Porphyromonas endodontalis에 대한 Polyphosphate의 항균기전에 관한 연구 (MECHANISM IN ANTIBACTERIAL ACTIVITY OF POLYPHOSPHATES AGAINST PORPHYROMONAS ENDODONTALIS)

  • Poly-P has been used to prevent decomposition of foods and has been shown to have inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of poly-P on the growth of Porphyromonas endodontalis, a gram negative obligate anaerobic rod, endodontopathic bacterium. P. endodontalis ATCC 35406 was in BHI broth containing hemin and vitamin K with or without poly-P. Inhibitory effect of each poly-P which was added at the beginning(lag phase) or during(exponential phase) the culture, MIC(minimum inhibitory concentration) was determined by measuring the optical density of the bacterial cell at 540nm. Viable cell counts were measured to determined whether poly-P has a bactericidal effect. Leakage of intracellular nucleotides from P. endodontalis was determined at 260nm and morphological change of P. endodontalis was observed under the TEM(transmission electron microscope). Binding of 32P-labeled poly-P to P. endodontalis was examined. SDS-polyacrylamide gel electrophoresis and zymography were performed to observe the changes in protein and enzyme profiles of P. endodontalis, respectively. The results from this study were as follows : 1. The minimal inhibitory concentration(MIC) of poly-P to P. endodontalis appeared to be 0.04~0.05%. 2. Poly-P added to the P. endodontalis culture during the exponential phase of P. endodontalis was as much effective as poly-P added at the begining of the culture, suggesting that the antibacterial effect of poly-P is not much dependent on the initial inoculum size of P. endodontalis. 3. Poly-P are bactericidal to P. endodontalis, demonstrating the decrease of the viable cell counts. 4. Intracellular nucleotide release from the P. endodontalis, was not increased in the presence of poly-P and was not reversed by the addition of divalent cations like $Ca^{2+}$ and $Mg^{2-}$. 5. Under the TEM, it was observed that fine electro-dense materials were prominent in the poly-P grown P. endodontalis, appearing locally in the cell, and the materials were more abundant and more dispersed in the cell as the incubation time with poly-P increased. In addition, highly electron dense granules accumulated in many poly-P grown cells, most of which were atypical in their shape. 6. Binding of 32P-labeled poly-P to P. endodontalis appeared to be 32.8 and 45.5 and 53.4% at 30 minutes, 1 hours and 2 hours, respectively. 7. In the presence of poly-P. the synthesis of proteins with apparent molecular masses of 25, 27, 35, 45 was lost or drastically decreased whereas expression of a protein with an apparent molecular mass of 75 was elevated. 8. Proteolytic activity of P. endodontalis was decreased by poly-P. The overall results suggest that use of poly-P may affect the growth of P. endodontalis, and the anti-bacterial activity of poly-P seems largely bactericidal. Changes in shape, protein expression, and proteolytic activity of P. endodontalis by poly-P may be directly and indirectly attributed to the antibacterial effect of poly-P. Further studies will be needed to confirm the effect of poly-P.

알코올-물 혼합용액을 이용하는 Solvothermal 법에 의한 나노크기의 TiO2 제조 (Solvothermal Preparation of Nanocrystalline TiO2 Using Alcohol-water Mixed Solvent)

  • 본 연구에서는 $TiCl_4$ 출발물질 및 알코올과 물의 혼합용액을 이용한 solvothermal 법을 통하여 나노 크기의 $TiO_2$를 제조하였다. 이 때 혼합용액 중의 알코올의 종류 및 그 조성이 생성되는 $TiO_2$ 입자들의 결정상 혹은 응집상태에 미치는 영향에 대해 조사하였다. 생성물들의 물성은 X-선 회절법과 투과 및 주사전자 현미경을 이용하여 분석하였다. 1-부탄올/물의 부피 비가 각각 다른 혼합용매(1-부탄올/물; 100/0, 75/25, 50/50, 25/75, 0/100)를 이용한 solvothermal 법에 있어서 얻어진 루틸상의 $TiO_2$ 입자들의 응집상태는 1-부탄올/물의 비에 따라 변하였으며, 1-부탄올/물의 75/25 부피 비에서 잘 분산된 $TiO_2$ 나노 입자가 얻어졌다. 알코올과 물의 비를 75/25로 고정시킨 혼합 용매를 이용한 solvothermal 법에 있어서 알코올의 종류에 따라 생성되는 $TiO_2$ 입자들의 결정상이 변함을 확인하였다. 즉 메탄올, 에탄올 및 이소프로필 알코올의 혼합용액을 사용한 경우에는 아나타제상의 입자가 얻어지며 1-부탄올 혼합용액을 사용하면 루틸상의 $TiO_2$ 입자가 생성되었다. 이상의 결과들로부터 출발물질로는 $TiCl_4$를 그리고 반응용매로 알코올과 물의 혼합용액을 이용하며 더 이상의 첨가제를 사용하지 않는 solvothermal 법에 있어서는, 단순히 혼합용액의 조성 또는 알코올의 종류를 변화시킴으로써 생성되는 $TiO_2$ 입자들의 분산성 향상 및 결정상 조절이 가능한 것을 확인할 수 있었다.

창작 뮤지컬 '혜초'의 무대의상 연구 -코러스(Chorus)의상을 중심으로- (A Study on Stage Costumes of Creative Musical 'Hyecho' - Focus on the Costumes of the Chorus -)

  • This study examines the stage costumes of the creative musical 'Hyecho', which was performed six times in the Chung Ang Art Center hosted by Chung-Ang University's performance & media agency from December 20 to 22 in 2006. The stage costumes were made based on these basic design elements of stage costumes that were dyed : line, silhouette, material, and color. First, the creative musical 'Hyecho' presents a new form of fusion theatre, which blends Korean traditional music and dance with videos. Thereby, the musical has opened a new phase in Korea's creative performing arts. Second, in musical performances that combines dance and songs, stage costumes should be made not only to attract the audience's eyes, but also not to interrupt actors' vocalizations and movements by taking their physical features into account. Also, costumes should be made in consideration of their relationships with visual elements, including stage settings and lighting. Third, the musical features fusion-style stage costumes, which combines Hanbok, the Korean traditional costume, and Indian traditional costumes with modern costumes. For the line and silhouette, costumes show the beauty of curves through the curves of Hanbok and India's traditional costumes and also through irregular pleats of pants. Also, by using cotton materials, which is easy to dye and not readily deformed, costumes feature colors that are found in nature through a gradation dyeing technique. In doing so, it offers visual amusement to the audience by making stage costumes look like a beautiful painting. Fourth, the stage costumes of the chorus feature costumes that use lining and pleated skirts using belts, and various accessories, including necklaces that use strings in order to express evil spirits. Since there is not much time to change costumes during a performance, using such items are helpful to show the unique characteristics of actors effectively during the limited time. Also, coordinating with the lighting director allows the costume designer to make better costumes for the chorus and make the performance more dramatic. Finally, it was not necessary to wash the costumes of the chorus of the fusion musical Hyecho 2006 since it was performed only six times. However, when using dyed costumes for the long-term performance, it might cause problems such as bleaching that result from the washing of costumes and low durability that can result in the deformation of costumes. As performing arts are made in various forms and are diversified, it is needed for stage costumes to change accordingly through new attempts and various ways of expression.

Ni-Al-$ZrH_2$ 연료극을 사용한 용융탄산염 연료전지의 온도의 영향 (Effect of operating temperature using Ni-Al-$ZrH_2$ anode in molten carbonate fuel cell)

  • Fuel cell is a device that directly converts chemical energy in the form of a fuel into electrical energy by way of an electrochemical reaction. In the anode for a high temperature fuel cell, nickel or nickel alloy has been used in consideration of the cost, oxidation catalystic ability of hydrogen which is used as fuel, electron conductivity, and high temperature stability in reducing atmosphere. Most MCFC stacks currently operate at an average temperature of $650^{\circ}C$. There is some gains with decreased temperature in MCFC to diminish the electrolyte loss from evaporation and the material corrosion, which could improve the MCFC life. However, operating temperature has a strong related on a number of electrode reaction rates and ohmic losses. Baker et al. reported the effect of temperature (575 to $650^{\circ}C$). The rates of cell voltage loss were 1.4mV/$^{\circ}C$ for a reduction in temperature from 650 to $600^{\circ}C$, and 2.16mV/$^{\circ}C$ for a decrease from 600 to $575^{\circ}C$. The two major contributors responsible for the change in cell voltage with reducing operation temperature are the ohmic polarization and electrode polarization. It appears that in the temperature range of 550 to $650^{\circ}C$, about 1/3 of the total change in cell voltage with decreasing temperature is due to an increase in ohmic polarization, and the electrode polarization at the anode and cathode. In addition, the oxidation reaction of hydrogen on an ordinary nickel alloy anode in MCFC is generally considered to take place in the three phase zone, but anyway the area contributing to this reaction is limited. Therefore, in order to maintain a high performance of the fuel cell, it is necessary to keep this reaction responsible area as wide as possible, that is, it is needed to keep the porosity and specific surface area of the anode at a high level. In this study effective anodes are prepared for low temperature MCFC capable of enhancing the cell performance by using zirconium hydride at least in part of anode material.

우수 골퍼의 아이언 클럽 스윙동작에 대한 운동학적 분석 (Kinematical Analysis of Swing Motion with Golf Iron Clubs Used by Elite Golfers)

  • 본 연구의 목적은 3명의 우수 골프선수를 대상으로 아이언 클럽(3, 6, 9번)의 스윙 동작을 3차원적으로 비교 분석하고, 이의 분석에 따른 요인들을 정량적으로 제시하여 선수 및 일반 골퍼들이 바람직한 아이언 스윙 동작을 배우는데 기초자료를 제공하는데 있다. 3차원 영상분석을 실시하여 다음과 같은 결론을 도출하였다. 1. 각 클럽별로 전체 스윙 시간에는 큰 차이를 보이지 않았으며, 각 국면별 시간도 거의 유사하게 나타났다. 2. 각 클럽별로 어드레스와 임팩트시 머리위치 변화는 차이가 없었다. 3. 클럽의 길이에 따른 차이를 제외하면 각 클럽별로 오른 무릎관절 각도변화는 유사하게 나타났다. 4. 각 클럽별로 고관절의 회전각도 변화는 약간의 차이가 나타났다. 5. 각 클럽별로 어드레스와 임팩트 동작시 어깨관절의 회전각도와 동일하게 만들어 주어 스윙의 정확도를 높이고, 탑스윙시 어깨회전 각도를 $90^{\circ}$이상 유지하여 스윙의 회전력을 높이는 것으로 나타났다. 6. 클럽이 짧을수록 상체전경 각도가 약간의 차이로 커지는 것으로 나타났으며 어드레스와 탑스윙, 임팩트 동작시상체전경 각도가 $30-36^{\circ}$의 형태를 유지하면서 스윙을 하였다.

선박용 디젤엔진 SCR 시스템에 NOx 환원제로 사용되는 암모늄 카바메이트의 물질 성분 분석 연구 (Analysis of Ammonium Carbamate Used as a NOx Reducing Agent for the SCR System of Marine Diesel Engines)

  • 선박용 디젤엔진의 NOx 환원제로 액체 우레아를 사용하는 SCR 기술이 널리 사용되고 있다. 하지만 액체 우레아 대신에 고체상의 암모늄 카바메이트를 NOx 환원제로 사용하면 저온 NOx 저감율 및 암모니아 저장용량 측면 등에서 다양한 장점이 있다. 이에 따라 본 연구에서는 암모늄 카바메이트를 EA, FTIR, XRD 방법으로 분석하여 순도를 관리하는 방법을 제시하고자 하였으며, 다양한 온도와 압력 조건에 암모늄 카바메이트가 노출되었을 때의 물질 변화 특성을 고찰하고자 하였다. 본 연구를 통하여 암모늄 카바메이트의 순도를 EA 분석을 통해 효과적으로 관리 할 수 있음을 알 수 있었으며, 선박용 디젤엔진의 SCR 시스템에 적용될 것으로 예상되는 열분해 온도 조건에서 가열과 냉각을 반복한 암모늄 카바메이트에 대한 FTIR 분석결과, 물질 특성은 변화하지 않는 것을 확인하였다. 또한, 대기 중에 장기간 노출된 암모늄 카바메이트는 암모늄 카보네이트로 물질 변화함을 알 수 있었다.

IRIS형 방사선검출기 콜리메이터 제작 및 MCNP 코드를 이용한 성능평가 (Development of the IRIS Collimator for the Portable Radiation Detector and Its Performance Evaluation Using the MCNP Code)

  • 방사선검출기를 이용한 고방사성물질의 측정이나 방사선사고 등의 신속한 대응을 위하여 주위의 선량률 준위에 따라 크기별로 여러 종류의 콜리메이터들을 구비해야 하며, 이는 무거운 콜리메이터의 특성상 효율적인 현장 측정에 심각한 장애가 될 수 있다. 본 연구에서는 콜리메이터의 모양을 카메라의 렌즈 조리개 형식으로 제작하여 사용자가 직접 카메라 렌즈를 돌려 초점을 맞추듯이 콜리메이터의 내경을 조절하고 방사선의 감쇄율을 쉽게 알아볼 수 있도록 IRIS형 콜리메이터를 제작하였다. 먼저, 콜리메이터를 위상을 달리한 2 중의 텅스텐 셔터 구조로 제작하여 기계적 공차에 의한 방사선의 침투를 차단하고자 하였다. 그리고 셔터의 재질별로 콜리메이터 내경에 따른 방사선 감쇄율을 MCNP 코드를 이용하여 계산함으로써 이론적인 성능평가를 수행하였다. 계산된 내경의 크기별 감쇄율을 콜리메이터 외부 눈금링에 표시함으로써, 카메라 렌즈에 표시된 배율과 같은 방법으로 사용자가 해당 표시지점으로 콜리메이터 내경을 조절하였을 때, 방사선의 세기가 얼마만 큼 감소되는지 쉽게 알아볼 수 있도록 구현하였다. 끝으로 개발된 IRIS형 콜리메이터를 장착한 소형 방사선검출기를 현장 측정에 활용할 경우, 콜리메이터의 교체 없이 주위 방사선의 세기에 따라 콜리메이터 내경을 적절한 크기로 신속히 교체가 가능하며, 방사선 세기의 감쇄 정도를 쉽게 알아보게 함으로써 신속하고 정확한 대처가 가능할 것으로 판단된다.

(La, Sr)MnO3을 이용한 가변 방사율 소재에 관한 연구 (Experimental Investigation of Variable Emittance Material Based on (La, Sr)MnO3)

  • 가변 방사율 라디에이터는 우주환경에서 작동하는 장비의 온도가 낮을 때는 방사율이 낮아져 시스템의 외부로의 열 손실을 줄이고, 반대로 온도가 높은 때는 방사율이 높아져 장비의 열을 외부로 방출할 수 있어 효율적인 열제어가 가능하게 한다. 가변 방사율 소재로 보편적으로 이용하고 있는 것 중 하나인 감온변색 장치(thermochromics device)는 추가적인 장비 필요 없이 열제어가 가능하기 때문에 효율적이고 고장이 발생하지 않는 장점을 가지고 있다. 본 연구는 이와 같은 이유로 감온변색 장치의 소재 중 하나인 LSMO($La_{1-x}Sr_xMnO_3$)채택하여 졸-겔(sol-gel)방법을 이용하여 제작하고, 제작된 샘플의 가변방사 성능을 실험적으로 규명하였다. 또한, 진공환경과 극저온 환경에 반복 노출시켜 시편이 극심한 환경 변화 이후에도 방사율 값이 일정함을 확인하여 우주적합성을 판단하였다.

생활한복 이미지를 활용한 캐주얼웨어 디자인 개발 (A Study on the Casual Wear Design based on the Image of the Modernized Korean Costume)

  • Costume is a formative art expressed by active human unlike the field of other living formativeness, and an expression of social actions based on a style of culture in a period. Therefore, costume has a deep connection with a mode of living and is recognized as a 'culture for living', and is more characteristic culture than all metaphysical cultures including architecture, craft, painting, and sculpture. Therefore, it expresses wearer's status or social standing, and furthermore, ideas and values of the times with aesthetic features in their form, color, materials, and pattern, so it is expressed as a mirror reflecting the phases of the times as. Korean costume had been dressed until the period of the Joseon Dynasty without a great change and started to be simplified in a simple style on the grounds of inconvenience in behaviors with the opening of an interchange of Western culture in the civilized period. And, this movement had been continued and Korean costume had been applied as an everyday dress under the name of 'Reformed Korean Costume'. Since the middle phase of 1980s, it aroused many people's interests with the introduction of designs focused on activity and convenience. In 1990's, many people had taken a growing interest in Korean costume with development of various designs keeping pace with the internationalization period and Korean Costume had been revitalized under the name of 'The Modernized Korean Costume'. And, since the 21st century, the advanced communication and full-scaled import of Western fashion have made the introduction of many fashion information in the world into Korea, affected greatly the fashion market, led consumers' sensitivity on a trend to be increased. Therefore, a design accepting a trend 'The Modernized Korean Costume' with fashions has risen. Second, this study is an attempt to suggest a revitalization method of domestic casual Korean costume brands by developing and suggesting competitive and highly value-added products with connection of practicality, variety, and highly sensitive fashion styles. For theoretical study, domestic and foreign literatures, academic journals, professional monthly magazines, and newspapers were examined. And, a process of change and features of the Korean fashion since the civilization period, and concept, features and images of casual Korean costume were analyzed, On the basis of analyzing image, features, and consumers' preference of the modernized Korean costume, a design development plan was established and 10 suits of costume were designed and made.

일측성 성대마비 환자에서 음성치료 효과를 예측할 수 있는 인자 (Factors Predictive of Voice Therapy Outcome in Patients with Unilateral Vocal Fold Paralysis)

  • Background and Objectives : Unilateral vocal fold paralysis is generally treated using injection laryngoplasty or voice therapy. However, the decision of treatment method is dependent on clinician's preference and hospital facilities without specific criteria. The purpose of the study was to examine factors predictive of voice therapy outcome in patients with unilateral vocal fold paralysis. Materials and Method : 38 patients diagnosed as unilateral vocal fold paralysis, aged from 24 to 81 years and undergone voice therapy more than 1 month were included. After 3 to 12 (mean 5.1) sessions of voice therapy, subjects had divided into responder group (RG, 28 patients) and non-responder group (NRG, 10 patients) according to G scale change. Paramters of perceptual assessment, acoustic and aerodynamic measure, and videostroboscopy were compared between two groups, and factors predictive of voice therapy result were analyzed. Results : RG patients showed significantly reduced rough, breathy, asthenic voice after voice therapy. Change of MPT and MFR was more substantial in RG than in NRG. By videostroboscopy, RG patients showed significantly more mucosal wave symmetry, glottal closure, reduced glottal gap index during the closed phase of phonation, while NRG patients showed more occurrences of abnomal supraglottic activities during phonation (p < 0.05). Poor outcome of voice therapy significantly associated with increased asthenic scale, short MPT, and less glottal closure (p=0.02). In addition, 90% of patients with MPT more than 5 seconds were in RG, whereas 56% of patients with MPT less than 5 secondes were in RG. Conclusion : Voice therapy is useful for large proportion of patients with unilateral vocal fold paralysis as an initial treatment method. However, patients with large asthenia scale, large glottic gap or MPT less than 5 seconds tend to have poor voice therapy outcome, and early injection laryngoplasty maybe recommended for these patients.
