Calcium Absorption and Growth Characteristics of Agrocybe cylindracea Mycelia in Submerged Culture (액체배양 버들송이 균사체의 칼슘흡수 및 생육특성)

  • The present study examined the effects of different calcium (Ca) sources and concentrations on the growth and Ca absorption of Agrocybe cylindracea mycelia grown in submerged cultures. The dry weights of the mycelia were not significantly different (significance level of 5%) according to the type of Ca added, and increased with increasing Ca concentration until 500 mg/L, and then decreased at concentrations of 1000 mg/L or greater. The Ca contents of groups were significantly different according to the various concentrations of the Ca source, in which the Ca content of the control group cultured without added Ca was 198.3 mg/kg, and in the treatment groups, Ca content increased to a minimum of 273.7 mg/kg (1.4 times) and a maximum of 67246.0 mg/kg (339.1 times) the Ca contents of the groups generally increased with increasing Ca concentration. According to the number of culture days, growth rates were highest during days 8 through 12, and remained relatively high until day 16. In addition, Ca contents per unit dry weight were higher in young mycelia with a shorter culture period than in mature mycelia with a longer culture period. According to pH, the most active growth and highest Ca content occurred in MCM liquid medium at pH 7.0. In conclusion, in order to produce Agrocybe cylindracea mycelia with high Ca content, it is considered most efficient to culture them in MCM liquid medium without a pH adjustment and containing 1,000 mg/L of Ca-lactate, which is commonly used as a Ca additive in food, as well as to use mycelia between 12-16 days of culturing.

Curcumin Inhibits Expression of Inhibitor of DNA Binding 1 in PC3 Cells and Xenografts

  • Inhibitor of DNA binding 1 (Id1) plays an important role in genesis and metastatic progression of prostate cancer. We previously reported that down regulation of Id1 by small interfering RNA could inhibit the proliferation of PC3 cells and growth of its xenografted tumors. Curcumin, the active ingredient of turmeric, has shown anti-cancer properties via modulation of a number of different molecular regulators. Here we investigated whether Id1 might be involved in the anti-cancer effects of curcumin in vivo and in vitro. We firstly confirmed that curcumin inhibited cell viability in a dose-dependent fashion, and induced apoptosis in PC3 cells, associated with significant decrease in the mRNA and protein expression of Id1. Similar effects of curcumin were observed in tumors of the PC3 xenografted mouse model with introperitoneal injection of curcumin once a day for one month. Tumor growth in mice was obviously suppressed by curcumin during the period of 24 to 30 days. Both mRNA and protein levels of Id1 were significantly down-regulated in xenografted tumors. Our findings point to a novel molecular pathway for curcumin anti-cancer effects. Curcumin may be used as an Id1 inhibitor to modulate Id1 expression.

Chronic Wound Treatment Using rh PDGF (재조합 인체 혈소판 성장인자(rh PDGF)를 이용한 만성 창상의 치료)

  • Wound healing is the result of interaction of normal cellular and biochemical responses that restore the interrupted anatomical structure in limited period. When any response of them is impaired, it results in chronic wound. The factors that influence the wound healing process is not only limited to the fundamental disease of the individual but also the local factors, especially various growth factors secreted from the various cells involved in tissue regeneration have important role. Recent reports that the chronic wounds are depleted of these growth factors have led active studies on the alteration of local wound environment with manipulation of the growth factors and the its application in management of chronic wounds. We investigated the effect on the chronic wounds in 10 patients with various pathologic conditions to suggest the appropriate application and guideline of the indication. The chronic wounds resulting from various causes in 10 patients were treated with rhPDGF gel and good wound care. All the chronic wounds were located on the lower extremity and the average diameter was 2.5 cm. 7 patients were completely cured within 8 weeks, however the patient who received previous radiation therapy the healing was delayed to 14 weeks. Two patients with vascular ulcer were not cured with rhPDGF alone. Local application of rhPDGF has yielded complete cure of the chronic wound in 70% of the patients within 2 months. The author suggests that it would be an effective alternative treatment modality of chronic wound when it is applied with good wound care and appropriate indication.

  • The purpose of this study was to find out some relationship between the positional change of mandible during treatment and the prognosis after treatment of ClassIII malocclusion. The patients selected for this study were sucessfully treated two patients, and three patients who wert also sucessful in active treatment but showed relapse during observation. Serial Cephalograms, photos, models, treatment record of these patients were analysed in each term of treatment and observation period. The results might be summarized as follows: The patient with swing back type in which mandible rotated posteroinferiorly during active treatment showed good dental md skeletal relationship during observation. Among the patients with Y-axis type, one who had harmonious growth between maxilla and mandible showed stable dental relationship during observation, the other who had not showed relapse. The swing back type was not changed during and after treatment but the patients who had Y-axis type were unchanged in some patients, and changed into forward type in the others. During active treatment, was lower facial height(ANS-Me) increased in all 5 patients, and in observation period the patients showing increase in lower facial height had stable occlusion but the others having decrease showed unstable dental relationship.

Study on the feasibility of metallic saggar for synthesizing NCM cathode active materials-I (NCM 계 양극활물질 합성용 금속질 내화갑 가능성 연구-I)

  • In this study, nickel, a pure metal material, was proposed as a saggar for synthesizing NCM [Li(NixCoyMnz)O2] cathode active material. Nickel is known as a metal that is resistant to oxidation and has a high melting point. Nickel is one of the main components of NCM cathode material and was expected to be free from problems with contamination from saggar during cathode material synthesis. We sought to confirm the possibility of nickel as a saggar for synthesizing NCM cathode active materials. When a Ni metal crucible and Ni0.8Co0.1Mn0.1(OH)2 (NCM 811) precursor material were reacted at 900℃ for a long time, the change in the reaction layer on the surface of the crucible over time was analyzed. The nickel crucible reaction layer formed during heat treatment at 900℃ was nickel oxide, and is thought to have been created by simultaneous oxygen diffusion from the cathode precursor oxide and reaction with oxygen in the atmosphere. The change in thickness of the oxide layer appears to slow down after 480 hours, which suggests that the rate of oxygen diffusion from the precursor is reduced. It remained combined without falling out of the crucible until 480 hours. However, it was confirmed that the oxide layer falls off after 720 hours, so it is thought that it can be used as saggar for NCM synthesis only for a certain period of time.

Behavioral characteristics of Hanwoo (Bos taurus coreanae) steers at different growth stages and seasons

  • Objective: This research analyzed behavioral characteristics of Hanwoo (Bos taurus coreanae) steers during each season and growth stage to enable measurement of the animals' welfare level for precision livestock farming. Methods: A hundred-eight beef steers were divided into three equal groups at a Hanwoo farm according to their growth stage: growing stage (GS), 8 months; early-fattening stage (EFS), 19 months; and late-fattening stage (LFS), 30 months. Twelve behavioral categories were continuously recorded for 13 day-time hours in each four seasons with three replications. Results: Time spent standing was found to be significantly longer in summer at all growth stages (p<0.05). Hanwoos at the GS spent significantly longer standing time in spring and summer than those at the EFS and LFS (p<0.05). Lying time in summer was the shortest for all growth stages (p<0.05). Steers at the LFS spent significantly longer lying time than that at the GS (p<0.05) in summer. For GS and EFS, time spent eating in spring and autumn were longer than in summer and winter (p<0.05). Eating time was the longest for the GS in spring, autumn, and winter, excluding for the LFS in winter (p<0.05). Regarding ruminating, steers at the LFS spent significantly shorter time than those at other stages in all seasons (p<0.05). GS and EFS steers showed the longest walking time in summer compared with other seasons (p<0.05). At GS and LFS, drinking time in summer was the longest of all seasons (p<0.05). Sleeping time was significantly shorter in summer compared with the other seasons (p<0.05). Self-grooming time was the longest in winter for all growth stages (p<0.05). Conclusion: Steers were found to have more variable behavioral patterns during summer and the GS and less active behaviors during the LFS, thus extra care seems necessary during the GS, LFS, and summer period.

Effect of Ethanolic Extract of Schizandra chinensis for the Delayed Ripening Kimchi Preparation (오미자(Schizandra chinensis) 추출물이 김치의 과숙억제에 미치는 영향)

  • This study mainly focused on to investigate the effects of Schizandra chinensis on the growth of a bacterium, CS6 which was isolated from kimchi. CS6 was final]y identified to lactobacillus plantarum that caused acidification of kimchi. The ethanolic extract of Schizandra chinensis(EES) inhibited the growth of L. plantarum. Minimum inhibition concentration of crude EES on L. plantarum was 62.5mg/$m\ell$. In broth culture, 5$\mu\textrm{g}$/$m\ell$ of EES completely inhibited the growth of L. plantarum during fermentation. The addition of 0.4% of EES has no apparent effect on quality including the taste and color on kimchi. It was expected that EES-containing kimchi could extend the period of preservation. Analysis of organic acids in water fractions of EES was carried out by HPLC. It is apparent that antimicrobial active fractions contained the highest concentration of succinic acid, a little tartaric acid and malic acid. Among these organic acids, succinic acid showed the strong inhibitory effect against L. plantarum CS6 in vitro. Succinic acid-containing kimchi with a concentration of 0.4 and 0.5% had the inhibitory effect on growth of L. plantarum. Inhibitory effect of EES on amylase, cellulase and pectinase was also tested. In conclusion, the present experiment demonstrated that EES inhibited the growth of L. plantarum, and various enzyme activity. EES-containing kimchi was sustained the hardness, and initial acidity during fermentation. EES was considered as the possible additive of kimchi process and EES added in kimchi increase the quality, and storage period of kimchi.

The Growth of Ginseng Industry and the Activities of Ginseng Cultivators in the 1930s: Focusing on Non-Government-Contract Cultivation Areas (1930년대 인삼업의 성장과 삼포민의 활동 -특별경작구역 이외 지역을 중심으로-)

  • This article focuses on changes in the ginseng industry in the 1930s in areas other than the Government Contract Cultivation (GCC) zones. A major characteristic of the ginseng industry in the 1930s was the rapid increase in the area covered by ginseng gardens: the area was about 212 ha in 1929 and 252 ha in 1930 and 1931 but soared to around 441 ha in 1938. This occurred because the non-GCC areas increased significantly during this period. Until the early 1930s, the ratio of GCC to non-GCC areas was 70:30. By the late 1930s, however, the ratio had changed to 53:47. The reason for this change was that the area of the newly established ginseng gardens in the GCC zones had decreased, while that of the non-GCC ginseng gardens had steadily increased. Due to the Japanese invasion of China, China boycotted red ginseng, and exports were sluggish, so the GCC areas were reduced. On the other hand, the non-GCC ginseng gardens were not affected, and the area they covered steadily increased. As a result, in the 1930s, the ginseng industry outside of the GCC areas grew rapidly. The region that led the growth of the ginseng industry outside of the GCC zone was Jeonbuk. By the late 1930s, Jeonbuk dominated the other provinces and accounted for more than 50% of the non-GCC farming zone. Gyeongbuk and Gangwon-do followed Jeonbuk in terms of ginseng cultivation areas. While Gyeonggi-do, Gyeongnam, and Chungbuk were also active in ginseng cultivation, Jeonnam and Chungnam were not active. In the 1930s, the growth of the ginseng industry outside of the GCC zones was driven by the efforts of ginseng farmers and the support of local governments. An examination of Yecheon-gun in Gyeongbuk, Ganghwa-gun in Gyeonggi, and Jecheon-gun in Chungcheongbuk-do showed that ginseng farmers organized cooperatives as the ginseng industry steadily developed in these regions, and these cooperatives worked systematically to cultivate and sell ginseng. In the case of Ganghwa-gun, activities were carried out to incorporate the GCC zone. The Deoksan Ginseng Association in Jecheon-gun determined that financing for cultivation was key and requested subsidies from the provincial government. Administrative authorities also supported the activities of the ginseng farmers. The activities of the farmers and the support of the administrative authorities together led to the growth of the ginseng industry during this period.

Effect of Pile Temperature Control on Changes of Nutritional and Microbilological Parameters of Composted Poultry Waste (육계분의 콤포스터 처리시 내부온도 조절이 생산물의 영양학적, 미생물학적 성상에 미치는 영향)

  • In an attempt to recycle as feed or fertilizer, broiler litter containing rice hull was manually composted under the control of peak temperature of piles(uncontrolled or controlled below $70^{\circ}C$ or below $60^{\circ}C$) in each of three $1.0\;{\times}\;1.0\;{\times}\;1.2m$ dimensional facilities. Changes of nutritional and microbiological parameters were evaluated throughout the 8 weeks of processing period. The initial content of crude protein(29.6%) decreased to 17.8% after 8 weeks of composting. The rapid nitrogen(N) loss observed in the early phase was attributed mainly to non-protein-N(NPN) loss. The initial content of ash(19.1%) increased to $26{\sim}29%$ after 8 weeks. For toxic heavy metals, Cr, Pb, and Hg contents of final composts were far less than the maximum tolerance levels allowed in food or compost. Bacterial growth was rather depressed until the second week, increased thereafter, and reached to peak($10^{12}cfu$ level) at the 4th week of composting. With composting, actinomycetes were active at the level of $10^7\;to\;10^9$. Fungi were active during the first to third week of composting. In general, control of pile temperature below $70^{\circ}C$ did not remarkably alter the nutritional and microbiological parameters of broiler litter compost, compared to that of pile temperature below $60^{\circ}C$. Further researches on prevention from the rapid loss of NPN in the early phase of composting are required for more effective recycling of broiler litter.

  • The female image has been changed incessantly with age and has been eminently represented in cosmetic advertisement. The female images are changed from the classical images to the active and professional images with the historical current of cyber, digital, and fusion. These changes in the expression of female image are largely due to the spread of post-modernism, feminism, de-construction (Kim, 1994), digital information revolution, and increase of income. The female images in cosmetic advertisement have been expressed very variously with fashion, marketing target, and characteristics of articles. (Im, 1997) The cosmetic advertisements of pure and graceful images were popular in the past. But nowadays individual image, womanly image, and unchanged beautiful image are in vogue. Individual image is very popular in young generation with very short fashion period. Active career woman image represents passion and beauty with extension of women' social roles. Unchanged beautiful image in modern industrial pollution stands for the desire of keeping the beauty in youth. Brand is very important factor to consumers in purchasing. Brand is no larger the simple concept in the past, and accepted as reflection of the image, social status, service, and life-style(Lee, 1998) consumers are very favorable to foreign cosmetics with the increase of overseas travels and import. This phenomenon is considered as very natural, especially in young generation. To create a successful brand image, the harmony of quality, psychological preference, advertisement, and reasonable price are required. According to the questionnaire research implemented by the subject of college women students in five universities in Seoul, the majority of college women students purchase domestic cosmetic brands in cosmetic specialty stores and depend on the tips of acquaintances. Quality, skin-trouble, and brand are considered as special regards for purchasing. Especially, internet shopping in purchasing marks high growth rate and preference for foreign cosmetics is very ardent. It can be expected that the 21st century is the epoch of various small production different from the mass-production in the 20th century. Female image will be probably expressed with individual, emotional, and professional image in the media of digital, fusion, cyber, and technology culture. It can be said that the tendency of cosmetic purchasing in the future will be more focused on brand image and life style.
