The collaborative study for verification of analytical results and assurance confidences for pesticide residue (분석결과 검증 및 신뢰성 확보를 위한 실험실간 협력 실험)

  • The residual study of pesticide has been used in various areas, such as food safety, environmental protection, establishment of tolerance, and explaining the pathway and reaction mode of pesticides, and its importance was expected to increase further more. The aspect of food safety, the pesticide residue survey have been practiced at many organizations, but there were no verification of analytical results at present. In this experiment, we focused on instrumental stability, including response of each instrument and the recovery ratio of each organization's method. As samples for this experiment, we prepared cucumber and sesame, and chose 4 pesticides (bifenthrin, chlorpyrifos, diazinon, and ethoprophos), which were mostly detected from pesticide residue survey and widely used for each crop. The standard deviation of peak areas in the chromatogram of each pesticide were under 1.212 %, so it showed that most instruments were stable. The relationship of recovery ratio of each organization were over 0.996 for every pesticide and each organization. Finally, the analytical results for pesticide residue from each participated organization were not statically significant and we could put confidence in the result from each organization.

Neutron Induced Capture Gamma Spectroscopy Sonde Design and Response Analysis Based on Monte Carlo Simulation (Monte Carlo 시물레이션에 기초한 포획모드 중성자-감마 스펙트럼 존데 설계 및 반응 분석)

  • For efficiently designing neutron induced gamma spectroscopy sonde, Monte Carlo simulation is employed to understand a dominant location of thermal neutron and classify the formation elements from the energy peak of capture gamma spectrum. A pulsed neutron generator emitting 14 MeV neutron particles was used as a source, and flux of thermal neutron was calculated from the twelve detectors arranged at each 10 cm intervals from the source. Design for reducing borehole effects using shielding materials was also applied to numerical sonde model. Moreover, principal elements and quantities of numerical earth models were verified through the energy spectrum analysis of capture gamma detected from a gamma detector. These results can help to enhance the signal-to-noise ratio, and determine an optimal placement of capture gamma detectors of neutron induced gamma spectroscopy sonde.

  • Three-dimensional porous scaffolds play an important role in tissue engineering strategies. They provide a void volume in which vascularization, new tissue formation, and remodeling can occur. Like any grafted materials, the ideal scaffold for bone tissue engineering should be biocompatible without causing an inflammatory response. It should also possess biodegradability, which provides a suitable three-dimensional environment for the cell function together with the capacity for gradual resorption and replacement by host bone tissue. Various scaffolds have already been developed for bone tissue engineering applications, including naturally derived materials, bioceramics, and synthetic polymers. The advantages of biodegradable synthetic polymers include the ability to tailor specific functions. The purpose of this study was to examine the osteogenic activity of periosteal-derived cells in a polydioxanone/pluronic F127 scaffold. Periosteal-derived cells were successfully differentiated into osteoblasts in the polydioxanone/pluronic F127 scaffold. ALP activity showed its peak level at 2 weeks of culture, followed by decreased activity during the culture period. Similar to biochemical data, the level of ALP mRNA in the periosteal-derived cells was also largely elevated at 2 weeks of culture. The level of osteocalcin mRNA was gradually increased during entire culture period. Calcium content was detactable at 1 week and increased in a time-dependent manner up to the entire duration of culture. Our results suggest that polydioxanone/pluronic F127 could be a suitable scaffold of periosteal-derived cells for bone tissue engineering.

The Relationship between FEV1 and PEFR in the Classification of the Severity in COPD Patients (만성 폐쇄성 폐질환 환자의 중증도 분류시 FEV1과 PEFR의 연관성)

  • Background : Measurement of the $FEV_1$ and PEFR in COPD patients is a significant indicator of the disease severity, the response to treatment and the acute exacerbation. However, it is not known if PEFR can be used to determine the severity of COPD because the agreement between PEFR and $FEV_1$ in COPD patients is not well known. Methods : From September, 2003 to August, 2004, 125 out patients with COPD who were treated at the pulmonary clinic in KonKuk University Hospital were enrolled in this study. The $FEV_1$ and PEFR of each patient were measured and all the data was analyzed using SPSS. Results : The average predicted $FEV_1$ % and PEFR % was $56.98{\pm}18.21%$ and $70{\pm}27.60%$, respectively. There was linear correlation between the predicted $FEV_1$ % and predicted PEFR %. There was no correlation between age of the COPD patients and the predicted PEFR %. There was correlation between dyspnea, which is a subjective symptom of the patients, and the predicted PEFR %. Conclusion : In COPD patients, the classification of the severity by PEFR tends to underestimate the state of the disease compared with the classification of the severity by the $FEV_1$. Therefore, the classification of the severity by PEFR should be interpreted carefully in patients with severe symptoms. Once the classification of the severity has made, the follow-up examination may use the PEFR instead of the $FEV_1$.

  • 일반적으로 건물 구조물에 전달되는 기계진동을 감소시키기 위해서 기계와 기초사이에 유연한 방진소자를 삽입하여 기계가진력(exciting force)의 전달 률을 줄인다. 또한 구조물의 고유진동수와 진동원의 가진주파수가 일치할 경 우, 가진주파수를 변화시키거나, 구조물의 동특성을 변화시키는 방법을 사용 한다. 어떠한 방안을 선택하든 효과적이고 정량적인 방진 시스템을 구성하고 구조물의 정확한 진동상태를 예측하기 위해서는 진동원의 가진특성과 구조 물의 동특성에 대한 정보가 요구된다. 일반적으로 방진설계를 위해 필요한 진동원의 가진특성은 제조회사의 사양이나 측정을 통하여 비교적 쉽게 얻을 수 있다. 복합재료, 다양한 경계조건, 복잡한 대형구조물등은 수치해석을 이 용하여 해석적인 방법으로 동특성을 구할 경우, 신뢰성 있는 정보를 얻기에 는 많은 노력이 요구된다. 더우기 현장에서 발생하는 진동문제는 대부분 복 잡하고 시간적으로 시급히 해결해야 하기 때문에 효율적인 절차를 구성하여 구조물의 동특성을 해석하는 방법을 사용할 필요가 있다. 구조물의 동특성은 실험적인 방법을 통하여 구하고 그 외의 필요한 계산들은 해석을 통하여 얻 는 것이 효율적일 수 있다. 실험적 동특성해석은 입력하중에 대한 응답의 크 기와 위상 비를 주파수별로 나타내는 전달함수를 측정하는 방법으로서 가진 장치 및 여러 측정/분석 장비가 필요하며, 철교, 교량, 건물의 철골 및 콘크 리트 슬라브등 다양한 중대형의 구조물을 Signal/Noise비가 좋도록 가진 시 켜야 할 필요성이 있다. 본 연구에서는 이러한 실험적 방법의 현장 적응성과 신뢰성을 확보하기 위해 대형충격기(large impact hammer, max, peak force 약 10000N, time duration 약 20ms)를 제작하고 실험/분석 시스템 및 구조물 의 진동제어를 위한 절차를 Fig.1과 같이 구성하고 이를 철근콘크리트 건물 에 설치한 기계식 주차설비의 진동제어에 적용하였다.force response simulation)를 수행하여 임의의 좌표 공간에 대한 진동수준을 해석적으로 예측할 뿐만 아니라 구조물의 진동제어 를 위한 동적인자를 변경시킬 수 있는 정보를 제공하며 장비를 방진할 경우 신뢰성 있는 전달률을 결정할 수 있다. 실험적으로 철교, 교량이나 건물의 철골구조 및 2층 바닥 등 대,중형의 복잡한 구조물에 대항 동특성을 나타내 는 모빌리티를 결정할 경우 충격 가진 실험이 사용되는 실험장비 측면에서 나 실험을 수행하는 과정이 대체적으로 간편하다. 그러나 이 경우 대상 구조 물을 충분히 가진시킬수 있는 용량의 대형 충격기(large impact hammer)가 필요하게 된다. 이러한 동적실험은 약 길이 61m, 폭 16m의 4경간 교량에 대 하여 동적실험을 수행하여 가능성을 확인하였다. 여기서는 실험실 수준의 평 판모델을 제작하고 실제 현장에서 이루어질 수 있는 진동제어 구조물에 대 한 동적실험 및 FRS를 수행하는 과정과 동일하게 따름으로써 실제 발생할 수 있는 오차나 error를 실험실내의 차원에서 파악하여 진동원을 있는 구조 물에 대한 진동제어기술을 보유하고자 한다. 이용한 해마의 부피측정은 해마경화증 환자의 진단에 있어 육안적인 MR 진단이 어려운 제한된 경우에만 실제적 도움을 줄 수 있는 보조적인 방법으로 생각된다.ofile whereas relaxivity at high field is not affected by τS.   • To explore the influence of coal thickness on the mechanical behavior and the failure characteristics of rock-coal-rock (RCR) mass, the experimental investigation of uniaxial compressive tests was conducted first and then a systematic numerical simulation by particle flow code (PFC2D) was performed to deeply analyze the failure mechanical behavior of RCR specimens with different coal thicknesses in conventional compression tests. The overall elastic modulus and peak stress of RCR specimens lie between the rock and the coal. Inter-particle properties were calibrated to match the physical sample strength and the stiffness response. Numerical simulation results show that the deformation and strength behaviors of RCR specimens depend not only on the coal thickness, but also on the confining pressure. Under low confining pressures, the overall failure mechanism of RCR specimen is the serious damage of coal section when the coal thickness is smaller than 30 mm, but it is shear failure of coal section when the coal thickness is larger than 30 mm. Whereas under high confining pressures, obvious shear bands exist in both the coal section and the rock section when the coal thickness is larger than 30 mm, but when the coal thickness is smaller than 30mm, the failure mechanism is serious damage of coal section and shear failure of rock section.

  • The analytical response properties of two types of continuous flow-through electrode system as fulfide ion detectors are examined and directly compared their reponse characteristics under the optimal conditions. In both detection systems, observed peak potentials are logarithmically related to the sulfide ion concentration and at least twenty samples per hour can be determined. In the pH electrode method, the pH of the flowing recipient stream leaving the dialyzer was monitored. The designed system involves the use of continuous flow gas dialyzer in conjunction with the tubular polymer membrane electrode. In this method, optimal experimental conditions are recipient of mixture of $5.0 {\times} 10^{-5} M NaOH + 5.0 {\times} 10^{-3} M$ NaCl and diluent of 0.10 M $H_2SO_4$, and all flow rates of recipient stream, diluent stream, and sample are 1.0 ml/min. In the sulfide ion electrode method, a commercially available sulfide ion-selective electrode was used to detect sulfide ion in the flow-through cell. The optimal flow rates of sulfide anti-oxidant buffer (3.5 g ascorbic acid and 7.6 g $Na_2EDTA$ dissolved in 1.0 M NaOH solution 1 l) and sample were 1.4 ml/min and 1.0 ml/min, respectively.

The Effect of IGF-1 on ALP Activity of MC3T3-E1 Cell (MC3T3-E1세포의 ALP activity에 대한 IGF-I의 영향)

  • Polypeptide growth factors belong to a class of potent biologic mediators which regulate cell differentiation, proliferation, migration and metabolism. IGF-I is polypeptides secreted by skeletal cells and is considered as regulators of bone formation. The purpose of this study is to evaluate the effects of IGF-I on bone nodule formation and alkaline phosphatase activity of MC3T3-E1 cells. MC3T3-E1 cells were seeded at $1{\times}10^4$ cells/well, $1{\times}10^5$ cells/well in alpha-modified Eagle medium containing 10% fetal bovine serum, 10 mM ${\beta}-glycerophosphate$ and $5O{\mu}g/ml$ of ascorbic acid. Before 48 hours of indicated time, medium were changed with serum free medium. After 24 hours, 0.1, 1, 10 ng/ml IGF-I were added to the cells and cultured for 3, 7, 14, 21, 28 days. And histochemical analysis was done and ALP activity was measured and was expressed as nmol/min/mg of protein. The bone nodule formation in MC3T3-E1 cells of IGF-I was seen at 21, 28 days, but there were no difference between control group and experimental groups. The ALP activity decreased when it is compare to control 2 group except for 1 ng/ml, 10 ng/ml IGF-I of 21-day-groups and 1 ng/ml IGF-I of 28-day-groups. Dose response effects of IGF-I of ALP activity in MC3T3-E1 cells were seen the highest ALP activity at 1ng/ml until 21days and the highest ALP activity at 10 ng/ml of 28 daygroups. The peak times were seen at 7-day group, 14-day group on control group and experimental group respectively, and 1 ng/ml group was the highest ALP activity, From the above results, IGF-I was not seen notable effect on bone nodule formation and decreased ALP activity of MC3T3-E1 cells but the use of IGF-I to mediate biological stimulation of MC3T3-E1 cells shows promise for future therapeutic application.

Reaction of Mast Cells and Goblet Cells in the Small Intestine of C57BL/6 and C3H/HeN Mice Infected with Echinostoma hortense

  • Mast cells and goblet cells have been known to protect the host against parasites. In this study, we examined the response of the mast cells and goblet cells over a period of 6 weeks in the duodenum, jejunum and ileum of C3H/HeN and C57BL/6 mice infected with Echinostoma hortense (E. hortense). In addition, we investigated whether the worm recovery rate of uninfected mice (the control group) or E. hortense-infected mice (the experimental group) was associated with the number of mast cells and goblet cells. The worm recovery rate was higher in the C3H/HeN mice than in the C57BL/6 mice. The number of goblet cells significantly increased in the experimental group of the C3H/HeN and C57BL/6 mice compared with the control group of both strains (P<0.005). Worm recovery peaked 3 weeks after the infection of the C57BL/6 mice and at 2 weeks after the infection of the C3H/HeN mice, and it was higher in the duodenum than in the jejunum and ileum. However, the infected site in the intestine had no relation with worm expulsion. In the C3H/HeN and C57BL/6 mice, the number of goblet cells in the experimental group was significantly higher than that in the control group (P<0.005). The number reached a peak 2 weeks after the infection and it even increased in duodenum, jejunum and ileum. The increased number of goblet cells was retained 6 weeks after infection. The number of goblet cells was higher in the C3H/HeN mice than in the C57BL/6 mice (P<0.01). These results indicate that goblet cells are related with the worm expulsion. Furthermore, immunohistostaining of the antral intestinal walls for lectin showed the significant increase of the number of goblet cells in the experimental group (P<0.001). The high infection rate in the duodenum was found during the early infection. An increased infection rate in the jejunum and ileum was found 3 weeks after infection and the infection rate was higher in the C3H/HeN mice than in the C57BL/6 mice. Taken together, the present study indicates that goblet cells, rather than mast cells, may play critical roles in parasite expulsion.

Relationship between Radiation Induced Activation of DNA Repair Genes and Radiation Induced Apoptosis in Human Cell Line A431 (인체세포주 A431에서 방사선 조사 후 DNA수선 유전자 발현과 세포고사와의 관계에 관한 연구)

  • Purpose: The purpose of this study was to evaluate the relationship between radiation-induced activation of DNA repair genes and radiation induced apoptosis in A431 cell line. Materials and Methods: Five and 25 Gys of gamma radiation were given to A431 cells by a Cs-137 cell irradiator. Apoptosis was evaluated by flow cytometry using annexin V-fluorescein isothiocyanate and propidium iodide staining. The expression of DNA repair genes was evaluated by both Northern and Western blot analyses. Results: The number of apoptotic cells increased with the increased radiation dose. It increased most significantly at 12 hours after irradiation. Expression of p53, p21, and hRAD50 reached the highest level at 12 hours after 5 Gy irradiation. In response to 25 Gy irradiation, hRAD50 and p21 were expressed maximally at 12 hours, but p53 and GADD45 genes showed the highest expression level after 12 hours. Conclusion: Induction of apoptosis and DNA repair by ionizing radiation were closely correlated. The peak time of inducing apoptosis and DNA repair was 12 hours in this study model. hRAD50, a recently discovered DNA repair gene, was also associated with radiation-induced apoptosis.
