6-Bromobenzo[4,5]imidazo[$1,2{\alpha}$pyridin-8,9-dione 유도체들의 Photosystem II 저해활성에 관한 3D-QSAR 분석 (3D-QSAR Analysis on the Photosystem II Inhibition Activity of 6-Bromobenzo[4,5]imidazo[$1,2{\alpha}$]pyridin-8,9-dione Analogues)

  • 기질 분자로서 6-bromobenzo-[4,5]imidazo[$1,2{\alpha}$]pyridin-8,9-diones 유도체의 photosystem II의 저해활성에 관한 3차원적인 정량적 구조와 활성과의 관계(3D-QSARs)들을 비교 분자장분석(CoMFA)과 비교분자 유사성 지수분석(CoMSIA) 방법으로 각각 검토하였다. CoMFA 모델은 CoMSIA 모델보다 높은 예측성과 상관성을 갖는 모델이었다. 또한 최적화된 CoMFA 2 모델의 기여도는 입체장(90.4%)에 가장 의존적이었다. CoMFA 등고도 분석으로부터, 치환기(R)로서 모체로 연결되는 탄소원자(ipso carbon)에 곁가지를 가진 치환체가 도입될수록 저해활성이 증가할 것으로 보인다. 또한, imidazol 고리와 pyridine 고리 사이의 위치에 양하전을 가질수록 활성이 높을 것으로 예상되며 친수성을 띄는 선 형태의 치환기가 도입되는 경우에 활성이 개선될 것으로 판단되었다.

시금치(Spinacia oleracea L.) 엽록체의 광합성 전자전달 활성에 미치는 iso-Butanol의 영향 (Effects of iso-Butanol on Photosynthetic Electron Transport Activity in Isolated Spinach Chloroplasts)

  • iso-Butanol이 엽록체의 광합성 전자전달 활성에 미치는 영향을 연구하기 위하여 암조건하에서 시금치의 엽록체에 iso-butanol을 처리한 후 대조구와 비교하여 광계 II 활성과 광계 I 활성을 조사하였으며, 또한 iso-butanol이 틸라코이드 막단백질에 미치는 영향을 알아보기 위해 전기영동으로 틸라코이드 막단백질의 변화를 조사하였다. 광계 I 활성은 iso-butanol이 저농도(1-4%)일 때는 증가하였으나 고농도(5-9%) 에서는 감소되었다. 광계 II 활성은 iso-butanol의 농도가 0.6, 0.8, 1%에서는 각각 75%, 55% 및 25%로 활성이 감소하였다. Butanol 중에서 탄소수는 같으나 OH 기의 위치가 다른 1-butanol, sec-butanol, tert-butanol, iso-butanol이 광계 II의 전자전달계에 미치는 영향을 비교해 본 결과 OH 기를 중심으로 탄소의 배열이 직선상 일 때 저해효과가 크다는 것을 알 수 있었다. iso-butanol이 1차적으로 광계 II에 미치는 억제 부위는 0.8% iso-butanol 처리구에 DPC 첨가했을 때 DCIP 환원율이 93%로 증가하는 것으로 보아 광계 II의 산화 부위임을 알 수 있다. 0.8% iso-butanol 처리구에 외부에서 망간과 칼슘을 첨가하였을 때 광계 II 활성이 각각 83%와 79%로 보호된 것으로 보아 광계 II 활성 억제는 망간의 소실과 관계 있는 것으로 생각된다. 또한 1% iso-butanol 처리한 엽록체를 20배 희석하였을 때 활성이 78%로 회복되었다. iso-Butanol은 1% 이하의 저농도에서 광계 II 활성을 감소시킨데 비해 1% iso-butanol 처리구의 틸라코이드 막단백질의 SDS-PAGE에 의한 band pattern은 대조구와 유사하며 2% 처리구에서는 52 Kd 부근에서 미세한 band pattern의 차이가 있으며 5%의 iso-butanol 처리구의 틸라코이드 막단백질의 band는 전체적으로 많이 소실되었다. 이것으로 보아 iso-butanol은 가역적으로 광계 II의 산화부위에 관여하는 단백질에 영향을 주어 $Ca^{2+}와\;Mn^{2+}$ 이온의 친화력을 저하시켜 광계 II 활성을 억제시키고 또한 알코올의 R-group의 틸라코이드 막에 침투하여 막구조를 변형시키므로 광계 I 활성이 증가하며 고농도(5-9%)에서는 비가역적으로 막구조를 변경하여 광계 I 활성이 감소한 것으로 사료된다.

시금치 엽록체의 광합성 전자전달 활성에 미치는 $Cd^{2+}$의 저해 효과

  • 분리한 시금치 엽록체에 $Cd^{2+}$을 처리하였을 때 광계 I에 비해서 광계 II의 활성이 낮은 농도의 $Cd^{2+}$에 의해 억제되었다. 그러므로 $Cd^{2+}$에 의해 1차적으로 광계 II 활성이 저해되었다는 것을 알 수 있다. 광계 II의 전자공여체인 diphenylcarbazide를 첨가했을 때 대조구에 비해 활성이 크게 증가하였다. $Cd^{2+}$ 처리시 $Mn^{2+}과\;Ca^{2+}을$ 외부에서 첨가했을 때 광계 II 활성은 회복되었지만 회복되는 정도가 15%로 낮았다. $50\;\mu\textrm{M}\;Cd^{2+}$ 처리구에서 광계 II 활성은 크게 감소하였으나 Fv/Fm는 미소하게 감소하였으며 $100\;\mu\textrm{M}\;Cd^{2+}$ 처리구에서 Fv/Fm는 크게 감소하였다. 광계 I활성은 $Cd^{2+}$의 처리 농도가 증가함에 따라 감소하였으나 세척하였을 경우 0.2 mM의 $Cd^{2+}$ 처리구에서 대조구보다 활성이 증가하였으며 농도가 높아짐에 따라 활성의 회복 정도가 컸다.

Structural Changes of the Spinach Photosystem II Reaction Center After Inactivation by Heat Treatment

  • The structural changes in the electron donor side of the PSII reaction center have been monitored since heat treatment ($45^{\circ}C$ for 5 min) of thylakoids is known to decrease the oxygen evolving activity. In heat-treated spinach chloroplast thylakoids, the inhibitory effect of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) on the electron transport activity of the PSII reaction center from diphenyl carbazide to dichlorophenolindophenol became reduced approximately 3.8 times and [$^{14}C$]-labeled DCMU binding on the D1 polypeptide decreased to 25~30% that of intact thylakoid membranes, implying that the conformational changes of the DCMU binding pocket, residing on the D1 polypeptide, occur by heat treatment. The accessibility of trypsin to the $NH_2$-terminus of the cytochrome b-559 ${\alpha}$-subunit, assayed with Western blot using an antibody generated against the synthetic peptide (Arg-68 to Arg-80) of the COOH-terminal domain, was also increased, indicating that heat-treatment caused changes in the structural environments near the stromal side of the cytochrome b-559 ${\alpha}$-subunit, allowing trypsin more easily to cleave the $NH_2$-terminal domain. Therefore, the structural changes in the electron donor side of the PSII reaction center complexes could be one of the reasons why the oxygen evolving activity of the heat-treated thylakoid membranes decreased.

MERCURY-INDUCED ALTERATIONS OF CHLOROPHYLL a FLUORESCENCE KINETICS IN ISOLATED BARLEY (Hordeum vulgare L. cv. ALBORI) CHLOROPLASTS

  • Effects of HgCl$_2$-treatment on electron transport, chlorophyll a fluorescence and its quenching were studied using isolated barley (Hordeum vulgare L. cv. Albori) chloroplasts. Depending on the concentration of HgCI$_2$, photosynthetic oxygen-evolving activities of photosystem II (PS II) were greatly inhibited, whereas those of photosystem I (PS I) were slightly decreased. The inhibitory effects of HgCl$_2$ on the oxygen-evolving activity was partially restored by the addition of hydroxyamine, suggesting the primary inhibition site by HgCl$_2$2-treatment is close to the oxidizing site of PS tl associated with water-splitting complex. Addition of 50 $\mu$M HgCI$_2$ decreased both photochemical and nonphotochemical quenching of chlorophyll fluorescence. Especially, energy dependent quenching (qE) was completely disappeared by HgCl$_2$-treatment as observed by NH$_4$CI treatment. In the presence of HgCI$_2$, F'o level during illumination was also increased. These results suggest that pH gradient across thylakoid membrane can not be formed in the presence of 0 $\mu$M HgCl$_2$. In addition, antenna pigment composition might be altered by HgCl$_2$-treatment.

Anabaena의 광합성 전자전달 활성에 미치는 Simazine의 억제효과 (Inhibitory Effect of Simazine on Photosynthetic Electron Transport Activity in Anabaena inequalis)

  • Effects of simazine [2-chloro-4,6-bis(methylamino)-s-triazine] on the photochemical reactions of isolaed spinach chloroplasts and crude thylakoids of Anabaena inequalis UTEX B-381 were compared. Simazine inhibited photosynthetic O2 evolution and increased the chlorophyll fluorescence in whole cells of Anabaena. The electron transfer from diphenylcarbazide to 2,6-dichlorophenolindophenol was inhibited by simazine treatment in spinach chloroplasts, but not in crude thylakoids of Anabaena. In spinach chloroplasts, the chlorophyll fluorescence was increased by simazine treatment in the presence of diphenylcarbazide and ferricyanide, but not in the presence of diphenylcarbazide and silicomolybdate. In crude thylakoids of Anabaena, simazine treatment did not increase the chlorophyll fluorescence in the presence of either diphenylcarbazide and silicomolybdate, or diphenylcarbazide and ferricyanide. There results suggest that the inhibitory site of simazine on photosynthetic electron transport chain of anabaena is different from that of spinach chloroplasts. And there may be a possiblity that the inhibition site of simazine in Anabaena lies on the donor side of photosystem II, before the site of electron donation by diphenylcarbazide.

보리 엽록체의 광계 II에서 수은, 구리 및 아연의 저해효과 비교 (Comparison of Toxic Effects of Mercury, Copper and Zinc on Photosystem II of Barley Cholroplasts)

  • The room temperature fluorescence induction of chloroplasts was utilized as a probe to locate the site of inhibition by mercury, copper and zinc on PS II by mercury. Inhibitory effect of Hg2+ on electron transport activity was notable as compared with Cu2+ and Zn2+. At concentrations of HgCl2 over 50 $\mu$M, activities of PS II and whole-chain electron transport decreased more than 70%, while that of PS I decreased about 10~30%. This suggests that PS II is more susceptible to Hg2+ than PS I is. In the presence of diphenylcarbazide (DPC), 50 $\mu$M HgCl2 inhibited the reduction of dichlorophenolindophenol (DCPIP) about 50%. Addition of heavy metals induced marked decrease in maximal variable fluorescence/initial fluorescence [(Fv)m/Fo], but no changes in Fo. With various concentrations of heavy metals, changes of chlorophyll a fluorescence emitted by PS II showed gradual decrease in photochemical quenching (qQ), which indicates an increase in reduced state of electron acceptor, QA. Especially, the addition of HgCl2 caused a notable decrease of qQ. In the presence of 50 $\mu$M CuCl2, energy-depended quenching (qE) was completely reduced, whereas in the presence of 50 $\mu$M CuCl2 and ZnCl2 it was still remained. The above results are discussed on the effects of mercury in relation to water-splitting system and plastoquinone (PQ) shuttle system.

광 및 저온처리한 벼잎 절편에서 분이한 엽록체의 전자전달 활성 (Electron Transport Activities of Chloroplasts Isolated from the Detached Rice Leaves Stored under Low Temperature with Illumination)

  • The electron transport activities of choloroplasts isolated from hte detached rise (Oryza sativa L. cv. Chucheong) leave stored under low temperature(4$^{\circ}C$) with light illumination were investigated to understand the role of light in the low temperature inhibition of photosynthesis in the chilling-sensitive plants. Chlorophyll content of the detached leaves upon incubation at 28$^{\circ}C$ and 4$^{\circ}C$ in the dark was also measured. The rice seedlings were grown with Hoagland medium in the growth chamber of 28$^{\circ}C$ temperature and 400 ft.c fluorescent light with the photoperiod of 16 h. Although chlorophyll content of the detached leaves stored in the dark declined by 61.7% after 28$^{\circ}C$ treatement, there occurred only 5.2% decrease of chlorophyll with 4$^{\circ}C$ treatment. Low temperature treatment(4$^{\circ}C$) for 6 days brought about decreases in total photosystem(PS II+PS I) activities by 35.2% and 73.6% in te presence and absence of light, respectively, while after 28$^{\circ}C$ treatment of the detached leaves for 6 days in the dark there was only 27.6% decrease in PS II+PS I activity. PS II activities were also decreased by 35.6% and 72.2% in the light and dark, respectively. PS I activities were decreased slightly, however, by 7.6% and 16.2% in the light and dark, respectively. Investigations into DPClongrightarrowDCPIP and NH2OHlongrightarrowDCPIP activities revealed that low temperature inhibition of PS II activities was not due to the inactivation of the water oxidation capacity at low temperature. It was concluded that light protects the electron transport activities of isolated rice chloroplasts from the inhibitory effect of low temperature in the detached leaves.
