Degradation of Phospholipids of Yeast after Freeze-Thawing

  • As an index of freeze-injury of yeast, the leakage of intracellular substances from yeast cells after freeze-thawing was investigated. It was found that much more ultraviolet-absorbing substances leaked out from non-freeze tolerant yeast (NETY) than from freeze-tolerant yeast. Furthermore, the rate of leakage of cellular substances form NFTY during incubation exceeded that of FTY, indicating that NFTY is more susceptible to freeze-injury than FTY during frozen-storage. An apparent degradation of phospholipid was observed during incubation of perfermented frozen-cells of NFTY, while little change of phospholipid occurred in FTY, These results suggested that the difference in the sensitivity of yeast might be due to the strength of cell membrane in terms of the degradation of phospholipid by enzymes, phospholipases, attached to cell membranes.

  • In order to study effects of ${\ulcorner}Persicae\;Semen{\lrcorner}$ on the hyperlipidemia rats induced by Triton WR, we have made hyperlipidemia rats by administering Triton WR-1339 for 3 days, then have administered. The results were summerized as follows: 1. The total cholesterol, triglyceride and phospholipid by hypercholesteremic rats injected by Triton WR-1339 of the sample group were decreased as compared with those of the control group, 2. The HDL-cholesterol by hypercholesteremic rats injected by Trion WR -1339 were increased as compeared those of control group. According to the above results, Persicae Semen showed inhibition on increase of serum total cholesterol, triglyceride and phosholipid contents as well as inhibition on decrease of HDL-cholesterol contents in hyperlipidemia.

Encapsulation of Bromelain in Liposome

  • Bromelain has been used as a meat-tenderizing agent in food processing. To increase the availability of bromelain, microencapsulation into liposome was carried out by the dehydration and rehydration method. Small unilamellar vesicles prepared by sonication treatment showed higher encapsulation efficiency (EE) than by the French press method. In the preparation of liposome, the effect of pH and centrifugal force on EE was also investigated and it showed a higher EE at acidic pH than at alkaline pH with centrifugation at 80, 000$\times$g. The actual EEs except unencapsulated bromelain which bound on the outside surface of liposome by electrostatic interaction also were investigated, and the optimal EE was at pH 4.6, at 0.6 of a ratio of bromelain to phosholipid(18.2%, w/w). Release of bromelain from liposomes was stimulated as the temperature increased at neutral pH.

Physical and Chemical Characteristics of Sesame Oils by Kinds of Sesame (참깨 종류에 따른 참기름의 이화학적 특성)

  • This study was investigated to viscosity, color, chemical properties, and lipid and fatty acid composition for examination of physical and chemical characteristics of sesame oils. Yield of sesame oil was higher in Ansan sesame than that of Chinese and Dambaek sesame. Viscosity of sesame oils was higher than that of shame oils from Dambaek and Chinese sesame, but turbidity and sedimentation rate were higher in Dambaek's sesame oil than those of Ansan's and Chinese sesame oils. In the acid value, saponification value and iodine value of sesame oils, and chemical characteristics of Dambaek's sesame oil were better than those of the others. The neutral, glycolipid and phosholipid contents of sesame oils were 91.1∼92.1, 2.5∼3.5 and 5.5∼6.4%, respectively. The major fatty acids of sesame oils were oleic, linoleic palmitic and stearic acid. The content of oleic acid was higher in Dambaek's and Ansan's sesame oil than that of Chinese sesame oil, and the content of linoleic acid was higher in Chinese sesame oil than the others. The fatty acid composition of neutral lipid, glycolipid and phospholipid were similar to those of total lipid. The ratio of unsaturated fatty acid and saturated fatty acid was higher in Korean sesame oils than those in Chinese sesame oil. The mineral(Mg, Cu, Fe, In, Al, Mn) content of Korean sesame oils was higher than that of Chinese sesame oil. and heavy metals(Cu, Ag, Pb, Cd, As) were not detected.

  • Objective : In order to study effects of ${\lceil}Persicae\;Semen{\rfloor}$ on the hyperlipidemia rats induced by Triton WR, we have made hyperlipidemia rats by administering Triton WR-1339 for 3 days, then have administered. The Total cholesterol, triglyceride, LDL-cholesterol, Free fatty acid, phospholipid and CRF(Cardiak Risk Factor) by hypercholesteremic rats injected by Triton WR-1339 of the herb-Acupuncture group were decreased as compared with those of the control group, especialy hypercholesteremic rats injected by Triton WR- 1339 of the herb-Acupuncture+Acupuncture group were some more decreased as compared with those of the control group. 2. The HDL-cholesterol by hypercholesteremic rats injected by Trion WR -1339 of the herb-Acupuncture group were increased as compeared those of control group, especialy hypercholesteremic rats injected by Triton WR-1339 of the herb-Acupuncture+Acupuncture group were some more increased as compared with those of the control group. According to the above results, Persicae Semen showed inhibition on decrease of serum total cholesterol, triglyceride and phosholipid contents as well as inhibition on increase of HDL-cholesterol contents in hyperlipidemia.

The Role of Membranes and Intracellular Binding Proteins in Cytoplasmic Transport of Hydrophobic Molecules : Fatty Acid Binding Proteins and Long Chain Fatty Acids (세포내 소수성 물질 이동에서 막과 세포내 결합단백질의 역살 : 지방산 결합 단밸직과 장쇄 지방산)

  • Path of a small hydrophobic molecule through the aqueous cytoplasma is not linear. Partition may favor membrane binding by several orders of magnitude : thus significant membrane association will markedly decrease the cytosolic transport rate. The presence of high concentration of soluble binding proteins for these hydrophobic molecules would compete with membrane association and thereby increase transport rate. For long chain fatty acid molecules, a family of cytosolic binding proteins collectively known as the fatty acid binding proteins(FABP), are thought to act as intracellular transport proteins. This paper examines the mechanism of transfer of fluorescent antyroyloxy-labeled fatty acids(AOFA) from purified FABPs to phosholipid membranes. With the exception of the liver FABP, AOFA is transferred from FABP by collisional interaction of the protein with a acceptor membrane. The rate of transfer increased markedly when membranes contain anionic phospholipids. This suggests that positively charged residues on the surface of the FABP may interact with the membranes. Neutralization of the surface lysine residues of adipocyte FABP decreased fatty acid transfer rate, and transfer was found to proceed via aqueous diffusion rather than collisional interaction. Site specific mutagenesis has further shown that the helix-turn-helix domain of the FABP is critical for interaction with anionic acceptor membranes. Thus cytosolic FABP may function in intracellular transport of fatty acid to decrease their membranes association as well as to target fatty acid to specific subcellular sites of utilization.

The Effect of Combined Oral Contraceptive Steroids on Serum Levels of Lipids (복합경구피임제 복용이 혈청 지질함량에 미치는 영향)

  • Increased incidence of myocardial infarction, arteriosclerosis and hypertension in oral contraceptive steroids users has been recognized as serious problem. Evidence have been accumulated that oral contraceptive steroids cause an increase in serum lipid levels, mainly triglyceride as well as an increase in serum cholesterol concentration in some women. Effect of oral contraceptives on serum lipids have been predominantly confirmed in Europian and American women. It should be emphasized that effects of contraceptives may he influenced by differences in nutrition, climate, socioeconomic status and race. The present study was, therefore, attempted to determine the effect of oral contraceptive steroids on serum lipid levels in Korean women and also to demonstrate whether the duration of pill treatment may affect serum lipid levels. Ten women, who had never taken contraceptive steroids, served as a control. One hundred women on combined oral contraceptive steroids were grouped according to the duration of pill use: duration of less than one year(14 women), 1-2 years(12 women), 2-5 years(29 women), 5-10 years(33 women) and more than 10 years(13 women). Serum levels of cholesterol, phosholipid and triglyceride were measured by enzymatic method, Fiske-SubbaRow method and Sugiura method respectively. The results obtained are as follows; 1) Oral contraceptive steroids increased serum cholesterol concentration by 4.8% as compared with cholesterol value of control group. But this increase was not statistically significant. 2) As compared with control value, concentration of serum phospholipid in pill-treated women decreased by 14.3%, which was statistically significant (p<0.05). 3) Concentration of serum triglyceride in pill-treated women increased by 10.6% over control value, but this increase was not statistically significant. 4) Duration of pill-treatment did not affect serum concentrations of cholesterol, phospholipid and triglyceride. From the results mentioned above, it may be concluded that combined oral contraceptive steroids, at least used for subjects involved in this study, decreased serum phospholipid significantly, thereby influencing serum lipid concentration.

Changes of Flavor Components and Lipid Contents in Tomato Fruits during Storage : Changes of Lipid Condents and Its Correlation with Flavor Components (감압저장중 Tomato 과실의 향기 및 지질성분의 변화 -저장중 지질성분의 변화와 향기성분과의 상관관계-)

  • Total lipid contents of tomato fruits were 97.6mg% and composed of neutral lipid(45.1mg%), phospholipid(31.2mg%) and glycolipid(12.4mg%). The contents of neutral lipid were slowly increased from the period of climacteric rise, but those of glycolipid and phospholipid were slowly decreased at the end of storage period. Major fatty acids in all lipids were identified to be palmitic, stearic, linoleic and oleic acids. The contents of linoleic acid in all lipids at $25^{\circ}C$ and those in neutrallipid at $15^{\circ}C$ were decreased, while those in phospholipid were slightly increased during storage. The contents of palmitic acid in neutral lipid were decreased, whereas those in glycolipid and phospholipid have a tendency to increase during storage. As for normal atmospheric pressure-normal temperature(NAP-N) condition, volatiles from homogenated tomato fruits were positively correlated with palmitic acid of neutral lipid, whereas negatively correlated with linoleic acid. As for subatmospheric pressure-low temperature(SAP-L) condition, the relationship between volatiles and fatty acids of neutral lipid was similar to NAP-N condition. Volatiles were positively correlated with linoleic acid of glycolipid and stearic acid of phosholipid, whereas negatively correlated with oleic acid of glycolipid and palmitic acid of phospholipid, respectively.
