감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level.   • The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. Salmonella typhimurium Strain TA98, 100에서 감잎차, 녹차, 우롱차 추출물의 돌연변이 억제 효과 (Antimutagenic Effects of Water Extracts of Persimmon Leaf Tea, Green Tea and Oolong Tea on Reversion and Survival of Selected Salmonella Tester Strains)

  • Water extracts of persimmon leaf tea(PLTE), green tea(GTE) and oolong tea(OTE), at the con centration used for human consumption, were examined for inhibitory effects on the mutagenicity of major classes of dietary and environmental mutagens including indirect acting mutagens, B[ ]P (benzo[ ]pyrene), IQ(2 amino 3 methylimidazo[4,5 f]quinoline), 2 AA(2 aminoanthracene) in the presence of S9 mix and direct acting mutagen, 4 NQO(4 nitroquinoline 1 oxide) without S9 mix, using the modified Ames Salmonella/microsome assay. PLTE, GTE and OTE showed very potent and concentration dependent antimutagenic effects against indirect acting mutagens B[ ]P and IQ. At the maximum concentration(16,200 g/plate) of each tea extract, number of colonies decreased in a dose dependent manner up to 82~100%. Similar inhibition of PLTE, GTE and OTE were seen at higher concentration in the mutagenicity of the 2 AA following an initial increase in the activity at lower concentration. However, the mutagenicity of the direct acting mutagen 4 NQO were not suppressed at lower concentration of the three tea extracts, and higher concentration of the tea extracts enhanced mutagenic activity of the mutagen. There were no differences in the mode of antimutagenesis between PLTE, GTE, and OTE, in both Salmonella typhimurium TA98 and TA100 strains against the same mutagen. In conclusion, the water extracts of persimmon leaf tea, green tea and oolong tea possess marked antimutagenic potential against a variety of important dietary and environmental indirect acting mutagens, but the activity was not observed against the direct acting mutagens. These results suggest that the mode of inhibitory action may not have resulted from direct interaction between tea extracts and the mutagens, but rather from indirect metabolic inactivation of mutagens by tea extracts.

마우스 소핵(Micronuclei, MN) 시험방법을 이용한 감잎차 추출물의 돌연변이 억제효과 (Antimutagenic Effects of Persimmon Leaf Tea Extract (PLTE) in Mice Using Micronucleus Induction (MN) Test)

  • The antimutagenic effects of persimmon leaf tea extract (PLTE) at concentration levels consumed by human were examined in mice using micronuleus induction with MMC(mitomycin C) or 4-NQO (4-nitroquinoline-1-oxide). When mice received oral gavage of 10 equivalent to PLTE 24 hr and 6 hr before, and 5 equivalent to PLTE 6 hr before and 3 hr after intraperitoneal injection of MMC, a significant decrease in the frequency of micronuclei were observed. The induction of micronuclei by 4-NQO was suppressed by oral dosage of PLTE at 5 equivalent to PLTE 6 hr before and 3 hr after, 10 equivalent to PLTE 3 hr before and 3 hr after intraperitoneal injection of MMC. Though the components of PLTE have not been analyzed so far, our present results suggest the existence of several bio-antimutagens and/or desmutagens in PLTE, beside catechin, well-known antimutagen.

감잎차, 녹차, 우롱차 추출물이 돌연변이 물질로 유발된 Sister Chromatid Exchanges 빈도에 미치는 영향 (Effects of Persimmon Leaf Tea Extract, Green Tea Extract and Oolong Tea Extract on the Frequencies of Mutagen-Induced Sister Chromatid Exchange in Chinese Hamster Ovary Cells)

  • 배양 CHO cell에 SCE법을 이용하여 실제로 음용하는 조건에서 추출한 시판 감잎차, 녹차, 우롱차 추출물의 돌연변이 억제효과를 보기 위하여 실험하였다. 돌연변이 물질로 사용한 MMC에 의하여 유발된 SCE 빈도에 미치는 각 차 추출물의 돌연변이 억제 효과를 본 결과, 감잎차는 MMC 처리 후 S9 mix와 함께 고농도$(1000\;{\mu}g/mL)$로 세포에 처리되었을 때 SCE 빈도를 유의하게 감소시켰다. S9 mix 없이 감잎차 추출물만을 후처리한 경우는 저농도$(20{\sim}80\;{\mu}g/mL)$에서 SCE 유발빈도를 유의하게 감소시켰다. 우롱차는 MMC처리 후 S9 mix와 함께 저농도$(10{\sim}20\;{\mu}g/mL)$로 처리 시 SCE 유발빈도를 감소시키는 효과가 있었으며, 녹차는 MMC 처리 후 S9 mix와 함께 추출물 농도 $160\;{\mu}g/mL$로 처리 시 SCE 유발빈도를 감소시키는 효과가 있었다. 감잎차, 녹차, 우롱차 모두 농도는 다르나 각 추출물을 S9 mix와 함께 세포분열 주기 중 G1기에 후 처리 되었을 때에 SCE 빈도를 감소시키는 효과가 있었으며, 감잎차는 S9 mix 없이 단독으로 후처리 되었을 때에도 용량 상관성은 없지만 SCE 빈도를 감소시키는 효과가 있었다. 결론적으로, 시판 감잎차, 녹차, 우롱차 추출물에는 MMC로 유발된 돌연변이를 억제시키는 효과가 있는 것을 확인할 수 있었으며, 감잎차의 경우 S9 mix 없이도 SCE 빈도를 감소시키는 효과가 있었던 것으로 보아 다른 두 차와는 다른 기전의 돌연변이 억제 작용을 하는 성분이 있는 것으로 사료된다.

단감 잎 추출물의 생리 활성 (Physiological activities of water extracts from sweet persimmon leaves)

  • 단감연구소에서 출하 시기 조절을 위해 육성된 중생종 단감 신품종 홍추와 기존 재배품종 상서조생, 서촌조생, 부유 3품종의 잎 추출물을 제조하여 생리활성을 비교하였다. 그 결과, 홍추의 잎 추출물의 총페놀 함량, Vit. C 함량, DPPH 및 ABTS 라디칼 소거능, ADH 활성에 미치는 영향이 다른 추출물에 비하여 유의하게 높다는 결과를 얻을 수 있었다. ADH 활성도는 부유의 잎 추출물에서 다른 품종에 비해 비교적 높게 나타났다. 이상의 결과는 신품종 단감 홍추의 잎을 이용한 고품질 감잎차의 제조가 가능함을 시사한다.

감잎의 광노화 피부 개선에 미치는 효과(1) (Effects of Persimmon leaf on the Photoaging Skin Improvement(1))

  • This study was performed to investigate the anti-photoaging effects of Persimmon leaf tea(PLT) in hairless mice(SKH-1) exposed to UVB irradiation. The animals were divided into non-treated group (normal, N) and UV-radiated groups. UV-radiated groups were divided into only UV-radiated group(control, C) and UV-radiated and PLT treated experimental groups[first extraction treated group(PLT-I), second extraction treated group(PLT-II), and third extraction treated group(PLT-III)]. Three PLT treated experimental groups of mice were treated with both oral administration(300 mg/Kg B.W./day) and topical application (100 ul of 2% conc./mouse/day) for 4 weeks. Anti-photoaging effects of Persimmon leaf were evaluated by anti oxidative reaction, stereomicroscopic and microscopic observations. The expression of photoaging skin related factors including mast cell tryptase, proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) was examined by immunohistochemical staining. Treatment of PLT-I, -II, -III prevented the wrinkle formation as well as epidermal hyperplasia, inflammatory cells, disruption of collagen in photoaged skin induced by UVB radiation. It also reduced the PCNA and VEGF expression in the UVB irradiated dorsal skin. Furthermore, it significantly decreased the number of mast cells in the UVB irradiated dermis(p<0.05 and p<0.01). On the effects of oxidative stress and antioxidant function on the treatment with water extract from Persimmon leaf tea(PLT), the activity of superoxide dismutase(SOD) was significantly increased in PLT-III group(p<0.05), and catalase(CAT) was significantly increased in PLT-I and PLT-III groups(p<0.05), and PLT-II group(p<0.001). These extracts showed relatively antioxidant activity and protective effect on UVB-induced oxidative stress in hairless mice(SKH-1). Our results suggest that Persimmon leaf tea may serve as an useful radical scavenging antioxidant and anti-photoaging skin agents in the UVB irradiated skin.

감잎차 추출액의 Sister Chromatid Exchange(SCE) 방법에 따른 항돌연변이 효과 (Antimutagenic Effects of Persimmon Leaf tea Extracts in Sister Chromatid Exchanges(SCE) Assay System)

  • 돌연변이 유발 물질인 mitomycin C(MMC)를 처리하여 배양한 Chinese hamster ovary(CHO) cell에 대한 감잎차 추출액의 항돌연변이 효과를 자매 염색 분체 교환(sister chromatid exchange, SCE) 시험법을 사용하여 측정하여 보았다. 감잎차 추출액 자체는 CHO 세포의 SCE 빈도수를 변화시키지 않았으며, 세포의 분열 주기중 S phase에 S9 mixture 없이 감잎차 추출액이 처리되었을 경우 MMC로 유도된 SCE 빈도수를 감소시키지 않았다. 그러나 S9 mixture 존재하에 $G_{1}$ phase에서 MMC 처리 후 감잎차를 처리하는 후처리 방식으로 감잎차 추출액을 처리하였을 때, 저농도($\leq$40$\mu\textrm{g}$/ml)에서 MMC로 인해 유발된 SCE 빈도수가 낮아지는 것을 볼 수 있었다. 이에 비해 고농도(>40$\mu\textrm{g}$/ml)에서는 SCE 빈도수의 감소 효과가 없었다. 본 연구결과, MMC 처리된 CHO 세포에 대한 감잎차 추출액의 항돌연변이 효과를 볼 수 있었고, 이 효과는 S9 mixture 존재하에서 저농도의 감잎차 추출액이 $G_{1}$ phase에 처리되었을 때 나타났다. 감잎차 추출액의 이러한 항돌연변이의 효과의 기전은 감잎차 추출액의 대사산물이 MMC 처리된 CHO 세포에 대한 DNA-excision repair activity를 촉진시키기 때문인 것으로 생각된다.

납투여한 흰쥐의 헴합성과 적혈구 중의 항산화효소 활성에 미치는 녹차, 감잎, 홍화 열수추출물의 영향 (Effect of Water Extract of Green tea, Persimmon Leaf and Safflower Seed on Heme Synthesis and Erythrocyte Antioxidant Enzyme Activities in Lead-Administered Rats)

  • 한국전통차로 알려진 녹차, 감잎 및 홍화의 열수추출물이 납투여된 흰쥐의 헴합성과 적혈구 중의 항산화효소 활성도 변화에 미치는 영향을 구명하기 위하여 체중 kg당 25 mg의 납을 매주 1회 경구투여 하였다. 녹차잎, 감잎 및 홍화 열수추출물은 매일 일정시간에 체중 kg당 1.26 g 수준이 되도록 4주간 경구투여하여 사육한 결과 헤마토크릿치, 헤모글로빈 함량 및 적혈구 수는 납 단독투여군이 정상군에 비하여 유의적인 감소를 보였으나 녹차, 감잎 및 홍화 열수추출물 급여로 감소 정도가 완화되었다. 혈액과 간조직 ALAD 중의 활성은 정상군에 비하여 납 단톡투여 군이 유의 적으로 감소되었으며, 한국전통차 열수 추출물 급여시 활성이 정상 수준 가까이 회복되었는데 그 효능은 혈액학적 성상 변화와 유사한 경향을 나타내었다 혈장 중 AST, ALT활성은 정상군에 비하여 납 단독투여군에서 유의적으로 증가되었으며 각각의 열수추출물 급여시 납에 의한 활성 증가가 현저하게 억제되어 정상수준으로 회복되었으며 한국전통차 종류에 따른 차이는 유의적이지는 않으나 감잎과 홍화가 효과적인 것으로 관찰되었다. 적혈구 중의 SOD와 CAT활성 및 GSH 함량은 납 단독투여시 유의적으로 증가되고 GSH-Px 활성은 감소된 반면, 녹차, 감잎 및 홍화 열수추출물 급여시 납으로 인한 적혈구의 항산화효소 활성변화가 완화되었다
