  • In vitro cultrue systems for the growth of sma-II oocytes and for meiotic maturation are expected to provide a new source of a large population of oocytes as well as assistance in basic physiological studies of oogenesis. Mouse oocytes mid-growth phase can complete grovvth and acquire full developmental capacity in vitro. On the other hand, growing pig oocytes need some other factors. FSH at a low concentration maintains the viability of both oocytes and granulosa cells, and hypoxanthine promotes the meiotic competence of the oocytes during culture period. Considerable improvement in the culture systems for growth of pig oocytes, suggested from mouse studies, and for oocyte maturation could help to develop this technology in larger species.

Impact of In-vitro Fertility and Matrix Metalloproteinases Activation of Spermatozoa by Supplement of Tea-N-Tris to Sperm Cryopreservation of Miniature Pig (미니돼지 정자 동결 보존에 Tea-N-Tris의 첨가가 체외 수정 및 MMPs 활성에 미치는 영향)

  • The main purpose of this study is to estimate the effect of adding Tea-N-Tris (TES) to the freezing buffer for miniature pig sperm. In particular, we attempted to identify the association between the MMPs expression and the fertility and viability of frozen sperm from each extender (LEY (Lactose Egg-Yolk), TLE (TES + LEY), TFGE (TES + Fructose + Glucose Egg-Yolk)). In accordance with this, Hypoosmotic Swelling Test (HOST) respond test was the lowest among sperms frozen in LEY while the highest HOST respond was observed among sperms frozen in TLE. Furthermore, we observed MMPs expression in all sperm groups, with pro-MMP showing lower expression than active MMPs. The expression of MMP-9 and MMP-2 was the highest in sperms frozen in LEY, Meanwhile, sperms from the TFGE and TLE group showed lower level of MMP-9 and MMP-2 expression in the order of TLE being the lowest. LEY group showed lower rate of blastocyst development than the TES supplement group, although the difference was not statistically significant. Meanwhile the rate of blastocyst development appeared similar when sperms from TLE and TFGE group were used for IVF. Together, these results indicate that adding Tea-N-Tris to the sperm freezing buffer only suppresses MMPs protein activation but also maximize in-vitro fertility, providing a means to improve the success rate in the in vitro manipulation of miniature pig sperm.

  • To search of a new porcine pheromonal odorant for biostimulation control system technologies to offer a potentially useful and practical way to improve reproductive efficiency in livestock species, the two dimensional quantitative structure-activity relationship (QSAR) models between physicochemical parameters as descriptors of 2-cyclohexyloxytetrahydrofurane (A), 2-phenoxytetrahydrofurane (B) analogues and binding affinity constant ($p[Od.]_{50}$) for porcine odorant-binding protein (pOBP) as receptor of pig pheromones were derived and disscused. The statistical quality of the optimized 2D-QSAR model is good ($r^{2}=0.964$) and accounts for 96.4% of the variance in the binding affinity constants. It was found that the binding affinity constants were dependent upon the optimal value, $(SL)_{opt.}=1.418$ of substituent lipole (SL) in molecules. Therefore, the SL constant was very important factor for binding affinity.

Study of the Experimental Dermatophyte Infection in Animals (실험동물에 유발시킨 피부사상균증에 대한 연구)

  • Experimental dermatophyte infections are essential for studying dermatophytosis. Induction of standard infections depends on control of three factors-spore dose, scarification, and species of the experimental animals. The authors evaluated the three factors in the experimental infection models, which were inoculated with quantitated spore solution on N. gypsea "+" and A. benhomiae "+" in rabit, guinea pig, rat, and mouse. The results were follows. 1. Infection was correlated with concentration of inoculum. 2. In traumatization method, abrasion with knife was the most effective for inoculation, followed by pricking, epilation, and shaving of hair in decreasing order. 3. Rabbit and guinea pig were more susceptable to dermatophyte infection rather than the rat and mouse. However, the mouse was not infected at all. 4. Guinea pig was the proper animal model for experimental dermatophytosis in susceptabillity, degree of clinical response, and duration of the infection. 5. A. benhamiae "+" showed more severe inflammation and shorter course than N. gypsea "+".

A Gene-Tagging System for Monitoring of Xanthomonas Species

  • A novel chromosomal gene tagging technique using a specific fragment of the fatty acid desaturase-like open reading frame (des-like ORF) from the tox-argK gene cluster of Pseudomonas syringae pv. phaseolicola was developed to identify Xanthomonas spp.released into the environment as biocontrol agents. X. campestris pv. convolvuli FB-635, a pathogen of Convolvulus arvensis L., (bindweed), was chosen as the organism in which to develop and test the system. A 0.52 kb DES fragment amplified from P. syringae pv. phaseolicola C-199 was inserted into pGX15, a cosmid clone containing a 10.3 kb Eco RI-HindIII fragment derived from the xanthomonadin biosynthetic gene cluster contained in plasmid pIG102, to create a pigG::DES insertion. The 10.8 kb EcoRI-BamHI fragment carrying the pigG:: DES insertion was cloned into pLAFR3 to generate pLXP22. pLXP22 was then conjugated into X. campestris pv. convolvuli FB-635 and the pigG::DES insertion integrated into the bacterial chromosome by marker exchange. Rifampicin resistant, tetracycline sensitive, starch hydrolyzing, white colonies were used to differentiate the marked strain from yellow pigmented wild-type ones. PCR primers specific for the unique DES fragment were used for direct detection of the marked strain. Result showed the marked strain could be detected at very low levels even in the presence of high levels of other closely related or competitive bacteria. This PCR-based DES-tagging system provides a rapid and specific tool for directly monitoring the dispersal and persistence of Xanthomonas spp.released into the environment.

A Comparison of the Intestinal Absorption of Amino Acids in Piglets When Provided in Free Form or as a Dipeptide

  • Three 28 day-old $Duroc{\times}Large$ $White{\times}Landrace$ litter mate gilts weighing an average of 6.5 kg were used to study the intestinal absorption of amino acids when provided in dipeptide form or in the form of a free amino acid mixture. The pigs were given one of three treatments. The control involved a duodenal infusion containing no amino-acids (phosphate buffer plus 5% sorbitol) while the remaining two treatments involved either a duodenal infusion containing a glycine-lysine dipeptide (1 g) or a mixture of the free amino acids glycine and lysine at the same concentration as in the dipeptide. Blood was drawn from a cannula inserted in the portal vein, at 5 to 20 minute intervals, for two hours following infusion. The concentration of intact dipeptide as well as free glycine and lysine in the portal blood was determined by high performance liquid chromatography. The intact dipeptide was never detected in the portal blood at any time after infusion. Lysine appeared in the portal blood more rapidly after infusion of dipeptide than after infusion of free lysine and the concentration of lysine in portal blood was higher in the pig infused with the dipeptide than after infusion of free lysine at almost all time points measured. The cumulative absorption of lysine and glycine from the intestine during the two hour period after infusion was greater in the pig infused with dipeptide than in the pig infused with free amino acids. The results suggest that although intact dipeptide did not reach he portal circulation, a special transport mechanism for absorption of dipeptide by intestinal cells appears to be present in pigs similar to that observed in other species.

Computational approaches for prediction of protein-protein interaction between Foot-and-mouth disease virus and Sus scrofa based on RNA-Seq

  • Foot-and-Mouth Disease (FMD) is a highly contagious trans-boundary viral disease caused by FMD virus, which causes huge economic losses. FMDV infects cloven hoofed (two-toed) mammals such as cattle, sheep, goats, pigs and various wildlife species. To control the FMDV, it is necessary to understand the life cycle and the pathogenesis of FMDV in host. Especially, the protein-protein interaction between FMDV and host will help to understand the survival cycle of viruses in host cell and establish new therapeutic strategies. However, the computational approach for protein-protein interaction between FMDV and pig hosts have not been applied to studies of the onset mechanism of FMDV. In the present work, we have performed the prediction of the pig's proteins which interact with FMDV based on RNA-Seq data, protein sequence, and structure information. After identifying the virus-host interaction, we looked for meaningful pathways and anticipated changes in the host caused by infection with FMDV. A total of 78 proteins of pig were predicted as interacting with FMDV. The 156 interactions include 94 interactions predicted by sequence-based method and the 62 interactions predicted by structure-based method using domain information. The protein interaction network contained integrin as well as STYK1, VTCN1, IDO1, CDH3, SLA-DQB1, FER, and FGFR2 which were related to the up-regulation of inflammation and the down-regulation of cell adhesion and host defense systems such as macrophage and leukocytes. These results provide clues to the knowledge and mechanism of how FMDV affects the host cell.

Effect of Cover Crop Species and Liquid Manure Application Rate on Green Manure Production, Leaf Mineral Content, Fruit Quality and Soil Chemical Properties in Pear Orchard

  • Cover cropping and liquid manure application are considered as effective ways to replace the use of chemical fertilizer in orchard. This study was conducted to investigate the effect of cover crop species and liquid manure application rate on green manure production, leaf mineral content, fruit quality and soil chemical properties in pear orchard. The treatments include rye and hairy vetch as cover crops, two liquid manure application levels based on N and $K_2O$ requirement on each cover crop species, and chemical fertilizer as control. Green manure production was higher in hairy vetch than in rye. K content of pear leaves and soil exchangeable K content increased in N based liquid manure application treatments. The yield was higher in rye + liquid manure and fertilizer treatments, and fruit quality was not different between the treatments. Taking all of these into account, rye + $K_2O$ requirement-based liquid manure application is recommended in pear orchard for not only sufficient nutrient supply but also prevention of any problem related with soil $K_2O$ accumulation in pear orchard in long-term perspective.

Toxicological Evaluation of Median Lethal Dose $(LD_{50})$ ($LD_{50}$의 독성학적 고찰)

  • This paper reviews the toxicological role of median lethal dose ($LD_50$) based on animal and human data. Animal oral $LD_50$ values of eighty seven chemicals were collected and comparatively evaluated with human minimum toxic dose ($TD_50$). In general, animal $LD_50$ values were much higher than human $TD_50$. The ratios between $LD_50$ and TDlo were ranged from 0.01 and over 1000, suggesting safety factor of up to 1000 between humans and animals in the case of acute toxicity data. However, about 40% of chemicals investigated were within the ratio of 10. Although the cases (N=20) were small, $LD_50$ values of guinea pig were closer to human TDlo than those of other animal species. In interanimal species (rat, mouse, rabbit, dog), the ratios of $LD_50$ values were between 0.1 and 5 (up to 50-fold difference). When the data are analyzed by chemical strut-ares, human $TD_50$ values were very close to rat oral $LD_50$ values. These data suggest that rat oral $LD_50$ value might be a useful parameter predicting human TDlo and one animal species could be sufficient for acute toxicity test.

  • The contents of histidine dipeptides, a metabolic products of muscle protein, were investigated to compare muscle composition among the 9 domestic animals including cattle. In major domestic animals, analyzed the effects of age, part and sex of the animals on their muscle composition. Large amounts of carnosine($68.63{\pm}17.41{\mu}mol/g$) were detected in cattle and it was higher than other animals. Whereas the content of anserine showed high level in order of turkey, chickens and duck. The ratio of carnosine and anserine(C/A ratio) was different depending on the animal species. Statistical data indicated that difference among species was significant(p < 0.05). The contents of histidine dipeptides in heated muscle by boiling for 40 minutes at $110^{\circ}C$ was similar to thoes of raw muscle. C/A ratio in heated muscle was not different from that of raw muscle, indicating that no change has been made after heating process. The contents of carnosine and anserine were different according to the parts of their muscle. Especially chuck of the mammalian showed extremely low level of histidine dipeptides compared with other parts, while C/A ratio maintained certain level regardless of age, part, sex. Therefore, based on the content of histidine dipeptides, could be found the difference of muscle composition among the species. Also C/A ratio of horse, pig, cattle, duck, sheep and turkey were characteristic respectively.
