재래종(在來種)갓의 Anthocyanin 색소(色素)에 관(關)한 연구(硏究) -[제2보(第二報)] Anthocyanin의 정량(定量)- (Studies on the Anthocyanins in Brassica juncea -Part II. Quantitative Determination of Anthocyanins-)

  • 우리나라 재래종(在來種)갓의 anthocyanin 색소(色素)의 구명(究明)을 위(爲)한 실험(實驗)에서 제일보(第一報)의 구조추정(構造推定)에 이어서 이들 색소(色素)의 함량(含量)을 측정(測定)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 채취시기(採取時刻)의 구별(區別)에 의한 전(全) anthocyanin의 함량(含量)은 peonidin-3-glucoside로 표시(表示)하여 봄갓이 175.5mg% 가을갓이 192.7mg%로 가을갓이 약간(若干) 더 많은 함량(含量)을 보였다. 2, 착색정도(着色程度)의 구별(區別)에 의한 전(全) anthocyanin의 함량(含量)은 peonidin-3-glucoside로 표시(表示)하여 잎의 양면착색(兩面着色)갓이 290.2mg%, 한면(面)만의 착색(着色)이 73.6mg%, 양면(兩面)모두 무착색(無着色)갓이 40.0mg%이었다. 3. 신선(新鮮)한 갓에서 peonidin-3-glucoside로 표시(表示)하여 40mg%이하의 anthocyanin 함량(含量)으로는 육안(肉眼)으로서는 전혀 갓특유(特有)의 착색(着色)을 감지(感知)할 수 없었다. 4. 갓에 존재(存在)하는 이종(二種)의 anthocyanin의 조성비(組成比)는 채취시기(採取時期)및 착색(着色)의 정도(程度)에 따라서도 거의 일치(一致)하였으며 그 조성(組成)은 peonidin-3-glucoside가 55.3% 그리고 peonidin-3-galactoside가 44.7%이었다.

  • 우리나라에서 채소(菜蔬)로 또 조미품(調味品)으로 많이 이용(利用)되고 있는 재래종(在來種)갓의 anthocyanin 색소(色素)의 구명(究明)을 위(爲)한 실험(實驗)에서 얻어진 결과(結果)를 요약(要約)하면 다음과 같다. 1. T.L.C.에 의(依)해 이종(二種)의 anthocyanin을 분리(分離)하였다. 2. 이들 anthocyanin의 구조구명(構造究明)을 위(爲)하여 Robinson 시험(試驗), Spectrophotometry, T.L. chromatography, 부분가수분해등(部分加水分解等)의 방법(方法)으로 각각 peonidin-3-glucoside와 peonidin-3-galctoside 임을 확인(確認)하였다.

자색고구마(자미) Anthocyanin 색소의 성분 분석 (Analysis of Anthocyanin Pigments from Purple-Fleshed Sweet Potato (Jami))

  • 자색고구마 (자미) anthocyanin 색소의 분석을 위하여 1% HCl 함유 methanol로 추출한 자색고구마 색 소액을 Amberlite IRC-50 양이온교환수지관을 통과시켜 정체하고 paper chromatography로 개별색소를 분리, 3개의 band를 얻었다. Paper chromatography에 의해 분리된 각 개별색소 에 대해 부분가수분해, aglycone의 확인, acyl group의 확인, 결합당의 확인, paper chromatography 그리고 HPLC 등을 기초로 한 각종 기기분석 결과 자색고구마의 anthocyanin 색소는 caffeic acid와 ferulic acid로 acylation된 peonidin-3-diglucoside-5-glucoside의 구조를 나타냄을 알 수 있었다.

  • Anthocyanin pigments of purple sweet potato roots (Ipomoea batatas L.) were extracted with 0.5% TFA (Trifluoroacetic acid) in 95% EtOh, and further isolated an purified by Amberlite XAD-7 and ODS column chromatography, and final preparative HPLC. Among nine anthocyanins isolated, the structure of three major anthocyanins were identified as 3-O-(6-O-trans-caffeyl)-2-O-(6-O-trans-caffeylglucopyranosyl)-$\beta$-D-glucopyranosyl-5-O-($\beta$-D-glucopyranosyl)-peonidin, 3-O-(6-O-trans-caffeyl)-2-O-(6-O-trans-feruloyl-glucopyranosyl)-$\beta$-D-glucopyranosyl-5-O-($\beta$-D-glucopyranosyl)-peonidin, and 3-O-(6-O-trans-caffeyl)-2-O-(6-O-p-hydroxylbenzoylglucopyranosyl)-$\beta$-D-glucopyranosyl)-peonidin, by using UV-visible absorption spectra, $^1$H-NMR and FAB-MS analysis.

작약꽃 가수분해물의 항산화, 항염 및 광노화 억제 효능 (Antioxidant, Anti-inflammatory and Anti-photoaging Activities of Hydrolyzed Peony (Paeonia lactiflora Pall.) Flower)

  • 본 연구에서는 작약꽃을 가수분해하여 플라보노이드계 화합물인 peonidin 배당체를 비배당체로 전환하여 생리 활성을 증대시키고 신규 화장품 소재로 이용하고자 하였다. 이를 위해 품종별 작약꽃을 추출하여 유효성분 함량이 가장 높은 품종을 선정하였으며, 해당 품종을 이용하여 가수분해를 진행하였다. 이를 통해 고농도의 hydrochloric acid (HCl) 처리가 가수분해에 가장 적합함을 확인하였으며, 효소를 이용한 생물학적 가수분해는 효과가 없음을 확인하였다. 작약꽃 가수분해물에는 붉은색을 나타내는 안토시아닌계 화합물인 peonidin 배당체가 peonidin으로 전환되어 63.3 ppm 함유되어 있음을 분석을 통해 확인하였다. 작약꽃 가수분해물의 생리 활성을 확인해본 결과, 가수분해물이 일반 작약꽃추출물 대비 2,2-diphenyl-1-picrylhydrazyl (DPPH) 라디칼을 소거하는 항산화 활성이 우수하며, nitric oxide (NO) 생성을 저해하는 항염 효과가 증대하였음을 확인할 수 있었다. 또한, ultraviolet A (UVA)에 의한 피부 섬유아세포 손상을 감소시켜 광노화를 억제함을 확인하였다. 따라서 본 연구를 통해 작약꽃 가수분해물이 화장품 산업에서 신규 항노화 소재로 활용이 가능할 것으로 기대된다.

Rapid Isolation of Cyanidin 3-Glucoside and Peonidin 3-Glucoside from Black Rice (Oryza sativa) Using High-Performance Countercurrent Chromatography and Reversed-Phase Column Chromatography

  • Anthocyanins are water soluble plant pigments which are responsible for the blue, red, pink, violet colors in several plant organs such as flowers, fruits, leaves and roots. In recent years, anthocyanin-rich foods have been favored as dietary supplements and health care products due to diverse biological activities of anthocyanins including antioxidant, anti-allergic, anti-diabetic, anti-microbial, anti-cancer and preventing cardiovascular disease. High-performance countercurrent chromatography (HPCCC) coupled with reversed-phase medium pressure liquid chromatography (RP MPLC) method was applied for the rapid and efficient isolation of cyanidin 3-glucoside (C3G) and peonidin 3-glucoside (P3G) from black rice (Oryza sativa L., Poaceae). The crude black rice extract (500 mg) was subjected to HPCCC using two-phase solvent system composed of tert-butyl methyl ether/n-butanol/ acetonitrile/0.01% trifluoroacetic acid (TBME/B/A/0.01% TFA, 1 : 3 : 1 : 5, v/v, flow rate - 4.5 mL/min, reversed phase mode) to give enriched anthocyanin extract (37.4 mg), and enriched anthocyanin extract was sequentially chromatographed on RP-MPLC to yield C3G (16.5 mg) and P3G (8.7 mg). The recovery rate and purity of isolated C3G were 76.0% and 98.2%, respectively, and those of P3G were 58.3% and 96.3%, respectively. The present study indicates that HPCCC coupled with RP-MPLC method is more rapid and efficient than multi-step conventional column chromatography for the separation of anthocyanins.

흑미(현진주벼)유래 안토시아닌의 항산화능 탐색 (Isolation of Anthocyanin from Black Rice (Heugjinjubyeo) and Screening of its Antioxidant Activities)

  • 유색미는 회색 빛 외에 붉은색 흑은 보라색 계통의 색소를 겨 부분에 함유하고 있다. 특히, 흑미는 예로부터 건강식품으로 여겨져 왔다. 흑미(흑진주벼)는 최근에 anthocyanin포함하는 생리활성 물질원으로서 관심이 증대되고 있다. Anthocyanin은 수용성의 색소로서 flavonoid계통의 대표 색소로 알려져 왔다. 흑미에 함유되어있는 Anthocyanin으로는 cyanidin 3-O-glucoside와 peonidin 3-O-glucoside, malvidin 3-O-glucoside, pelagonidin 3-O-glucoside, delphinidin 3-O-glucoside등이 알려져 있다. 본 연구에서는, 흑미로부터 amberlite XAD7 chromatography방법을 사용해서 anthocyanin을 추출 후 HPLC와 UV-Vis spectrophotometer를 이용하여 정량분석 및 정성분석을 실시하였다. 그 결과 흑미의 anthocyanin 추출물 중 95%가 cyanidin 3-O-glucoside, 5%가 peonidin 3-O-glucoside임을 확인하였다. 그리고 in vitro상의 여러 가지 실험방법을 통하여 anthocyani의 항산화 효과를 측정하였다. 그 결과 100g/ml 농도의 anthocyanin추출물은 90%이상 linoleic acid emulsion의 과산화를 억제하였고, 55.20%의 DPPH 자유 라디칼 제거효과, 54.96%의 superoxide 자유 라디칼 제거효과, 72.67%의 과산화수소 제거효과를 보였다. 그리고 또한 높은 환원 능력을 확인할 수 있었다. 표준 항 산화 물질로는 BHA, BHT, ${\alpha}-tocopherol$을 사용하였다. 이 결과로 보아 anthocyanin추출물은 새로운 항산화 물질로서 이용될 수 있을 것이다.

Antioxidative Activity and Varietal Difference of Cyanidin 3-glucoside and Peonidin 3-glucoside Contents in Pigmented Rice

  • The content of anthocyanin, cyanidin 3-glucoside (C3G) and the peonidin 3-glucoside (P3G) from 591 rice cultivars and the antioxidative activities of MeOH extract from 8 rice cultivars were evaluated. Among them, C3G content of pigmented rice were ranged from 0 to 451.9mg in 100 g brown rice, while the P3G contents were in the range from 0 to 42.7mg in 100g brown rice. There was no correlation between C3G and P3G content. Total anthocyanin content ranged from 0 to 475.1mg in 100g brown rice. The antioxidative activity, the scavenging activity on DPPH (2,2-diphenyl-picryl-hydrazyl) radical of MeOH extracts from rice grain, were different according to cultivars. The activity of blockish purple pericarp rice cultivars was twice stronger than that of white pericarp cultivar. Especially, the antioxidative activity of Heugjinjubyeo was four times stronger than that of white pericarp cultivar. The scavenging effect on DPPH radical in rice extracts was related to the total anthocyanin contents of the extracts.

포도파괴 Anthocyanin 색소의 분리 및 안전성 (Isolation and Stability of Anthocyanin Pigments in Grape Peels)

  • Anthocyanins were isolated and identified from grape peels which were wasted much in Korea, and their characteristics were as follows .Isolated pigments from grape peels were 11 varieties such as 5 varieties of 3, 5-diglucoside (DG), 6 varieties of 3-monoglucoside (MG), and acylated pigment was 2 varieties of them. Malvidin was 4 varieties , petunidin , peonidin and delphinidin 2 varieties in each, and cyanidin 1 variety of 11 vareities. Malvidin -3, 5-diglucoside and peonidin -3, 5-diglucoside of anthocyanins were above 48% in total anthocyanins content of 114.99mg/g in dried skins. Breakdown of anthocyanins was higher become intimate neutral pH, but stable to stroage period for 7-days. Hyperchromic effects were showed when sugars were added in pigment extract of grape peels, the highest value was glucose and the next ordor was fructose and sucrose. Breakdown velosity of anthocyanins was higher when ascorbic acid was added, but its velocity was reduced in anaerobic state . Absorption degree by organic acid treatment was higher than control, and anthocyanins were stable to storage period for 7 days.

A Comparative Study on the Functional Compounds of Color Potatoes

  • This study was carried out to obtain a basic information for the improvement of human health and the development of variety through analysis of organic compounds, contents of three CQA(3-caffeoylquinic acid, 4-caffeoylquinic acid and 5-caffeoylquinic acid) and five anthocyanin (petunidin-3-p-cumaroylrutinoside-5-glucoside, pelargonidin-3-p-cumaroylrutin-oside-5-glucoside, peonidin-3-p-cumaroylrutinoside-5-glucoside, pelargonidin-3-p-feruloyl-rutinoside-5-glucoside and peonidin-3-feruloylrutinoside-5-glucoside)to color potatoes is Hong-young(HY) and Ja-young(JY). The analytical results on organic compounds in color potatoes were shown as follow, The contents of CQA and Anthocyanin of JY variety were shown to be higher than HY, while CQA and Anthocyanin were appeared to be highest in peel of JY. Overall, JY had higher amount of physicochemical properties than HY. The results of this study reveal the quantitative analysis of functional compounds seperated from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers.
