Enhancing the Moisturizing Ability of the Skin Softener using Nanoemulsion Based on Phospholipid Liposome

  • In this paper, we present the improvement in low moisturizing ability and stability that existing skin softeners have due to the low oil content, by developing skin softener using nanoemulsion of phospholipid liposome, based on the properties of nanoemulsion in cosmetic formulation. In this study, two types of oil; dimethicone (DC 200/6cs) or medium chain triglyceride (MCT), and two kinds of lecithin; unsaturated or saturated were respectively applied to produce nanoemulsion. In the particle size analysis of nanoemulsion, the droplet size of nanoemulsion containing DC200/6cs and unsaturated lecithin was the smallest, and all nanoemulsion showed high stability in the measurement of zeta potential. Therefore, with the smallest particle size and high stability, moisture contents and trans epidermal water loss(TEWL) were measured using the skin softener of DC200/6cs and unsaturated lecithin contained nanoemulsion, and the measurement was compared with the non-oil skin softener and the skin softener with only small amount of oil. The results showed that the moisture content of the skin softener using nanoemulsion increased greatly than other two skin softeners, showing high hydration ability and water retention capacity, and TEWL decreased greatly, therefore preventing the evaporation of moisture from the skin. As a result, the oil content and stability of the skin softener was improved by utilizing nanoemulson based of phospholipid liposome, and it is expected to be used in various ways in cosmetic industry.

Effects of Oral Taurine Supplementation on Plasma Total and Phospholipid Fatty Acid Patterns in Healthy Female Adults (타우린 복용이 정상 성인여성의 혈장 총지방산 및 인지질 지방산패턴에 미치는 영향)

  • Effect of oral taurine supplementation on plasma total and phospholidpid -fatty acid profiles and their metabolism were evaluated in healthy female adults. Among twenty five female volunteers(23.6$\pm$0.3 years old ) participated in the taruine supplementation program(6g taurine /day), twenty four subjects succesfully completed the 2 week program , and only nine subjects continued to take taurine for another 2 weeks. Levels of plasma fatty acids and taruine were measured by gas-liquid chromatobraphy and an automated amino acid analyzer based on ion exchange chromatography, respectively. Plasma taurine concentration s of the subjects were 108. 7$\pm$3.4 , 184.2$\pm$8.2 and 235.9$\pm$77.0$\mu$emol/L at 0 , 2 and 4 weeks of taurine supplementation. Fatty acid compositions and elongation and desaturation indices of polyunsaturated fatty acids (PUFA) in plasma total lipids were not influenced by oral taurine supplementation. However, fatty acid compositions and their metabolism in plasma phospholipids were significantly affected by taurine supplementation in female adults. Compared to the values for 0 week, the percentage of saturated fatty acids (SFA) in plasma phospholipid was significantly lowered at 2 weeks, but elevated at 4 weeks of taurine supplementation. In contrast , the percentage of phospholipid PUFA significantly increased at 2 weeks and decreased at 4 weeks of taurine supplementation from to the values for 0 weeks. Foru weeks of oral taurine supplementation signifinatly elevated the eongation index(20 : 4$\omega$6 ⇒22 : 4 $\omega$6, p<0.01), and decreased the desaturation index (20 : 3 $\omega$6 ⇒20 : 4 $\omega$6 , p<0.01) of $\omega$6 fatty acids in plasma phospholipids. Plasma taurine concentration was positively correlated with the percentage of 14 : 0 fatty acids and the enlongation index o f$\omega$3 fatty acids(20 : 5 $\omega$3 ⇒22 : 5 $\omega$3), and thenegatively correlated with the percentage of 20 : 0 in plasma phospholipids. These results indicate that oral taurine supplementation for 4 weeks signidicantly elelvated the percentage of SFA, and lowered the percentage of PUFA in plasma phospholipids with no influence on plasm total fatty aicd composition in healthy female adults.

  • Chlorella ellipsoidea cells were cultured in an iron, copper, zinc, manganese, molybdenum or boron-free medium. Biosynthetic activities of nucleic acids, protein and phospholipid in chlorella cells, which were growing in a microelement deficient medium were compared with those of the normal cells by measuring the contents of phosphate, amino acids or UV-absorbing substances in the various cell fractions. When the algae were grown in a molybdenum-free medium, the amounts of phosphate in the acid-soluble fraction of the cells increased, whereas the amounts of alkali-stable protein and RNA decreased compared with the normal cells showing that the synthesis of protein and RNA from the early products of photosynthesis was inhibited. When the algae were grown in a boron-free medium, amounts of alkali-labile protein and phospholipid of the cells decreased, while the amount of phosphate in acid-soluble fraction increased compared with the normal cells showing that the biosynthesis of protein and phospholipid from the early products of photosynthesis was retarded. In general, amounts of protein and RNA in the microelement deficient cells significantly decreased compared with those of the normal cells. Phosphate content in the acid-soluble fraction of the algal cell grown in an zinc, copper, molybdenum, or boron-free medium increased considerably, whereas that of the algal cell grown in an iron or manganese-free medium decreased remarkably compared with that of the control. It is considered, therefore, that molybdenum, zinc, copper and boron etc. play an important role in the biosyntbesis of macromolecule from acid-soluble phosphate compounds, in contrast to the principal action of iron and manganese on the photosynthetic process itself.

A Study of Serum Lipid in Korean (한국인(韓國人)의 혈청지질(血淸脂質) 연구(硏究))

  • In Med Dept. Daehan Education Insurance, Total 690 cases(male 376 cases, female 314 cases) were inspected to study the change of serum lipid value. The objects of investigation were total cholesterol, HDL cholesterol, triglyceride, phospholipid and total lipid value. The results are as follows: 1) In the normal subjects, with the relation of serum lipid value and the age. triglyceride, phospholipid and total lipid value showed the increasing tendency by 70 age, but total cholesterol value and HDL cholesterol value showed the increasing tendency by 59 age and the decreasing tendency after 59 age. 2) In the normal subject the difference of serum lipid value between sexes did not make the significant difference of level statistically in all items which can be inspected. 3) The weight change and serum lipid value are very significantly related. Total cholesterol, HDL cholesterol and total lipid value are high in female if the weight is light. But they are high in male the weight is heavy phospholipid value is opposite to the above result. Triglyceride value is high in male regardless the weight, but the significant difference happened in the high weight group. 4) As serum lipid value of hypertension patients is compared with that of the normal subjects, HDL cholesterol, triglyceride, phospholipid and total lipid value are significantly high. 5) The doubtful group of the fatty infittration of liver among the hypertension pafients have very significant difference in serum lipid value compared with not only the value of the normal subject but also that of the obesity group. 6) The doubtless group of the fatty infitration of liver among the hypertension patients have the significant difference in all serum lipid value compared with that of the normal subject, but compared with the obesity group males had the significant difference of HDL cholesterol value and both sexes had the significant difference in total lipid value, but had no significant difference in total cholesterol, triglyceride and phosprolipid value. 7) The ratio of total cholesterol per HDL cholesterol had the significant difference in the doubtful hypertension patients of the fatty infittration of liver and the relative weight, but it had no significant difference compared with the normal subject in the doubtless hypertension patients.

  • The results of previous paper(Lee et al, 1987), indicated that the role of polyunsaturated carbonyls from the polar lipid seemed more important than those of the nonpolar lipid in the browning reaction. In this study, the role of phosphoipid in lipid oxidative browning reaction was investigated. In fatty acid composition of hair tail fish, neutral lipid and glycolipid contained the higher percent of saturated and monoenic acids while phospholipid contained more of polyenoic acids. When this fish was dried and stored, polyunsaturated fatty acids of phospholipid and glycolipid more rapidly decreased than that of neutral lipid. The browning was developed more rapidly in phospholipid than in glycolipid and neutral lipid. In the test of phospholipids, the phosphatidylcholine which fractionated by $CHCl_3-MeOH(1:1)$ solution was the largest in quantity, tut the phosphatidylethanolamine which fractionated by $CHCl_3-MeOH(4:1)$ was more active in the development of browning.

Purification and Physicochemical Characterization of a Recombinant Phospholipid Hydroperoxide Glutathione Peroxidase from Oryza sativa

  • Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes. In the present work, the entire encoding region for Oryza sativa PHGPx was expressed in Escherichia coli M15, and the purified fusion protein showed a single band with 21.0 kD and pI = 8.5 on SDS- and IFE-PAGE, respectively. Judging from CD and fluorescence spectroscopy, this protein is considered to have a well-ordered structure with 12.2% $\alpha$-helix, 30.7%$\beta$-sheet, 18.5% $\delta$-turn, and 38.5% random coil. The optimum pH and temperature of the enzyme activity were pH 9.3 and 27$^{\circ}C$. The enzyme exhibited the highest affinity and catalytical efficiency to phospholipid hydroperoxide employing GSH or Trx as electron donor. Moreover, the protein displayed higher GSH-dependent activity towards t-Butyl-OOH and $H_2O_2$. These results show that OsPHGPx is an enzyme with broad specificity for hydroperoxide substrates and yielded significant insight into the physicochemical properties and the dynamics of OsPHGPx.

A Study on the Stability of Ultra-Thin Film Mixed with Stearic Acid and Phospholipid (스테아르산과 인지질 혼합물의 농도변화에 대한 유기초박막의 안정성에 관한 연구)

  • In this study, we investigated the stability with respect to the Langmuir-Blodgett(LB) monolayer films of stearic acid and phospholipid(L-${\alpha}$-dimyristoylphosphatidyl choline, DMPC) mixture. LB films of stearic acid and DMPC monolayer were deposited by the LB method on the indium tin oxide(ITO) glass. The electrochemical properties measured by cyclic voltammetry with three-electrode system in $0.05N\;NaClO_4$ solution, -0.3 V initial, 1650 mV switching potential and -1350 mV final potential. As a result, monolayer LB films of stearic acid and phospholipid mixture was appeared on irreversible process caused by the oxidation current from the cyclic voltammogram. Diffusion coefficient (D) of stearic acid and DMPC mixture(molar ratio 1:1, 1:2, 1:3) was calculated $1.4{\times}10^{-3}$, $1.7{\times}10^{-3}$ and $1.6{\times}10^{-3}(cm^2/s)$ in $0.05N\;NaClO_4$ solution, respectively.

Development of a Burnt Beef Flavor by Reaction Flavor Technology (Reaction flavor 기술을 이용한 구운 쇠고기향 개발)

  • To develop a burnt beef flavor by reaction flavor technology, hydrolyzed vegetable protein (HVP) was reacted with precursors. Ribose, cysteine, furaneol, thiamin, methionine, garlic powder, and phospholipid were selected as suitable precursors for producing a burnt beef flavor. HVP and the selected precursors were reacted in a high pressure reactor to optimize reaction parameters, such as temperature, time, and water content. Optimum reaction conditions were $130^{\circ}C$, 1 hr, and 7.5% water addition. A burnt beef flavor was generated without pH adjustment. On the basis of an omission test, cysteine, furaneol, thiamin, and garlic powder were evaluated for optimization using response surface methodology. The optimum composition of precursore was determined to be 7.7% cysteine, 7.3% furaneol, 2.1% thiamin, and 6.9% garlic powder. Based on these results, optimum reaction conditions for the production of a burnt beef flavor from HVP were 5% ribose, 5% methionine, 5% phospholipid, 7.7% cysteine, 7.3% furaneol, 2.1% thiamin, 6.9% garlic powder, 7.5% water addition, $130^{\circ}C$ reaction temperature, and 1hr reaction time.

Effect of Different Dietary Fats on Colonic Epithelial Cell Phospholipid and Phosphatidyl Inositol Composition in DMH-treated Rats (서로 다른 종류의 식이지방이 1,2-Dimethylhydrazine으로 처리한 쥐의 대장점막 인지질 및 Phosphatidyl Inolsitol의 지방산조성에 미치는 영향)

  • The study was designed to observe the effects of different dietary fat consumed in Korea with those of three other fats on colonic epithelial cell phospholipid and phosphatidyl inositol composition, which were known as biomarker for colon cancer. Male Sprague Dawley rats, at 7 weeks of age, were divided into control and 1,2-dimethylhydrazine (DMH) -treated group that was again subdivided into four groups. The experimental diets contained one of four dietary fats at 15%(w/w) level, those were, blend fat(BF), beef tallow(BT), corn oil (CO) or perilla oil (PO) At the same time, each rat was injected nth saline for control group or DMH twice a week for 6 weeks to five total dose of 180 mg/kg body weight. Dietary fatty acid composition influenced the fatty acid compositions of tissues. Proportions of C18:2 colonic mucosal phospholipid well reflected dietary level of C18:2 showing in decending CO>BF>PO> BT. The percentage of C20:4 in phospholipid was the higher in CO and BT groups and the lowest in PO groups. Incorporation of -linolenic acid in colonic mucosal lipid In perilla oil group was negatively correlated to the content of C20:4. Therefore, $\omega$3-linolenic acid rich in perilla oil could be a very important dietary source in controlling arachidonic acid level in colon epithelial cell. Therefore it could be recommend to use more perilla oil in meal preparation to reduce the risk factor against colon cancer.

  • The concentration of free fatty acids and fatty acid composition as well as cholesterol supersaturation in bile may be an important factor in the gallstone formation. Therefore, we simultaneously determinded 23 fatty acids in bile by selected ion monitoring (SIM) method of gas chromatography-mass spectrometry (GC-MS). Biliary fatty acids were extracted by aminopropyl column and the extracts with (phospholipid fraction) or without (free fatty acid fraction) alkaline hydrolysis of phospholipid were derivatized with MSTFA/TMCS (N-methyl-N-trimethylsilyl-trifluoroacetamide/trimethylsilylchloride) mixture in order to be detected on the GC-MS. The recovery range of this method was 61.1-99.0% and the RSD value of within-a-day and day-to-day test were 3.1-25.6% and 3.8-27.0%, respectively. Using this method, biliary profile was investigated in the bile of normal controls and patients with gallstones. The amounts and their distribution of free and phospholipid fatty acids showed different pattern between normal subjects and patients.
