Genealogical Relationship between Pedigree and Microsatellite Information and Analysis of Genetic Structure of a Highly Inbred Japanese Black Cattle Strain

  • Japanese Black cattle of Hyogo prefecture (Tajima strain) are famous for its ability to produce high-quality meat and have been maintained as a closed system for more than 80 years. In order to assess the usefulness of microsatellite markers in closed cattle populations, and evaluate the genetic structure of the Tajima strain, we analyzed representative dams of the Tajima strain comprised of the substrains Nakadoi and Kinosaki. Genetic variability analyses indicated low genetic diversity in the Tajima strain. In addition, a recent genetic bottleneck, which could be accounted for by the high level of inbreeding, was detected in both substrains. In phylogenetic analyses, relationship coefficients and genetic distances between individuals were calculated using pedigree and microsatellite information. Two phylogenetic trees were constructed from microsatellite and pedigree information using the UPGMA method. Both trees illustrated that most individuals were distinguished clearly on the basis of the two substrains, although in the microsatellite tree some individuals appeared in clusters of different substrains. Comparing the two phylogenetic trees revealed good consistency between the microsatellite analysis tree and the pedigree information. The correlation coefficient between genetic distances derived from microsatellite and pedigree information was 0.686 with a high significance level (p<0.001). These results indicated that microsatellite information may provide data substantially equivalent to pedigree information even in unusually inbred herds of cattle, and suggested that microsatellite markers may be useful in revealing genetic structure without accurate or complete pedigree nformation.   • This paper propose G-code and G-Pedigree system that are development based on the e-Pedigree and EPC, and RFID solutions for logistics management is proposed. The proposed G-Pedigree system to include the forward logistics and reverse logistics all logistics and management systems are appropriate for the event, with security features, security and accessibility of critical data was strengthened. The proposed G-code systems and the EPC code of the existing RFID readers, tags can be applied to, the more you can manage the logistics-related information.

  • 전 세계적으로 유통되고 있는 가짜의약품으로 인하여 제약사들은 판매수익과 기업브랜드 이미지에 큰 피해를 입고 있으며 가짜 의약품을 복용한 소비자들은 생명에 위협을 받고 있는 실정이다. 이러한 가짜의약품의 유통을 근절시키기 위하여 세계 각국에서는 의약품의 생산에서부터 소비자에게 판매될 때까지, 즉 의약품 유통과정을 추적하여 가시성있는 이력관리를 할 수 있도록 RFID 및 barcode 등 IT기반의 e-Pedigree(electronic pedigree)를 법제화시키고 있지만 구체적이고 실질적인 구축방법이 제시되지 못하고 있다. 따라서 본 연구는 국내 제약산업의 유통합리화와 소비자 안전성을 제고시키기 위하여 RFID 기반의e-Pedigree 적용방안을 제시하는데 그 목적이 있고 더 나아가 세계 각 국가의 e-Pedigree 구축을 위한 참조모델 개발에 일조하고자 한다.

  • 콜드체인은 생산지로부터 소비지까지 저온상태로 상품을 신선하게 유지하여 안전하게 소비자에게 공급하는 것을 목적으로 한다. 그러나 유통 중 저온 유지가 안될 경우가 발생하고 이에 따라 변질된 상품을 소비자가 구매할 수 있다. 그러므로 소비자는 냉장 유통과정에 대해 입증할 수 있는 정보를 제공받기를 원한다. 따라서 본 논문에서 제안하는 Pedigree 시스템은 콜드체인 상품을 주고 받을 때 거래에 관한 정보인 Pedigree 데이터를 교환하는 시스템으로, 매 분배 단계에서 냉장 온도 및 물품의 상태를 체크하여 이상이 없을 경우에만 확인자의 서명정보를 추가하여 다음 분배지로 유통시키고 이상이 있을 경우 유통을 중지하여 변질된 상품이 유통되는 것을 막는다. 본 시스템의 아키텍처는 기존의 EPCIS(EPC Information Services)를 확장하여 사용하도록 설계하였고, Pedigree 처리를 위한 API 를 제공하는 Pedigree Library 를 제안하여 EPCIS 환경에 대하여 확장이 용이하다.

  • Facing the counterfeiting acts towards various products, many manufacturers implement ePedigree system to secure their supply chain. Using ePedigree, a distribution history including a valid product identifier from the manufacturer until the final retailer is recorded. And this ePedigree is signed by each involved supply chain party using digital signature. With this digital signature, any unauthorized alteration to the ePedigree document would generate a failed verification process. If there is a counterfeit product using a fake ePedigree document, it wouldn't be able to pass the verification process either. Hence, there wouldn't be any counterfeit product that could enter the legal supply chain and bought by the consumer. We are proposing to use ECDSA instead of RSA since it has faster performance and shorter key size. At a certain same security level, ECDSA only needs 163 bits, while RSA needs 1024 bits.

Selection of Young Dairy Bulls for Future Use in Artificial Insemination

  • Relationships of breeding values of sires for first lactation milk yield with pedigree information or indices were examined to identify the optimal criteria of selecting young dairy bulls for future use in artificial insemination (AI). Records of performance data on 1087 crossbred daughters (Holstein - Friesian, Jersey and Brown Swiss with Hariana) of 147 sires, generated at Livestock Production Research (Cattle and Buffaloes) Farm, IVRI, Izatnagar, U.P., during 1972 - 1995 were used to obtain the estimates of sire's breeding values (EBV) using the Best Linear Unbiased Prediction Procedures. The correlations between young bull's EBV and the dam's first lactation milk yield was non-significantly different from zero. However, the young bull's EBV was negatively and significantly related (r = - 0.275 ; P < 0.05) to the dam's best lactation milk yield, suggesting that the selection of young dairy bulls from high yielding elite dams is not a suitable criteria for genetic improvement. The correlations of sire's and paternal grandsire's EBV's with young bull's EBV were high and positive (0.532, 0.844; P < 0.01). The maternal grandsire's EBV was positively but non-significantly related to grandson's EBV. The pedigree index incorporating dam's milk records and sire's EBV's showed a negative and non-significant correlation with young bull's EBV. However, the correlation of a pedigree index $(I_3)$ combining information on sire's and paternal grand-sire's EBV's with young bull's EBV's was considerably high and positive (0.797; P < 0.01). The regression coefficients of young bull's EBV on pedigree index $I_3$, was higher than those on other pedigree information. These results revealed that there was no advantage in basing selection on dam's performance or maternal grand-sire's EBV and that sire's and paternal grandsire's EBV's were reliable pedigree information for selection of young dairy bulls for future use in AI.

  • 글로벌 시장환경체제에서 유통구조가 복잡해짐에 따라 제조사에서 유통업체 및 소비자까지 아우르는 전체 공급망에서 제품에 대한 이력추적관리는 투명한 유통구조를 형성시킬 뿐만 아니라 더 나아가 소비자들의 안전에 지대한 영향을 미친다. 세계보건기구(WHO)에 따르면 선진국에서 부정 전문의약품의 유통비중은 전체 의약품의 1% 정도이며 이를 미국에 적용할 경우(2006년 기준), 미국에서 유통된 34억 개 전문의약품 중 3,400만 개의 전문의약품이 가짜일 가능성이 있는 것으로 보고되고 있다. 이로 인해 미국 캘리포니아주에서는 유통망의 투명성과 소비자의 안전을 보고하기 위하여 2D 바코드나 RFID(Radio Frequency IDentification)를 통해 의약품 제조에서 약국까지의 유통과정 계보를 추적할 수 있도록 제약기업에 2015년부터 제품에 시리얼번호를 부착한 이력장치(serialized e-pedigree)를 시행할 예정이다. 따라서 본 연구에서는 국내 외적으로 물류/유통과정에서 제품의 위 변조를 예방하기 위한 e-pedigree에 현황 및 동태를 파악하고 국내 e-pedigree에 대한 방향성을 고찰하는데 그 목적이 있다.

  • Pharmaceutical companies over the world are exposed to risks of lessened sales and negative impact on corporate image due to circulation of counterfeit drugs, which eventually threatens safety of patients. thus, in order to eradicate distribution of counterfeit drugs, many countries are taking initiatives to legalize and set serialization through e-Pedigree, which is based on RFID technology or barcode system to track and trace distribution of drugs from point of manufacturing to usage. therefore, this study contemplates domestic and international management system of medical drugs and suggests application methods of e-Pedigree based on RFID technology that promotes safety of patients and distribution of drugs.

  • Sharing of product and process information with partners is a basic activity and key requirement which ensures success of distribution. ePedigree that encapsulates all of the event data from manufacturer to retail shop provides a flexible mechanism of storing and sharing traceable information to the partners of supply chain and credibility of shared information through digital signature based on Public Key Infrastructure (PKI). To generate the signature that can be authenticated through PKI, the partners of supply chain should pay for PKI certificates from Certificate Authority (CA). In case of agrifood cooperatives which consist of petty merchants or farmers, it is hard to pay for the PKI certificate for all members and is a big obstacle for the ePedigree to be applied to the supply chain. This paper proposes a private certification method of ePedigree for cooperatives. Cooperatives can apply the ePedigree using the proposed method to all the members at small cost and the proposed method can assure the credibility of information at the same level of the previous ePedigree.

Influence of Amount of Pedigree Information and Parental Misidentification of Progeny on Estimates of Genetic Parameters in Jeju Race Horses (제주마 집단의 혈연 정보량과 정보 오류가 유전 모수 추정치에 미치는 영향)

  • The pedigree information and race records of 1,000 m finishing time of Jeju race horses at KRA were used to study the effect of amount of pedigree information and parental misidentification on the estimates of genetic parameters. The modified data sets were made at the range of 2.5 to 25% parental misidentifications or loss of parental information of individuals with an increment of 2.5 percent. For each incremental level, 20 randomly replicated data sets were obtained and analyzed by single-trait analysis with a DF-REML(AI) algorithm. As the rate of misidentification increased or the amount of pedigree information decreased, the estimates of fraction of additive genetics variance component gradually decreased almost linearly (p<0.05), while the estimated fractions of error variance and permanent environmental variance components gradually increased for the finishing time. Regression coefficients of the percentage amount of both parents' information loss and incorrect pedigree information on additive genetic variances were -0.079 and -0.114, respectively (p<0.01). The estimate of heritability decreased by 0.92% for one percent loss of both parents' information and 1.39% for one percent increase of both parental misidentifications of progeny (p<0.01). For the consideration of probable incorrect and missing parent information of progeny in this early population of Jeju horses, the estimates of additive genetic parameters would be biased downward about ten percent. This results indicate that the amount of pedigree information loss and misidentification of progeny would severely affect estimates of genetic parameters and would reduce genetic gains for selection in Jeju horse population.
