All-optical mach-zehnder interferometric wavelength converter monolithically integrated with loss-coupled DFB probe source (Loss-Coupled DEB LD집적 Mach-Zehnder 간섭계형 파장 변환기)

  • We report the first demonstration of 10 Gb/s wavelength conversion in a Mach-Zehnder interferometric wavelength converter monolithically integrated with a loss-coupled DFB probe source. The integrated device is fabricated using a BRS (buried ridge stripe) structure with an undoped InP clad layer on the top of a passive waveguide to reduce high propagation loss. The device exhibited a static extinction ratio of 11 dB. Good performance at 10 Gb/s is obtained with an extinction ratio of 7 dB and a power penalty of 2.8 dB at a 10$^{-9}$ bit error rate.

Genetic Diversity and Relationship in Soybean MDP (Mutant Diversity Pool) Revealed by TRAP and TE-TRAP Markers

  • Mutation breeding is the useful tool to improve agronomic traits in various crop species. Soybean is most important crop and is rich in protein and oil contents. Despite of the importance as economic value and various genetic resource of soybean, there have been limited studies of genetic relationship among mutant resources through radiation breeding. In this study, the agronomical phenotype for selecting various genetic resources was evaluated in 528 soybean mutant lines. As a result, 210 soybean mutants with their original cultivars were selected with various traits. We named 210 selected lines as Mutant Diversity Pool (MDP). The genetic diversity and the relationship of the MDP were investigated using TRAP and TE-TRAP markers. In TRAP analysis, sixteen primer combination (PC)s were used and a total of 551 fragments were amplified. The highest (84.00%) and the lowest (32.35%) polymorphism levels were showed in PC MIR157B+Ga5 and B14G14B+Ga3, respectively. The mean of PIC values was 0.15 ranging from 0.07 in B14G14B+Sa12 to 0.23 in MIR157B+Sa4. Phylogenetic and population structure analysis indicated that the 210 MDP lines dispersed to four groups among the wild types and their mutants. The highest genetic diversity among populations was observed between lines Paldal and 523-7 (Fst=0.409), whereas the lowest genetic diversity was between population KAS360-22 and 94seori (Fst=0.065). AMOVA showed 11.583 (21.0%) and 43.532 (79.0%) variations in inter and intra mutant population, respectively. Overall, the genetic similarity of each intra mutant populations was closer than that of inter mutant population. A total of 408 fragments were amplified in the 210 MDP using twelve PCs of TE-TRAP markers that were obtained from a combination of three TIR sequence of transposable elements (MITE-stowaway; M-s, MITE-tourist; M-t, PONG). The highest (77.42%) and the lowest (56.00%) polymorphism levels were showed in PONG+Sa4 and PONG+Sa12, respectively. The mean of PIC values was 0.15 ranging from 0.09 in M-s+Sa4 and M-s+Ga5 to 0.21 in M-t+Ga5. AMOVA of M-s showed 2.209 (20%) and 8.957 (80%) variations in inter and intra mutant population, respectively. AMOVA of M-t showed 2.766 (18%) and 12.385 (82%) variations in inter and intra mutant population, respectively. AMOVA of PONG showed 3.151 (29%) and 7.646 (71%) variations in inter and intra mutant population, respectively. According to our study, the PONG had higher inter mutant population and lower intra mutant population. This mean was that for aspect of radiation sensitivity, M-s and M-t showed higher mobility than that of PONG. Our results suggest that the TRAP and the TE-TRAP markers may be useful for assessing the genetic diversity and relationship among soybean MDP and help to improve our knowledge of soybean mutation/radiation breeding.

Regulation of Chicken FABP4 Transcription by Toll-Like Receptor 3 Activation in DF-1 Cells

  • Long-chain fatty acids (LCFAs) are vital in cellular compartments, primarily regulating lipid metabolism. Fatty Acid-Binding Proteins (FABPs) facilitate LCFA transport, lipid synthesis, storage, and act as signaling molecules influencing various pathways, including inflammation. FABP4, in particular, is linked to vascular and cardio-related diseases, and it plays a role in macrophage-mediated inflammatory responses. Previous studies have identified FABP4 as not only a representative biomarker for lipogenesis but also as having correlations with immune responses. This study aims to investigate the regulation of the chicken FABP4 (chFABP4) gene by toll-like receptor 3 (TLR3) activation and determine the signaling pathways that are involved in chFABP4 transcriptional regulation. We analyzed the transcriptional regulation of chFABP4 in TLR3-stimulated DF-1 cells. The results showed that chFABP4 was up-regulated upon stimulation with polyinosinic-polycytidylic acid (PIC), a TLR3 ligand. Notably, chFABP4 transcription was independently regulated in the NF-κB signaling pathway. It was up-regulated in p38 inhibition, demonstrating that the p38 signaling pathway might suppress the transcription of chFABP4 within TLR3-activated DF-1 cells. In contrast, chFABP4 expression was down-regulated in JNK signaling pathway inhibition, suggesting the positive regulation of JNK signaling pathway for chFABP4 transcription in DF-1 cells in response to TLR3 activation, consistent with findings in macrophages. MEK pathway inhibition resulted in a similar regulation to NF-κB signaling. These results suggest that each MAPK contributes differentially to the transcriptional regulation of chFABP4 by in DF-1 cells in response to TLR3 activation.

Effects of Chromium Supplementation and Lipopolysaccharide Injection on the Immune Responses of Weanling Pigs

  • Sixteen specific pathogen free 4-wk-old crossbred weanling pigs were allotted into a $2{\times}2$ factorial experiment to evaluate the effects of chromium (Cr) on the immune responses after lipopolysaccharide (LPS) injection. Two factors included (1) no Cr or 400 ppb Cr supplementation from chromium picolinate (CrPic) and (2) LPS injection ($200{\mu}g/kg$ BW, intraperitoneally) on day 21 (d 21) and 35 (d 35) as compared with saline application. Plasma samples were obtained from all piglets before (0 h) and at 2 h, 4 h, 8 h, and 24 h after LPS injection. The changes in tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and leukocyte populations after LPS injection were not significant on d 21. On d 35, the plasma $TNF-{\alpha}$ level was increased at h 2 postinjection, and supplemental Cr reduced the $TNF-{\alpha}$ level. The leukocyte populations had changed profoundly and lymphocyte subsets of $CD2^+$ and $CD8^+$ were reduced at 8 h postinjection. The blood granulocytes were increased and the percentage of $CD2^+$ was reduced in the Cr-fed group on d 35. Furthermore, Cr supplementation decreased the blastogensis of concanavalin A-stimulated peripheral blood mononuclear cells (PBMC) on d 21. These results suggest that 400 ppb Cr supplementation from CrPic in diets may modulate the immune responses in weanling pigs during LPS injection.

A Basic Study of Hexapod Walking Robot (6족 보행로봇에 관한 기초연구)

  • A hexapod walking robot had been developed for gathering information in the field. The developed robot was $260{\times}260{\times}130$ ($W{\times}L{\times}H$, mm) in size and 14.7 N in weight. The legs had nineteen degrees of freedom. A leg has three rotational joints actuated by small servomotors. Two servomotors placed at ankle and knee played the roles of vertical joint for up and down motions of the leg and the other one placed at coxa played the role of horizontal joint for forward and backward motions. In addition, a servomotor placed at thorax between the front legs and the middle legs played the role of vertical joint for pumping the two front legs to climb stair or inclination. Walking motion of the robot was executed by tripod gait. The robot was controlled by manual remote-controller communicated by an infrared ray. Two controllers were equipped to control the walking of the robot. The sub-controller using PIC microcomputer (Microchips, PIC16F84A) received the 16 bit command signal from the manual remote controller, decoded it to 8bit and transmitted it to the main microcomputer (RENESAS, SH2/7045), which controlled the 19 servomotors using the PWM command signals. Walking speeds were controlled by adjusting the period of command cycle and the stride. Forward walking speed were within 100 cm/min to 300 cm/min. However, experimental walking speed had the error of 4-40 cm/min to compare with the theoretical one, because of slippage of the leg and the circular arc motion of servomotor of coxa.

Effects of Chromium Supplementation and Lipopolysaccharide Injection on Physiological Responses of Weanling Pigs

  • Sixteen specific pathogen free 4-wk-old crossbred weanling pigs were allotted into a $2{\times}2$ factorial design to evaluate chromium picolinate (CrPic) on growth and physiological responses. Two factors included (1) no Cr or 400 ppb Cr supplementation from chromium picolinate and (2) lipopolysaccharide (LPS) injection on day 21 (d 21) and 35 (d 35) compared to saline application. Plasma samples and rectal temperature were obtained from all piglets before (h 0) and at 2 h (h 2), 4 h (h 4), 8 h (h 8), and 24 h (h 24) after LPS injection ($200{\mu}g/kg$ BW, intraperitoneally). The rectal temperature on d 21 was significantly decreased (p<0.05) of about $0.36^{\circ}C$ with Cr supplementation before LPS injection. After LPS injection, the daily gain of piglets was decreased during d 35-38. Supplementation of Cr had no effect in general on growth performance particularly after LPS injection. The plasma glucose, triglycerides and urea nitrogen concentrations were changed in different ways after LPS injection. Plasma cortisol level was significantly elevated at h 2 after LPS injection on d 21 and d 35. The supplementation of Cr in the diet can delayed plasma cortisol release on d 35. The results suggest that 400 ppb Cr supplementation from CrPic may modulate the physiological response during immune stress in weanling pigs.

Genetic Diversity Measured by RAPDs in Korean Barley Germplasm Pools

  • Molecular-based genetic diversity for a set of 141 accessions of Korean barley cultivars and 24 accessions of foreign exotic cultivars were analyzed using random amplified polymorphic DNAs (RAPDs). Different level of genetic variability was observed with 30 random decamer primers in the Korean barley varieties and breeding lines which were preliminarily classified by morphological (hulled & hulless barley) and end-use (malting barley) and/or by the released periods. A total of 74 RAPD bands were scored, and the number of bands per primer varied from 1 to 7 with an average of 2.74. The hulled barley pool had one more marker genotype per primer than the hulless barley pool. The polymorphic information content (PIC) values based on the band pattern frequencies among genotypes varied depending on genetic pools where mean PICs of hulled, hulless and malting barleys were 0.62, 0.57, and 0.43, respectively. Certain genomic loci amplified by opR04, opF01, opB05, and opC13 were highly polymorphic with PIC>0.8. Patterns and temporal trends of genetic diversity assessed over the period from 1970s to 1990s had a tendency to increase, and in particular, this upward slant was quite clear and significant for the hulless barley pool. In the cluster analysis using genetic similarity matrix calculated from RAPD profiles, two major groups and several small subgroups were classified. Major grouping of materials was not affected by the presence of the husk but by their genetic background and the spike-row type. The validity of information on the genetic diversity and relationships between genotypes will have been reviewed to predict their yield potential.

  • Mutation breeding by radiation is useful for improving various crop species. Up to now, a total of 170 soybean mutant varieties have been released in the world, which is the second most registered varieties after rice. Despite the economic importance of soybean, there have been no TRAP marker system studies on genetic relationships between/among mutant lines. To develop a strategy of Mutant Diversity Pool (MDP) conservation, a study on the genetic diversity of 210 soybean mutant lines (8 cultivars and 202 mutants) was performed through a TRAP analysis. Sixteen primer combinations amplified a total of 551 fragments. The highest (84.00%) and lowest (32.35%) polymorphism levels were obtained with primers MIR157B + Ga5 and B14G14B + Ga3, respectively. The mean PIC values 0.15 varied among the primer combination ranging from 0.07 in B14G14B + Sal2 to 0.23 in MIR157B + Sa4. Phylogenetic, principal component analysis (PCA) and structure analysis indicated that the 210 lines belong to four groups based on the 16 combination TRAP markers. AMOVA showed 21.0% and 79.0% variations among and within the population, respectively. Overall, the genetic similarity of each cultivar and its mutants were higher than within other mutant populations. Our results suggest that the TRAP marker system may be useful for assessing the genetic diversity among soybean mutants and help to improve our knowledge of soybean mutation breeding.

  • 본 논문은 CDMA 시스템을 근간으로 향후 서비스 될 3세대 이동통신 시스템에 선택적으로 적용될 수 있는 다중 사용자 간섭 제거 기법을 제안한다. 제안 방식은 다단계 간섭 제거기의 각 단계에 직교 다항식의 근사화 기법을 적용하여 제거될 간섭 신호의 양을 결정하고, 이를 상호 보완적으로 제거한다. 사용자 용량의 포화도에 따른 최적해로의 수렴성을 각 방식의 스펙트럼 반경의 분석을 통해 비교하였으며, 제안한 방식이 병렬 간섭 제거 기법(Parallel Interference Cancellation, PIC)을 사용한 방식과 비교하여 2배 이상의 적은 단계를 사용했음에도 거의 같은 정도의 오차 성능을 보였으며, 실험을 통해 비트 오율이 $10^{-5}$을 기준으로 약 30% 정도의 사용자 수용 능력이 증대되었다. 또한 전력 제어의 적용 여부에 대한 시스템의 비트 오율 측면의 성능 비교, 원근 효과의 관점에서 성능을 비교하여 제안된 간섭 제거 기법이 비적응형 가중치를 적용한 병렬 간섭 제거 기법보다 우수함을 보였다.

  • 간단한 회로구조의 소형 LED 백라이트 제어기의 구현을 위하여 2.5W LED 4개를 각 섹터로 구분지어서 섹터별로 제어를 하는데 목적을 두었다. LED의 전류를 제어하기 위한 스위칭 구동회로 부분을 설계, 구현하고 스위칭을 구동하기 위한 PWM(Pulse Width Modulation)을 마이크로컨트롤러를 이용하여 제어하였다. 섹터제어 회로 부분을 구현하기 위하여 저가의 8비트 마이크로컨트롤러인 PIC16C73B를 사용하였다. 또한, RS-232 통신을 이용하여 각 섹터의 LED 제어 및 휘도를 제어하였다. 이러한 구성을 바탕으로 제어기를 제작 및 실험을 하였으며, 본 논문을 통하여 실험의 타당성을 증명하도록 한다.
