A quantitative GC-MS spectrometric assay was used for the determination of residual N, N-dimethylaniline as a contaminant in commercial penicillin derivatives from various sources. The assay utilizes selective ion focusing to monitor in a GC effluent the molecular ions of DMA generated by electron impact ionization. This method includes dissolution of the sample in alkaline solution, extraction of organic base with cyclohexane and injection into GC-MS with a 3% OV-17 column. Levels of 50 ppb of DMA were easily measured with a coeffecient of varation less than 5 % and recoveries from spiked samples exceeded 97 %. The results of the determinations of DMA in various commercial penicillins were relatively free of this contaminant.
Penicillins and cephalosporins are biosynthesized from L-.alpha.-aminoadipic acid, L-cysteine and L-valine. A tripeptide, LLD-$\delta$-($\alpha$-aminoadipyl)cysteinylvaline(LLD-ACV) was isolated from fermentation broths of Cephalosporium acremonium as well as of Penicillium chrysogenum and it was proved that the LL-$\delta$-($\alpha$-aminoadipyl cysteine was formed first in mycelia, to which valine would be connected to give LLD-ACV. However, several points are still unsolved; first, what mechanism is involved in the configurational change from L-valine to D-valine, second, what kind of cyclization mechanism gives a $\betha$-lactam ring and a thiazolidine ring and third, what is the pathways for the ring expansion from penicillins to cephalosporins. At present, it seems clear that LLD-ACV is cyclized to give isopenicillin N, which is transformed to penicillin N and further to cepbalosporin C. Other hydrophobic penicillins, including benzyl penicillin and penicillin V, are formed from isopenicillin N by acyl-exchange reactions catalyzed by penicillin transferase, rather than by acylation reaction on 6-aminopenicillanic acid(6-APA), which was isolated from the fermentation broth of P. chrysogenum and which would be formed by hydrolysis of $\delta-(\alpha$-amincadipyl)amido moiety at the C-6 position in isopenicillin N or penicillin N by penicillin acylase. Acylation of 6-APA is catalyzed also by penicillin acylase, but the reaction is proved not to be involved in penicillin biosynthesis. Understanding the biosynthesis of penicillins and cephalsoporins would provide solutions to increase in fermentation yields of penicillins, especially of cephalosporins and a solution to biological production of 7-aminocepbalosporanic acid (7-ACA) which is of importance in pharmaceutical industry. Still regulation mechanisms in penicillin and cephalosporin biosynthesis are unveiled at all.
Penicillin antibiotics such as penicillin G, ampicillin and amoxicillin have been widely used in the pig industry to control salmonellosis, bacterial pneumonia, and urinary tract infections. Extensive use of antibiotics in veterinary clinics has resulted in tissue residues and bacterial resistance. To prevent unwanted drug residues entering the human food chain, extensive control measures have been established by both government authorities and industries. The demands for reliable, simple, sensitive, rapid and low-cost methods for residue analysis of foods are increasing. In this study, we established a rapid prediction test for the detection of pigs with unacceptable tissue residues of penicillins. The recommended therapeutic doses of three penicillins, penillin G (withdrawal time, 7 days), ampicillin (withdrawal time, 7 days) and amoxicillin (withdrawal time, 14 days), were administered to three groups of 20 pigs each. Blood was sampled before drug administration and during the withdrawal period. The concentration of penicillins in plasma, determined by a semi-quantitative ELISA, were compared to that of internal standard, 4 ppb, which corresponded to the Maximum Residue Limit in milk. The absorbance ratio of internal standard to sample (B/Bs) was employed as an index to determine whether drug residues in pig tissues were negative or positive. That is, a B/Bs ratio less than 1 was considered residue positive, and larger than 1 negative. All 60 plasma samples from pigs were negative to three penicillins at pretreatment. Penicillin G could be detected in the plasma of the treated pigs until day 4 post-treatment and ampicillin until day 2, whereas amoxicillin could be detected until day 10 of its withdrawal period. The present study showed that the semi-quantitative ELISA could be easily adapted to detect residues of penicillin antibiotics (penicillin G, ampicillin and amoxicillin) in live pigs.
The in vitro activity of penicillins(penicillin G, ampicillin, carbenicillin, methicillin and cloxacillin) and gentamicin to Staphylococcus aureus and coagulase-negative staphylococci isolated from bovine mastitic milk samples was determined. The growth of all isolates of staphylococci tested was inhibited by cloxacillin and methicillin at a concentration of $0.78{\mu}g/ml$ and by gentamicin at a minimum inhibitory concentration (MIC) of $1.56{\mu}g/ml$. Of the 140 strains of Staphylococcus aureus isolated 99(70.7%) gave positive reactions for penicillinase on the starch-iodine test. Of 121 isolates of coagulase-negative staphylococci, 58(47.9%) showed penicillinase production, but all the Staph. xylosus lacked the ability to produce penicillinase. MIC of penicillin G of the penicillinase-positive strains was $0.2{\mu}g/ml$ or more. The combination of cloxacillin with gentamicin showed a synergistic effect by inhibiting regrowth of the tested organism.
The study was designed to determine the estrogenic effect of some penicillins on endocrine function in adult Japanese medaka (Oryzias latipes). Vitellogenin (Vtg) produced in male fish has been used for a biomarker to study endocrine disrupters. $17\beta-estradiol\;(E_2)$ was used a positive control that was induced Vtg in male fish. Result of total protein qantification and ELISA for female and male fish were exposed to $17\beta-estradiol$ 10ng/ml for $3\sim5$ days. As a result, male fish exposed to amoxicillin respectively appeared 0.75, 0.23, 8.21 and $9.36\%_{\circ}$ of 1, 10, 100 and 1000 ppm respectively, that value was elevated compared with control male fish. Male fish exposed to ampicillin respectively appeared 1.85, 4.68, 0.85 and $39.59\%_{\circ}$ of 1, 10, 100 and 1000 ppm respectively, that value was elevated compared with control male fish. This study is one of the first reports suggesting potential endocrine disruption of some penicillins in aquatic ecosystem. These results suggest that vitellogenin and estrogen receptor induction patterns alter in male medaka treated with selected estrogenic compounds, and that these results may be useful molecular biomarkers for screening estrogenic EDCs (endocrine-disrupting chemicals) in the shortest possible time.
Objective: This study was to define the clinical effect on the clinical decision support system (CDSS) for prescribing antibiotics integrated with the order communication system in a National Hospital. Method: We extracted data collected before integrating the CDSS of 4,406 adult patients in 2007 and data collected after integrating the CDSS of 4,278 adult patients in 2009. These patients were 50.4% and 45.2% of all patients admitted in 2007 and 2009, respectively. The clinical effect was defined as the proportion of prescribed antibiotics, the length of antibiotics use, and the DDDs (defined daily doses) of antibiotics per 1,000 patient-days using these retrospective data. Results: There were a significant change in the proportion of patient prescribed penicillins with extended spectrum (OR=0.55, p=001), penicillins included beta-lactamase inhibitors (OR=0.75, p<.001), 3rd cephalosporin (OR=1.47, p<.001). The mean of the length of antibiotics use was decreased statistically from $6.09{\pm}5.48$ to $5.85{\pm}5.51$ days (p=.003). The DDD of glycopeptides was decreased from 24.43 DDD to 19.55 DDD per 1000 patient-days. The DDD of 3rd cephalosporins was also decreased from 15.88 to 11.65. Conclusion: Therefore, the clinical decision support system for prescribing antibiotics was effective for the clinical outcomes.
페니실린계 항생물질은 ${\beta}$-lactam 계열의 항생물질로서 인간과 동물에게 널리 사용되어지는 항생물질이다. 이러한 항생항균물질들의 오남용은 소비자가 항생제에 노출될 위험성을 증가시킨다. 본 연구에서는 국내 유통 중인 축 수산물을 대상으로 LC-MS/MS를 이용하여 페니실린계 동물용의약품 7종인 아목시실린, 암피실린, 벤질페니실린, 클록사실린, 디클록사실린, 나프실린, 옥사실린에 대한 잔류량을 조사하였다. 특이성, 정밀성, 회수율, 직선성을 통해 시험법의 유효성을 확인하였으며 아목시실린을 제외한 나머지 6종은 모두 76% 이상의 회수율을 보였고, RSD는 10% 이하로 나타내었다. 시료는 80% 아세토니트릴로 추출한 후 고체상 추출 카트리지를 사용하여 정제 한 후 분석하였다. 축 수산물 225건을 대상으로 잔류량을 조사한 결과 벤질페니실린이 축산물에서 12건, 수산물에서 7건으로 총 19건이 검출되었고 나머지 6개 항목인 아목시실린, 암피실린, 클록사실린, 디클록사실린, 나프실린, 옥사실린은 검출되지 않았다. 벤질페니실린이 검출된 축산물은 우유와 알을 제외한 소고기, 돼지고기, 닭고기이었으며 검출 수준은 0.001~0.009 mg/kg으로 잔류허용기준(MRL)인 0.05 mg/kg 이하로 검출되었다. 수산물에서 검출된 7건은 잔류허용기준인 0.03 mg/kg 이하로 검출되었다. 본 연구사업에서 수행한 모니터링 결과 해당항목의 경우 대부분 기준치 이하로 검출되어 비교적 안전한 수준인 것으로 나타났으나, 향후 지속적인 잔류실태조사와 제도적인 관리지도를 통하여 동물용의약품의 오남용에 대한 안전관리가 이루어져야 할 것으로 사료된다.
Since ampicillin, one of the important semi-synthetic derivatives of penicillins, is restricted in use because of its unfavorable physico-chemical and pharmacological properties, the author synthesized a new derivative, methampicillin lysinate and its physical and characteristics were determined. The antibacterial activity of this compound against ten species of Gram positive and negative bacteria in vitro showed that it was more active to Gram negative bactria than the positive and that was three times more potent than ampicillin trihydrate.
It is certainly a difficult work to find out novel-type "practical" antibiotic from natural sources. In fact, chemically modified antibiotics, such as synthetic penicillins, cephalosporins, derivatives of tetracycline, lincomycin, rifamycin and kanamycin are most widely used in medical practice at present. However, as the possibility of success gained by chemical modification is limited, it would be the buty of our applied microbiologists to offer novel antibiotics to chemist and physician without interruption.
