These studies were carried out to select the active grow hairy root lines induced from various ginseng(Panax ginseng C. A. Meyer) parts. Hairy roots were induced in root explants, stem and petiole in vitro by A. rhizogenes R1000 or A. rhizogenes $A_4$. These hairy roots could be grown on the phytohormone free medium, and PCR analysis of rol C and vir C gene fragments confirmed that hairy roots were transgenic tissues. We have selected 11 hairy root lines with active growing characters among 300 hairy root lines selected based on growth and morphological characteristics on 1/2MS solid media with 250 mg/L carbenicillin. Morphological characteristics of selected 11 hairy root lines were thickness and thiness of main roots, and many projection for lateral roots, active grow of lateral roots. Among selected 11 hair root lines prominent characteristics of hairy roots with active growing characters were thiness of main roots and active grow of lateral roots. But characteristics of low growing hairy roots were thickness of main roots and low grow of lateral roots. Finally we have selected actively growing hairy roots, KGHR-1, KGHR-5, KGHR-8 among 11 hairy root lines.
Volatile flavor components in leaf and petiole of fresh Pleurospermum kamtschaticum H$\_$OFFM/ were extracted by SDE (simultaneous steam distillation and extraction) method using diethyl ether as solvent. Essential oils were analyzed by gas chromatography (GC) and combined gas chromatography-mass spectrometry (GC-MS). Identification of volatile flavor components was based on the Rl of GC and mass spectrum of GC-MS. A total of 31 components, including 15 hydrocarbons, 4 aldehydes, 1 ketone, 5 alcohols, 2 esters, 3 acids and 1 oxide were identified in the essential oils. (Z)-${\beta}$-Farnesene, (Z, E)-${\alpha}$-farnesene and farnesene were the major volatile flavor components in fresh Pleurospermum kamtschaticum. Volatile flavor patterns of Pleurospermum kamtschaticum were analyzed using electronic nose. Sensor T30/1 and PA2 that were sensitive to alcohols had the highest resistance for fresh Pleurospermum kamtschaticum. Resistance of six metal oxide sensors was decreased in dried sample compared with fresh one.
This study was conducted to determine the optimal planting distance in cultivation of yacon (Polymnia sonchifolia Poeppig & Endlicher). Plug seedlings were planted with 6 different plant densities of 80 $\times$ 60, 80 $\times$ 50, 70 $\times$ 50, 70 $\times$ 45, 70 $\times$ 40 and 60 $\times$ 45 cm. The plant height and the petiole length were increased with increasing the planting distance. The tallest plant height of yacon was 165.4cm with the plant density of 80 $\times$ 60 cm. However, branch number per plant, leaf number on main stem and stem diameter were not significantly difference among planting densities. Tuberous root was harvested 31.42 tons/ha in 70 $\times$ 50 cm spacing. The ratio of heavier tuberous roots than 200 g to total tuberous roots decreased significantly according to increase of planting density. Fresh weights of shoot and root, contain the crown bud, were decreased, as planting distance was shorter. Tuberous root number was fewer but its weight was heavier in wide planting than in dense planting. We think that optimal planting density is about 30,000 plants/ha, if it were to be 70 cm row spacing, intrarow spacing should calculate about 47cm.
We describe culture conditions for a high-efficiency in vitro regeneration system of Papaver nudicaule through somatic embryogenesis and secondary somatic embryogenesis. The embryogenic callus induction rate was highest when petiole explants were cultured on Murashige and Skoog (MS) medium containing 1.0 mg $1^{-1}$${\alpha}$-naphthaleneacetic acid (NAA) and 0.1 mg $1^{-1}$ 6-benzyladenine (BA) (36.7%). When transferred to plant growth regulator (PGR)-free medium, 430 somatic embryos formed asynchronously from 90 mg of embryogenic callus in each 100-ml flask. Early-stage somatic embryos were transferred to MS medium containing 1.0 mg $1^{-1}$ BA and 1.0 mg $1^{-1}$ NAA to germinate at high frequency (97.6%). One-third-strength MS medium with 1.0% sucrose and 1.0 mg $1^{-1}$$GA_3$ had the highest frequency of plantlet conversion from somatic embryos (91.2%). Over 90% of regenerated plantlets were successfully acclimated in the greenhouse. Secondary somatic embryos were frequently induced directly when the excised hypocotyls of the primary somatic embryos were cultured on MS medium without PGRs. Sucrose concentration significantly affected the induction of secondary embryos. The highest induction rate (89.5) and number of secondary somatic embryos per explant (9.3) were obtained by 1% sucrose. Most secondary embryos (87.2-94.3%) developed into the cotyledonary stage on induction medium. All cotyledonary secondary embryos were converted into plantlets both in liquid and on semisolid 1/3-strength MS medium with 1.0% sucrose.
Gray mold caused by Botrytis cinerea occurred on Astragalus sinicus in an experimetal rice field of Gyeongsangnam-do Agricultural Research and Extension Services in Korea from 2008 to 2009. The symptoms appeared on the leaves, petiole and stems of infected plants. and then started with water-soaked, rotted and eventually died blight on plants. The colony were grayish brown and the optimum temperature for sclerotial formation was $20^{\circ}C$ on potato dextrose agar. The conidia were one celled, mostly ellipsoid or ovoid in shape, and were colorless or pale brown in color. The conidia were $7{\sim}16{\times}5{\sim}10{\mu}m$ in size and conidiophores were $14{\sim}28{\mu}m$ in length. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Botrytis cinerea Persoon: Fries.
$\beta$-Glucuronidase (GUS) gene of Escherichia coli was introduced into sweet potato (Ipomoea batatas (L.) Lam.) cells by particle bombardment and expressed in the regenerated plants. Microprojectiles coated with DNA of a binary vector pBI121 carrying CaMV35S promoter-GUS gene fusion and a neomycin phosphotransferase gene as selection marker were bombarded on embryogenic calli which originated from shoot apical meristem-derived callus and transferred to Murashige and Skoog (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxyacetic acid and 100 mg/L kanamycin. Bombarded calli were subcultured at 4 week intervals for six months. Kanamycin-resistant calli transferred to MS medium supplemented with 0.03 mg/L 2iP, 0.03 mg/L ABA, and 50 mg/L kanamycin gave rise to somatic embryos. Upon transfer to MS basal medium without kanamycin, they developed into plantlets. PCR and northern analyses of six regenerants transplanted to potting soil confirmed that the GUS gene was inserted into the genome of the six regenerated plants. A histochemical assay revealed that the GUS gene was preferentially expressed in the vascular bundle and the epidermal layer of leaf, petiole, and tuberous root.

This study was carried out to investigate the growth responses of medicinal plants such as Glehnia littoralis, Cryptotaenia japonica, Angelica gigas, Reynoutria japonica, Saururus chinensis, Houttuynia cordata, Ligularia fischeri, and Aralia cordata under 70%-shading and full sunlight conditions on a rooftop condition using artificial soil as a growth media. Glehnia littoralis, Cryptotaenia japonica, Ligularia fischeri, Saururus chinensis, and Houttuynia cordata showed better growth on plant height, leaf length, leaf width, petiole length and leaf number under full sunlight conditions on a rooftop condition. Cryptotaenia japonica and Houttuynia cordata had high value of Hunter's a (red-green) under full sunlight, while had lower value of L(lightness) and b(blue-yellow) than those of 70% shading condition. As an index of plant stress response, Glehnia littoralis, Reynoutria japonica, Houttuynia cordata, Ligularia fischeri showed Fv/Fm values 0.79~0.84 under full sunlight in August, indicating low stress on plant growth. Therefore they seemed to be suitable medicinal plats for rooftop conditions. Ligularia fischeri and Reynoutria japonica showed better growth under 70% shading treatment in August, while showed high growth response under full sunlight conditions in September. The 70% shading treatment was effective for the growth of Angelica gigas, Reynoutria japonica, and Aralia cordata. Angelica gigas and Reynoutria japonica, however, showed better plant growth under full sunlight during summer. The results showed that Glehnia littoralis, Cryptotaenia japonica, Ligularia fischeri, Saururus chinensis, and Houttuynia cordat seemed to be suitable medicinal plants for rooftop garden where there is a full sunlight condition.
To develop a new propagation method of Korean ginseng (Panax ginseng C.A. Meyer) by cutting, stem cuttings with leaflets obtained from ginseng seedlings were planted in rooting media treated with three levels of NAA concentration. NAA solution of 1.0 ppm was the most effective for the rooting of ginseng stem cuttings as well as for root growth after initiation of the roots from cuttings. A mixture of sand and leaf compost as a rooting medium for ginseng stem cuttings showed the best results in rooting percentage of cuttings and root growth after rooting. An acril film cap covering was very effective to prevent water loss by transpiration from the leaflets of stem cuttings and for rooting and root growth after rooting. Cuttings of leaf petiole with 5 leaflets from 2 year old ginseng plants showed good rooting as well as root growth after rooting in a mixture of sand and leaf compost treated with 1.0 ppm NAA.

To understand effect of inoculum size, cell density, sucrose concentration and concentrations of MS basal on suspension culture and protoplast isolation of wild viola(Viola patrinii DC.) callus from petiole segments this experiment was conducted. In the lot of 30 mesh inoculum size, two observations were; One was that a considerable increase in the fresh and dry weight of callus was determined. Another was that the callus mass was relatively compact compared with others. A recommendable cell density was 0.4g for 20ml culture medium and the higher sucrose concentration, the higher fresh and dry weight were obtained. The dilution of MS basal salt was differently affected on fresh and dry weight; the highest fresh weight was found in 1x MS salt, while the higest dry weight was in 1/3x dilution. The addition of casein hydrolysate(3g/L) was more effective to increase of both fresh and dry weight. THe contents of protein was great in the inoculum lots with larger inoculum sizeand higher concentration of MS basal salts contenting 3g/L of casein hydrolysate and higher sucrose compared with others. The greatest protoplasts were isolated from the lot of 10 mesh size treated with 1%pectinase SE-150 and 2% cellulase YC. In general, for optimal protoplast isolation the followingconditions were recommended; 1) Use of smaller cell size cultured for 2-5 weeks, and 2) more than 5 hours incubation using the combined mixture of the enzymes with proper concentrations.
A black scab disease occurred on sword bean (Canavalia gladiata) in plastic film houses around Chinju area during the spring season of 1999. The disease started from flower bud, then moved to flower stalk, pod, petiole, cirrus, stem and leaves. The lesions started with small dark brown spots then were gradually expanded. Severely infected plants reached 37.4% of whole plant covered with scab. Numerous conidia were produced on the diseased flower disk, pod, floral axis, stem and leaves. Most of the conidia were appeared to be readily dispersed in the air, but the mycelia were not suggested causing of sooty mold by ectoparasitism. A fungus was isolated from the diseased stem, and inoculated to healthy plants to satisfy the Koch's postulates and proved the fungus was the causal agent of the disease. The isolated fungus grew on potato dextrose agar, forming greenish black to pale brown colonies. Conidia were ellipsoidal, fusiform or subspherical, mostly one-celled but occasionally septated. The conidia were $3.9{\sim}34.1{\times}2.7{\sim}5.1\;{\mu}m$ in size and formed in long branched chains on the erected conidiophores which were pale olivaceous brown and variable in length between $7.2{\sim}210.7\;{\mu}m$ in size. Ramoconidia were $7.6{\sim}29.2{\times}3.2{\sim}14.4\;{\mu}m$ in size. The fungus was identified as Cladosporium cucumerinum based on the above morphological characteristics. The optimum temperature for mycelial growth and conidial formation was about 15 to $25^{\circ}C$. Cladosporium scab of sword bean caused by the fungi has not been reported in Korea previously.
