Plant Leave as an Indicator for Pollution by Hydrocarbons and Heavy Metals in Al-Zubair City, Southern Iraq

  • The potential sources and spatial distribution of heavy metals and polycyclic aromatic hydrocarbons (PAHs) were investigated in the leaf plants of Al-Zubair city. A total of 14 samples of conocarpus lancifolius plant leaf were collected and analyzed for their heavy metals and PAHs content using inductively coupled plasma mass spectrometry (ICP-MS) and a 7890 Agilent capillary gas chromatograph (GC) respectively. Bioaccumulation factor calculation revealed the highest pollution of heavy metals , due to the activity of a petrochemical in the area. The diagnostic ratio of Ant/(Phe+Ant), BaA/BaA+Chr), In/(In+BghiP), Flu/Pyr, FlA/FlA+Pyr), FlA/FlA+Pyr), ∑LMW/∑HMW are commonly used for determining the origin and source of PAHs in various environmental media. The diagnostic ratio indicated the anthropogenic origin. PAHs with five-to-six membered rings were dominant in the plant leaf, which likely results from anthropogenic activities. The leaves of C. lancifolius have a preponderance of high molecular weight PAHs compared to low molecular weight PAHs, indicating a combustion origin (car exhaust, petroleum emissions, and fossil fuel). C. lancifolius leaves are a reliable indication of atmospheric PAHs absorption. The background level of heavy metals in the city (or the near environment) is in the order of Fe > Cu > Ni > Cr. On the other hand, the bioaccumulation in plant leaves showed greater tendencies as follows: Co>Cd>Zn=As>Cu>Mn>Ni>Pb>Cr>Fe. Cobalt showed high bioaccumulation, indicating strong uptake of Co by plant leaves. These findings point to human activity and car emissions as the primary sources of roadside vegetation pollution in Al-Zubair city.

Effects of Diet Complexity and Fermented Soy Protein on Growth Performance and Apparent Ileal Amino Acid Digestibility in Weanling Pigs

  • Two experiments were conducted to evaluate the effects of diet complexity and fermented soy protein on growth performance and amino acid digestibility. In Exp. 1, a total of 120 crossbred weanling pigs ($5.68{\pm}0.80\;kg$ BW) were randomly allocated into 4 treatments. Each treatment had 6 replicate pens comprising 5 pigs in each replicate. Experimental diets consisted of simple (soybean meal as protein source) and complex (soybean meal, rice protein concentrate, potato protein concentrate and fish meal as protein sources) diets; each diet contained 0 or 5% fermented soy protein (FSP), respectively. Dietary treatments included: i) simple diet; ii) simple diet with 5% FSP; iii) complex diet; iv) complex diet with 5% FSP. Pigs were provided each experimental diet for 20 d (phase 1) and then fed the same common diet for 10 d (phase 2). During days 0-10, pigs fed FSP diets had greater ADG than those fed non-FSP diets (p<0.05). G/F in FSP treatments was significantly higher than that in non-FSP treatments (p<0.05) from days 0 to 10. Throughout the overall period, G/F was greater in FSP treatments compared with non-FSP treatments (p<0.05). On d 10, N digestibility was higher in pigs fed FSP diets than in those fed non-FSP diets (p<0.05). Diet complexity did not affect growth performance and nutrient digestibility (p>0.05) in this experiment. In Exp 2, 12 ileal-cannulated, weanling barrows were housed in individual metabolism crates and randomly assigned to 1 of 4 treatments (same as Exp. 1) by using a $4{\times}4$ Latin square design. Among the essential amino acids, apparent ileal digestibility (AID) of Met and Val were increased in pigs fed FSP diets compared with those fed non-FSP diets (p<0.05). AID of Met, Phe and total essential amino acids were higher in pigs fed complex diets than in those fed simple diets (p<0.05). Among the non-essential amino acids, AID of Ala in FSP treatments was greater than that in non-FSP treatments (p<0.05). In addition, Asp, Cys, Glu, Pro, Ser and total non-essential amino acid digestibilities in pigs fed complex diets were higher compared with those fed simple diets (p<0.05). Interaction was observed in AID of Met, Asp and Pro. In conclusion, these results indicated that feeding of 5% FSP to nursery pigs improved feed efficiency and AID of amino acids, and diet complexity did not maximize the growth performance of pigs in the subsequent phase.

주류의 풍미 및 품질 향상을 위한 야생 효모의 분리 및 특성분석 (Isolation and Characterization of Wild Yeasts for Improving Liquor Flavor and Quality)

  • non-Saccharomyces 효모는 야생효모로서, 다양한 효소를 세포 밖으로 분비하여 와인의 향기 성분 증가에 중요한 영향을 준다고 알려져 있다. 본 연구에서는 한국의 발효식품에서 분리된 효모 1,007주의 세포외 효소 활성을 조사하였다. 그 결과, β-glucosidase 566, glucanase 45, protease 401로 나타났으며, AA decarboxylase 활성 균주는 각 아미노산 별로 His 69, Tyr 306, Phe 171, Trp 23, Lys 197, Leu 198 균주로 나타났다. Cerulenin 과 TFL 내성 균주는 각각 563, 610주로, 15% 에탄올 내성 균주는 307주로 나타났다. 황화수소 생성균은 500주로 조사되었다. AA decarboxylation와 황화수소 저생성 균주 중 유용 효모 12 균주를 선발하여 26S rDNA 염기서열을 분석한 결과, C. tropicalis, H. opuntiae, H. uvarum, P. kudriavzevii, S. cerevisiae, W. anomalus로 각각 동정되었다. 12주의 효모 중 당 내성 우수 균주는 9주이며, 알코올 내성 우수 균주는 5 균주로 나타났다. 알코올 발효능은 Saccharomyces 효모는 8% 이상으로 나타났으며, non-Saccharomyces 효모는 3% 내외로 나타났다.

Cordycepin 강화 한우고기 생산에 관한 연구 I. 곡립기주 동충하초 균사체의 화학적 성분 및 안전성 조사 (Development of cordycepin fortified meat production in Hanwoo steers I. Determination of the chemical composition and safety of Cordyceps militaris from grain or pupae and Paecilomyces tenuipes)

  • 본 연구는 곡립 밀리타리스 동충하초 균사체의 사료영양적 가치를 알아보기 위하여, 세 종의 동충하초 즉, 곡립기주 밀리타리스 동충하초(Cordyceps militaris) 균사체, 번데기(pupae) 기주(곤충) 밀리타리스 동충하초 균사체 및 눈꽃 동충하초(Paecilomyces tenuipes) 균사체를 선정하여 일반성분, 아미노산, 지방산, cordycepin 및 중금속함량을 분석하였다. 본 연구를 통하여 얻어진 결과를 살펴보면, 조단백질함량은 곡립 및 곤충 밀리타리스 동충하초균사체에서 각각 76.16과 75.45%로서 눈꽃동충하초 균사체의 57.21%보다 유의하게 높았다(P<0.05). 에테르추출물함량은 눈꽃동충하초, 곤충 밀리타리스 동충하초, 곡립 밀리타리스 동충하초 순으로 유의하게 낮아 졌고(P<0.05), 곡립이 0.96%로 곤충의 9.04%보다 에테르추출물함량이 유의하게 낮았다(P<0.05). 조섬유함량은 곡립 밀리타리스 및 눈꽃동충하초 균사체가 각각 12.24 및 13.93%로 곤충 밀리타리스 동충하초균사체 보다 유의하게 높았다(P<0.05). 수분, 조회분 및 가용무질소물 함량은 처리구간에 유의한 차이가 나타나지 않았다. 세 종류의 동충하초의 아미노산 조성은 모든 처리구에서 공히 글루탐산(Glu)비율이 가장 높았다. 글리신(Gly)은 곤충 밀리타리스 동충하초 균사체가 곡립 균사체보다 유의하게 높았고(P<0.05), 눈꽃동충하초 균사체는 밀리타리스 균사체와는 달리 타이로신(Tyr) 비율이 유의하게 높았다(P<0.05). 곡립 밀리타리스 동충하초균사체는 트레오닌(Thr), 세린(Ser), 글루탐산(Glu), 알라닌(Ala), 류신(Leu), 페닐알라닌(Phe) 및 프롤린(Pro)을 곤충 밀리타리스 및 눈꽃동충하초에 비하여 유의하게 높은 수준으로 함유하였다(P<0.05). 곡립 밀리타리스 동충하초균사체는 리놀레산(C18:2)의 비율이 다른 처리구에 비하여 현저하게 높았으나(P<0.01), 곤충 밀리타리스 및 눈꽃동충하초 균사체군은 리놀렌산(C18:3)이 유의하게 높았다(P<0.01). 눈꽃동충하초 균사체는 팔미트산(C16:0)과 스테아린산(18:0)의 비율이 다른 처리군에 비하여 유의하게 높았다(P<0.05). 곡립 밀리타리스 균사체내 cordycepin 함량은 건물기준 1.64%를 함유하여 곤충 밀리타리스의 0.68%보다 유의하게 높았다(P<0.01). 하지만, 눈꽃동충하초 균사체는 cordycepin이 검출되지 않았다. 세 종류의 동충하초 내 중금속(Cr, Pb, Cd, Hg)은 농림부 허용수준이하로 안전하였다.

Effects of Replacing Soy Protein Concentrate with Fermented Soy Protein in Starter Diet on Growth Performance and Ileal Amino Acid Digestibility in Weaned Pigs

  • For Exp. 1, 120 ((Yorkshire${\times}$Landrace)${\times}$Duroc) weaned pigs (7.96${\pm}$0.01 kg average initial BW, 21 days weaning) were used in a 28 d-growth assay to determine the effects of replacing soy protein concentrate (SPC) with fermented soy protein (FSP) in a starter diet (d 0 to 7) on the growth performance, apparent fecal amino acid digestibility and subsequent performance in weaned pigs. Dietary treatments included: i) FSP0 (basal diet; whey-skim milk powder-SPC based diet); ii) FSP5 (replacing SPC with 5% FSP); iii) FSP10 (replacing SPC with 10% FSP). Pigs were fed the phase I diet for 7 days, and then each group was fed a common commercial diet for 21 days to determine the effect of previous diet on subsequent performance. Average daily gain (ADG) from d 5 to 7 (linear effect, p = 0.01) and d 7 to 14 (linear effect, p<0.001) were increased as FSP level increased. The pigs fed with FSP was heavier than the pigs fed with SPC at d 5 to 7 and d 7 to 14 after weaning (p<0.05). In the entire period (d 0 to 28), there were no significant differences in weight gain and final weight between SPC and FSP diets (p>0.05). Average daily feed intake (ADFI) was higher in pigs fed with the 5% FSP diet than those fed with the other diets at d 0 to 2 post-weaning (quadratic effect, p = 0.05). Also, for the entire period of phase I (d 0 to 7), pigs consumed more 5% FSP diet compared to other treatments (quadratic effect, p = 0.03). Gain/feed (G/F) was not affected by dietary SPC or FSP in phase I and subsequent periods, but G/F from d 5 to 7 after weaning was improved linearly (p = 0.04) as dietary FSP level increased. Pigs fed with 10% FSP also improved G/F compared with those fed only SPC (p<0.05). At d 7, there were linear increments in fecal dry matter (DM) (p<0.1) and nitrogen (N) (p<0.01) digestibilities as the dietary FSP level increased. The digestibilities of fecal essential and total amino acids were increased as the FSP level increased (linear effect, p<0.1). For Exp. 2, three ((Yorkshire${\times}$Landrace)${\times}$Duroc) weaned barrows (average initial BW of 7.32 kg) were surgically fitted with a simple T-cannula approximately 15 cm prior to the ileo-cecal junction. The experimental designs were 3${\times}$3 latin squares with pigs and periods as blocking criteria. Dietary treatments and composition were the same as in Exp. 1. Apparent ileal N digestibility was increased as FSP level was increased (linear effect, p<0.05). The dietary treatments (SPC and FSP) did not affect apparent ileal DM digestibility (p>0.05). Among essential amino acids, apparent digestibility of ileal arginine (Arg), lysine (Lys), methionine (Met) and phenylalanine (Phe) were improved as the FSP level increased (linear effect, p<0.1). Also, apparent ileal total essential, non-essential and total amino acid digestibilities were increased linearly (p<0.1). In conclusion, replacing SPC with fermented soy protein appeared beneficial in growth performance, N and amino acid digestibility during the early 7 days after weaning, and an equivalent effect showed on growth performance in subsequent period of 7 to 28 days after weaning.
