Geochemical Characteristics of Deep Granitic Groundwater in Korea (국내 화강암질암내 심부지하수의 지구화학적 특성)

  • As a part of study on geological disposal of radioactive waste, hydrogeochemical characteristics of deep granitic groundwater in Korea were investigated through the construction of a large geochemical dataset of natural water, the examination on the behaviour of dissolved constituents, and the consideration of phase stability based on thermodynamic approach. In granitic region, the contents of total dissolved solids increase progressively from surface waters to deep groundwaters, which indicates the presence of more concentrated waters at depth due to water-rock interaction. The chemical composition of groundwater evolves from initial $Ca^{2+}$-(C $l^{-}$+S $O_4$$^{2-}$) or $Ca^{2+}$-HC $O_3$$^{-}$ type to final N $a^{+}$-HC $O_3$$^{-}$ or N $a^{+}$-(C $l^{-}$+S $O_4$$^{2-}$) type, via $Ca^{2+}$-HC $O_3$$^{-}$ type. Three main mechanisms seem to control the chemical composition of groundwater in the granitic region; 1) congruent dissolution of calcite at shallower depth, 2) calcite precipitation and incongruent dissolution of plagioclase at deeper depth, and 3) kaolinite-smectite or/and kaolinite-illite reaction at equilibrium at deeper depth. The behaviour of dissolved major cations (C $a^{2+}$, $K^{+}$, $Mg^{2+}$, M $a^{+}$) and silica is likely to be controlled by these reactions.

Glass Inclusions in Quartz Phenocrysts of Tuff from Sunshin Au Mining Area, Haenam, Jeonnam. (전남 해남의 순신 금광산 지역에 산출하는 응회질암에 포획된 유리포유물)

  • Clear and homogeneous glass inclusions are well preserved at the rim of the quartz phenocrysts of tuff from Sunshin epithermal Au deposit, Haenam, although the host rocks experienced extensive silicification and argillic alteration. Glass inclusion vary in size from $5\;{\mu}m$ to larger than $200\;{\mu}m$ consisting of glass(60~80 vol%) + vapor bubble(15~30 vol%) $\pm$ daughter crystals(<10 vol%). Most of glass inclusions are cubic to rectangular in shape, indicating that the host quartz grew in the stability field of $\beta$-quartz. All the glass inclusions appear to be primary. Glass inclusions are composed of highly evolved high-K calc-alkaline rhyolites, which can represent the final liquidus phase of the magma system. The $Au_2O_3$ concentration (<0.30 wt%) is trivial in the glass, indicating there was no enrichment in the final residual melt. Textural characteristics suggest that magma was water-saturated shortly before or during the eruption. $H_2O$ content of the glass (ca. 2-4 wt%) suggests a water saturation pressure($P_{H2O}$) of about 300-900 bars. This pressure implies a minimum depth of 0.8-2.5 km for the magma chamber.

Simultaneous Determination of B Group Vitamins in Supplemented Food Products by High Performance Liquid Chromatography-Diode Array Detection

  • A simple HPLC-DAD method was developed and validated to determine B group vitamin content (thiamine, riboflavin, nicotinamide, pantothenic acid, pyridoxine and folic acid) in supplemented food samples, i.e., infant formula, cereal, low-calorie food, a multi-vitamin pill and a vitamin drink. In this study, the most significant advantages were simultaneous determination of the six B group vitamins in various food matrices and a small number of sample treatment steps that required only an organic solvent, acetonitrile. Moreover, this method prevents reduction of column durability, because the mobile phase does not contain ion-pairing reagents. Analytes were separated on a Develosil RPAQUEOUS $C_{30}$ (4.6 mm ${\times}$ 250 mm, 5 ${\mu}M$ particle size) column with a gradient elution of acetonitrile and 20 mM phosphate buffer (pH 3.0) at a flow rate between 0.8 and 1.0 mL/min. Detection was performed at 275 nm, except for that of pantothenic acid (205 nm). The calibration curves for all six vitamins showed good linearity with correlation coefficients ($r^2$) higher than 0.995. The developed method was validated with respect to linearity, intra- and inter-day accuracy and precision, limit of quantification (LOQ), recovery and stability. The method showed good precision and accuracy, with intra- and inter-assay coefficients of variation less than 15% at all concentrations. The recovery was carried out according to the standard addition procedure, with yields ranging from 89.8 to 104.4%. This method was successfully applied to the determination of vitamin B groups in supplemented food products.

Thermoresistant properties of bacterioferritin comigratory protein against high temperature stress in Schizosaccharomyces pombe (Schizosaccharomyces pombe에 존재하는 bacterioferritin comigratory protein의 고온 스트레스에 대한 열저항적 성질)

  • The Schizosaccharomyces pombe structural gene encoding bacterioferritin comigratory protein (BCP) was previously cloned using the shuttle vector pRS316 to generate the BCP-overexpressing plasmid pBCP10. The present work aimed to evaluate the thermoresistant properties of BCP against high temperature stress using the plasmid pBCP10. When the S. pombe cells were grown to the early exponential phase and shifted from $30^{\circ}C$ to $37^{\circ}C$ or $42^{\circ}C$, the S. pombe cells harboring pBCP10 grew significantly more at both $37^{\circ}C$ and $42^{\circ}C$ than the vector control cells. After 6 h of the shifting to higher incubation temperatures, they contained the lower reactive oxygen species (ROS) and nitrite content, an index of nitric oxide (NO), than the vector control cells. After the temperature shifts, total glutathione (GSH) content and total superoxide dismutase (SOD) activities were much higher in the S. pombe cells harboring pBCP10 than in the corresponding vector control cells. Taken together, the S. pombe BCP plays a thermoresistant role which might be based upon its ability both to down-regulate ROS and NO levels and to up-regulate antioxidant components, such as total GSH and SOD, and subsequently to maintain thermal stability.

Numerical Study on Freezing and Thawing Process in Modular Road System (모듈러 도로시스템의 동결-융해에 대한 수치해석적 연구)

  • In order to understand response of geo-structures to the freezing-thawing process in the ground, it is necessary to consider phase change of the pore water of the ground and also to understand soil interaction with structures. In this study, numerical analysis was carried out for freezing and thawing effect on the modular road system. Neumann's theoretical equation for freezing-thawing processes in porous media can be used to estimate frozen depth and heaving from basic soil properties and ground and surface temperature, but its application is limited to the case for the sediment with fully saturated condition and zero unfrozen water content. Numerical analysis of the modular road system was performed on various soil types and different ground water table as the varying freezing index. The amount of heaving in the silty soil was much larger than those in granite weathered soil or sandy soil, and lowering groundwater level reduced ground heaving induced by freezing. Numerical analysis for temperature history of the ground surface predicted residual heaving near the surface by the freeze-thaw process in silty soil. It ought to reduce stiffness and bearing capacity of the ground so that it will impair stability and serviceability of new road system. However, the amount of residual heaving was insignificant for the road system installed in weathered soil granite and sandy soil. Since modular road system is a pavement structure mounted on the supporting substructure unlike the prevalent road pavement system, strict criteria should be applied for uniform and differential settlement of the pavement system.

Effect of Surface Treatment on Hydrogen Production of Cadmium Sulfide Particulate Film Electrodes (수소제조용 CdS 입자막 전극의 표면처리 효과)

  • To improve the photochemical energy conversion efficiency and the stability of CdS particulate film electrode which is used to produce hydrogen from the aqueous $H_2S$ solution photoelectrochemically, surface treatment of this film was carried out using $TiCl_4$ solution. CdS particles for preparation of the films were synthesized by precipitation reaction of $Cd({NO_3})_2{\cdot}9H_2O$ and $Na_2S{\cdot}4H_2O$. Then, the CdS sol was hydrothermally treated for 12hr in an autoclave with the variation of treatment temperature to control the crystalline phase of particles. CdS film electrode was thus prepared by annealing at $400^{\circ}C$ for 12hr of the wet-film cast at room temperature, and subsequently surface treated with $TiCl_4$ solution. The electrodes were characterized using XRD, SEM, and the photocurrent meter. The photocurrents of Cds film electrodes prepared with surface treatment were up to two times higher than the electrodes without surface treatment, indicating about $4.0mA/cm^2$. Hydrogen production rate in a continuous flow system using photoelectrochemical or photochemical cells prepared with surface treatment also increased in proportion to the increase of photocurrents.

Clinical presentation of a horse-derived biomaterial and its Biocompatibility: A Clinical Case Report

  • Purpose: The objective of this clinical presentation was to present a clinical case series report of socket preservation, sinus augmentation, and bone grafting using a horse-derived biomaterial. Methods: A horse-derived biomaterial was used in 8 patients for different indications including socket preservation following tooth extraction, osseous bone grafting, and sinus augementation procedures. Surgeries were performed by a well trained specialist and clinical radiographs were obtained at designated intervals. Biopsy cores of 2 ${\times}$ 8 mm prior to implant placement was obtained following a healing interval of 4 - 6 months. A clinical and histologic evaluation was performed to evaluate the clinical effectiveness and biocompatibility of the biomaterial. Results: All surgeries in 8 patients were successful with uneventful healing except for one case with membrane exposure that eventually resulted with a positive outcome. Radiographic display of the healing phase during different intervals showed increased radiopacity of granular nature as the healing time increased. No signs of adverse effect or infection was observed clinically and the tissues surrounding the biomaterial seemed well-tolerated with good intentional healing. The augmented sinuses healed uneventfully suggesting in part, good biocompatibility of the biomaterial. Dental implants placed following socket preservation were inserted with high initial torque suggesting good initial stability and bone quality. Conclusions: Our results show that at least on a tentative level, a horse-derived biomaterial may be used clinically in socket preservation, sinus augmentation, bone grafting techniques with good intentional healing and positive results.

Expression and Secretion of Recombinant Inulinase under the Control of GAL or GAP Promoter in Sacharomyces cerevisiae (Sacharomyces cerevisiae에서 GAL또는 GAP 프로모터 조절에 의한 재조합 Inulinase의 발현 및 분비)

  • To investigate the promoter effect on heterologous gene expression in S. cerevisiae, the recombinant plasmids pYI11, pYI12, pYI10-2, and pYIGP were constructed to contain the inulinase gene (INUI) as a reporter under the control of GAL10, GAL7, GAL1, and GAP promoters, respectively. When the yeasts transformants were cultivated on galactose-containing rich media, the cell growth reached to 36-39 OD600 at 72 hours of cultivation. The specific growth rates of the cells harboring the four different plasmids decreased similarly : they dropped from $0.24 h^{-1}$ during the glucose-consuming period to 0.04 -$0.10 h^{-1}$ during the galactose-consuming period (gene expression phase for GAL promoter system). After the depletion of glucose, the expression of inulinase gene was started and reached to maximal levels of 4.3(GAL1 promoter), 4.0(GAL10 promoter), 3.8(GAL7 promoter), and 1.6(GAP promoter) unit/mL at 72 hours of cultivation. Based on the maximal expression level and activity staining on the plate, the promoter strength was in the order of GAL1, GAL10, GAL7 and GAP promoter. While the GAL-promoter systems showed a high plasmid stabilities of more than 78%, the GAP-promoter plasmid revealed a lower plasmid stability of 55%. Most of inulinase activity (98%) was found in the extracellular medium, indicating that the secretion efficiency of inulinase is independent on the type of promoter.

Electrical Characteristics Measurement of Eddy Current Testing Instrument for Steam Generator in NPP (원전 증기발생기 와전류검사 장치의 전기적 특성 측정)

  • A steam generator in nuclear power plant is a heatexchager which is used to convert water into steam from heat produced in a nuclear reactor core, and the steam produced in steam generator is delivered to the turbine to generate electricity. Because of damage to steam generator tubing may impair its ability to adequately perform required safety functions in terms of both structural integrity and leakage integrity, eddy current testing is periodically performed to evaluate the integrity of tubes in steam generator. This assessment is normally performed during a reactor refueling outage. Currently, the eddy current testing for steam generator of nuclear power plant in Korea is performed in accordance with KEPIC & ASME Code requirements, the eddy current testing system is consists of remote data acquisition unit and data analysis program to evaluate the acquired data. The KEPIC & ASME Code require that the electrical properties of remote data acquisition unit, such as total harmonic distortion, input & output impedance, amplifier linearity & stability, phase linearity, bandwidth & demodulation filter response, analog-to-digital conversion, and channel crosstalk shall be measured in accordance with the KEPIC & ASME Code requirements. In this paper, the measurement requirements of electrical properties for eddy current testing instrument described in KEPIC & ASME Code are presented, and the measurement results of newly developed eddy current testing instrument by KHNP(Korea Hydro & Nuclear Power Co., LTD) are presented.

Synthesis and Properties of Di-azomesogenic Liquid Crystal Compounds with Terminal Substituents (말단에 치환기를 갖는 이-아조메소젠 액정화합물의 합성 및 성질)

  • Two series of symmetric dimesogenic compounds containing a butylene or 1-methylbutylene spacer as a flexible group were synthesized. The mesogenic groups of synthesized compounds consist of an azobenzene group with a terminal substituent. Chemical structures as well as, thermal, mesomorphic, and photochemical properties of the synthesized compounds were investigated using FT-IR, $^1H-NMR$, differential scanning calorimetry (DSC), polarizing optical microscopy (POM), and UV-visible spectrometry. P-H, P-F, and $P-OC_6H_{13}$ showed monotropic liquid crystal phases, whereas the others showed enantiotropic liquid crystal phases. Compounds with butylene group as a flexible spacer exhibited wider mesophase temperature ranges and higher thermal transition temperatures than compounds containing a 1-methylbutylene group. Compounds with a high absolute value of the Hammett substituent constant exhibited high thermal transition temperatures and improved stability in the liquid crystal phase. Furthermore, in the absence or presence of UV light illumination, terminal substituents of the azomesogenic group were important factors in deciding the maximum absorbance wavelength (${\lambda}_{max}$) and the rate of photoisomerization (K).
