An abattoir survey of incidence of pneumonia in slaughter pigs and an investigation of microbiology of affected lungs (도축돈의 폐렴병변 분포조사 및 폐렴병소로부터 호기성균의 분리동정)

  • The present study was conducted to investigate the incidence of pneumonic lesions with special regard to enzootic pneumonia and the microbiology of pneumoic lungs from 544 slaughter pigs during the period from October 1995 to September 1996. The incidence of enzootic pneumonic lesion was 76.3% (41s/s44) and pleurisy was detected from 7.9% of slaughter pigs. Seasonal prevalence of pneumonic lesions in slaughter pigs were in order of prevalence of 82.9% in spring, 76.8% in winter, 74.8% in autumn and 69.0% in summer, respectively. Pasteurella multocida, Streptococcus sp, Str suis, Corynebacterium sp, Actinobacillus pleuropneumoniae, Hemophilus parasuis, and Klebsiella pneumoniae were detected in order of prevalence from 16.9%, 15.9%, 7.5%, 6.0%, 1.4%, 1.0% and 0.5% of 415 pneumonic lungs, respectively. P multocida were susceptible to oxytetracycline, polymyxin-B, streptomycin, and vancomycin, while the majority of them were resistant to amoxicillin, ampicillin, cephalothin, kanamycin, and penicillin-G. Str suis were susceptible to amoxicillin, ampicillin, cephalothin, penicillin-G, although the majority of them were resistant to erythromycin, oxytetracycline, streptomycin, vancomycin. A pleuropneumoniae were susceptible to ampicillin, and cephalothin, but the majority of them were resistant to oxytetracycline.

Sensitivity of Bordetella bronchise ptica Isolated from Pigs Affected by Infectious Atrophic Rhinitis to Chemotherapeutic Agents (전염성위축성비염돈(傳染性萎縮性鼻炎豚)으로부터 분리(分離)한 Bordetella bronchiseptica의 화학요법제(化學療法劑)에 대(對)한 감수성시험(感受性試驗))

  • A total of 98 strains of Bordetella bronckiseptica isolated from pigs affected the infectious atrophic rhinitis(AR) during 1978 were surveyed for drug sensitivity to 26 chemotherapeutic agents, and minimum inhibitory concentration(MIC), incidence rate of resistant strain and resistant patern from the strains which were obtained from the different pig farm in Jeonnam province were examined. The results obtained are summarized as follows. 1. Most of the strains tested were resistant to Ampicillin (AB, PC), spiramycin(SPO, sulfa drugs (SD) (MIC:$400.0{{\mu}g/ml}$) and streptomycin(SM) (MIC:$200.0{{\mu}g/ml}$). Of the 75.0% of strains were also resistant to penicillin(PC) (MIC:$200.0{{\mu}g/ml}$) and of the 14.3 of strains were inhibited to grow to tetracycline(TC), chlortetracycline(CTC), oxytetracyc-line(OTC), erythromycin(EM), tylosin(TS), leucomycin (LM) and chloramphenicol (CP) (MIC:$6.25{{\mu}g/ml}$). On the other hand, most of the strains tested were inhibited to grow to kanamycin(KM), gentamycin(GM) neomycin(NM) (MIC:$25.0{{\mu}g/ml}$) and to colistin(CL) (MIC:$12.5{{\mu}g/ml}$). 2. Incidence rate of resistant strains to main chemotherapeutic agents was 100.0% of sulfa drugs, 96.4% of streptomycin, 85.7% of penicillin, tetracycline, chloramphenicol and erythromycin, 46.4% of gentamycin, 17.9% of colistin and 0.0% of kanamycin and nalidixic acid.

  • Tracheal bronchus is an aberrant, accessory or ectopic bronchus arising almost invariably from the right lateral wall of the trachea and may be related to inflammatory conditions affecting the lung, including recurrent pneumonia, bronchiectasis. Recently we experienced a case of tracheal bronchus associated with pulmonary actinomycosis. The 37-year-old male patient had suffered recurrent hemoptysis and had been medicated as a presumptive diagnosis of tuberculosis, but either clinical or radiologic improvement was not seen. Right upper lobectomy was performed and pulmonary actinomycosis was confirmed by the histologic examination. Postoperatively, the patient was medicated with penicillin and ampicillin for 3 months and completely recovered without any evidence of recurrence during the 6month follow-up period.

  • In order to develope a protoplast fusion system for industrial coryneform bacteria, the optimum conditions for the formation and regeneration of progoplast were examined for Brevibacterium flavum and Corynebacterium glutamicum and the protoplast fusion was performed. For the formation of the protoplast of B. flavum and C. glutamicum, the optimum time for penicillin G. treatment to obtain protoplast was mid-exponential growth phase ($O.D_{580}=0.6-0.8,\;8.0{\times}10^7-1.0{\times}10^8cell/ml$). At the optimum conditions (0.3units/ml penicillin G and $400{\mu}g/ml$ lysoyme for treatement), frequencies of protoplast formation and protoplast regeneration were 99% and 25%, respectively. Protoplast regeneration frequency was highest under the optimum conditions for the protoplast formation. Addition of 25mM $Mg^{2+}\;and\;50mM\;Ca^{2+}$ to the regeneration medium further increased the regeneration frequencies. The protoplast fusion frequencies of B. flavum and C. glutamicum in intraspecies fusion were $1.0{\times}10^{-8}\;and\;7.8{\times}10^{-4}$, of the regenerated protoplast respectively, when 33% of PEG (polythylene glycol) 6,000 was used as the fusing agent.

  • To investigate the epidemiological aspects of listeriosis, serotypes of L monocytogenes and antimicrobial susceptibilities of Listeria spp isolated from chicken carcases and chicken slaughter house environmental specimens were determined. Of 28 L monocytogenes strains, 12 strains(42.9%) were serotype 4, and the remaining 16 strains were untypable. Peak distributions of minimum inhibitory concentration$({\mu}g/ml)$ of the isolates were $0.78{\mu}g/ml$ for ampicillin, $0.39{\mu}g/ml$ for erythromycin and penicillin G, $1.56{\mu}g/ml$ for tetracycline and $6.25{\mu}g/ml$ or $12.5{\mu}g/ml$ for chloramphenicol, and $3.13{\mu}g/ml$ to > $100{\mu}g/ml$ for kanamycin and neomycin. Most of 214 isolates were sensitive to ampicillin and erythromycin, but 20. 1~78. 0% of the strains were resistant to tetracycline, kanamycin, penicillin-G and neomycin. Single or double drug resistance were observed in 75.8% of the resistant strains. The most common resistance patterns were Nm P-G(37.4%) in double pattern and P-G(23.7%) in single pattern.

Antimicrobial Susceptibility Patterns of Staphylococcus intermedius Isolated from Skin Lesions of Thoroughbred Horse. (더러브렛 말의 피부병변에서 분리한 Staphylococcus intermedius의 약제 감수성 양상)

  • Staphylococcus intermedius (S. intermedius) has been rarely isolated from horse. In this study, we investigated the antimicrobial susceptibility patterns of S. intermedius isolated from skin lesions of Thoroughbred horse in Daegu Equestrian Association. The skin lesions were showed with dehairing and slight purulent inflammation. Bacteria were isolated from skin lesions and identified as S. intermedius by biochemical tests and MicroLog (BIOLOG, California, USA). Antimicrobial susceptibility test of S. intemedius isolates was performed with 33 antimicrobial agents (BBL, Maryland, USA) by using the agar disk diffusion method. It showed a high sensibility in the amikacin, amoxycillin/clavuramic acid, ampicillin, cefoxitin, cephalothin, chloramphenicol, ciprofloxacin, enrofloxacin, florfenicol, kanamycin, neomycin, nitrofurantoin, norfloxacin, ofloxacin, oxacillin, penicillin G, tetracycline and vancomycin. The horse was treated with penicillin, and cured completely after two weeks. The present results showed valuable information for treatment and prevention of skin disease in horse.

  • Actinomycosis is a chronic suppurative and granulomatous bacterial infection characterized by contiguous spread, abcess formation and sinus tract formation. There are four clinical forms according to the lesional site, as 1) cervicofacial, 2) thoracic, 3) abdominal, and 4) disseminated form. Recently, we experienced a case of 54 year-old patient with left mandibular actinomycosis. The pathognomonic findings of actinomycosis is sulfur granule with multiple filaments in Gram-stain and the treatment of actinomycosis is surgical excision of mass or sinus tract with massive antibiotics(esp. Penicillin) therapy for 6 to 12 months.

Degradation of Clavulanic Acid During the Cultivation of Streptomyces clavuligerus; Instability of Clavulanic Acid by Metabolites and Proteins from the Strain

  • Clavulanic acid (CA) produced by Streptomyces clavuligerus is degraded during the bacterial cultivation. The degradation was examined in three different aspects, including physical, chemical, and enzymatic effects, in order to understand the degradation during the cultivation. The result showed that CA was unstable in the production medium containing ammonium salts and amino acids, owing to ammonium ions and amine groups. In addition, the degradation was not only due to instability of CA by metabolites and proteins, but also enzymes from S. clavuligerus such as $\beta-lactamase$ and penicillin-binding proteins. However, the degradation caused by these enzymes was not highly significant compared with the degradation during the cultivation, owing to irreversible reactions between CA and enzymes.

  • Monofunctional biosynthetic peptidoglycan transglycosylase (MBPT) catalyzes the formation of the glycan chain in bacterial cell walls from peptidoglycan subunits: N-acetylglucosamine (NAG) and acetylmuramic acid (NAM). Bifunctional glycosyltransferases such as the penicillin binding protein (PBP) have peptidoglycan glycosyltransferase (PGT) on their C terminal end which links together the peptidoglycan subunits while transpeptidase (TP) on the N terminal end cross-links the peptide moieties on the NAM monosaccharide of the peptide subunits to create the bacterial cell wall. The singular function of MBPT resembles the C terminal end of PBP as it too contains and utilizes a similar PGT domain. In this article we analyzed the infectious and non infectious protein sequences of MBPT from 31 different strains of bacteria using a variety of bioinformatic tools. Motif analysis, dot-plot comparison, and phylogenetic analysis identified a number of significant differences between infectious and non-infectious protein sequences. In this paper we have made an attempt to explain, analyze and discuss these differences from an evolutionary perspective. The results of our sequence analysis may open the door for utilizing MBPT as a new target to fight a variety of infectious bacteria.

Cytotoxicity of Ethanol Extract of Cephaxus koreana on Human Breast Cancer Cell Lines (유방암 세포주를 애용한 개비자 나무의 에탄올 추출물의 항암 활성 분석)

  • This study was designed to investigate the cytotoxic effect of Cephaxus koreana on human breast cancer cell lines (SCC-1395 and SK-BR-3) us ins MTT(3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. RPMII -1640 medium with $10\%$ FBS and $1\%$ of penicillin-streptomycin was used. The ethanol extrart of Cephaxus koreana after adsoption resin treatment showed higher anticancer activity against SCC- 1395 than the ethanol extract of Cephaxus koreana without adsoption resin treatment. SK-BR-3 showed higher sensitivity activity than SCC-1395 at the same concentration of 20000 ppm.
