  • The polysaccharide fraction from the rhizome of Trichosanthes kirilowii (Cu-curbitaceae) showed marked antitumor and cytotoxic activity with immunopotentiating activity. It was evidenced by the increase in the number of circulating leucocytes and peritoneal exudate cells and the recovery of lowered antibody forming activity in mice. The polysaccharide was mainly composed of glucose, galactose, fructose, manmose and xylose and a small amount of protein.

Studies on the Activities of Peritoneal Macrophages Induced by Yookmijihwangtang, Palmijihwangtang and Gamijihwangtang (육미지황탕(六味地黃湯), 팔미지황탕(八味地黃湯) 및 가미지황탕(加味地黃湯)이 생쥐의 복강대식세포(腹腔大食細胞) 활성(活性)에 미치는 영향(影響))

  • In order to investigate the effects of YOOKMIJIHWANGTANG, GAMIJIHWANGTANG and PALMIJIHWANGTANG on the various immune responses, the chemotactic, adherent, nitric oxide release ability of macrophages were studied in vivo and in vitro. The results obtained were follows : 1. The total number of peritoneal exudate cells from mice injected with three drugs were 2~3 times more than those from the PBS. 2. The administration of three drugs enhanced significantly the Fc receptor mediated activity(phagocytosis, rosette formation activity). 3. The administration of three drugs enhanced significantly the chemotactic activity of peritoneal macrophages. 4. The administration of three drugs enhanced significantly the adherent activity of macrophages and neutrophils. 5. The administration of GAMIJIHWANGTANG and PALMIJIHWANGTANG enhanced significantly the chemotatic activity of macrophages (p<0.01). And also the administration of YOOKMIJIHWANGTANG enhanced significantly the chemotatic activity of macrophages(p<0.05). 6. The administration of GAMIJIHWANGTANG and enhanced significantly the chemotatic activity of neutrophils(p<0.01). And also the administration of YOOKMIJIHWANGTANG and PALMIJIHWANGTANG enhanced significantly the chemotatic activity of neutrophils(p<0.05). From above findings, it is suggested that YOOKMIJDTWANGTANG, GAMIJIHWANGTANG and PALMIJIHWANGTANG seems to produce the increase of immune response such as the Fc receptor activity, chemotactic activity, adherent activity, nitrate release of macrophages. From the above results, the GAMIJIHWANGTANG among three drugs may be the most useful drug having immunostimulating effects.

  • The effects of peritoneal exudate cells(PEC) and sera of athymic nude and DS mice infected with Clonorchis sinensis metacercariae or sensitized by injection of metabolic products into footpad on transfer of immunity against the fluke to the syngeneic mice were studied. There was no significant difference in eggs per gram pattern between the sensitized and control groups, and between nude and DS mice. However, the worm burdens were slightly greater in nude mice than in DS mice. Also, a few plaque forming cells were found in only DS mice given PEC and serum from Group II DS mice. In the light of these results, it is likely that PEC and sera of nude or DS mice which are deficient, at least partially, in the cellular immune system are unable to transfer immunity against C. sinensis to syngeneic recipients.

Some In-Vitro and In-Vivo Biological Activities of Hot Water Extracts from Fruit Body and Cultured Mycelium of Hericium erinaceum (Hericium erinaceum 균사체와 자실체 열수 추출물의 몇몇 In-Vitro 및 In-Vivo 생물활성)

  • The water-soluble materials extracted from fruit bodies and mycelium of H. erinaceum were prepared. In-vitro anticancer activities on cancer cells and In-vivo proliferation effect on mouse peritoneal exudate cell and spleen cell of samples were investigated. Also, nitric oxide (NO) generation of peritoneal exudate cell, IL-2 production capacity of spleen cells and phagocytic activity of peritoneal macrophages were examined. The water extracts of H. erinaceum suppressed the proliferation of cancer cell (HeLa, Raw264.7, Jurkat, KATO3, EL4, LyD9) with concentration-dependent. The water extract from fruit body showed better suppression effect than that from mycelium in most of cancer cells used. The anticancer effect of water extract of fruits body in the range of 0.01 and 10 mg/ml for Raw 264.7 and EL4 cell lines were the same as the Taxol with one thousandth equivalent of fruit body concentration. Water extracts of fruit body and liquid-cultured products of H. erinaceum induced nitric oxide (NO) generation of peritoneal exudate cell and increased NO generation by stimulus of lipopolysaccharide. Water extracts alone did not induce the proliferation and IL-2 production capacity of spleen cells. However, spleen's proliferation and IL-2 production were induced significantly by the addition of lipopolysaccharide and Con A (concanavalin A) or Con A alone, and the effectiveness of mycelium extract with water were more active than those from fruit body.

  • To produce and characterize antineoplastic constituents in the submerged cultured­mycelia of Laccaria laccata, the mycelia were extracted with distilled water. Purification of the extract was carried out by acetone precipitation, by ion exchange chromatography using DEAE­Sephadex A-50, CM-Sephadex C-25 resins, and by gel filtration chromatography on Sephadex G-200. Each fraction obtained during the purification was examined for antineoplastic activity against sarcoma 180 in ICR mice. As the purification proceeded, the antineoplastic activity was markedly increased. The highly purified Fraction E showed 75% tumor inhibition ratio at a dose of 10mg/kg/day and contained 81% polysaccharide and 4% protein. The antitumor component of Fraction E stimulated an accumulation of peritoneal exudate cells including peritoneal macrophages, and is named laccaran.

Nonsaponin fraction of Korean Red Ginseng attenuates cytokine production via inhibition of TLR4 expression

  • Background: Ginsenosides of Korean Red Ginseng extracts (RGE) and its saponin components suppress secretion of inflammasome-mediating cytokines, whereas the nonsaponin fraction (NS) of RGE oppositely stimulates cytokine secretion. Although direct exposure of NS to macrophages in mice induces cytokine production, oral administration of NS has not been studied in inflammasome-related disease in animal models. Methods: Mice were fed RGE or NS for 7 days and then developed peritonitis. Peritoneal cytokines were measured, and peritoneal exudate cells (PECs) were collected to assay expression levels of a set of toll-like receptors (TLRs) and cytokines in response to NS ingestion. In addition, the role of intestinal bacteria in NS-fed mice was assessed. The effect of preexposure to NS in bone marrow-derived macrophages (BMDMs) on cytokine production was further confirmed. Results: NS ingestion attenuated secretion of peritoneal cytokines resulting from peritonitis. In addition, the isolated PECs from NS-fed mice presented lower TLR transcription levels than PECs from control diet-fed mice. BMDMs treated with NS showed downregulation of TLR4 mRNA and protein expression, which was mediated by the $TLR4-MyD88-NF{\kappa}B$ signal pathway. BMDMs pretreated with NS produced less cytokines in response to TLR4 ligands. Conclusion: NS administration directly inhibits TLR4 expression in inflammatory cells such as macrophages, thereby reducing secretion of cytokines during peritonitis.

  • This report describes a case of postpartum uterine rupture associated with retained placenta and sequential acute metritis, not dystocia. A 14-month-old Bichon Frise bitch presented with continuous vaginal bleeding. She delivered five puppies 3 days ago and expelled four placentas on the next day. After then, anorexia, acute depression with continuous vaginal haemorrhage developed. Plain radiography showed decreased serosal detail, and ultrasonography showed irregularly thickened uterine wall and ascites. Blood-tinged peritoneal fluid was an exudate with bacteria. Ovariohysterectomy was curative for the patient, and four full-thickness perforated holes were identified on both sides of the uterine horns.

Studies on the Effects of the Saponin Fraction of Panax Ginseng on the Immune System of the Mouse Fed with Protein Malnutritive Diet (인삼사포닌 분획이 단백부족 마우스의 면역체계에 미치는 영향)

  • The thymus weight of the mouse was 54.1% in protein malnutritive diet group and 39.2% in group treated with saponin fraction of Panax ginseng in comparison to normal diet group. This decreasing effects of protein malnutritive diet and saponin fraction on the thymus weight practically disappeared after four weeks. The saponin fraction showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total peritoneal exudate cells, content of total serum protein and albumin content of the mouse, each 45, 8 and 10% respectively in comparision to that of normal diet group. And these values in protein malnutritive diet group were 61.2, 83.6 and 87.0% respectively in comparision to that of normal diet group, and recovered to the level of normal diet group by the supplement of the saponin fraction. The electrophoregram of the serum protein of the mouse fed with protein malnutritive diet was different from that of the mouse fed with normal diet, but this difference practically disappeared by the supplement of the saponin fraction.

Antitumor Effects of Glycoportein Extracted from Sea Cucumber (Stichopus japonicus)

  • The anititumor and immunologic activities of the glycoproteins extracted from sea cucumber (Stichopus japonicus) on mice bearing sarcoma 180 cells were investigated . Maximum tumor suppression (64%) occurred at the dose of 100mg glycoprotein/kg. The highest prolongation ratio was achieved at the level of 100mg/kg an dincreased by 395 more than that of control. Glycoproteins from sea cucumber exhibited direct cytotoxic effect on the tumor cells. Dose dependent increase of leucocyte, peritoneal exudate cell and weights of immunoorgans revealed the improvement of immunity. When the glycoportein-administered group was compared with the control, a significant difference was not noted in the clinico-chemical values such as S-GOT, S-GPT , alkaline phoshatse activity, total protein, cholesterol, triglyceride, urea nitrogen and glucose levels in blood. These results suggests that the antitumor activity of sea cucumber glycoprotein is associated with activation of cells in the immune system.

  • Polysaccharide fractions were prepared from Forsythia Corea by several different extraction schedules. The fractions obtained were designated as ForCo A, ForCo B and ForCo C respectively. ForCo C was further purified through Sephadex G 200 column chromatography and obtianed two subfractions (Coreana A, Coreana B). ForCo showed marked antitumor activity against sarooma 180 and its activity was dose-dependent. Coreana A, purified from ForCo C, showed somewhat higher antitumor activity. ForCo C increased the number of circulating leucocytes and peritoneal exudate cells, but didn't show any significant effects on the phagocytic activity and total serum protein level. Chemical analysis showed that ForCo C was composed of glucose, galactose, xylose and arabinose. A close relationship between antitumor activities and polysaccharide contents waw observed. These results indicate strongly that antitumor active principle of Forsythia Corea is polysaccharide.
