도열병에 감염된 벼의 엽조직에서 Peroxidase의 활성 (Peroxidase Activity in Leaf Tissue of Rice Infected by Pyricularia oryzae)

  • 수도품종 농백, 진흥, 낙동, 태백 등 4품종과 도열병균 KJ-101과 KJ-301의 2race를 사용하여 peroxidase 활성증가와 저항성과의 관계를 관찰하였다. peroxidase 활성은 수도유균가 성장할수록 모든 품종에서 증가되는 경향은 있었으나 품종간 차이는 크지 않았다. 도열병균을 접종 후 시간이 경과함에 따라 이병 잎에서는 무접종 건전잎보다 효소활성이 높았고 접종된 균주에 비친화적인 품종보다 발병이 심한 친화적 품종에서 더욱 높았다. 전기영동법에 의한 peroxidase pattern은 이병잎에서는 도열병균의 peroxidase band를 관찰할 수 없었다. 질소시비량을 증가시켰을 지라도 peroxidase 활성은 변함이 없었다.

Selective Homologous Expression of Recombinant Manganese Peroxidase Isozyme of Salt-Tolerant White-Rot Fungus Phlebia sp. MG-60, and Its Salt-Tolerance and Thermostability

  • Phlebia sp. MG-60 is the salt-tolerant, white-rot fungus which was isolated from a mangrove forest. This fungus expresses three kinds of manganese peroxidase (MGMnP) isozymes, MGMnP1, MGMnP2 and MGMnP3 in low nitrogen medium (LNM) or LNM containing NaCl. To date, there have been no reports on the biochemical salt-tolerance of these MnP isozymes due to the difficulty of purification. In present study, we established forced expression transformants of these three types of MnP isozymes. In addition, the fact that this fungus hardly produces native MnP in a high-nitrogen medium (HNM) was used to perform isozyme-selective expression and simple purification in HNM. The resulting MGMnPs showed high tolerance for NaCl compared with the MnP of Phanerochaete chrysosporium. It was worth noting that high concentration of NaCl (over 200 mM to 1200 mM) can enhance the activity of MGMnP1. Additionally, MGMnP1 showed relatively high thermo tolerance compared with other isozymes. MGMnPs may have evolved to adapt to chloride-rich environments, mangrove forest.

수도의 세포간극 물질 추출 (Extraction of Intercellular Material from Rice Leaf Tissue)

  • 본 연구는 세포간극물질을 추출코자 실시하였다. 2. Vacuum flask에 물을 넣은 후 볏잎을 물에 잠기게 하고, 280mb로 기압을 줄여 벼조직의 세포간극내의 공기를 뺀 후 Vacuum을 끊고 증류수로 세포간극을 채운다. 볏잎에 묻은 증류수를 제거하고 본 연구에서 사용한 leaf holder에 끼워 3,000rpm으로 5분간 원심분리하면 세포간극물질만 채취할 수 있다. 3. 세포내물질과 세포간극물질의 성분분석 및 Peroxidase와 Easterase의 isozyme pattern을 비교한 결과 내용성분의 함량이 서로 다르고 효소의 활성과 isozyme pattern이 다르게 나타나 세포간극물질 추출액 속에는 세포내물질이 혼입되어 있지 않음을 증명하였다.

Ganoderma lucidum의 계통분류(系統分類) (Classification of Strains of Ganoderma lucidum)

  • Ganoerma lucidum의 인공재배(人工栽培)로 얻은 자실체(子實體)의 형태적(形態摘) 특성(特性)을 기초(基礎)로 계통(系統)을 분류(分類)하고 전기영동특성(電氣泳動特性)과의 관련성(關聯性)을 검토하였다. 영지균은 버섯의 형태(形態)와 윤문(輪紋)의 유무에 따라 4계통(系統)으로 분류(分類)되었으며 그중 편각 윤문형(輪紋形)이 가장 많았고 전형적 녹각형은 소수였다. 자실체(子實體)의 모양 즉 편각과 녹각, 갓 표면(表面)의 윤문(輪紋) 유무(有無)는 유전성(遺傳性)이었다. 영지균사(菌絲) 및 자실체(子實體)의 전기영동(電氣泳動)에 의한 단백질(蛋白質), Peroxidase, Esterase 및 Leucine Amino Peptidase Isozyme Pattern을 조사(調査)한 결과(結果) 한국자생영지균(韓國自生靈芝菌)은 유전형질(遺傳形質)에 다양성(多樣性)을 보였다. 그중 Esterase Isozyme Pattern은 편각과 녹각간에 차이가 컸고 동(同) 계통(系統)간에는 높은 유사도(類似度)를 보여 계통분류(系統分類) 결과(結果)와 일치하였다.

Identification of Different Species and Dultivars of Brassica by SDS-PAGE, Isozyme and Molecular Marker

  • Eighty-five different cultivars of Brassica rapa, B. juncea, B. nap us, B. carinata, B. oleracea and hexaploid Brassica collected from Bangladesh, Japan, China and Denmark were analyzed by SDS-PAGE for seed and leaf protein variations, using esterase, acid phosphatase and peroxidase isozyme analysis. Ten polymorphic bands were identified from seed protein however no identifiable polymorphic band was found in the leaf protein. Polymorphic markers clearly distinguished the different Brassica species as well as yellow sarson (YS) and brown seeded (BS) cultivars of B. rapa. The $F_1$ cross between YS and brown seeded cultivars showed the existance of all poly-morphic bands of the respective parents. The Bangla-deshi and Japanese cultivars of B. rapa differed in the amount of seed protein. In the case of isozyme analysis, esterase showed the highest number of polymorphic bands (13) followed by acid phosphatase (9) and peroxidase (5). These polymorphic markers were very effec-tive for classification of all the species studied in this experiment. In parentage tests using isozymes, the hybridity of intra-and-interspecific crosses of almost all the seedlings could be identified from their respective cross combinations. Esterase polymorphism showed a clear differentiation between YS and BS types of B. rapa. In addition, two esterase polymorphic markers were iden ified to differentiate some cultivars of B. juncea. Segregation patterns in these two esterase bands showed a simple Mendelian monohybrid ratio of 3:1 in $F_2$, 1:1 in test cross and 1:0 in back cross progenies. No polymorphic band was identified to distinguish different cultivars of the same species by acid phosphatase or peroxidase. Polymerase Chain Reaction (PCR) was carried out with seed coat color specific marker of B. juncea. The yellow seeded cultivars produced a strong band at 0.5 kb and weak band 1.2 kb. In the addition of these two specific bands, Japanese yellow-seeded cultivars expressed two more weak bands at 1.0 kb and 1.1 kb. Where the brown seeded cultivars generated a single strong band at 1.1 kb. In segregating population, the yellow seed coat color marker segregated at a ratio 15 (brown) : 1 (yellow), indicating the digenic inheritance pattern of the trait.

종자발아에 있어서 Protein Profile과 Isozyme에 관한 연구 (Studies on Protein Profiles and Isozymes in Germinating Seeds)

  • The purpose of this experiment was to study one side of germination physiology based on that protein profiles and protease relating to protein metabolism, that peroxidase, catalase, $\alpha$-amylase, $\beta$-amylase, and malate dehydrogenase involved in the carbohydrate metabolism of seed germination. All these experiments were divided into the two groups with and without acetone treatment, and were carried out. The protein bands of each germinating stage between the groups treated with and without acetone showed certain basic pattern in polyacrylamide gel disc electrophoresis. However, there was a little difference in the number of protein band, optical density, and migration velocity between two groups. The isozyme bands of peroxidase, and catalase between two groups in polyacrylamide gel disc electrophoresis did not show the numeral difference, but the optical density of certain germinating stage treated with acetone was higher than the group untreated with it and it showed their enzyme activity. The $\alpha$-amylase and $\beta$-amylase activities which involved in starch metabolism of seed germination were higher in the treated group than the other. On one hand, the protease activity of hydrolase occurred in the seeds for germination was also higher, more or less in the treated group than in the other. The isozyme band pattern of malate dehydrogenase in TCA cycle of energy metabolism pathway was very different between two groups growing for 72 hours with and without acetone treatment in cellulose acetate electrophoresis. It indicated that two isozyme bands of malate dehydrogenase was high. Consequently these experimental results mentioned above indicated that acetone treatment before sowing had an effect on dissolving certain complexed lipid substance involved in the seed coats, the activity of carbohydrate hydrolase increased with water absorption which was most comfortable in its germination, dissolved glycerin and fatty acid became certain energy source, and they stimulated the acceleration of respiration metabolism.

  • In an attempt to clarify the physiological functions of individual isoperoxidases, we have studied enzymatic and immunological properties as well as cellular distribution of isoperoxidases from tobacco callus and Korean radish. The gene expression patterns of isoperoxidases in shoot and non-shoot-forming tobbaco callus were also examined by rabbit reticulocyte lysatein vitro translation system. These results indicate that fraction of translatable poly(A)-isoperoxidase mRNA was increased considerably in shoots. At the present time, at least 6-7 isoperoxidases could be detected from the translation mixture of total cellular RNA, among which only one cell wall localized anodic isoperoxidase (named A3) mRNA was bimorphic mRNA. These data suggest the possible regulation of peroxidase activity during shoot formation by altering the polyadenylation state of mRNA. In case of Korean radish seedlings, poly(A)- peroxidase mRNA were also increased depending upon aging.

Effect of Metals on Anti-Oxidase Activity and Isozyme patterns in Brassica juncea

  • To study the effects of metal ions on the activity of anti-oxidase enzymes, the activity of superoxide dismutase (SOD) and peroxidase (POD) and isozyme patterns of Brassica juncea have been studied after treating with CD, Cu, Zn, and Al. The activity of SOD after treating with metal ions was higher than that of untreated control. SOD activity in leaves increased by treatment of 50 ppm of Zn and 500 ppm of Al. POD in stems gave highest activity after treating with 500 ppm of Cu. When the activity was compared by plant parts, lowest POD activity was observed in leaves in which protein content was higher than other tissues. When the activity was expressed as percentage of control, SOD activity was increased after treating with metal ions. SOD activity in leaves and roots of metal treated plant was significantly increased under the metal ions stress conditions. In the roots of 50 ppm of Zn treated plant, SOD activity was extremly high. POD activity was inhibited with Cd and Zn treatment in all parts of the plant. However, in leaves and stems, there was marked increase in activity after treating with Cu. The patterns of SOD isozyme after metal treatment show that two bands were stained in all metal ion treated and that no new band appeared. POD isozyme band intensity resulting from the treatment of metal ions was in order of roots > stems > leaves, but there was no significant difference.

동위효소 분석에 의한 느타리속의 종간 유연관계 (Interspecific Relationships within the Fungal Genus Pleurotus by Isozyme Analysis)

  • 국내외에서 수집된 Pleurotus속 13종 32균주와 종(species)이 밝혀지지 않은 4균주 등 모두 36균주의 동위효소 분석에 의한 종간 유연관계를 파악하기 위하여 본 실험을 수행하였다. Pleurotus속 균주들의 균사체에서 수용성 단백질을 추출하여 등전점 전기영동 isozyme polymorphic banding pattern법으로 여섯 종류의 동위효소 즉 esterase, glucosephosphate isomerase, leucine aminopeptidase, malate dehdrogenase, peroxidase, phosphoglucomutase를 분석하여 총 166개 밴드를 확인하였고 36개 느타리 균주의 종간 다형성을 관찰하였다. Esterase가 종간에 가장 다양한 밴드 pattern을 나타내어 종의 구별에 아주 유용한 동위효소라 생각되며, 다른 동위효소에서도 종간 다형성을 관찰할 수 있었다. 동위효소 분석 결과 유연관계가 가장 가까운 종은 P. florida와 P. sajor-caju로 유사도가 약 89%였으며, 연구에 이용된 총 13종간의 유사도는 약 77%였다. 분류상 논란이 많이 되는 P. ostreatus는 P. pulmonarius 종과 확실히 구별되었으며 P. florida도 독립된 종으로 판명되었다. 그러나 P. sapidus와 P. spodoleucus 균주들은 확실한 종의 판명이 어려웠다. Isozyme polymorphic banding pattern 결과와 현미경적인 형태 분류에 의한 종 판명 결과가 몇 종을 제의하고는 일치하였다. 동위효소를 이용한 분류는 매우 유용하다고 판단되었다.

Fusarium속 내의 section Elegans, section Liseola와 유사종의 Isozyme Patterns (Isozyme patterns of section Elegans, section Liseola and similar species in the genus Fusarium)

  • Fusarium 속내 종들의 분류학적 위치를 알아보기 위하여, 각 균의 $esterase-{\alpha},\;-{\beta},\;acid\;phosphatase,\;malate\;dehydrogenase,\;peroxidase,\;polygalacturonase$를 추출하여 전기영동의 방법으로 isozyme patterns를 비교하여 보았다. Banding patterns은 polygalacturonase만이 7종 모두에서 monozyme이었고, 나머지 효소는 종마다 약간씩 다르게 나타났다. Isozyme banding patterns을 바탕으로 genetic similarity를 산출하여 dendrogram으로 보았을 때, 동일한 section Liseola에 속하는 F. subglutinans와 F. moniliforme의 종간 유사도는 74.3%로 가장 높았으며, section Elegans와 section Liseola의 서로 다른 section 간의 유사도는 45.4%로 비교적 낮았다. F. napiforme와 F. nygamai의 유사도는 64.7%로 동일 section 내의 종간 유사도와 비슷한 수치를 나타내었다. 이들 두 종은 section Liseola에 대하여 55.2%, section Elegans에 대하여 45.4%의 유사도를 보여 분류학적 위치에 있어서 section Liseola에 더 가까운 경향을 나타내지만, 이는 다른 section 간의 유사도와 비슷한 수치로 하나의 독립된 section으로 추정된다. F. graminearum과 다른 6종 사이의 유사도는 28.2%의 낮은 수치를 나타내어 서로의 유사성이 매우 낮다는 것을 보여 주었다.
