Statistical Optimization of Medium Composition for Bacterial Cellulose Production by Gluconacetobacter hansenii UAC09 Using Coffee Cherry Husk Extract - an Agro-Industry Waste

  • During the production of grape wine, the formation of thick leathery pellicle/bacterial cellulose (BC) at the airliquid interface was due to the bacterium, which was isolated and identified as Gluconacetobacter hansenii UAC09. Cultural conditions for bacterial cellulose production from G. hansenii UAC09 were optimized by central composite rotatable experimental design. To economize the BC production, coffee cherry husk (CCH) extract and corn steep liquor (CSL) were used as less expensive sources of carbon and nitrogen, respectively. CCH and CSL are byproducts from the coffee processing and starch processing industry, respectively. The interactions between pH (4.5-8.5), CSL (2-10%), alcohol (0.5-2%), acetic acid (0.5-2%), and water dilution rate to CCH ratio (1:1 to 1:5) were studied using response surface methodology. The optimum conditions for maximum BC production were pH (6.64), CSL (10%), alcohol (0.5%), acetic acid (1.13%), and water to CCH ratio (1:1). After 2 weeks of fermentation, the amount of BC produced was 6.24 g/l. This yield was comparable to the predicted value of 6.09 g/l. This is the first report on the optimization of the fermentation medium by RSM using CCH extract as the carbon source for BC production by G. hansenii UAC09.

Histopathologic Observation of the Aborted Fetus from Pregnant Dairy Cows Naturally Infected with Neospora caninum (Neospora caninum에 자연 감염된 임신우로부터 유산된 태아의 병리조직학적 관찰)

  • This study examined the histopathologic and electron microscopic findings of aborted fetuses from pregnant dairy cows naturally infected with Neospora caninum (N. caninum) at four farms in Gongju city and Yeonki gun of Choongnam province. Systemic subcutaneous edema was observed in the aborted fetuses. The necropsy revealed considerable serosanguinous fluid in the body cavity of the aborted fetuses. Light microscopy showed the infiltration of many inflammatory cells consisting of macrophages, lymphocytes and mononuclear cells, accompanied by congestion, hemorrhage and necrosis of myocardiac cells and hepatocytes in the liver and heart of the aborted fetuses. In the liver, clusters of tachyzoites were formed in the cytoplasm of hepatocytes and the interstitial tissue. In the brain, many tissue cysts of various sizes were observed in the nerve cells and their adjacent areas. Tissue cysts had a round shape and contained a large amount of bradyzoite. In addition, there was diffuse gliosis accompanied by congestion and hemorrhage and focal necrosis in the brain. Infiltration of microglial cells were observed at the periphery of the focal necrosis and perivascular area in the brain. Electron microscopy showed that the tissue cyst wall had a thickness of approximately 1 ${\mu}m$ with an irregular shape. On the interior side, more than 100 bradyzoites with lengths of 2-5 ${\mu}m$ and widths of 1-2 ${\mu}m$ were observed. The nucleus of in the bradyzoites was located approximately 1-1.5 ${\mu}m$ anterior to the posterior tip of the zoite. In the cytoplasm between the nucleus and the posterior tip, there were many amylopectin granules, electron-dense small-sized and electron-thin large-sized round granules, homogeneously electron-dense rhoptries and micronemes oriented perpendicularly to the zoite pellicle. To summarize, tissue cysts were identified on electron microscopy from the aborted fetus from N. caninum seropositive pregnant cow by the ELISA. This led to the confirmed presence of N. caninum.

THE STUDY OF THE DENTAL PLAQUE FORMATION ON DIFFERENT IMPLANT SURFACES (임플란트 표면에 따른 인공치면세균막 형성에 관한 연구)

  • The purpose of this study was to evaluate the artificial dental plaque by Streptococcus mutans on 4 different implant surfaces. In this study, the specimens were divided into 4 groups according to implant surface treatment. Uncoated implant group(n=5) which has an uncoated, smooth surfaced implant(Osstem, Korea), SLA implant group(n=5) which has an sandblasted large grit and acid-etched surface implant(Bicon, USA). Oxidized implant group(n=5) which has an oxidized surfaced implant (Osstem, Korea), and RBM implant group(n=5) which has resorbable blasting media(RBM) surfaced implant(Osstem, Korea). Acquired pellicle by human saliva and dental plaque by Streptococcus mutans were made on each implant surface. To analyze the plaque condition on implants surfaces, cell count and optical density were taken as a microbiologic method, and SEM(Scanning Electronic Microscope) findings was also taken for evaluation of surface condition. The following results were obtained. 1. Cell counting results of artificial dental plaque were Uncoated group($658.0{\pm}102.0$), RBM group($878.0{\pm}170.0$), SLA group ($946.0{\pm}42.0$), Oxidized group($992.0{\pm}40.0$), and there was difference between Oxidized group and Uncoated implant group(p<0.05). In case of modified cell counting results by v/w% were RBM group($197.8{\pm}45.2$), Oxidized group($207.04{\pm}8.34$), Uncoated group($261.6{\pm}40.6$), SLA group($315.4{\pm}14.0$), and there was difference between RBM group and SLA group(p<0.05). 2. Optical density results of artificial dental plaque after ultrasonic treatment was that there was difference among groups, and optical density of RBM group was higher than that of Uncoated group(p<0.05). In case of modified optical density results by v/w%, there was difference among groups, and the modified optical density of Uncoated group and SLA group was higher than those of Oxidized group and RBM group(P>0.05). 3. SEM findings of artificial dental plaque on the surfaces of implant as follows; there were artificial dental plaque on the surfaces of all test implants. Streptococcus mutans and by-product were observed at 10,000 times magnified condition on all test implants. Adhesion area of artificial dental plaque was about 1/2 of total surface after 24 hours incubate at $37^{\circ}C$. These results showed that there were differences among implant surfaces on the growth of Streptococcus mutans, and bacteria and by-product were covered about 1/2 area of total implant surfaces at 24 hours incubate at $37^{\circ}C$.

A study for the standardization of dental hygiene terms (치위생 용어 표준화를 위한 방안 연구)

  • Objectives : The purpose of this study was to compare and analyse periodontal terminology of 16 dental hygiene textbooks including periodontology, dental prophylaxis for the standardization. Methods : 16 kinds of major publications were classified into the following criteria. - the standardization of translation, orthography, classification Results : 1. There are twelve cases, all told, of words with same origin used as different explanation and interpretation. Mucogingival Junction is most translated as '치은점막 경계' with high percentage of 53.8%, while Probe is translated as '치주낭 측정기' with 41.7% and Acquired Pellicle is translated as '획득피막' with 57.1%. In addition, Stippling is interpreted as '점몰' with high percentage of 70%, and Supine Position is interpreted as '수평자세' or '앙와위' with 33.3%. Yet, Pen Grasp is explained differently in all five books, whereas Modified Pen Grasp is explained as '변형 연필 잡기법' with 57.1%. The rest, Palm Thumb Grasp and Adaptation, have different interpretation in books, requiring unified interpretation of terms. 2. There are two cases of loanwords expressed according to orthography. Implant is expressed as '임플란트'(87.5%) and '임프란트'(12.5%). Scaling is expressed as '스켈링'(40.0%) and '스케일링'(60.0%). 3. There are four cases of same terms classified in a different way. Classification of 'Probing시 압력' and '치주인대섬유군' concurs only 50.0% in six books. '치석제거 시술자의 위치' is classified differently in all six books, while '정상치은 열구 깊이' concurred 45.5% in books. Conclusions : In conclusion, related academic majors and writers should provide cohesive explanation and interpretation of terms for unification of terminology in the field of periodontics as well as higher level of understanding and learning opportunities for students. Furthermore, description of all terms should be reviewed and analyzed comprehensively based on existing classification criteria in order to provide best standard explanation.

Inhibitory Effects of Galla Chinensis Extract on Cariogenic Properties of Streptococcus mutans (오배자 Galla Chinensis 추출물이 Streptococcus mutans의 우식활성 억제에 미치는 영향)

  • Streptococcus mutans (S. mutans) is one of the most important bacteria in the formation of dental plaque and dental caries. S. mutans adheres to an acquired pellicle formed on the tooth surface, and aggregates with many oral bacteria, and initiates plaque formation by synthesizing glucan from sucrose, which is catalyzed by glucosyltransferases. S. mutans metabolizes the dietary sugar to the organic acids. The organic acids demineralize tooth surface and result in dental caries. Galla Chinensis have been traditionally used for stopping bleeding of gingiva, removing edema and halitosis, drainage, fixing the teeth and as an antiphlogistic agent. In previous reports, antibacterial effects of Galla Chinensis have been investigated whereas anti-cariogenic effects is still not examined enough. Therefore we tested effects of ethanol extracts of Galla Chinensis on the cariogenic properties such as the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In the result, ethanol extracts of Galla Chinensis showed the inhibition of S. mutans growth and organic acids production over 0.031 mg/ml concentrations. The adhesion of S. mutans to Saliva-coated Hydroxyapatite beads S-HAs has decreased with the increase of concentration of ethanol extracts of Galla Chinensis. And it seems to have adhesion inhibitory effect in concentration of over 0.25 mg/ml. It gives us the result that Galla Chinensis have anti-caries effects. But ethanol extract of Galla Chinensis didn't have inhibitory effect on insoluble glucan synthesis. Preliminary phytochemical analysis of the ethanol extract of Galla Chinensis showed strong phenolic compounds, medium steroids & terpenoids and glycosides, and weak organic acids and peptides. These results suggest that the ethanol extracts of Galla Chinensis may have anti-cariogenic properties, which may be able to be related with strong phenolic compounds.

COMPARATIVE STUDY OF REMOVAL EFFECT ON ARTIFICIAL PLAQUE FROM RBM TREATED IMPLANT (RBM 처리된 임플란트 표면의 인공치태 제거 효과 연구)

  • Purpose: This study was to evaluate the removal effect on artificial plaque from RBM treated implant surfaces that are exposed due to peri-implantitis. Materials and methods: Artificial plaque with Streptococcus mutans and acquired pellicle adhered to RBM treated implant discs. Study materials divided into one control and six test groups. In test groups, physical and chemical methods used to remove plaques. Prophyflex, Professional Mechanical Tooth Cleaning (PMTC) and interdental brush as mechanical treatments and 0.1% Chlorhexidine, Citric acid, HCl tetracycline as a chemical treatment were used. To analyses the study, disc weight was measured for remaining plaque quantities and SEM(Scanning Electronic Microscope) findings was taken for evaluation of surfaces. Results: 1. In weight changes, there was significant difference between each treatment group and the control group (p<0.05). Therefore all treatment methods using this study have good ability for remove plaques. 2. In weight changes, there was no significant difference between mechanical and chemical group, and there were no significant differences between each groups (p>0.05). 3. SEM findings after mechanical treatment disclosed as follows; Prophyflex group looked like sound implant surface, and there were some paste on implant surface at PMTC group, and there were some artificial plaque at interdental brush group. 4. SEM findings after chemical treatment disclosed as follows; there were some dark lesions which were supposed as the product from Streptococcus mutans at Chlorhexidine, Citric acid and HCl tetracycline groups. Conclusion: All six methods using in this study have good ability to remove artificial plaque on RBM treated implant. According to SEM findings, prophyflex is a superior method for removing of dental plaque among test groups.

Studies on the Yeasts for the Brewing of Soy Sauce(3) -Isolation and Identification of Osmophilic Yeasts for Higher Concentration of NaCl (간장발효에 관여하는 효모에 관한 연구(제3보) -고농도 식염내성 효모의 분리동정-)

  • This experiment was carried out to isolate osmophilic yeasts for higher concentration of NaCl than 20 per cent may affect greatly the fermentation of soy sauce. 6 strains of yeasts were selected from the soy sauce mashes at the fermentating periods and identified observing their morphological characteristics. Their salt-resistance and effects upon the flavor of soy sauce were examined. The results obtained were as follows: (1) The strain $T_3,T_5,$ and $T_{11},\;T_8,\;T_9,\;and\; T_{10},$ selected were identified Saccharomyces cerevisiae group II, Debaryomyces nicotianae, Saccharomyces chevalieri, Saccharomyces rouxii and Saccharomyces rosei, respectively. (2) They were more salt-resistant on the liquid media than on the solid media in the case of cultures on the media containing more than 26 per cent of NaCl. (3) They were grown on the media containing 5-23 per cent of NaCl better than containing none of NaCl. (4) The strain $T_3,\;T_8,\;T_9,\;and\; T_{10}$ were grown on the media containing up to 28 per cent of NaCl, and $T_5\;and\;T_{11}$ up to 32 per cent of NaCl is the case of liquid cultures. (5) Among them, the strain $T_9$ showed the best alcohol fermentation ability and the best fermentative flavor, and $T_5\;and\;T_{11}$ formed moldy smell and pellicle.

Development of Fermented Acidic Beverage using Wild Grape Juice (산머루과즙을 이용한 발효산형음료 개발)

  • Wild grape juice was fermented by Gluconacetobacter hansenii TF-2 isolated from tea fungus, to develop a new acidic beverage (fermented wild grape beverage, WGB). Broth was prepared by fermentation of 11~17% (v/v) juice, and sweetened with sucrose (initial sucrose level: $10^{\circ}$ Brix). Fermentation was initiated by addition of 5% (w/v) seed gel (the pellicle of the tea fungus) which had been previously cultured in the same medium (freshjuice broth), and fermentation proceeded in the dark at $29{\pm}1^{\circ}C$ for about 15 days. The major acids produced were succinic acid, malic acid, and acetic acid. After 15 days of fermentation, the organic acid content (principally succinic acid) was 49.6 ppm in WGB 11 and 77.4 ppm in WGB 17. The free sugar content of WGB was 1063.6-1082.5 mg/mL, composed of unfermented fructose, glucose, and sucrose, in that order. The microbial inhibitory effects of the fermented beverage were most apparent when Gram-negative bacteria (Escherichia coli and Salmonella typhimurium) were tested; the inhibition rate was 34.46-88.00%. The new fermented beverage thus displays effective antimicrobial activity against some species of bacteria.

Adsorption of Salivary Proteins on Titanium Implants (타이태늄 임플란트 표면에 형성된 타액성 단백질에 관한 생체연구)

  • 치과용 임플란트 실패의 주요 원인은 임플란트 표면에 부착되는 세균의 침착의 결과로 생기는 임플란트 주위염이다. 구강 내에서 세균성 치태의 침착은 치태가 부착하는 기질 표면의 물리적 성상과 타액성 피막의 성분에 영향을 받으며 형성된 피막의 유기질 성분의 차이가 치태의 성분과 병원성에 영향을 미친다. 최근 연구에 의하면 생체재료의 표면에 침착되는 치태세균은 사용되는 재료에 따라 특이한 세균 침착을 보이며 이는 초기 타액성 피막의 차이에 의한 것으로 알려져 있다. 이 연구의 목적은 플라즈마분사법으로 표면 처리된 타이태늄 임플란트에 흡착되는 타액성 단백질 피막의 특성을 정성적인 방법으로 분석하는 데 있었다. 법랑질 조각과 플라즈마분사법으로 표면 처리된 타이태늄 임플란트를 스프린트에 치실을 이용하여 연결한 장치를 구강 내 장착하여 2시간 동안 피막이 침착되게 한 후 피막을 분리 추출하여 냉동 건조시켰다. 재수화 과정을 거치고 나서 전기 영동법과 Western transfer 분석을 통해 단백질 성분에 관한 분석을 시행하였다. 사람의 총 타액과 이하선 타액 및 악하선-설하선 타액을 수집기를 이용하여 채취하고 같은 방법으로 처리한 후 성분분석을 실시하였다. 피막 흡착 전후의 표면변화를 주사전자현미경을 이용하여 관찰하였다. 실험결과 타이태늄 임플란트에 흡착된 피막은 법랑질 표면의 피막과는 다른 단백질 성분을 가지고 있었으며, 주로 악하선-설하선 타액에서 유래하였다. 임플란트와 법랑질 표면 모두에서 흡착된 피막에는 아밀라제, 분비성 면역 글로불린A 및 락토페린이 존재함을 알 수 있었으나 법랑질의 경우는 blotting이 약하게 나타났다. 주사전자현미경 관찰결과 시편의 표면에 균질한 피막이 덮고 있었으며 세균의 부착은 거의 관찰되지 않았다. 이상의 실험 결과들을 통하여 플라즈마분사법으로 표면 처리된 타이태늄 임플란트 표면에 부착된 타액성 단백질 성분은 법랑질과는 차이가 있음을 알 수 있었으며, 이러한 차이는 치태세균의 종류 및 병원성에 영향을 미칠 것으로 생각된다. 법랑질과 타이태늄 임플란트는 기질과 표면구조가 다르므로 표면에 형성 되는 치태성분도 다르다는 사실과 본 연구 결과를 종합하여 볼 때, 타이태늄 임플란트 표면에 흡착되는 초기 타액성 단백질의 성불이 타이태늄 표면에 침착되는 미생물 군의 조절에 중요한 역할을 가지고 있으며, 임플란트 치료 시에 올바른 치태 관리법의 교육을 통하여 환자 스스로 적절한 관리를 하도록 함으로써 임플란트 치료의 성공률을 높일 수 있을 것으로 생각된다.

Generation of a monoclonal antibody against AgI/II, a cellular surface protein of Streptococcus mutans GS5 (Streptococcus mutans GS5의 세포막 단백질 Ag I/II에 대한 단항체의 생산)

  • Most of oral streptococci express the Antigen I/II (AgI/II) proteins, cell wall anchored adhesions. AgI/II protein binds to salivary agglutinin glycoprotein, a component of tooth pellicle and to ligands in other bacteria. These associations play important roles in bacterial colonization. Recently, it was reported that diverse host molecules also interact with AgI/II protein and that these interactions induce inflammatory responses from host cells. Among mutans streptococci containing -type hemolytic activity, Streptococcus mutans is a causative agent for dental caries. Compared with many other strains of S. mutans, GS-5 strain is unique in that this bacterium expresses truncated secretory AgI/II protein due to the nonsense mutation in the agI/II gene. This indicates that S. mutans GS-5 has a different clinical role and a recent report supported this idea based on the results from clinically isolated S. mutans strains. Previously, we had cloned agI/II gene from S. mutans GS-5 and generated recombinant N-terminal AgI/II protein. In this study, we further produced a hybridoma line expressing anti-AgI/II monoclonal antibodies named as 1C11A. This antibody showed high sensitivity to AgI/II protein in Western blot and ELISA. This new reagent will provide a basis for investigating the mechanisms of AgI/II-related diseases.
