Physicochemical Properties of Sweet Potato Starch Reclaimed from Sweet Potato Processing Sludge (고구마 가공 슬러지로부터 회수된 고구마 전분의 물리화학적 특성)

  • The physicochemical properties of sweet potato (SP) starches reclaimed from an SP-processing sludge without freezing (RC/NF) and with freezing (RC/FR) were investigated. Lab-isolated (LI) SP starch, as a control, were prepared from raw SP. RC/NF and RC/FR SP starches were recovered from SP-processing sludges by the repeated sieving and washing procedure. The total starch contents and amylopectin branch-chain distributions did not differ for three SP starches. Relative to LI and RC/NF SP starches (possessing similar physicochemical characteristics), the apparent amylose and phosphorus contents, swelling factor, and pasting viscosity were reduced for RC/FR SP starch. However, the freezing treatment altered X-ray diffraction pattern (at $5.5^{\circ}$, $11-12^{\circ}$, and $24^{\circ}$ $2{\theta}$) of RC/FR SP starch, which likely increased its gelatinization peak and completion temperatures. Its amorphous region in total diffractogram was reduced, resulting in the enhanced relative crystallinity. Overall results suggested SP starches recovered from an SP-processing sludge would have the potential to replace commercial SP starch products.

Reduction Factors of Pesticides with Different Physicochemical Properties under Washing and Cooking Conditions (물리화학적 특성이 다른 농약의 세척 및 조리방법에 의한 감소계수 산출)

  • The reduction factor of pesticides is getting more crucial these days. However, most studies have focused on the relationship between pesticides and commodities. This study was conducted to examine the pesticide reduction factor based on their physicochemical properties. Five pesticides were selected among 288 insecticides by considering the presence of an ionizable group, the log P, and boiling points. The correlation coefficients between log P and removal by tap water, 5% acetic acid, 20% ethanol, and 0.15% detergent were -0.835 (p<0.001), 0.336 (p=0.221), 0.659 (p<0.01), and -0.939 (p<0.001), respectively. Removal by blanching was affected by log P as it showed a positive correlation with a log P of 0.620 (p<0.05). Removal by frying showed a strong negative correlation with a log P of -0.913 (p<0.001). The results suggest that removing pesticides was affected largely by log P and by vapor pressure during cooking, whereas during washing, the matrix of the food also contributed to the reduction.

Physicochemical Characteristics of Rehmanniae Radix Preparata Powder by Milling with Pre-freezing Temperatures (예비동결 온도에 따른 숙지황 분말의 이화학적 특성)

  • Background: Rehmanniae radix preparata (RRP) has been used as a traditional medicine and is one of the most important oriental herbal medicines. However, the physical characteristics of RRP are not suitable for use in industry. The present study was under-taken to determined the preparation method of RRP powder and the physicochemical characteristics of RRP powder by milling under different pre-freezing temperatures. Methods and Results: Moisture content, powder yield, particle size, bulk density, compressive stress, extraction yield, and 5-HMF content of PRR powders by milling with pre-freezing temperatures (-20, -40, -60, and $-80^{\circ}C$) were analyzed, and correlation among these factors was determined. Powder yield increased and particle size decreased in a pre-freezing temperature-dependent manner from -20 to $-60^{\circ}C$. The Hausner ratio increased from 1.186 to 1.225 with decreasing temperature from -20 to $-80^{\circ}C$, whereas compressive stress showed the opposite trend. Extraction yield and 5-HMF content were not significantly different between RRP powder. Significant correlations were observed among pre-freezing temperature and physical characteristics (e.g., yield, particle size, Hausner ratio, and compressive stress). Conclusions: These results suggest that the pre-freezing temperature is an important factor affecting the physical characteristics of PRR powder and applicable to the industrial production of RRP powder.

Physicochemical Properties of an Anti-Yeast Substance Produced by Rahnella aquatilis Strain AY2000 (Rahnella aquatilis AY2000균주가 생산하는 항효모성 물질의 이화학적 특성)

  • Rahnella aquatilis strain AY2000 produces an anti-yeast substance (AYS), however activity of the AYS has a declining tendency during storage. To investigate what has been decreased activity of the AYS, the AYS was treated with various physicochemical agents in this paper. The activity of AYS was decreased by heat treatment. Thiol reagent such as $\beta$-mercaptoethanol or dithiothreitol was also another factor decreasing the activity of AYS. However, pH, EDTA, and NaCl were not factors decreasing the activity of AYS. Use of methanol to precipitate the AYS was also decreased the activity of AYS. The activity of AYS was not lost after Sepharose S-400 gel filtration. However, the AYS activity was completely lost, when a polysaccharide and a unknown substance (230 nm absorption) among components of the AYS was separated by DEAE-cellulose chromatography. MIC of the AYS against S. cerevisiae was usually determined at $7.8-15.6{\mu}g/ml$.

Physicochemical Properties and Anti-inflammatory Effects of Astragalus membranaceus (Fisch.) Bunge Fermented by Aspergillus awamori (Aspergillus awamori로 발효한 황기 열수 추출물의 이화학적 특성과 항염증 효과)

  • Background: Fermentation of medicinal plants increases their absorption rate and bioavailability in the body. Astragalus membranaceus has been used as a raw material, but research in its use as a food ingredient is lacking. Therefore, the purpose of this study was to identify the physicochemical characteristics and anti-inflammatory effect of fermented Astragalus membranaceus. Methods and Results: Astragalus roots were fermented using Aspergillus awamori for 4 days and their extracts prepared using hot water. The pH, total acidity (%), and reducing sugar (%) of the extracts were then investigated. The pH and total acidity decreased during fermentation. After fermentation, the pH and total acidity decreased, whereas the reducing sugar level increased. The active ingredients in fermented Astragalus were calycosin-7-O-ßd-glucoside, ononin, calycosin and formononetin. The calycosin contents was highest in the hot-water extracted samples fermented for 4 days. The other components were similar to those in control. Nitric oxide level was lower in the hot-water extracted samples fermented for 4 days than in lipopolysaccharide control group. The sample fermented for 4 days was confirmed to inhibit the production of tumor necrosis factor-α and interleukin-1β. Conclusions: Our results showed the physicochemical properties and anti-inflammatory effects of A. membranaceus after fermentation using Aspergillus awamori. These results indicated that fermented Astragalus membranaceus can be used as a functional food.

Physicochemical Properties of Red Wine using Active Dry Yeast Strains (활성건조효모를 이용한 양조주의 이화학적 특성)

  • In wine making, to perform yeast culture effectively that is the most important factor, 5 (Montrachet, Pasteur Champagne, Epernay II, prise de Mousse, Lalvin W15) representative active dry yeasts were selected. These are results about physicochemical properties. As a consequence of examining the survival rate in both 17% ethanol solution and 200ppm sulfite solution after 72 hours, Lalvin W15 strain was the highest among the 5 representative active dry yeasts. Moreover, in 1% citric acid solution, the survival rate of Pasteur Champagne after 72 hours was the highest one. As a result of experiment of red wine that is fermented by 5 different active dry yeasts, the highest one of total acidity was must of Lalvin W15. And then, the must of the Epernay II contains the highest ethanol content. Regarding of the content of organic acid, the wine that is fermented by Prise de mousse was the highest one and the order could be explained by Tartaric > Malic > Citric.

In vitro and in vivo pharmacokinetic characterization of LMT-28 as a novel small molecular interleukin-6 inhibitor

  • Objective: Interleukin-6 (IL-6) is a T cell-derived B cell stimulating factor which plays an important role in inflammatory diseases. In this study, the pharmacokinetic properties of LMT-28 including physicochemical property, in vitro liver microsomal stability and an in vivo pharmacokinetic study using BALB/c mice were characterized. Methods: LMT-28 has been synthesized and is being developed as a novel therapeutic IL-6 inhibitor. The physicochemical properties and in vitro pharmacokinetic profiles such as liver microsomal stability and Madin-Darby canine kidney (MDCK) cell permeability assay were examined. For in vivo pharmacokinetic studies, pharmacokinetic parameters using BALB/c mice were calculated. Results: The logarithm of the partition coefficient value (LogP; 3.65) and the apparent permeability coefficient values (Papp; 9.7×10-6 cm/s) showed that LMT-28 possesses a moderate-high cell permeability property across MDCK cell monolayers. The plasma protein binding rate of LMT-28 was 92.4% and mostly bound to serum albumin. The metabolic half-life (t1/2) values of LMT-28 were 15.3 min for rat and 21.9 min for human at the concentration 1 μM. The area under the plasma drug concentration-time curve and Cmax after oral administration (5 mg/kg) of LMT-28 were 302±209 h·ng/mL and 137±100 ng/mL, respectively. Conclusion: These data suggest that LMT-28 may have good physicochemical and pharmacokinetic properties and may be a novel oral drug candidate as the first synthetic IL-6 inhibitor to ameliorate mammalian inflammation.

Evaluation of Physicochemical Properties of Burley Leaf Tobacco during $2000{\sim}2004$ Crop Years at Various Growing Areas (생산 연도 및 지역별 버어리종 잎담배의 이화학성 평가)

  • To get the informations of burley leaf, the chemical constituents, quality indices, leaf color and the effects of climatic factors on the physicochemical properties of leaf produced from 2000 to 2004 crop years at 6 growing areas were analysed. The average leaf chemical contents for 5 years were as follows ; nicotine 2.87%, total nitrogen 4.55%, and chlorine 0.57%. The nicotine content was low while total nitrogen was slightly high as compared with recommended contents(nicotine ; $3.0{\sim}3.5%$, total nitrogen; $4.0{\sim}4.5%$). The variations of physicochemical properties among crop years were high while those of growing areas were relatively low. The nicotine contents of upper leaves were positively correlated to the sunshine hours in June and negatively correlated to the rainfalls in June and July according to crop years. However the total nitrogen content of upper leaves were positively correlated to the rainfalls in July according to growing areas. The tan-tended colored leaves were produced under the high air temperature, drought and long sunshine weather condition while the buff-tended colored leaves were produced under the contra교 condition according to crop years. It is considered that the increasing of nicotine content of Lugs may be available to decrease the nitrogen number(nitrogen/nicotine ratio) of leaf tobacco.

Evaluation of Physicochemical Properties of Flue-cured Leaf Tobacco during $2000{\sim}2004$ Crop Years at Various Growing Areas (생산 연도 및 지역별 황색종 잎담배의 이화학성 평가)

  • To get the information of flue-cured leaf, the chemical constituents, quality indices, leaf color and the effects of climatic factors on the physicochemical properties of leaf produced from 2000 to 2004 crop years at 6 growing areas were analysed. The average leaf chemical contents for 5 years were as follows ; nicotine 2.22%, total sugar 28.0%, total nitrogen 1.89%, ether extracts 6.37% and chlorine 0.38%. The nicotine and total nitrogen contents were low while the total sugar were high as compared with KT&G recommended contents(nicotine ; $2.5{\sim}3.0%$, total nitrogen; $2.0{\sim}2.5%$, total sugar ; below 25.0%). The variations of physicochemical properties among crop years were high while those of growing areas relatively low. The nicotine content of leaf was negatively correlated to the rainfalls in June and July sugar content was negatively correlated to the average air temperature in June and July according to crop years. The orange colored leaves were produced under the drought and long sunshine weather condition while the lemon colored leaves were produced under the contrary condition according to crop years. Blending the different crop year's leaves in the proper way may be beneficial to produce of uniform and consistent cigarettes. It is considered that the increasing of nitrogen fertilizer or improving of nitrogen uptake may be available to increase the nicotine and nitrogen and decrease the total sugar contents of flue-cured leaf tobacco.

Characterization of Mineralogical and Physicochemical Properties of Soils Contaminated with Metals at Gahak Mine (가학광산 주변 중금속 함유 토양입자의 이화학적·광물학적 특성연구)

  • Soil samples collected in an area of Gahak Mine were investigated for the characterization of mineralogical and physicochemical properties of contaminants in soils. It is found that soils in the study area are contaminated by lead (Pb), copper (Cu), zinc (Zn), cadmium (Cd), in which their concentrations are 595.3 mg/kg, 184.9 mg/kg, 712.8 mg/kg, and 10.64 mg/kg, respectively. All the concentrations exceed the concern criteria of Korean standard. Upon distribution patterns of metals identified by the sequential extraction procedure, our results show that more than 50% of metals are found as a residual type, and 30% are accounted for the association of Fe/Mn oxides. Interestingly, XRD results show that minium (Pb3O4) and cuprite (Cu2O) are identified in the soil samples, suggesting that the sources of the contaminants for Pb and Cu are these minerals. In SEM images, tens of µm of Pb oxides and Pb silicate-minerals are observed. We, therefore, note that the contamination of metals in the study area results from the direct influx and disturbance of tailings. Our findings indicate that the characterization of physicochemical and mineralogical properties of contaminated soils is a critical factor and plays an important role in optimizing recovery treatments of soils contaminated in mine development areas.
