Fenchone Ameliorates Constipation-Predominant Irritable Bowel Syndrome via Modulation of SCF/c-Kit Pathway and Gut Microbiota

  • In this study we sought to elucidate the therapeutic effects of fenchone on constipation-predominant irritable bowel syndrome (IBS-C) and the underlying mechanisms. An IBS-C model was established in rats by administration of ice water by gavage for 14 days. Fenchone increased the reduced body weight, number of fecal pellets, fecal moisture, and intestinal transit rate, and decreased the enhanced visceral hypersensitivity in the rat model of IBS-C. In addition, fenchone increased the serum content of excitatory neurotransmitters and decreased the serum content of inhibitory neurotransmitters in the IBS-C rat model. Meanwhile, western blot and immunofluorescence experiments indicated that fenchone increased the expressions of SCF and c-Kit. Furthermore, compared with the IBS-C model group, fenchone increased the relative abundance of Lactobacillus, Blautia, Allobaculum, Subdoligranulum, and Ruminococcaceae_UCG-008, and reduced the relative abundance of Bacteroides, Enterococcus, Alistipes, and Escherichia-Shigella on the genus level. Overall, fenchone ameliorates IBS-C via modulation of the SCF/c-Kit pathway and gut microbiota, and could therefore serve as a novel drug candidate against IBS-C.

Multi-locations and stability evaluation on growth character of the permata hybrid carp

  • The success of establishing the Indonesian growing fast hybrid carp, namely "Permata", on a controlled environmental test must be followed up with a large-scale test. This study aims to evaluate the phenotypic performance of the Permata hybrid carp in multi-locations with different cultivation systems. The test sites consisted of floating net cages, running-water ponds, semi-concrete ponds, earthen ponds, fully concrete ponds, and static net cages. For 90 days, fish were fed commercial pellets with a 28%-30% protein content. At the end of the test, all fish were harvested and counted. Data on length, weight, survival rate, and harvested biomass were used to analyze the effect of genotype, environment, and their interaction on the phenotypic performance. The growth based on final weight is used to analyze the stability performance in each test location. The results showed that the length and weight of common carp were significantly affected by genotype and the environment, but not by the interaction of both. The genotype, environment, and the interaction of both factors affected common carp's survival and harvested biomass. Common carp reared in floating net cages generally had the best performance, while carp reared in fully concrete tanks and static net cages had the lowest. The growth stability analysis showed that the common carp in this study were unstable genotypes but have a broad adaptability in term of different environments.

Effects of Medicinal Herb Extracts of Artemisia iwayomogi Kitamura and Angelica gigas Nakai on Disease Resistance in Whiteleg Shrimp Litopenaeus vannamei (양식 흰다리새우(Litopenaeus vannamei)의 혼합생약재 투여에 따른 항병력 조사)

  • Although aquaculture shrimp production has increased, disease mortality has also increased. Acute hepatopancreatic necrosis disease (AHPND) is an emerging disease that significantly reduces production. In this study, we have investigated the potential of medicinal herbs in enhancing the immune system of shrimp. A mixture of medicinal herb extracts (Han In-jin, Artemisia iwayomogi kitamura and Cham Dang-gwi, Angelicae gigas Nakai) was absorbed into pellets and fed to whiteleg shrimp Litopenaeus vannamei for 12 weeks. The weight gain, immune activity, and disease resistance effects of the shirmp were examined. The growth performance of shrimp fed the mixture of medicinal herbal extracts was higher than that of the control group. The prophenoloxidase (proPO) activity was found to be significantly higher at 8 weeks in the 0.1% medicinal herb extract fed group. The cumulative mortality when artificially infected with AHPND pathogens commonly decreased in the 8 weeks fed group when compared with the control group. Furthermore, the RPS (relative percentage survival) was improved. This study confirmed that a mixture of medicinal herbal extracts has a positive effect on growth performance, innate immunity and disease resistance in shrimp. The optimum concentration of the medicinal herb extract for shrimp feed was 0.1% .

Optimum Dietary Lipid Level and Feeding Rates of Extruded Pellets in Juvenile Flounder Paralichthys olivaceus during the Summer Season (여름철 넙치치어 배합사료의 적정지질함량 및 공급량)

  • We evaluated the optimum dietary lipid level and feeding rates of extruded pellets (EP) in juvenile flounder Paralichthys olivaceus during the summer season. The first experiment was conducted to determine the optimum dietary lipid level in juvenile flounder. Five isonitrogenous EP (52% crude protein) with increasing dietary lipids (6, 8, 10, 12 and 14%) were fed to satiety to triplicate groups of the juveniles ($18.4{\pm}0.11g$) twice a day for 6 weeks. Weight gain (WG) of fish fed EP with 10% lipid was significantly higher than those of fish fed EP with 6 and 14% lipid (P<0.05). Broken line model analysis suggested that the optimum dietary lipid level could be $9.08{\pm}0.37%$ for the maximum WG in juvenile flounder. The second experiment was conducted to determine the optimum feeding rate using experimental diet contained 10% lipid level that had the highest WG in first experiment. The feed intake of triplicate groups of the juveniles ($13.4{\pm}0.15g$) was restricted to four different feeding rates of 2.0, 2.5, 3.0 and 3.5% of their body weight $day^{-1}$ and to satiation using experimental diets. WG and feed efficiency (FE) of fish was affected by feeding rates. WG and specific growth rate of fish fed the diets increased with increasing feeding rate, however no significant differences (P>0.05) in WG and specific growth rate were observed between the fish fed 3.5% of their body weight $day^{-1}$ and to satiation. FE of fish fed 3.5% of their body weight $day^{-1}$ was significantly higher than those offish fed 2.0% of their body weight $day^{-1}$ and to satiation (P<0.05). Broken line model analysis suggested that the optimum dietary feeding rate could be $3.56{\pm}0.06%$ for the maximum WG in juvenile flounder. These results indicated that the optimum lipid level and feeding rate could be $9{\sim}10%$ (Energy: 4,774kcal and PIE ratio: 108mg protein/kcal in diet) and 3.5% of their body weight $day^{-1}$in juvenile flounder, respectively.

Effect of Extruded and Moist Pellets at Different Feeding Rate on Growth and Body Composition of Juvenile Flounder, Paralichthys olivaceus (부상 건조사료(EP) 습사료(MP)의 만복 및 제한 공급이 넙치 치어의 성장 및 체조성에 미치는 영향)

  • A 10-week feeding trial was conducted to determine the optimum feeding rate of two extruded pellets (EP1 and EP2) and raw fish-based moist pellet (MP) for juvenile flounder. Two replicate groups of the fish (average weight of 27 g) were fed EP and MP three times daily at different feeding rate (satiation and $85\%$ satiation). Weight gain of fish was not affected by diet, but affected by feeding rate. Weight gain with satiation feeding group showed :t tendency to higher compared to that of fish with $85\%$ satiation feeding at the same diet. Feed efficiency of fish was affected by diet, but not feeding rate. Feed efficiency of fish fed the EP1 and EP2 was higher than that of fish fed the MP at the same feeding rate. Daily feed intake was affected by diet and feeding rate. Daily feed intake of fish fed with satiation was higher than that of fish with $85\%$ satiation at same diet, and this variable of fish fed the MP was higher than that of fish fed the EP 1 and EP2. Protein efficiency ratio of fish fed the MP was significantly lower than that of fish fed the EP1 and EP2 regardless of feeding rate. Lipid content of whole body was affected by diet, and that of fish fed the EP2 and MP at satiation feeding was the highest. It can be concluded that satiation feeding will be good and EP is better than MP for growth of juvenile flounder grown from 27 g to 51 g.

Effects of Dietary Mulberry Leaf on Loperamide-Induced Constipation in Rats (식이뽕잎이 흰쥐의 Loperamide로 유도된 변비에 미치는 영향)

  • We investigated the effect of dietary mulberry leaf powder (MP) on loperamide-induced constipation in rats. Male Sprague-Dawley rats were given MP in their diets at a concentration of 0% 5% and 10% for 33 days. Rats were divided into 4 groups: normal diet group (NOR), normal diet and loperamide treated group (MPL0), 5% MP and loperamide treated group (MPL5), and 10% MP and loperamide treated group (MPL10). Constipation was induced by subcutaneous injection of loperamide (1.5 mg/kg body weight/day) for the final 5 days of the experiment Supplemental MP had no effect on the food efficiency ratio, but it reduced body weight gain and food intake in a concentration dependent manner. Administration of loperamide decreased food intake. MP had a concentration-dependent effect on decreasing total cholesterol, LDL-cholesterol, and triglycerides and on increasing HDL-cholesterol. Loperamide had no significant effect on serum lipid profiles. Loperamide decreased the number and wet weight of fecal pellets and fecal water content MP increased the number and wet weight of fecal pellets and fecal water content in a dose-dependent manner. In addition, MP increased gut transit time and transit speed, and the guts of mts treated with MP plus loperamide were longer than those of mts treated with loperamide alone. These results indicate that MP is an effective treatment for constipation.

Evaluation of Extruded Pellets as a Growing Diet for Adult Flounder Paralichythys olivaceus (넙치 성어기 배합사료 및 생사료의 사육효과 비교)

  • This study was conducted to evaluate extruded pellets (EP) for growth of adult flounder by comparing with raw fish-based moist pellet (MP). Two replicate groups of 150 fish per each tank (initial mean weight 329 g) were fed one of seven EP (EP1, EP2, EP3, EP4, EP5, EP6 and EP7) and a MP for 8 months. Survival of fish fed the MP was not significantly different from that of fish fed the EP1, EP5 and EP7, but significantly higher than that of fish fed the EP2, EP3, EP4 and EP6 (P<0.05). Weight gain of fish fed the MP was significantly lower than that of fish fed the EPI (P<0.05), but not significantly different from that of fish fed EP2, EP3, EP4, EP5, EP6 and EP7. Feed efficiency of fish fed the MP was significantly lower than EP1, EP3, EP4, EP5 and EP6 (P<0.05), but not significantly different from that of fish fed EP2 and EP7 Feed supply (kg/tank) of fish fed the MP was significantly higher than that of fish fed all EP (P<0.05). Condition factor of fish fed the MP was not significantly different from that of fish fed all EP. The contents of moisture, crude protein and lipid in dorsal muscle and whole body was not significantly different among the groups. It is concluded that the dietary formulation used in the EP1, EP3, TP4, EP5 and EP6 can be applied in the practical extruded pellet feeds for adult flounder (329-680g).

Evaluation of Muscle Quality of Olive Flounder (Paralichthys olivaceus) Fed Extruded Pellets Containing Different Protein and Lipid Levels, and Raw Fish-based Moist Pellet (단백질 및 지질함량이 다른 배합사료와 생사료로 사육한 넙치의 육질평가)

  • This study was conducted to investigate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) on flesh quality of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Moisture content of fish fed MP was significantly higher than that of fish fed all EP. Significantly higher contents of 16:1n-7, 20:4n-6, 20:5n-3 and lower 18:2n-6 were observed in fish fed EP4 and MP compared with fish fed EP1, EP2, EP3. The 22:6n-3 content was not significantly different among all groups. Taurine content of fish fed MP was significantly higher than that of fish fed all EP. Asparagine content of fish fed EP1 was significantly higher than that of fish fed EP2, EP, EP4 and MP. Significant difference were observed in lysine and serine contents of fish fed experimental diets. In nucleotides and their related compounds, ATP and AMP content was not affected by diets. Significant difference were observed in IMP contents of fish fed experimental diets. Textural properties, no significant difference was observed among the fish groups fed different diets.

Analysis of the Characteristics of Bulking Agents Used in Livestock Manure Composting (축산분뇨 퇴비화에 이용되는 수분조절제의 특성 분석)

  • This study analyzed the characteristics of 6 recyclable bulking agents which can replace sawdust and rice hulls that are commonly used in livestock manure composting. The content of all the hazardous chemicals found in the bulking agents used in this experiment was between 0.0 and 34.1 ppm, which was noticeably lesser than the maximum content levels of hazardous substances 5.0~900.0 ppm prescribed by the Rural Development Administration(RDA), which means that the bulking agents would be able to replace sawdusts and rice husks. The water content of the bulking agents ranged from a minimum of 12.4% to a maximum of 16.6% in the original state, which was much less than 60%, the optimal moisture content. These results indicate that they would be able to function fully as bulking agents. Their water absorption rate was in the range of minimum 31.9% ~ maximum 600.0%, which showed huge differences among the types of bulking agents. The most appropriate bulking agent in terms of water absorption rate was wood pellets. It was the highest in the wood briquette manufactured by A, which was followed by C and B in the order. The shear strength of the 5 types of bulking agents in the original state was 271.7 N on an average, the highest figure of all, except for the oak briquette whose shear strength could not be measured and the wood pellets in the absorption state. The shear strength in the dry and absorption states then followed, and were 78.0 N and 27.7 N on an average, respectively. The wood briquette of A recorded considerably lower shear strength than that of B and C. Overall, shear strength tended to increase according to lower water absorption rates. Since pine wood chips and oak cuts have relatively greater shear strength even in the absorbing state than the other materials, they will be able to endure some power in the case of stirring with pig droppings.

A Study on the Optimal Process Parameters for Recycling of Electric Arc Furnace Dust (EAFD) by Rotary Kiln (Rotary Kiln에 의한 전기로 제강분진(EAFD)의 재활용을 위한 최적의 공정변수에 관한 연구)

  • As a recycling technology for recovering zinc contained in large amounts in electric arc furnace dust (EAFD), the most commercialized technology in the world is the Wealz Kiln Process. The Wealz Kiln Process is a process in which components such as Zn and Pb in EAFD are reduced/volatile (endothermic reaction) in high-temperature Kiln and then re-oxidized (exothermic reaction) in the gas phase and recovered in the form of Crude zinc oxide (60wt%Zn) in the Bag Filter installed at the rear end of Kiln. In this study, an experimental Wealz kiln was produced to investigate the optimal process variable value for practical application to the recycling process of large-scale kiln on a commercial scale. Additionally, Pellets containing EAFD, reducing agents, and limestone were continuously loaded into Kiln, and the amount of input, heating temperature, and residence time were examined to obtain the optimal crude zinc oxide recovery rate. In addition, the optimal manufacturing conditions of Pellets (drum tilt angle, moisture addition, mixing time, etc.) were also investigated. In addition, referring to the SiO2-CaO-FeO ternary system diagram, the formation behavior of a low melting point compound, a reaction product inside Kiln according to the change in the basicity of Pellet, and the reactivity (adhesion) with the castable constructed on the inner wall of Kiln were investigated. In addition, in order to quantitatively investigate the possibility of using anthracite as a substitute for Coke, a reducing agent, changes in the temperature distribution inside Kiln, where oxidation/reduction reactions occur due to an increase in the amount of anthracite, the quality of Crude zinc oxide, and the behavior of tar in anthracite were also investigated.
