In vitro Formation of Cochliobolus nisikadoi, the Perfect State of Bipolaris coicis (Bipolaris coicis의 완전세대인 Cochliobolus nisikadoi의 배양기내 형성)

  • The perfect state of Bipolaris coicis, causing leaf blight of Job's tears, was in duced under in vitro conditions. Eighty nine isolates of the fungus were collected from 17 locations in Korea from 1994 to 1996. They were crossed on Sach's nutrient agar, on which a piece of rice straw was placed, and incubated at $25^{\circ}C$. Pseudothecia were produced only by certain combinations of compatible isolates. Although pseudothecia were usually produced on rice straw two weeks after incubation, asci and ascospores were observed only in a few pseudothecia examined. The pseudothecia were black and globose with protruding ostiolar beaks. The locules were filled with a mass of hyaline and filamentous pseudoparaphyses. Asci were cylindrical to clavate and straight or slightly curved. The ascus wall was bitunicate with short stipes. Ascospores were filiform, hyaline, and arranged parallel to slightly coiled in the asci, measuring $143-166.4{\times}2.6-3.8\;{\mu}m$. Perfect state of the fungus was identified as Cochliobolus nisikadoi (Tsuda, Ueyama & Nishihara Alcorn), based on the morphological characteristics.

A Study on the Economical Nutrition Supplement of Cereal Food for Improvement in our National Eating Habits (국민식생활(國民食生活) 향상(向上)을 위(爲)한 곡류제품(穀類製品)의 경제적( 經濟的) 영향강화(營養强化)에 관(關)한 연구(硏究))

  • I. Subject of the Study: Studies on the economical nutrition supplement of cereal foods for the improvement in our notional eating habits. II. Purpose and Importance of the Study: 1. Our nation is confronted with the situation that the rice, a principal food, short of some essential amino acids, lysine and threonine, leads to imbalanced meals insufficient in the nutrient of protein, to bring many difficulties in the elevation of our national physique. 2. The shortage of even the rice imperfect in the nutrient of protein makes the import of lots of foreign rice inevitable. It is considered that the protein supplement and decrease in the consumption amount, of rice, is a serious key to the solution of our food difficulty, and then a way of the proetin supplement of rice through the addition of essential amino acid is to be rarely applied in the view of the our present finance and situation. 3. In the present experiment, therefore, it aims to the suggestion of an aspect of the improvement in our national eating habits guiding in the nutrition elevation which our nation can afford economically through the development of first, a way of the protein supplement by the mixture of cereals producted plentifully in our country, and second, a way of the decrease in the consumption amount and the improvement in the nutrition of rice through the substitution of the other cereals for rice. III. Contents of Scope of the Study: 1. Objects of the study: Objects of the study are the following three items; a) The nutrition supplement of rice through the mixture of cereals. Our nation makes mainly rice as a principal food, but practically many kinds of cereal are produced in our country. They contain different levels and qualities of each nutrient and they are different from one another in the kinds of essential amino acid consisting protein. For that reason, the mutual complement efficeincy of insufficient nutrients is observed through the mixture of cereals. b) The nutrition supplement of rice through the addition of superior protein sources to rice, a principal food. The development of rice as superior foods in the sense of nutrition is conducted through the risement in protein quality by the addition of protein sources in good quality, for example, fish flour (anchovy flour), egg powder, milk powder, and so on, and through the supplement of vitamins and minerals. c) The decrease in the consumption amount of rice through the substitution of the other cereals for rice, as a principal food. The compensation for the short amount of rice is made by the reduction in the consumption of rice through the discovery of a way of substitution of the other cereals for rice, as a principal food and of the settlement of problems in nutrition and finance subsequent to this. 2. Contents of the study: a) An ideal mixture-ratio of cereals is established for rats by feeding mixed foods(rice-barely or rice-wheat) containing 5%, 15%, 25%, 35% and 45% level of either barely or wheat. b) The nutritive value is determined in the whole subsititution of other foods for rice, and then, a way of the complement of over and under nutrients is devised. c) The ideal combination is investigated for rats through feeding mixed foods of main food, rice and supplement foods of protein sources, soy bean, fish flour, egg powder and milk Powder. d) According to results from the above three experiments, the concise functional test for men and the examination of economical property are made. 3. Scope of the study: a) The observation of the effect of each diet on the growth rate for rats. The growth rate of rats was observed for 15 groups of mixed foods of a main food, rice, and wheat flour, barley powder or soy bean powder, respectively, and 12 groups of wheat flour diets supplemented with $1{\sim}3%$ milk powder, and rice or wheat flour diets supplemented with 5% of milk powder, egg powder, fish flour or soy bean powder, respectively. b) The determination of food consumption. The food consumption was determined at weekly intervals for 27 kinds of diet described in a) item. c) The determination of food efficiency rate. The food efficiency rate for each diet was determined by calculation from the gained body weight and the food consumption amount at the same intervals described in b) item. d) The determination of protein efficiency rate. The protein efficiency rate for each diet was determined by calculation form gained body weight and the protein amount of the food consumption amount at the same intervals described c) item. e) The determination of the body component. The hematocrite and hemoglobin levels in the blood, total nitrogen in the serum, blood sugar, and lipids and glycogen in the liver were determined. f) The observation of nitrogen balance. As a means of the observation of nitrogen balance, the total nitrogen in the urine was determined. g) The analysis of economical property. The economical property was analyzed as the gained body weight to the amount equivalent to one won through the conversion of the food consumption amount into money. h) The functional test for men. The concise functional test for men was made in order to establish if the best diet for experimental animals can be applied to men. IV. Results of the Study: The national food product plan, nationwide nutritive enlightment and the improvement activities in our country eating habits, especially, mixed and powder food problems are to be significantly referred, and the following results must be applied. a) In the mixed foods of cereals, the mixed food of the rice-barley containing $5{\sim}15%$ level of barley is best in terms of nutrition. b) The addition of superior protein sources, egg, Bilk, soy bean, or fish, respectively to either rice or wheat flour makes a great risement in the nutritive value. c) The animal protein is more effective in the elevation of nutritive value of cereals. d) Rice takes the most nutritive operation and has the highest preference, among rice, wheat flour and barley. e) Wheat flour is more economical than rice in evaluation of the gained body weight to the regular money, and the addition of fish or soy bean is more economical than that of any other supplement food. But the above results are true of the range of nutrition and economical property. f) The study on the nutrition composition and barley will lead to the improvement in our national eating habits as mixed food of the rice-barley containing $5{\sim}15%$ level of barley is more nutritive. g) This study on the nutrition only for the growing animal can not be considered as a perfect and entire evaluation. Consequently, the perfect data for our national nutrition can be obtained from the experiment similar to this for the much longer period examining, in details, the growth rate, change of physical strength, mental and bodily change, average life span, and resistance ability to infectious diseases.

Effects of Damage by Brown Planthopper at Ripening Stage on Yield and Quality of Rice (벼 등숙기 벼멸구 피해가 수량 및 품질에 미치는 영향)

  • This study was carried out to investigate the change yield and grain quality in rice caused by damages of brown planthopper (Bph) on major agronomic traits at the ripening stage. A major agronomic traits such as culm length, panicle length, number of panicle and number of grain per panicles was no-significant difference compared hopperburn area (HBA) and moderately damaged area (MDA) by brown planthopper with non-damaged area (NDA). but, the ripeing ratio and yield in HBA and MDA was significantly decreased. The perfect rice of brown rice in HBA and MDA was also lowered than the NDA and the immatured grain among imperfect rice in damaged area by BPH was significantly increased than in the NDA. Among the traits related with palatability of cooked rice, the amylose content was not affected by damage of brown planthopper, whereas, the protein content was significantly increased in the HBA and MDA. additionally, Peak viscosity in HBA and MDA is lower than the NDA, and setback was increased. In most cases, there was negative on yield and grain quality by the damage of brown planthopper at the ripening stage. Thus, the rice product harvested in damaged area should be separated with that in non damaged area for grain market quality of milled rice.

Changes in Soil Properties and Rice Production as Influenced by the Consecutive Application of Liquid Swine Manure in Paddy Field (돈분 액비의 연용이 벼의 수량과 토양에 미치는 영향)

  • The application of animal manure on farm fields is one of the most economical ways. However, the continuous application of manure in paddy fields might change soil properties influencing the growth of rice plant. Thus, this study was conducted to investigate the changes in selected chemical and biological properties of soils and rice production as affected by the applications of two different fertilizer sources, which were the consecutive applications of liquid swine manure (LSM) and chemical fertilizer (CF), during the three experimental years, from 2012 to 2014. Application amount of LSM was based on 100% of nitrogen fertilizer recommendation rate for rice cultivation estimated by soil testing. Plant height and tiller number in rice at the first year of liquid swine manure manure plot were lower than those of chemical fertilizer plot. Height and tillers of rice in liquid swine manure plot were higher than those of rice in chemical fertilizer plot after consecutive application for 3 years. Rice yield In the first year of application was decreased by 7% than that of chemical fertilizer, but the yield of rice in the third year of application in LM 100% plot was increased by 8% compared to the chemical fertilizer. Toyo-taste value of milled rice in LM 100% was decreased by increasing of protein contents and decreasing rate of perfect grain. The K and Zn contents in the soil were increased in the plots of consecutive LSM application. The results implied that the liquid manure may neither decrease the yield of rice and nor increase soil properties except K and Zn in the soil, and decrease rice quality.

Effects of leaf-defoliation and different nitrogen, phosphorus and potassium application on maturity and yield of rice (질소 인산 가리의 시용량의 차이 및 전엽처리가 수도의 등숙 및 수량에 미치는 영향)

  • Field experiment was conducted to disclose some effects of leaf defoliation in heading time and the application of different combinations of nitrogen, phosphorus and potassium on the maturity and yield of paddy rice. The results obtained are as follows; 1.All were components considered were increased as the amounts of nitrogen, phospgorus and potassium applied were increased, except the maturity rate had the tendancy to be decreased with the increase of the above fertilizer applications. 2.As the rate of cutting leaves went higher, the weight of rough rice, the maturity rate and the weight of 1, 000 kernels of perferct brown rice were decreased, and there were significant differences among the treatments of leaf blade cutting. 3. The contribution of each leaf to yield through the maturity, number of spikelets per panicle and 1, 000 kernel weight were 61.87-66.18-65.61% respectively for the flag leaf, 17.50-20.65-23.41%for the 2nd leaf, 12-71-12.73-10.11%for the 3rd lead and 7.93-6.44-0.87%for the 4th leaf respectively. This simply indicared that the effects of the leaves to the yield were decreased as their position went down. This tendency was specially emphasized in the case of the weight of 1, 000 kernels of perfect brown rice.

Effects of Development Time and Severity of Rice Ear Blight on Rice Yield and Quality (벼 이삭마름병 발병시기와 정도가 수량 및 품질에 미치는 영향)

  • Grain discoloration, caused by several microorganisms, is a minor disease, but it is gaining importance in almost all rice growing areas of the world in recent years. Such grains are of low market value and consumption appeal in addition to their abnormal performance as seed of source of nutrient value. When the casual agents were inoculated on rice plant,'Ilmi-byeo'(at right before heading time and on heading time), the incidence of rice ear blight caused by Alternaria alternate and Nigrospora oryzae was higher in a plot inoculated at right before heading time than (in n plot inoculated) on heading time. Meanwhile, the incidence of rice ear blight caused by Bipolaris oryzae and Curvularia lunate was higher in a plot inoculated on heading time than in a plot inoculated at right before heading time. In rice quality, the ratio of imperfect grains was increased at right before heading time or on heading time by Nigrospora oryzae or Alternaria alternata, respectively. As discoloration rate of rice grains increased on 'Ilmi-byeo', the percentage of perfect grains decreased proportionally and the percentage of damaged rice grains increased remarkably.

Influence of Silicate Application on Rice Grain Quality (규산 시용이 미입의 품질에 미치는 영향)

  • To evaluate the characters related to rice grain quality as affected by silicate application, the ripened color, appearence and physicochemical properties of rice grain which were produced at the silt-loam paddy field of National Yeongnam Agricultural Exeperiment Station with a little poor drainage for trials on long-term effects of continuous application of same fertilizer were analyzed comparing with the NPK fertilizer an the NPK+compost in 1995. The whiteness value and color space value in ripened color of rice hull by silicate application were higher than those in the plots of NPK and NPK+compost, because of low percentage of rusty grain and sooty grain, The percentage of perfect grain in brown rice and in milled rice by the silicate application increased by 6.1~7.5% and by 1.8~3.5% respectively, as compared with the NPK application and white core and white belly of milled rice were decreased, The amylose content, protein content and starch-iodine blue value of milled rice by silicate application were lower than those in the plots of NPK and NPK+compost, while the peak point and breakdown viscosity of gelatinized rice flour on amylograph, and tastes value evaluated as a indicater of pannel test by nireco tester were higher. By silicate application these factors could be affected toward better in eating quality.

Mapping of the Reduced Culm Number Trait in Rice (Oryza sativa L.) rcn10(t) Mutant

  • In rice, tillering is an important trait determining yield. To study tillering at the agricultural and molecular aspects, we have examined a spontaneous rice mutant that showed reduction in the number of culms. The mutant was derived from a $F^6$ line of the cross of Junambyeo*4 / IR72. It could produce, on average, 4 tillers per hill in the paddy field while wild-type plants usually have 15. Except the reduced culm numbers, they also show pale green phenotypes. The phenotypes of this mutant were co-segregated as the monogenic Mendelian ratio (${\chi}^b=0.002$, p=0.969). In order to locate a gene responsible for the rcn phenotype, the mutant with the japonica genetic background was crossed with Milyang21 of the indica background. Bulked segregant analysis was used for rapid determination of chromosomal location. Three SSR markers (RM551, RM8213, and RM16467) on chromosome 4 were genetically associated with the mutant phenotype. Each of the 217 $F_2$ plants was genotyped with simple sequence length polymorphisms. The data showed that RM16572 on chromosome 4 was the closest marker that showed perfect co-segregation among the $F_2$ population. We suggest the new rcn gene studied here name as $rcn10^t$ because there was no report which exhibit a rcn phenotype with a pleiotropic effect of pale green (chlorophyll deficiency), and mapped at same position on chromosome 4.

Influence of Coated Urea Complex Fertilizer Application on Growth and Grain Quality of Paddy Rice (피복요소복합비료(被覆尿素複合肥料)의 시용방법(施用方法)이 벼 생육(生育)과 미질(米質)에 미치는 영향(影響))

  • This study was conducted to evaluate coated urea complex fertilizer (CUC) application for rice growth and its grain quality on clay loam (Deogpyeong series) and sandy loam (Gangseo series) in southern region of Korea, 1992. The coated urea complex fertilizer used in the experiment was CUC I and II. The CUC I is a complex fertilizer consisting of 18-12-13 for $N-P_2O_5-K_2O$ including 50% of coated urea as nitrogen source, and CUC II is the same complex fertilize as mentioned above but it contains 25% of coated urea. Seventy percent of conventional fertilizer for nitrogen level was applied as basal dose of side-band placement (SBP) and whole plowlayer placement (WPP). Greenish degree of rice leaf color at the late growth stage was high in CUC plots regardless of application method and soil series. Amount of nitrogen uptaken by the rice plant at harvesting stage was mush more in the CUC treatment compared to convential fertilizer, and also much more in CUC I than that of CUC II. In the WPP on clay loam, harvest index of CUC treatment was showed very low level, which was reduced more in CUC II plot. In the CUC treatments, perfect rice grain ratio decreased remarkably, and the notched-belly rice kernel ratios among imperfect rice grain increased drastically. In rice grain, Hon-value(Mg to K.N ratio) decreased, protein and amylose contents increased in CUC application plots in WPP. In addition, gel consitency and alkali spreading value related to eating quality in CUC plots were getting longer and lower, respectively. The yield in CUC plot was similar to conventional fertilizing, but significant decreased in SBP. As a result, the rice grain quality became poor with the coated urea complex fertilizer application, but it could be improved by incorporation with fast-release fertilizer.

Quality Comparison of Static-culture and Commercial Brown Rice Vinegars (정치배양 및 시판 현미식초의 품질특성 비교)

  • The quality of brown rice vinegar that was produced via static culture (A) was compared with the quality of three types of domestic commercial brown rice vinegar (B, C, and D) and of three types of Japanese brown rice vinegar (E, F, and G). The results showed titratable acidity levels of 6.39%, 4.52-6.32%, and 4.51-4.89% in the static-cultured brown rice vinegar, the domestic commercial brown rice vinegars, and the Japanese brown rice vinegars, respectively. The pH levels were 3.28, 2.58-2.97, and 3.03-3.27 in the static-culture brown rice vinegar, the domestic commercial brown rice vinegars, and the Japanese brown rice vinegars, respectively, which show similar values of the static-culture brown rice vinegar and the Japanese brown rice vinegars. The total nitrogen (TN) values of the static-culture brown rice vinegar, the domestic commercial brown rice vinegars, and the Japanese brown rice vinegars were 0.24, 0.03-0.16, and 0.12-0.17, respectively, with the highest value for the static-culture brown rice vinegar, substantial differences among the domestic commercial brown rice vinegars, and similar contents among the Japanese brown rice vinegars. For free sugar, glucose was either detected only in a small quantity or not detected at all in the static-culture brown rice vinegar and the Japanese brown rice vinegars, which showed perfect fermentation. The glucose and maltose contents were higher in the domestic commercial brown rice vinegars. The organic acid content of the static-culture brown rice vinegar was similar to that of the Japanese brown rice vinegars. Therefore, the total acidity content, TN value, sensory property, and quality of the static-culture brown rice vinegar (A) were superior to those of the domestic and Japanese brown rice vinegars.
