Quinolone계 항균제의 일종인 pefloxacin을 넙치에 투여한 후, HPLC를 이용하여 약물의 분포도를 살펴보았다. 100 mg/kg의 pefloxacin을 넙치에 이틀 연속 경구투여한 후, 넙치의 혈액, 근육 및 간을 포함한 8개의 장기를 분석한 결과, pefloxacin은 장을 통하여 흡수되며 담낭에 축적됨을 알았다. 약물 투여 후 45분에 $4.70{\mu}g/ml$로 혈액내 농도가 최대치를 나타냈으며, 11일째에는 $0.31{\pm}0.01{\mu}g/ml$이 검출되었다. 근육조직에서는 10일째까지 $0.29{\pm}0.02{\mu}g/g$ tissue가 잔류하였다.
The photodegradation of pefloxacin, photolabile fluoroquinolone antibacterial agent, was studied. In the presence of $N_2$, photodegradation of pefloxacin was suppressed. The singlet oxygen and free radical generated in the reaction media proceeded photochemical reaction. The photodegradation of peflxacin was sensitized by benzophenone, a triplet state sensitizer.
Pharmacokinetic interaction of theophylline with pefloxacin following i.v. administrations was investigated in rabbits. Pefloxacin was coadministrated at doses of 10 and 20 mg/kg or previously administered for 6 days 10 and 20 mg/kg. Plasma concentration and AUC of theophylline were increased significantly (p<0.05) and the renal clearance $(CL_r)$, total body clearance $(CL_r)$ and the volume of distribution $(Vd_{ss})$ were decreased significantly (p<0.005) by the pretreatment. It demonstrates that adjustment of dosage regimen of theophylline should be considered when concomitant administration of pefloxacin is prescribed.
Pefloxacin을 15분간 분쇄한 분쇄품에 있어서는 $D_10$, $D_50$ 및 $D_90$은 각각 2.79 $\mu$m, 15.94 $\mu$m 및 39.75 $\mu$m로 나타나 표준품에 비해서 입도가 현저하게 작아짐을 알수 있었으며, 이 보다 분쇄시간이 길어질 경우 입자가 점점 더 커지는 경향을 나타내어 분쇄에 의해 입도가 점점 작아지다가 정점을 지난 후부터는 재응집에 의해 입자가 다시 커지는 경향을 나타냄을 알 수 있었다. 분쇄한 peflocacin의 XRD패턴, 열분석 측정 및 IR-Spectrum 관측 결과 분쇄의 유무와 관계없이 패턴의 양상이 변하지 않음을 알 수 있었으며, 분쇄라는 물리적인 조작에 의해 pefloxacin의 물성과 안정성에는 특이한 변화가 없음을 알 수 있었다. 이상의 결과들은 분쇄에 의한 pefloxacin의 화합물질 변화나 기능의 변화, 원자간의 결합상태, 분자의 구조 등에 있어서 물성적 변화의 차이가 없음을 나타내고 있다. Pefloxacin 표준품에 비해서 분쇄를 하였을 경우 용해도가 개선됨을 알수 있었으며, 특히 15 분간 분쇄한 분쇄품의 경우에 있어서 가장 양호한 용해도 곡선을 얻을 수 있었다. 즉, 난용성 항균제 pefloxacin은 분쇄라는 조작에 의해 입도가 작아지고, 비표면적의 증가로 인한 그 물리적인 특성중의 하나인 용해도가 개선되므로 속효성 제형으로의 전환이 가능함을 시사하였다[1,11].
The effect of antioxidants on photochemical reaction of pefloxacin by UVB (290~320 nm) was investigated and the possible mechanism of phototoxicity on the skin was also studied. The photo-degradation of pefloxacin by UVB was suppressed by cysteine, reduced glutathione and ascorbic acid, but was promoted by ${\alpha}-tocopherol$. Squalene, accounts for more than 10% of skin surface lipids, was peroxidized by pefloxacin through both radical and singlet oxygen mechanism.

Antibiotics have been routinely used to control the disease of farm-raised animals in the aquaculture facilities without any criterion based on a pharmacokinetic study. This lack of information on the effective usage of antibiotics would have brought the farmers to use excessive and/or less dosages, causing the advent of drug-resistant bacteria as well as economic loss and possible contamination of the local farming area. Until recently, few studies on a detailed manual for the antibiotic usage including chemotherapy procedure, dosage, and treatment schedule of the aquatic antibiotics have been conducted throughout the world. To the worse, there is no available criterion for optimal usage of aquatic antibiotics to control diseases in aquatic farms in this country because every country has its own aquacultural system. Therefore, based on the previous studies on the usage of the various antibiotics, our studies are to focus on the development of optimal method for the detection of various antibiotics on the fate of antibiotics applied to the fish, including absorption, circulation, and secretion physiology. Pharmacokinetic study were to sep up the optimal detective condition against residual antibiotics of fish by HPLC. The grinding pefloxacin for 15 min is most effective in dissolution test and pharmacokinetic parameters. Pharmacokinetic parameters were satisfactory for 15 min-grinding products and they can be explained as one-compartment model.
어류 및 갑각류 중에 잔류하는 fluoroquinolone계 항균제를 조사하기 위하여 HPLC-FLD와 LC-MS를 이용하여 분석을 실시하였다. HPLC-FLD를 이용하여 분석하였을 때 검출한계와 정량한계는 각각 0.0030-0.0083, 0.0090-0.0252 mg/kg이었고, LC-MS는 각각 0.0019-0.0040, 0.0059-0.0122 mg/kg으로 나타났다. ofloxacin, norfloxacin, pefloxacin, ciprofloxacin and enrofloxacin의 회수율은 76.4-104.6% 범위로 분석되었다. 총 268건에 대하여 fluoroquinolone계 항균제의 잔류량을 조사한 결과 7.5%의 검출율을 나타내었고, 동자개 1건에서 enrofloxacin과 ciprofloxacin의 합이 허용 기준치인 0.1 mg/kg보다 높게 검출되었다. Ofloxacin, norfloxacin과 pefloxacin은 모든 시료에서 검출되지 않았고 ciprofloxacin과 enrofloxacin은 각각 3.4%와 6.7%의 검출율을 보였다. 어종별 검출율은 장어(2.6%), 미꾸라지(2.2%), 동자개(1.5%), 붕어(0.7%), 가물치(0.4%) 순서로 나타났다.
-4-Quinolone antibiotics (pefloxacin, ciprofloxacin, norfoxacin, ofloxacin and enoxacin) were tested for mutagenicity in Salmonella typhimurium TA98, TA100, TA1535, TA1537, TA1538 and TA102, for chromosomal aberrations in cultured Chinese hamster lung (CHL) cells, and for wing somatic mutations and recombinations (wing spot) in Drosophila. Five 4-quinolones did not show any mutagenicity in Salmonella typhimurium TA98, TA100, TA1535, TA1537 and TA1538. However, they were mutagenic inSalmonella typhimurium TA102 with and without metabolic activation in both plate incorporation method and preincubation method. Ciprofloxacin induced structural chromosome aberrations in CHL cells both with and without metabolic activation, and the frequencies were 6% and up to 28%, respectively. Pefloxacin showed equivocal evidence, however, norfloxacin, ofloxacin and enoxacin did not induce the structural chromosome aberrations both in the presence and absence of metabolic activation. In the wing spot assay in Drosophila, ofloxacin increased the frequency of small single spots significantly in a dose-dependent manner but there was no dose-dependent increase of single or twin spots in the others.
The modern integrated fish-seaweed mariculture has been tested to reduce the environmental impacts of an intensive fed culture. To obtain the best seaweed bioremediation performance, the effects of therapeutants used for fish disease control on the selected seaweed species should be considered. As a selected seaweed, Porphyra yezoensis was tested with six commercial antibiotics including erythromycin thiocyanate_A, erythromycin thiocyanate_B, oxytetracycline, doxycycline, pefloxacin, and amoxicillin trihydrate under the batch incubation at a photon flux density of 10 $\mu$mol ${\cdot}m^{-2}\;{\cdot}\;s^{-1}$ at 15$^{\circ}C$. Among the tested commercial antibiotics, erythromycin thiocyanate_A, erythromycin thiocyanate_B, oxytetracycline, and doxycycline showed decreases in Fv/Fm, the photochemical efficiency of photosystem II, with a dose-dependant and time-dependant manner. From the quenching analysis of chlorophyll fluorescence, three differential patterns were observed in the antibiotics-treated Porphyra: (1) high nonphotochemical quenching (NPQ) and low photochemical quenching (qP) in the cases of Erythromycin thiocyanate_B and amoxicillin trihydrate, (2) high NPQ and high qP in the case of pefloxacin and (3) low NPQ and low qP in the case of oxytetracycline. These results indicated that antibiotics affected in various ways on the photosynthetic apparatus, reflecting differential lesion sites of antibiotics. In addition, the rates of ammonium uptake also decreased with a decrease of Fv/Fm in P. yezoensis thalli treated with erythromycin thiocyanate_B and oxytetracycline. Therefore, the four antibiotics mentioned could affect the bioremediation capacity of the selected seaweed species in the integrated fish-seaweed mariculture system due to the decrease of photosynthetic activity and the simultaneous decrease of ammonium uptake.
