Hydrogen Permeation of SiC-CeO2 Composite Membrane by Dip-coating Process

  • A SiC-$CeO_2$ composite membrane was successfully fabricated using an ally-hydridopolycarbosilane (AHPCS) binder and treated by dip-coating at 60 times with a $CeO_2$ sol solution. The dip-coated SiC membrane was calcined at 773 K and then sintered at 1173 K under an air atmosphere. The coated membrane was characterized by X-ray diffraction (XRD), field-emission scanning electron microscopy (FE-SEM) and a BET surface analysis. The difference in permeation performance between $H_2$ and CO gases was measured by varying the temperature. The permeation flux of $H_2$ on the SiC membrane with layered $CeO_2$ was obtained as $8.45{\times}10^{-6}\;mol/m^2sPa$ at room temperature. The CO permeation flux was $2.64{\times}10^{-6}\;mol/m^2sPa$ at room temperature. The reaction enthalpy (${\Delta}H^{\circ}$) for the hydrogen permeation process was calculated as -7.82 J/mol by Arrhenius plots.

The Effect of Vehicles and Pressure Sensitive Adhesives on the Percutaneous Absorption of Quercetin through the Hairless Mouse Skin

  • To investigate the feasibility of developing a new quercetin transdermal system, a preformulation study was carried out. Therefore, the effects of vehicles and pressure-sensitive adhesives (PSA) on the in vitro permeation of quercetin across dorsal hairless mouse skin were studied. Among vehicles used, propylene glycol monocaprylate (PGMC) and propylene glycol mono-laurate were found to have relatively high permeation flux from solution formulation (i.e., the permeation fluxes were 17.25$\pm$1.96 and 9.60$\pm$3.87 $\mu\textrm{g}$/$\textrm{cm}^2$/h, respectively). The release rate from PSA formulations followed a matrix-controlled diffusion model and was mainly affected by the amount of PSA and drug loaded. The overall permeation fluxes from PSA formulations were less than 0.30 $\mu\textrm{g}$/$\textrm{cm}^2$/h, which were significantly lower compared to those obtained from solution formulations. The lower permeation fluxes may be due to the decrease of solubility and diffusivity of quercetin in the PSA layer, considering the fact that the highest flux of 0.26 $\mu\textrm{g}$/$\textrm{cm}^2$/h was obtained with the addition of 0.2% butylated hydroxyanisole in PGMC-diethyl-ene glycol monoethyl ether co-solvents (80-85 : 15-20, v/v). Taken together, these observations indicate that improvement in the solubility and diffusivity of quercetin is necessary to realize fully the clinically applicable transdermal delivery system for the drug.

Permeation Characteristics of the Tubular Membrane with Continuous Air Cleaning System (연속식 공기세정 관형막 투과특성)

  • This study was carried out for microfiltration tubular membrane module equipped with self-designed air injection nozzle in order to determine the permeate flux due to the effect of membrane fouling reduction. The 0.1 wt% yeast particle solution was used as a feed solution and permeation tests were performed for the cases with and without air injection. Permeation fluxes were measured and analyzed to examine the effect of membrane fouling reduction. While the permeation flux without air injection decreased continuously, that with air injection was improved more than 30 percent than that of no air injection case.

  • Artemisiae Capillaris Herba is a dried aerial part of Asteraceae capillaris Thunb.(Compositae), which has been used in Korean traditional medicine for the treatment of various diseases. It has a variety of pharmacological activities and has been evaluated for potential as an active ingredient in cosmeceutical products. In the cosmetics industry, animal experiments is besides the major concern of ethics, there are few more disadvantages of animal experimentation like demand of skilled manpower, time consuming protocols and high cost. Therefore, various alternatives to animal experiments have been proposed. The purpose of this study was to investigate the skin permeation characteristics of chlorogenic acid and dimethyleculetin, which are constituent of Artemisiae Capillaris Herba by using Franz diffusion cell. As a result, skin permeability was characterized by flux(penetration rates) and $K_p$(permeability coefficient) value, chlorogenic acid had lower flux and $K_p$ than dimethylesculetin. According to the definitions of Marzulli, chlorogenic acid and dimethylesculetin would be classified as 'Moderate' and 'Very fast' respectively. In conclusion, skin permeation characteristics of chlorogenic acid and dimethylesculetin were confirmed through Franz diffusion cell, and suggests the direction of alternative method for skin permeation of natural compounds.

Permeation and Enzymatic Degradation of Aspalatone in Gastrointestinal Tract of Rabbit (아스팔라톤의 토끼 위장관 점막 투과 및 효소적 분해)

  • To evaluate the site-specific permeation of aspalatone (acetylsalicylic acid maltol ester, AM) through gastrointestinal tract, the enzymatic degradation and permeation studies were carried out using gastric, duodenal and jejunal mucosae of rabbits. It was found that $15.2{\pm}11.4%$, $11.6{\pm}5.2$ and $0.8{\pm}0.6%$ of the donor dose of AM, salicylmaltol (SM) and aspirin (ASA) permeated through the upper gastric mucosa after 8 hr of permeation, respectively. After 8 hr of AM permeation, SM and ASA were measured to be $15.0{\pm}1.7$ and $2.6{\pm}0.8%$ of the dose in the donor solutions, respectively, and salicylic acid (SA) was not detected even after 6 hr, suggesting a very low gastric damage. For the gastric mucosa, the increase of donor dose from 100 to $1,000\;{\mu}g/ml$ increased the permeation flux dose-dependently (r=0.9905). For the duodenal and jejunal mucosae, however, AM was fully degraded into SM and SA due to the esterase activities within 30 min. AM and ASA were not detected in the receptor solution. This result indicates that AM is not a prodrug of ASA. Addition of potassium fluoride (0.5%) into the donor solution delayed the degradation of AM, but did not allow the permeation through duodenal mucosa even by the inhibition of esterase activity. The addition of $dimethyl-{\beta}-cyclodextrin$ and $2-hydroxypropyl-{\beta}-cyclodextrin$ (5%) into the donor solutions also did not show favorable effects on the permeation of AM through various mucosae. In comparison of permeation rates of AM and ASA through the upper gastric mucosa, the flux of ASA was 4.2 times faster than AM based on the molar concentration. ASA also was fully degraded in the donor solutions faced with duodenal and jejunal mucosae within 2 hr, and was not detected in the receptor solution, suggesting a slower metabolism compared with AM.

  • Radiation-induced grafting of 2-hydroxyethyl methacrylate(HEMA), acrylic acid(AAc) and methacrylic acid(MAAc) onto polypropylene microfiltration membrane has been studied. The effect of grafting conditions such as solvent composition(MeOH and $H_2O$) and monomer concentration on the grafting yield in investigated. The highest degree of grafting is obtained at a solvent composition of 25% $H_2O$:75% MeOH for HEMA, pure water for AAc and 50% $H_2O$:50% MeOH for MAAc. Modification of the PP membranes with hydrophilic monomers is shown to cause an increase in the water permeation flux of the membranes. It is found that HEMA is the best monomer to increase the water permeation flux and the highest water permeation flux is obtained at 99% degree of grafting. The water permeation flux of AAc-grafted PP membrane and MAAc-grafted PP membrane is very sensitive to environmental pH and $Cu^{2+}$ ion, but the water permeation flux of HEMA-grafted PP membrane scarcely depends on pH and $Cu^{2+}$ ion.

  • Microporous polypropylene (PP) membranes have the high chemical and corrosion resistance, the good mechanical properties and the thermal stability under high temperatures, but its application is restricted within narrow limits due to hydrophobicity of membranes. In order to impart permanent hydrophilicity to the PP microfiltration membrane, the radiation-induced graft of 2-hydroxyethyl methacrylate (HEMA) and acrylic acid (AAc) containing hydrophilic functional group onto the membrane has been studied. The effect of graft conditions such as reaction time, total radiation dose, reaction temperatures, acid compositions on graft yield was investigated. Modified PP membranes were shown to cause an increase in the gas flux. Oil emulsion permeation flux of both original PP membrane and modified PP membrane was examined.

Evaluations of Coagulation Process for Membrane Pre-treatment using Floc Growth Rate Analyzer (응집 플록 성장률 측정기를 이용한 멤브레인 공정의 전처리 응집공정 평가)

  • In this study, we have investigated to find optimal pre-treatment flocculation condition by analyzing the floc growth rate with mixing conditions and the membrane permeation flux for pre-treatment step of the membrane process. The higher mixing intensity showed a constant floc size index (FSI) values, and lower mixing intensity increased the degree of dispersion of the FSI values. Results of comparing the distribution characteristics of the FSI value and the permeation flux were more effective in increasing flux when the FSI values were 0.2 or higher. The degree of dispersion of FSI was relatively large in 40 rpm mixing condition compared to 120 rpm. In 40 rpm mixing condition, it decreased the permeation flux compared to 120 rpm because various sizes of flocs were distributed. Coagulation-UF membrane process enhanced 30%~40% of the flux rate compare to UF alone process, and the coagulation-MF process increased up to 5% of the flux rate compare to MF alone process. Pre-treatment, that is, coagulation process, has been found to be less effects on relatively larger pore size for MF membrane. For UF membrane, the flux was a little bit same when applying only the rapid mixing process or rapid mixing with slow mixing processes together. In case of MF membrane, the flux was improved when rapid mixing process applied with slow mixing process together.

Formulation of Caffeine Nasal Sprays and Its Enhanced Permeation through Rabbit Nasal Mucosa (카페인의 비강 분무액의 제제설계 및 점막 투과 증진)

  • This study was aimed to investigate the feasibility of nasal delivery of caffeine for the elimination of sleepiness. The effects of various vehicles, solubilizers, and enhancers on the permeation of caffeine through rabbit nasal mucosa was observed. The permeation study was carried out using a Franz-type permeation system at $37^{\circ}C$, and the amount of caffeine permeated though the rabbit nasal mucosa was determined by a validated HPLC. The apparent solubility and phys iochemical stability of caffeine in various nasal formulations were was determined. The effect of hydrotropes and modified cyclodextrins on the solubility of caffeine in water was determined by equilibrium solubility method. The solubility of caffeine in water was 29 mg/mL at $30^{\circ}C$. The addition of sodium benzoate and nicotinamide at 10% improved the solubility of caffeine (115 and 132 mg/mL, respectively) in aqueous solution. The flux of caffeine though the nasal mucosa from aqueous solution was $2.1{\pm}0.26\;mg/cm^2/hr$. The addition of sodium benzoate reduced its permeation $(1.4{\pm}0.01\;mg/cm^2/hr)$, but sodium benzoate with 5% $2HP{\beta}CD$ and 0.03% monoterpenes increased its permeation $(2.4{\pm}0.04\;mg/cm^2/hr)$ markedly. The addition of nicotinamide also increased also increased its permeation $(2.5{\pm}0.36\;mg/cm^2/hr)$. markedly. As the concentration of caffeine in nasal formulation increased, the permeation flux increased linearly. Caffeine was stable physicochemically and enzymatically in the nasal mucosa extract at $37^{\circ}C$. These results suggest that caffeine can be efficiently delivered nasally and the development of nasal formulation will be feasible.

  • To increase skin permeability of LMWH (Low Molecular Weight Heparin), ultradeformable liposomes were developed. Ultradeformable liposomes were developed by Egg phosphatidylcholine (Egg-PC) and edge activator. Entrapment efficiency, vesicle size and zeta potential of vesicles were determined and characterized for deformability and stability. Transepidermal permeation of LMWH was compared to saturated aqueous control in vitro. The steady-state flux and its maximum time were calculated from the flux curves. (omitted)
