New Approach for Herbal Formula Research: Network Pharmacology (방제 연구를 위한 새로운 접근: 네트워크 약리학)

  • It is a fact that the existing pharmacological research method is difficult to explain the effect and mechanism of action of herbal formula of Korean medicine. We are now very pleased with the development of modern science and the development of a methodology for studying herbal formula characterized by network targets and multi-component therapeutics over the human body. In this review, systems pharmacology or network pharmacology is demonstrated how these are applied to explain the effectiveness of herbal medicine. The post-genomic era provides a unique opportunity for the two fields to understand and benefit from each other. In particular, recent research trends, research methodology, useful databases and results of research on herbal formula are introduced. China already has a policy of scientific development of traditional chinese medicine (TCM) and the development of Chinese medicine industry with a focus on herbal formula research at national level, and in Korea, it is urgent to support and nurture the methodology appropriate to the characteristics of the herbal formula in order to study the safety and efficacy of Korean medicine.

Effect of Quercetin on the Activity and mRNA Expression of Antioxidant Enzymes and Physiological Responses in Olive Flounder (Paralichthys olivaceus) Exposed to Cadmium

  • We investigated the antioxidant efficacy of quercetin (0% Diet 1, 0.25% Diet 2, and 0.5% Diet 3) pretreatment for 30 and 60 days in response to cadmium (Cd) toxicity in the olive flounder, and measured the plasma lysozyme activity to understand the immune effects of quercetin. The lysozyme activity with Diets 2 and 3 was higher than with Diet 1. Based on this result, to examine the immune ability and antioxidant role of quercetin, we exposed olive flounder fed quercetin to Cd and then measured the expression and activity of antioxidant enzymes (superoxide dismutase (SOD) and catalase (CAT)) and lipid peroxidation (LPO). With Diets 2 and 3, the expression and activity of antioxidant enzymes and the $H_2O_2$ concentration were lower than with Diet 1. In addition, the LPO levels were lower than with Diet 1, which protected the cell membrane. Therefore, quercetin removed the reactive oxygen species (ROS) produced by Cd, indicating that quercetin has antioxidant ability. In addition to its antioxidant ability, quercetin has immune effects.

Electrical Acupuncture Combined Bee Venom Therapy for Pain and Disability Induced Intervertebral Herniated Disc of L-spine;A Pilot Study

  • To examine the efficacy of electrical acupuncture combined bee venom in controlling low back with leg pain induced Herniated Intervertebral Disc (HIVD). 13 patients (men 8, women 5; age $41.30\;{\pm}\;17.27$) diagnosed as HIVD by CT or MRI had low back and leg pain as a chief complaint and diseases such as spinal tumor, infection and fracture were excluded. They were treated by electrical acupuncture (8 Hz) combined bee venom therapy three times a week in Wonkwang University Hospital, and pain intensity with VAS and functioning with Roland-Morris Disability Qestionnaire(RDQ) were measured at the baseline(B), and 4 weeks after treatment. The data were analyzed with Wilcoxon signed ranks test(SPSS12.0) and the level set for statistical significance was P<0.05. The pain was significantly reduced by two combined treatments (=0.002;$B7.69{\pm}2.06$,4Wks$4.70{\pm}2.72$). The function was also significantly improved by both treatments(=0.001;$B12.15{\pm}6.85$,4Wks$6.85{\pm}5.15$). The results suggest that a combination of acupuncture and bee venom therapy may be beneficial for low back pain from HIVD. In future, randomized clinical trials including follow-up are needed to examine the effects of these therapies.

Treatment of Damp-heat Type Liver Disease With Yugan-tang (임상관찰관우유간탕치료습열형간병(臨床觀察關于愈肝湯治療濕熱型肝病))

  • The aim of the present study is to determine clinical efficacy and adverse reaction of yugan-tang. We compared symptoms and liver function test results of 160 patients with damp-heat type liver disease in before and after taking yugan-tang. The patients took 200ml yugan-tang two times daily for three months. Then we analyzed these datas statistically. The overall therapeutic effect was "markedly improved" in 70 of 160 patients (43.75%) and "improved" in 75 of 160 patients (46.88%). yugan-tang markedly improved damp-heat type liver disease, especially jaundice, abdominal distention, hypochondriac pain, nause, vomiting and anorexia. Alanine aminotransferase (ALT) level and total bilirubin (TBIL) level were within normal limits at baseline. But albumin-globulin ratio (A/G) level was not changed markedly. I think that yugan-tang is worth applying widely to clinic.

Effects of Mageum-tang on the Hyperlipidemia in Rats (마금탕(麻芩湯)이 흰쥐의 고지혈증 병태에 미치는 영향)

  • This study was designed to investigate the hyperlipidemia-healing effect of liquid extract from Mageum-tang(麻芩湯), it was performed on the hyperlipidemia of rats induced by high cholesterol diet and Triton WR-1339. We prepared two types of hyperlipidemia model in rats induced by high cholesterol diet and Triton WR-1339. The amount of total cholesterol, HDL-cholesterol, LDL -cholesterol, triglyceride, phospholipid, free fatty acid, the activity of superoxide dismutase(SOD), and glutathione were measured. The liquid extract from Mageum-tang showed significant decreasing effects on total cholesterol, LDL- cholesterol, triglyceride, phospholipid, free fatty acid in the both model. And it showed significant increasing effects on HDL-cholesterol, the activity of SOD in both model and amount of glutathione in Triton WR-1339 model. These results suggest that liquid extract from Mageum-tang has healing efficacy on hyperlipidemia induced by high cholesterol diet and Triton WR-1339.

Protective Effects of Mundongcheongpye-eum on Lung Injury Induced by Elastase

  • This study aimed to evaluate the protective effects of Mundongcheongpye-eum (MCE) on elastase-induced lung injury. The extract of MCE was treated to A549 cells and elastase-induced lung injury mice model. Then, various parameters such as cell-based cyto-protective activity and histopathological finding were analyzed. MCE showed a protective effect on elastase-induced cytotoxicity in A549 cells. This effect was correlated with analysis for caspase 3 levels, collagen and elastin contents, protein level of cyclin B1, Cdc2, and Erk1/2, and gene expression of TNF-${\alpha}$ and IL-$1{\beta}$ in A549 cells. MCE treatment also revealed the protective effect on elastase-induced lung injury in mice model. This effect was evidenced via histopathological finding including immunofluence stains against elastin, collagen, caspase 3, and protein level of cyclin B1, Cdc2, and Erk1/2 in lung tissue. These data suggest that MCE has a pharmaceutical properties on lung injury. This study would provide an scientific evidence for the efficacy of MCE for clinical application to patients with chronic obstructive pulmonary disease.

  • Paeony flowers to separate substances contained in the ETOH extract the polyphenol materials sympathetic to the HPLC, anti-aging and whiting used in the cosmetics extract and comparing the results follows. Extracted by a different way to keep the flowers found in the experiment, after 1 year when stored frozen material and did not show changes in appearance. Concentrated extract by HPLC, investigated the content of the type of polyphenol naringin, sinapic acid, methyl gallate, syringic acid were detected, including 11 kinds. Learn anti-aging effect in the experiment for the EtOH 80% extract showed excellent efficacy in the remaining conditions were low. The whitening effect Peony Flower extract higher than arbutin paeoniflorin was the main ingredient.

Effects of Angelicae Gigantis Radix on Noise Stress-induced c-Fos Expression in Rats

  • Previous studies reported that exposure to noise during pregnancy adversely influenced the development of the fetus and neonate. In Oriental medicine, medications based on Angelicae gigantis radix have been known to be of efficacy in the treatment of various diseases. c-Fos, an immediate early gene whose expression is sometimes used as a marker for stimulus-induced changes in the metabolic activity of neurons. In the present study, the influence of postnatal Angelicae gigantis radix administration on c-Fos expression in the each region of hippocampus of offspring rats with prenatal noise stress during pregnancy was investigated. From the present results, exposure to the prenatal stress during pregnancy enhanced c-Fox expression, whereas exposure to postnatal Angelice gigantis radix suppressed c-Fos expression in the offsprings with prenatal noise stress during pregnancy. Based on the present study, Angelicae gigantis radix may provide new therapeutic opportunities as an agent to counteract the effects of prenatal noise stress- induced hippocampal dysfunction, and may be useful in the treatment of psychiatric problems in children of mothers who have experienced noise stress during pregnancy.

Effectiveness of a Yeonkyopaedok-san Extract in the Treatment of The Common Cold : Results of a Community-based, Double Blind, Randomized Placebo Controlled Trial (감기에 대한 연교패독산의 유효성 평가를 위한 community-based 이중맹검 위약 대조군 연구)

  • Yeonkyopaedok-san(YPS) extract is widely used to treat the common cold. The purpose of this study was to evaluate the efficacy of YPS on the common cold. Two hundred Patients with common colds of recent onset were randomized to the double blind, placebo-controlled study. They received 800 mg YPS extract or placebo in capsules, orally dissolved 3 times a day for 7 days. The total symptom score was assessed by the physician, using a 5-point scale on start and finish. Resolution of cold symptoms based on subjective daily symptoms. Total symptom score was significantly decreased in YPS groups in comparison to that in placebo group (p=0.027). Headache (p=0.012), loss of appetite (p=0.037), eyeball discomfort (p=0.002) were more affected. Time to resolution of cold symptoms did not show significant effect (p=0.592). Adverse effects were less in the YPS group than placebo group (2% vs 3%). In this community-based, randomized controlled trial, YPS were effective in treating cold symptoms in college students.

Clinical Report about the Efficacy of Herbal Medicine in Comparison with Western Medication Mixed for an Acute Liver Injured CVA Patient (급성 간손상을 동반한 CVA환자에서 한약, 양약 복합투여 시와 한약 단독 투여 시의 간기능 개선 효과에 관한 증례 보고)

  • This patient has been acute hepatic injured three times(2005.11.14-11.30, 2006.2.25-3.8, 2006.3.21-4.10) while he hadn't taken herb medicine or anything special. During these periods, his liver function tests showed a sharp elevation of AST, ALT and ALP level. The 1st period by western medication and herb medicine mixed and the 2nd, the 3rd by herb medicine only, he'd been treated. Each of it got significant results in short haul. It maybe means that herb medicine is not dangerous even for the acute hepatic injured person, moreover it can be more effective in comparison with western treatment.
