Studies on the Nuclease (Part 1) Phosphodiesterase and Phosphomonoesterase Producing by Streptomyces sp. (핵산분해효소에 관한 연구 (제1보) Streptomyces속 균주가 생산하는 Phosphomono, diesterase)

  • This paper deals with investigation on the various effects of phosphodiesterase production of Streptomyces sp. and several properties of the enzyme. The results were as follows. 1) As a carbon source, sucrose was most effective PDase production when it was added. to the basal medium at 3% concentration. 2) These enzymes were remarkably activated by $Ca^{2+}$, Co$^{2+}$ and Mn$^{2+}$ but inhibited by Cu$^{2+}$. It was observed that concentration of metal ions, 0.1% of $Ca^{2+}$, 0.01% of Co$^{2+}$ and 0.04% of Mn$^{2+}$ were effective on the production of phosphodiesterase and phosphomonoesterase. 3) In case of the effect of aeration volume, 25 ml was very effective, that is, the more sufficient aeration, the better enzyme activity. Enzyme activity was to be found effective at 3% of inoculation volume, and comparatively more effective at 2%, 4% of inoculation volume. 4) Initial pH was 8. The enzyme activity reached to the maximum at 48 hours of cultivation time. 5) The optimum pH of phosphodiesterase was about 8 and that of phosphomonoesterase was about 9. The optimum temperature of phosphodiesterase and phosphomonoesterase was 6$0^{\circ}C$ and 5$0^{\circ}C$ respectively. respectively.

Effect of Oyster Shell Meal on Improving Soil Microbiological Activity (굴패화석 비료 시용이 토양의 생물학적 활성에 미치는 영향)

  • The effect of oyster shell meal, which is made of a simple crushing and alkaline calcium materials, on soil microbial properties, microbial biomass C, N and P contents, and enzyme activities were evaluated in silt loam soil. The oyster shell meal fertilizer was added at the rates of 0, 4, 8, 12 and $16Mg\;ha^{-1}$. Microbial biomass C, N and P contents were significantly increased with increasing application of oyster shell meal. Soil enzyme activities, such as urease, ${\beta}$-glucosidase and alkaline phosphomonesterase were increased significantly by shell meal application, due to increased soil pH towards neutral range and increased nutrient availability in soil. In particular, the increased microbial biomass P content and phosphomonoesterase activities were strongly correlated with available P content in soil. Conclusively, oyster shell meal fertilizer could be a good supplement to improve soil microbial activities.

Effects of Composted Pig Manure Application on Enzyme Activities and Microbial Biomass of Soil under Chinese Cabbage Cultivation (돈분퇴비의 시용이 토양의 미생물체량 및 효소활성에 미치는 영향)

  • To elucidate the effects of composted pig manure on soil biochemical properties, composted pig manure was amended in a sandy loam soil and Chinese cabbage was grown. Composted pig manure treatments included 8, 29 and $57Mg\;ha^{-1}$ for CM-08, CM-29, and CM-57 plots, respectively. Biomass contents and enzymes activities in the non-rhizophere soil were measured. Activities of protease, phosphomonoesterase and dehydrogenase in the plot CM-57 increased to 2.3, 1.6, and 2.4 fold as compared with those of the control plot. Soil microbial biomass contents increased in proportion to the application rates of compost and biomass C, N, and P in the plot CM-59 were 4.3, 3.4, 2.8-fold higher than those of control p1ot(no fertilizer), respectively. During cultivation of Chinese cabbage, biomass C and N were higher in the middle growth stage, although biomass P was the highest in the early growth stage. The average ratio of biomass C:N:P was 11:2:1, and proportion of biomass C and N in the soil organic C and N was 1.1 and 3.6%, respectively. Activities of protease and dehydrogenase had significant correlations with biomass C and P.

Evaluation of Bioavailability of Phosphorus Accumulated in Arable Soils (농경지 토양에 집적된 인산의 생물이용가능성 평가)

  • BACKGROUND: Soil utilization pattern can be the main factor affecting soil physico-chemical properties, especially in soil phosphorus (P). Understanding the distribution and bioavailability of P is important for developing management to minimize P release from arable soils to environment. This study was conducted to evaluate the potential bioavailability of soil organic P by using phosphatase hydrolysis method. METHODS AND RESULTS: Twenty-four soils from onion-rice double cropping and 30 soils from plastic film house were selected from Changyeong and Daegok in Gyeongnam province, respectively. The P accumulation pattern (total P, inorganic P, organic P, residual P) and water soluble P were characterized. Commercial phosphatase enzymes were used to classify water-extractable molybdate unreactive P from arable soils into compounds that could be hydrolysed by (i) alkaline phosphomonoesterase (comprising labile orthophosphate monoesters), (ii) a combination of alkaline phosphomonoesterase and phosphodiesterase (comprising labile orthophosphate monoesters and diesters), and (iii) phytase (including inositol hexakisphosphate). Available P was highly accumulated with 616 and 1,208 mg/kg in double cropping system and plastic film house, respectively. Dissolved reactive P (DRP) and dissolved unreactive P (DUP) had similar trends with available P, showing 24 and 109 mg/kg in double cropping and 37 and 159 mg/kg in plastic film house, respectively, indicating that important role of dissolved organic P in the environments had been underestimated. From the result of phosphatase hydrolysis, about 39% and 66% of DUP was evaluated as bioavailable in double cropping and plastic film house, respectively. CONCLUSION(S): Orthophosphate monoester and orthophosphate diester accounted for high portion of dissolved organic P in arable soils, indicating that these organic P forms give important impacts on bioavailability of P released from P accumulated soils.

Comparative Studies on Growth and Phosphatase Activity of Endolithic Cyanobacterial Isolates of Chroococcidiopsis from Hot and Cold Deserts

  • The growth and phosphatase (phosphomonoesterase) activity of Chroococcidiopsis culture isolated from the cryptoendoliths of the Antarctic were compared with a similar isolate from the Arizona hot desert. Such cyanobacteria living inside rocks share several features with the immobilized cells produced in the laboratory. This study has relevance because the availability of phosphorus is a key factor influencing the growth of these cyanobacteria in nature, in such unique ecological niches as the hot and cold deserts. Phosphatase activity therefore is of particular importance for these organisms if they are to survive without any other source of phosphorus availability. Also, there is paucity of knowledge regarding this aspect of study in cyanobacterial cultures from these extreme environments. The salient feature of this study shows the importance of specific pH and temperatures for growth and phosphatase activity of both cultures, although there were marked differences between the two isolates. The pH and temperature optima for growth and phosphatase activity (PMEase) of Chroococcidiopsis 1 and 2 were 9.5, $240^{\circ}C$ and 8.5, $40^{\circ}C$ respectively. The $K_m and V_max$ values of cultured Chroococcidiopsis 1 showed lower affinity of PMEase for the substrate compared to the enzyme affinity of the same organism when found within the rocks; Chroococcidiopsis 2 and Arizona rocks containing the same alga however showed similar affinity of PMEase for the substrate. An interesting observation was the similarity in response of immobilized Chroococcidiopsis 1 culture and the same organism in the Antarctic rocks to low light and low temperature stimulation of PMEase. This thermal response seems to be related to the ability of the immobilized Antarctic isolate and the rocks to either cryoprotect the PMEase or undergo a change to save the enzyme from becoming nonfunctional under low temperatures. The free cells of Chroococcidiopsis 1 culture however did not show such responses.

  • The chemical structure of glycolipid of Selenomonas ruminantium cell wall was to be elucidated. The bacterial cells were treated in hot TCA and the glycolipid fractions were extracted by the solvent $CHCl_3\;:\;CH_3OH$ (1 : 3). The extracted glycolipids fraction was further separated by acetone extraction. The acetone soluble fraction was named as the spot A-compound. The acetone insoluble but ether soluble fraction was named as the spot B-compound. These two compounds were examined for elucidation of their chemical structure. The results were as follows: 1. The IR spectral analysis showed that O-acyl and N-acyl fatty acids were linked to glucosamine moiety in the spot A-compound. However in the spot B-compound in addition to O and N-acyl acids phosphorus was shown to be attached to glucosamine. 2. It was recognized by gas liquid chromatography that spot A compound contained beta-OH $C_{13:0}$ fatty acid in predominance in addition to the fatty acid with beta-OH $C_{9:0}$, whereas the spot B compound was composed of the predominant fatty acid of beta-OH $C_{13:0}$ with small amount of beta-OH $C_{9:0}$. 3. According to the paper chromatographic analysis of hydrazinolysis products of the spot A compound, a compound of a similar Rf value as the chitobiose was recognized, which indicated a structure of two molecules glucosamine condensed. The low Rf value of the hydrazinolysis product of the spot B-compound confirmed the presence of phosphorus attached to glucosamine. 4. The appearance of arabinose resulting from. ninhydrin decomposition of the acid hydrolyzate of the spot A compound indicated that the amino group is attached to $C_2$ of glucosamine. 5. The amount of glucosamine in the N-acetylated spot A compound decreased in half of the original content by the treatment. with $NaBH_4$, indicating that there are two molecules of glucosamines in the spot A compound. The presence of 1, 6-linkage between two molecules of glucosamine was suggested by the Morgan-Elson reaction and confirmed by the periodate decomposition test. 6. By the action of ${\beta}-N-acetyl$ glucosaminidase the N-acetylated spot A compound was completely decomposed into N-acetyl glucosamine, whereas the spot B compound was not. This indicated the spot A compound has a beta-linkage. 7. When phosphodiesterase or phosphomonoesterase acted on $^{32}P-labeled$ spot B compound, $^{32}P$ was not released by phosphodiesterase, but completely released by phosphomonoesterase. This indicated that one phosphorus is linked to glucosamine moiety. 8. The spot A compound is assumed to have the following chemical structure: That is glucosaminyl, ${\beta}-1$, 6-glucosamine to which O-acyl and N-acyl fatty acids are linked, of which the predominant fatty acid is beta-OH $C_{13:0}$ fatty acid in addition to beta-OH $C_{9:0}$ fatty acid 9. The spot B compound is likely to have the linkage of $glucosaminyl-{\beta}-1$, 6-glucosamine to which phosphorus is linked in monoester linkage. Furthermore both O-acyl and N-acyl fatty acids contained beta-OH $C_{13:0}$ fatty acid predominantly in addition to beta-OH $C_{9:0}$ fatty acid.

Emerging Roles of Human Prostatic Acid Phosphatase

  • Prostate cancer is one of the most prevalent non-skin related cancers. It is the second leading cause of cancer deaths among males in most Western countries. If prostate cancer is diagnosed in its early stages, there is a higher probability that it will be completely cured. Prostatic acid phosphatase (PAP) is a non-specific phosphomonoesterase synthesized in prostate epithelial cells and its level proportionally increases with prostate cancer progression. PAP was the biochemical diagnostic mainstay for prostate cancer until the introduction of prostate-specific antigen (PSA) which improved the detection of early-stage prostate cancer and largely displaced PAP. Recently, however, there is a renewed interest in PAP because of its usefulness in prognosticating intermediate to high-risk prostate cancers and its success in the immunotherapy of prostate cancer. Although PAP is believed to be a key regulator of prostate cell growth, its exact role in normal prostate as well as detailed molecular mechanism of PAP regulation is still unclear. Here, many different aspects of PAP in prostate cancer are revisited and its emerging roles in other environment are discussed.

Effect of the Long-term Application of Organic Matters on Microbial Diversity in Upland Soils (유기물 장기 연용이 밭토양 미생물의 다양성에 미치는 영향)

  • To investigate the effect of long term application of organic matter in upland soils, plots for treatments of NPK, NPK+pig manure compost, rape seed cake, rice straw compost, and green manure were set up. Populations of Bacillus and Gram negative bacteria were high in the plot treated with green manure application, but microbial biomass was increased with chemical fertilizer or pig manure compost in upland soils. Activities of phosphomonoesterase and dehydrogenase were high with organic matter application comparing to control. Cluster patterns analysed using phospholipid fatty acid of plots treated with rice straw and or pig manure compost were clearly different comparing with other treatments. Dominant bacteria in upland soils were Bacillus flexus, B. subtilis and B. megaterium. And the strains isolated from upland soils had amylase, protease and lipase activities.

Effect of Soil surface Soil Management Practices on Microflora in Volcanic Ash Soils of Citrus Orchard (화산회토 감귤원의 표토관리방법이 토양 미생물상에 미치는 영향)

  • This study was conducted to investigate the effect of different surface soil management practices on soil microflora in volcanic ash soils of citrus orchard. Soil samples were collected from citrus orchards of clean cultivation, grass sod, and grass mulch system in May and September 1997. Soil chemical properties, populations of various microorganisms, enzyme activities, microbial biomass C were analyzed. Average soil pH were 4.7, and average nitrogen and organic matter contents were 6 and $140.2g\;kg^{-1}$, respectively. Aerobic bacteria were distributed at $26,2-47.3{\times}10^6cfu\;g^{-1}$ level. Among the aerobic bacteria Pseudomonas spp., Rhizobium spp., and thermophilic Bacillus spp. were dominant in most of the investigated orchard soils. Density of actinomycetes were low at $1.8-84.6{\times}10^5cfu\;g^{-1}$ level. Fungi were distributed at $26.4-182.1{\times}10^5cfu\;g^{-1}$ level and the density was higher in grass mulch and sward sites. In september, phosphomonoesterase activity was high at $239.6{\mu}g\;PNP\;g\;soil^{-1}\;h^{-1}$ in clean cultivated citrus orchards. Soil cellulase activity were higher at $602.6{\mu}g\;GE\;g\;soil^{-1}$\;24\;h^{-1}$ in grass sward cultivation than any other soil management practices. Soil microbial biomass C was higher in grass mulch cultivated orchards.

Studies on Eriophyes kuko KISHIDA and its Galls -On the Activity of Enzymes and the Chemical Constituents in Growing Galls- (구기자혹응애(Eriophyes ruro KISHIDA) 및 그 혹(Gall)에 관한 연구 II. 혹의 성장에 따르는 효소활성 및 화학성분에 대하여)

  • This work was carried out to study the chemical properties, the activity of several enzymes, chemical components and the respiratory intensity of mite galls on the leaves of Lycium chinense MILL. caused by Eriophyes kuke KISHIDA. The activities of catalase and peroxidase were higher in the gall tissue, when compared to the healthy tissue, and were increased as tile gall developed. The activity of phosphorylase of the healthy tissue seemed to be higher than that of the large gall l tissue, considering the competitive inhibition of phosphomonoeoterase and $\beta-amylase$, The activities of invertase and $\beta-amylase$ were about two times higher in the large sail tissue than those of the healthy tissue. The content of the crude protein was $20\%$ higher in the small gall tissue than that of the healthy leave tissue, and decreased as the gall matured. On the other hand, the reducing sugar level was less in the small gall tissue than that of the healthy leaves, but as the gall grew, the content of reducing sugar of the gall tissue was increased. The contents of phosphorus and tannin were increased gradually as the gall matured, and their content showed about two times higher than those of the healthy. The matured gall tissue showed higher $QO_2$ than the healthy tissue. On the other hand, the matured gall tissue presented lower $QCO_2$ than the healthy tissue, and the RQ of the healthy tissue was higher than that of the gall tissue.
