차즈기(Perilla frutescens var. acuta)의 생육에 미치는 광도의 영향 (Effect of Light Intensity on the Growth of Perilla frutescens var. acuta)

  • 차즈기의 관상가치 증진 및 광도에 따른 생육반응을 알아보기 위하여 실험을 실시한 결과 차광정도에 따라 생육상태가 매우 다르게 나타났다. 초장, 수관폭, 엽병장, 엽장, 엽폭, 줄기굵기, 건물중 등 대부분이 30% 차광 시에 높게 나타나 생육이 가장 좋았으며, 차광율이 높아질수록 모두 감소하였다. 엽록소함량도 30% 차광시에 가장 높았으며, 안토시아닌함량의 경우 70% 차광시에만 감소하였을 뿐 나머지 처리구에서는 큰 차이가 없었다. 또한 70% 차광 시에는 엽색이 암자적색에서 황록색으로 변하였고, 생육이 저조하였을 뿐만 아니라 식물체가 도복하여 차즈기 고유의 관상가치가 떨어지는 결과를 보였다.

식품부패 및 병원성 미생물에 대한 자소잎 추출물의 항균효과 (Antimicrobial Activities of Extracts of Perilla Frutescens Briton var. acuta Kudo on Food Spoilage or Foodborne Disease Microorganisms)

  • 자소잎중 물과 에탄올을 이용하여 얻은 추출물을 2종의 부패곰팡이(Aspergillus flavus KCTC 6143, Aspergillus jlavus KcrC 6961) 및 5종의 식중독세균 ( isteria monocytogenes ATCC 15313, Staphylococc따 aureus 196E ATCC 13565, Escherichia coli 0157:H7 ATCC 43895, Salmonella typhimurium ATCC 13311, Yersinia enterocolitica)에 대하여 항균효과를 조사하였다. 생육증식 저해농도를 disk method로 검색한 결과 에탄올추출물에서 그 효과가 인정되어 5종의 식중독 세균에 대하여 자소잎 에탄올 추출물을 농도별로 첨 가한 후 그 효과를 비교하였다. Gram양성균(L. monocytogenes, S. aureus)의 경우가 Gram음성균(E. coli 0157:H7, S. typhimurium, Y. enterocolitica)에 비하 여자소잎 에탄올추출물에 의한 증식억제효과가 추 출물 $1000\mu\textrm{g}$/mL 농도 첨가구에서 월둥히 컸으며 생 육증식억제효과의 크기는 L. monocytogenes, S. aureus, S. typhimurium의 순이었고, E. coli 0157:H7과 Y enterocolitica균에 대한 생육증식억제 효과는 농도첨 가별에 따라 생육증식이 비례적으로 억제되었다. 자 소잎 에탄올 추출물은 $121^{\circ}C$에서 15분간 가열처리한 후에도 항균력이 인정되었다.

Genetic diversity and population structure among accessions of Perilla frutescens (L.) Britton in East Asia using new developed microsatellite markers

  • SSRs were successfully isolated from the Perilla crop in our current study, and used to analyze Perilla accessions from East Asia. Analyses of the clear genetic diversity and relationship for Perilla crop still remain insufficient. In this study, 40 new simple sequence repeat (SSR) primer sets were developed from RNA sequences using transcriptome analysis. These new SSR markers were applied to analyze the diversity, relationships, and population structure among 35 accessions of the two cultivated types of Perilla crop and their weedy types. A total of 220 alleles were identified at all loci, with an average of 5.5 alleles per locus and a range between 2 and 10 alleles per locus. The MAF (major allele frequency) per locus varied from 0.229 to 0.943, with an average of 0.466. The average polymorphic information content (PIC) value was 0.603, ranging from 0.102 to 0.837. The genetic diversity (GD) ranged from 0.108 to 0.854, with an average of 0.654. Based on population structure analysis, all accessions were divided into three groups: Group I, Group II and the admixed group. This study demonstrated the utility of new SSR analysis for the study of genetic diversity and population structure among 35 Perilla accessions. The GD of each locus for accessions of cultivated var. frutescens, weedy var. frutescens, cultivated var. crispa, and weedy var. crispa were 0.415, 0.606, 0.308, and 0.480, respectively. Both weedy accessions exhibited higher GD and PIC values than their cultivated types in East Asia. The new SSR primers of Perilla species reported in this study may provide potential genetic markers for population genetics to enhance our understanding of the genetic diversity, genetic relationship and population structure of the cultivated and weedy types of P. frutescens in East Asia. In addition, new Perilla SSR primers developed from RNA-seq can be used in the future for cultivar identification, conservation of Perilla germplasm resources, genome mapping and tagging of important genes/QTLs for Perilla breeding programs.

Geographic Variation of Flowering Response to Daylength in Perilla frutescens var. frutescens in East Asia

  • We investigated the variations of the flowering response to daylength in Perilla crop (var. frutescens). Seventeen accessions of Perilla crop and one accession of weedy type of var. crispa from China, Korea and Japan were cultivated under three daylength conditions, i.e., short-days, natural daylength and long-days. Most accessions of Perilla crop from China, Korea and Japan were divided into three types, early maturing type, intermediate maturing type and late maturing type by their natural flowering habit. In most of the accessions used, the flowering habit was significantly accelerated by short-day conditions and was delayed by long-day conditions. All the accessions of Perilla crops flowered within 57 days under the 10 hrs light treatment, whereas they did not flower at all even at 170 days after sowing under the 16 hrs light treatment. Thus, this finding suggested that there is a relationship between the types of flowering response to daylength and the geographical distribution which determines the planting season in traditional cultivation practices of Perilla crops. Positive correlation was observed between days to flowering and plant height or internode number in both the short-day and natural daylength conditions. Whereas, correlation was negative between days to flowering and inflorescence length or floret number in natural daylength condition, but it was positive in the short-day condition. Therefore, the daylength condition is considered as the most important environmental factor for flowering habit and morphological characters of Perilla crops. Flowering habit is considered as an important key character for the study of geographical differentiation of Perilla crop in East Asia.

Morphological Variations Between Cultivated Types of Perilla Crop and Their Weedy Types in Korea and Japan

  • In order to better understand the morphological differentiation of the two cultivated types of Perilla crop and their weedy types in Korea and Japan, we studied the variation of 62 accessions by examining 15 morphological characteristics. By using ANOVA (one-way analysis of variance), we determined that var. frutescens and var. crispa showed significant morphological differences in terms of plant height and seed weight. Furthermore, cultivated var. frutescens and var. crispa could also be clearly discriminated from one another using PCA (principal component analysis). Specifically, quantitative and qualitative characteristics such as plant height, seed weight, degree of pubescence, shape of leaf, color of leaf, fragrance of plant, color of flower, color of stem and seed size greatly contributed to differences seen in the positive and negative direction on the first axis. In our study, most accessions of cultivated var. frutescens and those of its weedy type could be clearly discriminated from one another, however, most accessions of cultivated and weedy types of var. crispa were not clearly discriminated by the ANOVA and PCA analyses. These results indicated that cultivated var. frutescens can be considered to be a domesticated form, while the cultivated var. crispa can not be considered to be a domesticated form in Korea and Japan. It is our belief that our results concerning the morphological variations among cultivated types of Perilla crop and their weedy types in Korea and Japan will help ensure the long-term success of breeding programs and maximize the use of the germplasm resources in Korea.

인공산성비가 들깨의 생장 및 효소에 미치는 영향 (Effect of simulated Acid Rain on the Morphology and Enzyme of Perilla frutescens var. japonica Hara)

  • This experiment was performed with the purpose of finding out the effect of simulated acid rain at various pH levels on the morphology and enzyme of Perilla frutescens var. japonica hara. The pH levels of simulated acid rain ranged from pH 2.0 to pH 6.0. The experiment showed the anion concentrations in the order of $SO_4^{2-}$, Cl^-$, $NO_3^-$, and $F^-$, $SO_4^{-2}$ was found out to be the main factor which contributed to the rainwater acidification. A general decrease of growth in Perilla frutescens var. japonica hara growth was shown with the decreas of pH concentration. As acidity increases a definite reduction in the rates of germination, heigth of plant, malate dehydrogenase, and 6-phosphogluconate dehydrogenase was ovserved, but the density of spots on the leaf apex was increased.

  • The effects of monazite and fixing agents on cotton fabric dyed with Perilla frutescens var. acuta extract were investigated. The proper monazite treatment concentration, temperature and time were 10%(o.w.b.), $50^{\circ}C$ and 60minutes. By various fixing agent treatment, $FeSO_4$ showed a relatively high K/S value and the order of K/S value decreased as follows, cation surface active agents, soybean and NaCl. And the monazite and $FeSO_4$ fixing agent showed higher anion emissity than those of untreated cotton and other fixing agents. The cotton fabrics showed improved color fastness by monazite and fixing agents treatments with the exception of light fastness. And the cotton fabrics fixed with fixing agents were showed effective bacterial reduction with the exception of NaCl.

자소엽의 항알레르기와 항염증효과 (Antiallergic and Anti-inflammatory Effects of Perilla frutescens var. acuta)

  • In the present study, we investigated the effect of the water extract of Perilla frutescens var. acuta (Labiatae; WEPF) on the mast cell-mediated allergic reactions. WEPF was anally administered to mice for high and fast absorption. WEPF inhibited compound 48/80-induced systemic allergic reaction. WEPF attenuated immunoglobulin E (IgE)-mediated local allergic reaction. In addition, WEPF decreased the gene expression of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated HMC-1 cells. These results indicate that WEPF inhibits mast cell-mediated allergic reactions in vivo and in vitro.

Triterpenes from Perilla frutescens var. acuta and Their Cytotoxic Activity

  • Nine triterpenes were isolated from the petroleum ether and MeOH extract of Perilla frutescens var. acuta leaves. Their structures were determined to be arjunic acid (1), maslinic acid (2), oleanolic acid (3), euscaphic acid (4), tormentic acid (5), 3-O-trans-p-coumaroyltormentic acid (6), 28-formyloxy-$3{\beta}$-hydroxy-urs-12-ene (7), ursolic acid (8), and corosolic acid (9) by spectroscopic methods. The compounds 1, 2, 4, 6, and 7 were isolated for the first time from this plant and the Genus Labiatae. The isolated compounds (1-9) were tested for cytotoxicity against four human tumor cell lines (A549, SK-OV-3, SK-MEL-2, and HCT-15) in vitro using a Sulforhodamin B bioassay.

  • Perilla frutescens Britton var. acuta Kudo, an annual plant primarily cultivated in China, Japan, and Korea, has been used as a traditional medicine to treat inflammatory diseases, depression, and many anxiety-related disorders. Previously, we reported the inhibitory effects of n-hexane layer of P. frutescens var. acuta extract against beta-amyloid (Aβ) aggregation, and the isolation of asarone derivatives as active constituents from n-hexane layer. In this study, dichloromethane layer of P. frutescens var. acuta was applied to bioassay-guided isolation methods accompanied with Thioflavin T (Th T) fluorescence assay to investigate the inhibitory effect on Aβ aggregation and disaggregation. As the results, three triterpenoids including ursolic acid (1), tormentic acid (2) and corosolic acid (3) were isolated. All compounds reduced Aβ aggregation and increased disaggregation of preformed Aβ aggregates in a dose-dependent manner. However, the inhibitory effect of three compounds on Aβ aggregation was not correlated with antioxidant activity, which was measured by DPPH assay. Taken together, these results suggest that the triterpenoid derivatives from P. frutescens have the potential to be developed as good therapeutics or preventatives for AD.
