of cadmium to the bacterium.

수용성 황색색소를 분비하는 Bacillus sp. PY123균주의 분리 및 특성

  • To develop a yellow pigment for a food-additive, a strain producing a water-soluble yellow pigment was isolated from phylloplane of tree leaf. The strain PY123 was identified a Bacillus sp. based on morphological and biochemical characterization such as a bacillus form, mortility, spore formation, Gram positive, and catalase production. The pigment production of the strain PY123 was increased about 3 times in patato broth containing 1% sucrose and 0.1 mM CoCl$_{2}$ than in only potato broth after incubation at 30$circ$C, for 2 days. When 0.1 mM CoCl$_{2}$, was added in potato broth containing 1% sucrose at late log phase during incubation of the strain PY123, cell growth was not inhibited, and the period at maximal pigment production of the strain PY123 was about 12hrs faster than in potato broth containing 1% sucrose and 0.1 mM CoCl$_{2}$, simultaniously.

  • The semicontinuous production of blue pigment from gardenia fruit by immobilized cells of Bacillus subtilis KS-380 which excreted $\beta$-glucosidase was investigated in comparison with free cells, . The blue pigment produced higher productivity under the conditions of aeration of 0.2m$\ell$/min and 2mm diameter of gel beads by using 3.5% sodium alginate. Semicontinuous production by immobilized cell showed the highest productivity with replacement of fresh production medium in every 24hr for fourth fermentation cycle following the conditions of blue pigment productivity.
