  • Traditional pharmacological theory was not built in a day. It had been changing in the past, still changing in the present day and will be changing in the future. The transition was made by criticism, modification and complements through countless debates. Therefore, we can learn the systematic stream of traditional pharmacology and logical construction and make an estimate of the changes in the future, by looking at the debates and the transition of pharmacological theory.

A Study on the medical and pharmacological theory of Interior-Overheated-Disease of Taeumin (태음인(太陰人) 이열병(裡熱病)의 병증(病證) 약리(藥理)에 대한 연구(硏究))

Background and Purpose: I intended to understand the medical and pharmacological theory of Taeumin, through a study of the process through which Lee Je-ma discovered the Interior-Overheated-Disease of Taeumin and created the prescriptions for it. 2. Methods: I studied and analized the change in the medical and pharmacological theory, through a historical study on the quotations and prescriptions of "DongYi Soose Bowon". 3. Results: Through a literature study I could find that in the existing Oriental Medicine before Lee Je-ma, the difference of the Liver Febrile Disease of Taeumin and the Stomach Febrile Disease of Soyangin were recognized, and the prescriptions of the two diseases were a little distinguished, but the medical theories of those were not distinguished at all. And I found that the Liver Febrile Disease of Taeumin shows the pain in the eyes and the throat, and drying of the nose due to interior dry and heat, so it is different from the Stomach Febrile Disease of Soyangin that shows irritation of fever, headache, and the trouble in passing feces and urine. Also I could know that Radix Puerariae, Rhizoma Cimicifugae, Rhizoma Ligustici, Radix Angelicae Dahuricae, and Radix et Rhizoma Rhei are Taeumin's medicines because these medicines had been used for Taeumin's syndromes, and that Radix Scutellariae, Radix Platycodi, Semen Raphani, and Fructus Gleditsiae are Tae-Eum-In's medicines because these medicines are known as acting for lung.

Herbal and Pharmacological effects of Ginseng Radix and Strategy for Future Research (인삼의 본초 및 약리 효능과 향후 연구방향)

  • For the purpose of wide application of Korea Ginseng Radix (KGR) as a tonic and medicinal drug, bibliographical study on Ginseng Radix (GR) was done about its herbal and pharmacological effects known so far. Herbal effect of GR was studied in 28 classical books including Shennongbencaojing(신농본초경), the oldest herbal book in oriental medicine and also its pharmacological effects was compared with its herbal effect. In bibliographical study 38 kinds of efficacies of GR were referred in those books, for example, in fields of gastrointestinal tract, heart, psychology, body fluid, hormone and respiratory tract in order from statistical analysis. GR was chiefly used for supplementing Qi, vital energy in oriental medicine. However, experimental study on respiratory diseases was not reported except lung cancer yet, while GR was used for the treatment of asthma mostly mixed with other herb in oriental medicine. So far research with GR was performed chiefly for isolation of constituents as saponins and evaluation of its efficacy experimentally by KGTR (Korea Ginseng Tobacco Research Institute). From the above studies, to make KGR used widely than before, I suggest study with KGR should be done according to oriental principles such as Qi and tasters and combination theory between herbs excepting typical analytical research with saponins.

The application of recent taste theory in physiology for the Kimi theory of the 『ShinNongBonChoKyung(Divine Farmer's Classic of Materia Medica)』. (현대 생리학적 미각(味覺) 이론을 통한 『신농본초경(神農本草經)』 기미론(氣味論)의 재해석)

  • Objects : Recent taste theory has been advanced to the receptor and cell of taste. We tried to apply the taste theory to the Kimi theory (氣味論) of 『ShinNongBonChoKyung (神農本草經)』 for the purpose of resonance between the Kimi theory and the taste theory. Methods : The special issues in Current Opinion in Physiology of the year 2021 gave us much information. Based on it, we have searched for more papers using google scholars with the key words; taste, taste receptor, sweet, umami, bitter, salty, sour. Then, we analyzed and compared the Kimi theory of 『ShinNong BonChoKyung)』 with the research papers of physiological taste. Results : Three classification of the herbs in 『ShinNongBonChoKyung』 is very reasonable based on the Taste theory. There is umami and no spicy in the taste theory, and Sweet includes umami in the Kimi theory. Taste is such a complex and connective sensory reaction of the brain and body of human being. There are much more results to couple the Kimi with the taste theory. Conclusions : The pharmacological property of Kimi theory of 『ShinNongBonCho Kyung』 will help the taste theory much more develop and expand. In addition to qi(氣), the Kimi theory will also find a way to be in harmony with modern pharmacology in the respective of taste(味) with the help of taste theory.

Bibliographical Study on Sanghanron (I) (한의약학의 서지학적(書誌學的) 연구(I) -상한론(傷寒論)을 중심으로-)

  • Three important books in the oriental medicine are Huangjenaekyung (medical book), Sanghanron (pharmacological book) and Shinnongbonchokyung (herbal book). Chang, joong-kyung who is the writer of Sanghanron would be recommended as a saint physician and his book, Sanghanron is granted for a best book in this field by Chinese people. Also, Japanese physicians would estimate Sanghanron as the most magnificent book ever written by a human being. Sanghanron contains a lot of confirmations (證) of diseases, their progressing courses and exact therapeutic ways. Therefore modern physicians and pharmacists who are interested in the oriental medicine might study and practice more scientific approach of therapy for patients. The aim of this bibliographical study on Sanghanron is to explain systematically theories of this book and harmonize them with the modern medicinal study: In this study we have acquired three main results, the first is that we could get the coincidence between western & oriental way on the five therapeutic principles-sweating, vomiting, harmonizing, excreting and bleeding : the second, notice the exclusion of the Five Element Theory(the fundamental one in oriental medicine) and the third, found out the possibility of the immunological and neuroscientific explanation for pharmacological application to the oriental medicine.

Mitochondria: multifaceted regulators of aging

  • Aging is accompanied by a time-dependent progressive deterioration of multiple factors of the cellular system. The past several decades have witnessed major leaps in our understanding of the biological mechanisms of aging using dietary, genetic, pharmacological, and physical interventions. Metabolic processes, including nutrient sensing pathways and mitochondrial function, have emerged as prominent regulators of aging. Mitochondria have been considered to play a key role largely due to their production of reactive oxygen species (ROS), resulting in DNA damage that accumulates over time and ultimately causes cellular failure. This theory, known as the mitochondrial free radical theory of aging (MFRTA), was favored by the aging field, but increasing inconsistent evidence has led to criticism and rejection of this idea. However, MFRTA should not be hastily rejected in its entirety because we now understand that ROS is not simply an undesired toxic metabolic byproduct, but also an important signaling molecule that is vital to cellular fitness. Notably, mitochondrial function, a term traditionally referred to bioenergetics and apoptosis, has since expanded considerably. It encompasses numerous other key biological processes, including the following: (i) complex metabolic processes, (ii) intracellular and endocrine signaling/communication, and (iii) immunity/inflammation. Here, we will discuss shortcomings of previous concepts regarding mitochondria in aging and their emerging roles based on recent advances. We will also discuss how the mitochondrial genome integrates with major theories on the evolution of aging.

The life and medical idea of Jang Won-So (장원소(張元素)의 생규(生涯)와 의학사상(醫學思想))

  • Geum-Won dynasty is the era which totalized the every medical attainments of before Song dynasty and appeared some medical practioners who found new medical theory based upon this, and is important time for various developments of the oriental medical theory. At this time of the era, the representative practioners are called the four noted physician of Geum-Won dynasty. Jang Won-So who influenced the formation of the theory of the four noted physcian of Geum-Won dynasty, originated new theory by joining the theory of "Nae-Gyung" with his own medical experience. The results can be summarized as follows after studying his life and idea of medicine. 1. Jang Won-So, courtesy name is Gyeol-Go, came from Yeog-Joo and it is unknown that when he was born and dead, but he lived in 11C. At 27, after failed in a Jin-Sa examination, he started medical study, and widely spreaded his medical art by healing the Yoo Wan-So's Sang-Han disease. 2. There are many Jang Won-So's writings, such as "Eui-Hag-Gye-Won" which is the condensation of his idea of medicine, "Jin-Joo-Nang" which is greatly contributed to development of Herbology, "Jang-Boo-Pyo-Bon-Han-Yeol-Heo-Sil-Yong-Yag-Sig" which is the sample of Jang-Boo-Byun-Jeung-Lon-Chi, and "Yag-Joo-Nan-Gyung" "Gyeol-Go-Ga-Jin" "Gyeol-Go-Joo-Sug-Hwa-Mag-Gyeol" "Gyeol-Go-Bon-Cho" "San-Yog-Bo-Sang-Bang" which are known do not existing. 3. Jang Won-So's study about Jang-Boo-Byun-Jeung was influenced by "Nae-Gyung" "Jung-Jang-Gyung" "So-A-Yag-Jeung-Jig-Gyeol" etc. and outline by Han-Yeol-Heo-Sil and the change and prognosis of the state of disease can be decided by Saeng-Sa-Yeog-Soon, so he contribute to development of lang-Boo-Byun-Jeung-Lon-Chi of posterity. 4. Jang Won-So succeeded and develope the theory of the herbs of Oh-Jang-Go-Yog-Bo-Sa and about Ki-Mi-Seung-Gang-Boo-Chim etc. appeared on "Nae-Gyung", especially invented such as Kwi-Gyung-Lon, In-Gyeung-Bo-Sa-Seol, Jang Boo-Pyo-Bon-Yag-Sig and he enriched the content of pharmacological theory, so he motivated various development of herbalogical theory. 5. Jang Won-So's idea of On-Yang-Bi-Wi directly influence Lee Go's assertion of Bi-Wi-Lon, idea of Go-Geum-I-Gue-Seol presented theological basis of posterior medical practioners formation of prescription, so the way of drug usage greatly developed. 6. Jang Won-So's theory of the lang-Boo-Byun-Jeung and herbology directly influenced to Lee Go, Wang Ho-Go, Na Chen-Ik, lang Byeok etc. so he became the father of Yeog-Soo class and indirectly influenced to Seol Gi, Jo Hen-Ga, lang Gae-Bin, Lee Jung-Jae etc. of Myeng dynasty, so the On-Bo class has been formed. Like this, because, his idea of medicine developed at various aspect and greatly influenced to posterity, Lee Si-Jin said "Since the emergency of "Nae-Gyung", Jang Won-So is the only man who greatly enhance medical principle.", so it is clear that this is not unfounded expression at all.

Conformational and Molecular Dynamical Properties of Damaged DNA (손상된 핵산의 구조와 분자동력학적 특성)

  • Some of the benzopyrene (BP)-DNA adduct are known to build intercalated motif between flanking base pairs in damaged DNA depending on the structural condition. The size of benzopyrene itself is definitely not comparable with any of the DNA bases and thus the question whether the lesion of some base pair by insertion of benzopyrene can happen with or without a dramatic distortion of the helical structure is a highly interesting theme. In this work we used a molecular dynamics simulation based on the theory of molecular mechanics. The specific consequences about the structural properties of the intercalated structures and benzopyrene motif in minor groove of the double helix are deduced after 5 ns simulation time.

Drug research and development tend to hyperlipidemia (이상지질혈증과 치료제 연구개발 경향)

  • Most of the cholesterol is synthesized by liver in the body while about one of third is taken via dietary. The main functions of cholesterol is to protect membranes in cell surface, avoid the arterial bleeding by hypertension, and prolong the life of erythrocytes, and so on. However, overload of cholesterol leads to arteriosclerosis associated with leading death cause. Lack of physical activity, emotional and environmental stress, and low intake of protein or vitamin E induce the unbalance between HDL- and LDL-cholesterol so become a basis of ischemic disorders in heart, brain and elsewhere in the body. So far, four major classes of medications for hyperlipidemia are HMG-CoA reductase inhibitors (statins), bile acid sequestrants, nicotinic acid, and fibric acids. The statins can lower LDL and levels triglyceride, but may induce myopathy and an elevation of liver enzyme levels. The bile acid sequestrants lower LDL levels and raise HDL levels with no effect on triglyceride levels but side effects of gastrointestinal (GI) distress, constipation, and a decrease in the absorption of other drugs. Nicotinic acid and fibric acids lower LDL and triglyceride levels with showing flushing, hyperglycemia, hyperuricemia, GI distress, and hepatotoxicity dyspepsia, gallstones, myopathy, and unexplained noncardiac death as adverse effects. Above western drugs lower cholesterol by 15 to 30% while all have notable adverse effects. In traditional medicine, hyperlipidemia is regarded as retention of phlegm and fluid disease. Etiology and pathogenesis of hyperlipidemia is basically based on Spleen-Deficiency and Phlegm-Stagnation, accumulation and stasis of -heat, and Qi & blood stagnation induced by Phlegm-damp, water-dampness, and blood stasis. Thereby, strengthening Spleen and dissolving Phlegm, clearing away heat and diuresis, and supplementing Qi and activating blood circulation are commonly used therapeutic methods for hyperlipidemia. The traditional herbal medicine, have been used for patients with CVA, hypertension or hyperlipidemia in Oriental hospital or Oriental clinic. The lock and key theory is used to develop most of western medicine, however many diseases are caused by mixed factors in body-complex system. We expect that Oriental pharmacological theory could be newborn as a novel drug showing high advantage of blood levels of lipidsand QOL of performance without side effects.

Effect of KH-BaRoKer-SeongJangTang based on traditional medicine theory on longitudinal bone growth

  • KH-BaRoKer-SeongJangTang (KBS) is a recently developed formulation by using traditional drugs considering traditional medical theory of Oriental books such as ShinNongBonChoGyeong and JuRye, which has been used to improve the growth of child in Korea. Although KBS is usually prescribed to many children who are in retard for their age, its pharmacological effects have not been fully understood in experimental models. The aim of this study was to evaluate the effects of KBS on bone growth. Growth plate thickness and bone parameters such as bone volume/tissue volume (BV/TV), trabecular thickness (Tb.Th), trabecular number (Tb.N), connection density (Conn.D), and total porosity were analyzed by means of microcomputed tomography. Serum insulin-like growth factor-I (IGF-I) levels were measured by enzyme-linked immunosorbent assay. Hepatic IGF-I mRNA expression was analyzed by real-time polymerase chain reaction. Phosphorylation of signal transducer and activator of transcription5 (STAT5) was investigated using Western blot analysis and immunohistochemistry. The thickness of growth plate was increased by KBS. BV/TV, Tb.Th, TbN, Conn.D, and total porosity were improved by KBS. Hepatic IGF-I mRNA and serum IGF-I levels were elevated by KBS. Phosphorylation of STAT5 was increased with administration of KBS. These results suggest that KBS would be helpful to children who are in retard for their age through the elevation of IGF-I.
