Effects of Sclerophyllous Plant Leaves Addition on Fermentative and Sensory Characteristics of Kimchi (경엽식물 잎의 첨가가 김치의 발효 및 관능특성에 미치는 효과)

  • The effects of persimmon, mulberry and bamboo leaves addition on the fermentative and sensory characteristics of kimchi were investigated. Total polyphenol content, DPPH radical scavenging activity and lactic acid bacteria growth inhibition of EtOH extract from persimmon leaves were significantly higher than those from mulberry and bamboo leaves. From the sensory evaluation of kimchi added with three chopped plant leaves, kimchi added with chopped persimmon leaves gave the highest point in color, flavor, texture and overall preference significantly (p<0.05). When the chopped persimmon leaves were added at a ratio of 0.3% based on cabbage weight, the changes of pH and total acidity (TA) during the storage for 14 days were significantly lowered, compared to general kimchi (p<0.05). Total polyphenol content and DPPH radical scavenging activity of kimchi supplemented with chopped persimmon leaves at 0.3% also significantly increased after storage for 14 days (p<0.05), and this kimchi provided higher total polyphenol content and DPPH radical scavenging activity than general kimchi. These results suggest that the addition of perisimmon leaves have significant influences on the fermentation and sensory characteristics of kimchi.

Separation of Antioxidant compounds from Persimmon Leaves (감잎(시엽(枾葉))의 항산화 성분 분리)

  • This study was carried out to separate and identify the antioxidative substances in persimmon leaves. The antioxidative substances in persimmon leaves were extracted by methanol. The extract was fractionnated by SEP-PAK cartridge colum. From these results five fractions(F-I${\sim}$V) were obtained. Antioxidative activity of each fractions was examined by the DPPH methord. The F-II, III and IV showed antioxidative activity and among them F-II and F-III showed the strongest. Five frations were separated by TLC using ethylacetate : chloroform : formic acid : $H_2O$(8 : 1 : 1 : 1 v /v) as the solvent. From these results were obtained spots of Rf 0.71, 0.35 and 0.25. This spots were scraped from the plate and extracted by methanol. The extracts thuse obtained were used for examination of identify by TLC, UV /VIS-spectrophotometer and HPLC. Among them spot of Rf 0.71 were demonstrated to catechin and the spots of Rf 0.35 and 0.25 was suggested to polyphenol substances.

Physicochemical characteristics of hot-water leachate prepared from persimmon leaf dried after steaming or freezing treatment (스팀 및 동결 전처리가 건조 감잎 열수추출물의 이화학적 특성에 미치는 영향)

  • This study was conducted to develop a preservation technology that can induce changes in physicochemical properties to effectively utilize of persimmon leaves. The application effects of steaming or freezing technique were investigated. Astringent persimmon leaves were steam-blanched (100℃, 30 sec) or frozen (-20℃, 15 d), followed by hot-air drying (50℃). The physicochemical properties of the extract obtained by hot-water leaching from the dried leaves were compared. The extract of leaves dried without pretreatment was used as a control. L* value was higher in steamed than in control and frozen. a* value was highest in the control. The browning index was higher in the frozen and lower in the steamed than in the control. Soluble solids were the highest in the steamed and the lowest in the frozen. Sucrose content was relatively high in the steamed, and the glucose and fructose contents were relatively high in the frozen. Total polyphenol content and DPPH radical scavenging activity were higher in steamed and lower in frozen than in control. Thus, it was confirmed that steam or freeze pretreatment after harvesting persimmon leaves affects the extraction yield, color, antioxidant capacity and component changes of dried persimmon leaves. Unlike steaming, freezing pretreatment showed the effect of promoting decomposition and browning reactions, and it is considered useful when such an effect is needed.

Microscopic Observation of the Pseudothecial Development of Mycosphaerella nawae on Persimmon Leaves Infected by Ascospore and Conidia (감나무 둥근무늬낙엽병균 Mycosphaerella nawae의 자낭포자 및 분생포자에 감염된 이병엽 상에서 위자낭각 형성과정 관찰)

  • In order to illustrate the role of conidia of Mycosphaerella nawae as a secondary inoculum in nature, pseudothecial development on persimmon leaves was investigated microscopically. The fungal ascospores have been believed as the primary or only inoculum source in nature, however, pseudothecia were readily formed on persimmon leaves infected naturally and artificially by conidia. The pseudothecia of M. nawae were found to form in the tissues of infected leaves while the leaves were still hanging on the trees. The size of pseudothecia were approximately 51.0~122.4$\times$51.0~112.2 ${\mu}{\textrm}{m}$ (82.8 $\times$72.5 ${\mu}{\textrm}{m}$in average), the shapes were spherical, ovoid or occidental pear type. The sizes of asci were approximately 30.6~61.2$\times$8.2~10.2 ${\mu}{\textrm}{m}$(46.6$\times$9.4 ${\mu}{\textrm}{m}$ in average) and the shapes were cylinder or banana. The ascospores were mostly spindle type, and the sizes were 10.2~12.2$\times$3.1~4.1 ${\mu}{\textrm}{m}$ (11.4$\times$3.2 ${\mu}{\textrm}{m}$ in average)-like. The pseudothecial formation was initiated before defoliation and morphological characteristics of the pseudothecia, ascus and ascospores on the infected leaves were fully illustrated in this study. Results indicated that conidia of M. nawae induce circular leaf spot of persimmon as much as ascospores, and might play an important role of the disease epidemics in nature.

Development of a Transient ihpRNA-induced Gene Silencing System for Functional Analysis in Persimmon (Diospyros kaki Thunb.)

  • A transient ihpRNA-induced gene silencing system based on Agrobacterium-mediated injection infiltration has been established to evaluate candidate genes involved in proanthocyanidin (PAs) biosynthesis in persimmon (Diospyros kaki Thunb.). We chose DkPDS (phytoene desaturase) as a gene-silencing target to evaluate the newly developed transient gene silencing system. Our qRT-PCR analysis indicated that two ihpRNA constructs (pHG-PDS5' and pHG-PDS3') targeted DkPDS, which also led to significantly reduce expression of DkPDS in 'Mopanshi' persimmon leaves. To further confirm the reliability of the system, we successfully utilized it for DkLAR (leucoanthocyanidin reductase) gene silencing. The expression levels of DkLAR in 'Mopanshi' and 'Eshi 1' leaves were ca. 6-fold and ca. 5-fold lower than those in leaves harboring empty vector (pHG-GFP), respectively. DMACA (4-dimethylaminocinnamaldehyde) staining and the Folin-Ciocalteau assay showed that the accumulation of PAs was markedly inhibited in 'Mopanshi', 'Eshi 1' and 'Youhou' leaves. These results indicate that DkLAR plays an important role in the accumulation of PAs in persimmon. The transient ihpRNA-induced gene silencing method developed in this study is a highly efficient and useful tool for functional analysis of persimmon genes involved in PA biosynthesis.

Survey of Overwintering Inoculum Potential of Anthracnose of Sweet Persimmon Caused by Colletotrichum gloeosporioides (감나무 탄저병균(Collectotrichum gloeosporioides)의 월동 전염원 조사)

  • In 2006 to 2007, the potential inoculum source of the anthracnose of sweet persimmon caused by Colletotrichum gloeosporioides was surveyed. The infected twigs, buds, dead twigs, petiole, leaves, dropped fruits were collected and tested for their possibility as overwintering inoculum. The detection rates of the pathogen from various parts of sweet persimmon tree were varied. When the collected samples were examined in April. Over than 93.3% of infected twig samples were harbored mycelia of C. gloeosporioides, and 46.7% of infected buds, 36.7% of dead twigs, 23.3% of petioles, and 16.7% of leaves were beared pathogenic fungus. No pathogenic fungus were detecded from healthy twigs and buds. Infected twigs and bud was important overwintering sites and formed conidia actively in next spring. The infected twigs, leaves, petioles, and fruits in growing season produced great number of conidia and caused active dissemination of the anthracnose disease in sweet persimmon. In growing season, all of the infected parts, such as twigs, leaves, petioles, and fruits produced pathogenic fungus.

Changes of Chemical Components in Persimmon Leaves during Growth for Processing Persimmon Leaves Tea (감잎차 제조를 위한 감잎의 성장시기별 함유 성분의 변화)

  • As a foundational study for processing persimmon leaves tea, the physico-chemical characters were investigated in persimmon leaves from Chungdo Bansi, Sagoksi, Kyungsan Bansi and Hiratanenasi during growth. Flesh weights increased rapidly until the middle of May and then decreased slightly. Moisture contents decreased continuously from $79{\sim}81%$ at the beginning of May during growth. Water soluble tannin contents reached $1.55{\sim}2.25%$, maximum values at the middle of May and at the beginning of June, and increased again at the middle of July and then decreased. Contents of catechins, precursor of condensed tannin, indicated $12{\sim}27\;mg%$ at the middle of May and reached $17{\sim}34\;mg%$, maximum values at the middle of June. Contents of catechin were low in order of (+)-catechin, (-)-epicatechin, (-)-epicallocatechingallate, (-)-epigallocatechin and (-)-epicatechingallate. Sugars present in persimmon leaves were composed of sucrose, glucose, fructose, raffinose and mannitol. Sucrose increased continuously, glucose and fructose decreased during growth. Raffinose content was less than 0.1%. Glucose and fructose took more than 90% until the beginning of May, and then sucrose took up $60{\sim}80%$ of total sugar contents. Total vitamin C contents indicated maximum values at the middle of May and at the beginning of June in Chungdo Bansi, Sagoksi and Kyungsan Bansi, maximum vaule at the middle of July in Hiratanenasi. From the basis of these data It was suggested that proper period for picking persimmon leaves prior to processing persimmon leaves tea was from the middle of May to the beginning of June. Since maximum values for most of chemical components occurred at the middle of May and at the beginning of June and persimmon leaves thicken after the middle of June.

Seasonal Occurrence of Japanese Gall-forming Thrips, Ponticulothrips diospyrosi Haga et Okajima, and Its Damage Pattern (감관총채벌레(Ponticulothrips diospyrosj)의 발생소장과 피해 양상)

  • Seasonal occurrence of Japanese gall-forming thrips, Ponticulothrips diospyrosi Haga et Okajima, and its damage pattern on leaves and fruits were studied at sweet persimmon orchards in Gimhae (orchard A, B) and Changwon, Gyeongnam. Korea in 2002 and 2003. Monitoring adults by yellow sticky traps and inspecting each developmental stage in damaged rolled-leaves revealed that the overwintered adults moved to sweet persimmon orchard from late April to late May, and oviposited inside the rolled leaves. Adults developed from the eggs showed peak occurrence of the first generation adults in early to mid June. Inspection of rolled leaves indicated that the peak occurrences of eggs, nymphs, and pupae of the first generation took place in early to mid May, late May to early June, and early June, respectively. Each developmental stage showed the second small peaks in the late season. Results suggest that most thrips live a single generation per year, but a small portion may develop to the second generation in persimmon orchards. The percent of damaged leaves was highest in Changwon orchard at 9.7% in early June. Percentage of damaged fruits increased from 0.84% in early June to 30.2% in early September in Gimhae B orchard. It was found that the closer the persimmon trees were to the edge of the orchard, the worse damaged the leaves were. Appropriate timing for incorporation of control measures were discussed in relation to the seasonal occurrence of adults.

Effect of Dried Leaf Powders and Ethanol Extracts of Persimmon, Green Tea and Pine Needle on Lipid Metabolism and Antioxidative Capacity in Rats (감잎, 녹차, 솔잎의 건분 및 에탄올추출물이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.

Development of Granular Tea by Using Astringent Persimmon and Persimmon Leaves (떫은 감과 감잎을 이용한 과립차 개발에 관한 연구)

  • The purpose of this study was to find appropriate processing condition of astringent persimmon and persimmon leaf granular tea. Under the condition of 4 hour extraction and $50^{\circ}brix$, extraction yield and concentrating of astringent persimmon were the greatest. Under the condition of 2 hour extraction and $7^{\circ}brix$, extraction yield and concentrating of persimmon leaf were the greatest. The optimum condition of granular tea with astringent persimmon and persimmon leaf added was the best in astringent persimmon concentrate of 16% and persimmon leaf concentrate of 4%. It had the best taste after the condition in a brix value of 16.15% and a pH of 5.57. The L-value of chromaticity was -79.13, a-value 2.27, and b-value 2.41. In the sensory properties correlation analysis of astringent persimmon and persimmon leaf granular tea, sweet taste, color acceptability, odor acceptability and taste accept-ability were positively significant correlation with most sensory properties. However, astringent taste was no significant correlation with sensory acceptability. Bitter taste and after taste were negatively significant correlation with most sensory properties.
