Evaluation of a Ground Heat Exchanger Appropriate for the Site of the Third Stage Construction of Incheon International Airport (인천국제공항 3단계 건설부지에 적합한 지중열교환기 시스템 평가 연구)

  • In the present study, a ground heat exchanger was installed for each heat source in the system at the site to evaluate ground heat conductivity, constructability, and economic feasibility; the factors considered in the study included ground heat, groundwater, fillers (such as bentonite and pea pebbles) and the shape of the heat exchange pipe (e.g., U and D-U). The aim was to determine the ground heat exchanger appropriate for the geothermal system in the 3rd-phase construction of Incheon International Airport. A comparative cost analysis of the initial costs based on the above information showed that although the initial costs of the regular vertical closed loop-II and modified vertical closed loop were lower than those of the regular vertical closed loop-I, they could not be expected to deliver high economic efficiency from the viewpoint of constructability (filler injection, heat exchange pipe insertion). The initial costs proved to be higher in the case of Geohil.

Delia platura(Meigen): Bionomics and It`s Resistance to Host Plants (씨고자리파리의 생태 및 기주식물에 대한 저항성)

  • A series of experiments was undertaken to learn bionomics and gowt plant resistance of the seedcorn maggot, Delia platura(Meigen), under controlled(24$\pm$$2^{\circ}C$, RH70$\pm$5%, and LD 16:8h)and field conditions. The preoviposition period for the flies was 9 days. The females survived for an average of 50(3-77) and the males for 24(1-59) days. A greater proportion of flies emerged between 6:00 A.M. and 9:00 A.M., soon after the sun rise. After the over-wintering, adults started to emerge in mid-April from pupae located near the soil surface, and peaked in late April by others located deeper. The sex ratio was about 1:1 with total samples of 1,609 females and 1,641 males. Weight of pupae reared from onion was heavier than those from other diets in the laboratory, however its size was samller than that of natural flies. Considerably more eggs were laid near pea seeds than other hosts tested. Among beans, Bapmitkong with blue seed-coat and a cowpea bean strain were preferred for oviposition. 'Namcheon` cultivar was found to be susceptible to attack by the larvae in the laboratory.

Effect of Undersoil Heating on Growth and Mineral Contents of Turfgrasses in Simulated Athletic Field During Winter Season (겨울철 지하부의 가온처리가 경기장 잔디의 생육 및 무기성분 함량에 미치는 영향)

  • Studies were conducted to determine the effect of undersoil heating on growth and quality of turfgrasses including Kentucky bluegrass (Poa pratensis L.‘Nuglade’), perennial ryegrass (Lolium perenne L.‘Accent’), tall fescue (Festuca arundinacea Schreb.‘Pixie’), and Korean lawngrass (Zoysia japonica Steud.) in simulated athletic field during winter season in Korea. Mineral contents in clippings of turfgrasses grown at different soil mixtures and temperatures were also analyzed. Undersoil heating (approximately 20$\pm$2$^{\circ}C$) was effective in protecting turfgrasses except Korean lawngrass from freezing injury and discoloration of shoots due to extremely cold temperatures during midwinter. Among turfgrasses grown at undersoil heating zone, tall fescue and perennial ryegrass showed the highest clipping weights and chlorophyll contents, respectively. However, anthocyanin contents of shoots were higher in Kentucky bluegrass. There was little or no difference in clipping weights, chlorophyll contents, anthocyanin contents and greenness of shoots between turfgrasses grown at two soil mixtures composed of 80% sand+10% peat moss+10% soil (v/v/v) and 80% sand+20% pea moss (v/v). Contents of mineral K, Ca and Mg in clippings of cool-season turfgrasses were comparatively higher in a soil mixture composed of 80% sand+10% peat moss+10% soil, but little difference in contents of N and P was observed between two soil treatments. Results indicated that undersoil heating can improve quality of turf surface by thawing soil, melting snow, and maintaining shoot growth and greenness of turfgrasses in sports field during winter season.

A virus disease of sesame (Sesamum idicum L.) caused by watermelon mosaic virus (WMV) (참깨의 모자이크 증상에서 분리한 수박${\cdot}$모자이크 바이러스에 관한 연구)

  • This paper deals with the studies on the occurence of a new virus disease of sesame and the identification of the causal virus. The virus disease of sesame has been regarded as a widespread disease in the sesame-growing areas in the southern part of Korea. The disease was found to be caused by watermelon mosaic virus (WMV). During the years since 1978, stunting of sesame plants, with yellow mosaic, necrotic spot, and malformation, were collected from 17 different places. Virus isolates from 27 out of 32 samples were identified as WMV. Natural infection of squash, pumpkin, cucumber, and watermelon by WMV as well as sesame was proved. The virus is inactivated at temperatures of 55 to $60^{\circ}C$, at dilution of $10^{-3}\;to\;10^{-4}$, and in the aging of 10 to 14 days at about $20^{\circ}C$. Sesame, Chenopodium amaranticelor, pea, bean, as well as many plants of the Cucurbitaceae, are susceptible to the sesame-isolates of WMV. In negatively stained preparations, particles of the virus appear under the electron microscope as flexible filaments of about $750\~800nm$ in length. Cylindrical inclusions and virus particles were found in the cytoplasm of mesophyll cells by ultra-thin sections of WMV infected tissues.

Further Modifications to the Mobile Nylon Bag Technique to Determine Nutrient Digestibility for Swine

  • Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

Comparison of Nitrogen Removal in a Horizontal Subsurface-Flow Wetland Purifying Stream Water with and without Litter Layer on its Surface (하천수를 정화하는 수평흐름 여과습지의 표면 잔재물층 유무에 의한 질소제거 비교)

  • Abatements of TN and ${NO_3}^-$-N in a horizontal subsurface-flow wetland with litter layer on its surface were compared with those without one. The wetland was constructed in 2001 on floodplain of the Gwangju Stream which flows through Gwangju City in Korea. Its dimensions were 29m in length, 9m in width and 0.65m in depth. A bottom layer of 45cm was filled with crushed granites (15~40mm in diameter) and a middle layer of 10cm had pea pebbles. An upper layer of 5cm contained coarse sands. Reeds (Phragmites australis) growing in natural wetlands were transplanted on its surface. Water of the stream was channelled into the wetland by gravity flow and its effluent was discharged back into the stream. Average Litter layer of 12.2cm was formed on its surface in 2007. The layer and above-ground parts of reeds were eliminated in April 2008. Volumes and water quality of influent and effluent of the wetland were analyzed from May to November in 2007 and 2008, respectively. Inflow into the wetland both in 2007 and 2008 averaged approximately 40$m^3$/day and hydraulic residence time both in 2007 and 2008 was about 1.5days. Influent TN concentration in 2007 and 2008 averaged 3.96 and 3.89mg/L, respectively and average influent ${NO_3}^-$-N concentration in 2007 and 2008 was 2.11 and 2.05mg/L, respectively. With a 0.05 significance level, influent concentrations of TN and ${NO_3}^-$-N, temperatures and pH of effluent, and heights and stem numbers of reeds showed no difference between the wetland with litter layer and without one. TN retention in the wetland with litter layer and without one averaged 64,76 and 54.69%, respectively and ${NO_3}^-$-N removal averaged 60.83 and 50.61%, respectively. Both TN and ${NO_3}^-$-N abatement rates in the wetland with litter layer were significantly high (TN abatement: p<0,001, ${NO_3}^-$-N abatement: p=0.001) when compared with those without one. The subsurface-flow wetland having litter layer on its surface was more efficient for TN and ${NO_3}^-$-N removal.

Inhibition of Enzymatic Degradation of Leucine Enkephalin and $[D-Ala^2]$-Leucine Enkephalinamide in Various Rabbit Mucosal Extracts by Inhibitors (효소 억제제에 의한 토끼의 점막 추출액중 로이신엔케팔린 및 [D-알라$^2$-로이신엔케팔린아미드의 분해 억제)

  • To inhibit the enzymatic degradation of leucine enkephalin (Leu-Enk) and its synthetic analog. $[D-ala^2]$-leucine enkephalinamide (YAGFL), in the nasal, rectal and vaginal mucosal and serosal extracts of rabbits, effects of enzyme inhibitors such as amastatin (AM), puromycin (PM), thiorphan (TP), thimerosal (TM), EDTA, N-carboxymethyl-Phe-Leu (CPL), phenylethyl alcohol (PEA), phenylmercuric acetate (PMA), benzalkonium chloride (BC) and modified cyclodextrins, alone or in combination, were observed by assaying the pentapeptides staying intact during incubation. Mucosa extracts were prepared by exposing freshly-excised mucosal specimens mounted on Valia-Chien cells to isotonic phosphate buffer while stirring. The degradation of Leu-Enk and YAGFL followed the apparent first-order kinetics. The half-lives (mean) in the nasal, rectal and vaginal mucosal extracts were found to be 1.07, 0.33 and 1.14 hr for Leu-Enk, and 16.9, 6.2 and 6.8 hr for YAGFL, respectively. AM or PM, which is an aminopeptidase inhibitor, did not show a sufficient inhibition of Leu-Enk $(50\;{\mu}g/ml)$ degradation in all kinds of extracts. $Dimethyl-{\beta}-cyclodextrin\;(DM-{\beta}-CyD)$ decreased the degradation rate constants of Leu-Enk about 2 or 3 times, comparing with no additive. However, the use of mixed inhibitors of AM $(50\;{\mu}M)$/TM (0.25 mM)/EDTA (5 mM) resulted in a full stabilization of Leu-Enk by decreasing the degradation rate constants 67.3, 161.3 and 113.8 times far the nasal, rectal and vaginal mucosal extracts, respectively, comparing with no inhibitor. With mixed inhibitors, Leu-Enk remained intact more than 90% after 6 hr-incubation. In the stabilization of YAGFL, hM, TP or CPL alone showed little efffct, and some additives demonstrated a considerable inhibition of YAGFL degradation in the rank order of TM > BC > EDTA. However, the addition of mixed inhibitors such as TM (0.5 mM) and EDTA (5 mM) into the extracts protected YAGFL from the degradation by more than 85% even after 24 hr-incubation, suggesting almost complete inhibition of YAGFL degradation in the extract. On the other hand, $DM-{\beta}-CyD\;or\;hydroxypropyl-{\beta}-cyclodextrin$ (10%) were also found to retard enzymatic degradation rates of YAGFL markedly, and resulted in staying intact more than 80% of YAGFL in the nasal and vaginal mucosal extracts, and more than 60% in the rectal mucosal extract after 16 hr-incubation.

Stratification Variation of Summer and Winter in the South Sea of Korea (한국 남해의 여름과 겨울철 성층 변동)

  • In order to illustrate the variation cf stratification and to know the effects of the temperature and the salinity on the stratification in the South Sea of Korea, the stratification parameter defined as potential energy anomaly (PEA, $V(J/m^3)$) introduced by Simpson and Hunter (1974) was used. The oceanographic data were obtained in August 1999 and February 2000 by National Fisheries Research and Development Institute (NFRDI). V in August is generally high in offshore and low in near shore. However, in February, V in the near shore is higher than that cf the offshore due to the vertical temperature gradient between surface and bottom layer caused by the expansion of South Korean Coastal Waters (SKCW). In summer, the increase of the atmospheric heating acts on the stratification as the buoyancy forcing. In most cases, the effect cf the temperature on the stratification is stronger than that of the salinity. The temperature effect is predominantly due to the extent of the intrusion of Tsushima Warm Current into the study area. However, at stations where V is high the effect of the salinity is also significant. In winter, V is very low due to the decrease cf the buoyancy forcing, but some stations show the relatively high V due to the expansion of SKCW and salinity in winter unlike that in summer makes the stratification weak.

  • Protoplasts of Pisum sativum L. were isolated and cultured from leaf mesophyll tissue. The successful yield of protoplast was obtained in an enzyme solution of 2% 'Onozuka R-10' and 2 % 'Macerozyme R-10' contained 6mM $CaCl_2{\cdot}2H_2O$ within 4 hours. They were divided in B5 culture medium supplemented with 2mg/l kinetin, 1mg/l 2, 4-D and 0.2% Difcobacto agar. Divisions of the protoplasts were continued and led to colony formation for 1 months. The colony from protoplasts of pea mesophyll tissue was formed to callus after subculture in a medium contained macronutrients and amino acids of BII medium and micronutrients and vitamins of B5 medium, and also supplemented with 2mg/l kinetin 2mg/l NAA.

Effect of Root Zone Cooling on Growth and Mineral Contents of Turfgrasses in Simulated Athletic Field during Summer Season (여름철 근권부의 냉온처리가 경기장 잔디의 생육 및 무기성분 함량에 미치는 영향)

  • This study was conducted to determine the effect of root zone cooling on growth and quality of turfgrasses including Kentucky bluegrass (Poa pratensis L.‘Nuglade’), perennial ryegrass (Lolium perenne L.‘Accent’), tall fescue (Festuca arundinacea Schreb.‘Pixie’), and Japanese lawngrass (Zoysia japonica Steud.) in simulated athletic field during summer season in Korea. Mineral contents in clippings of turfgrasses grown at different soil mixtures and temperatures were also analyzed. Root zone cooling (approximately 4~6$^{\circ}C$ lower than that of untreated-control) resulted in good uniformity, little disease incidence and higher level of chlorophyll contents in cool-season turfgrasses. The effectiveness of root zone cooling in protecting disease incidence from high temperature stress was the most manifest in perennial ryegrass compared to others. Fresh clipping weight in treatment of root zone cooling was increased approximately 2 times in Kentucky bluegrass and perennial ryegrass, and 2.5 times in tall fescue compared to those of control. There was higher growth rate in a soil mixture composed of 80% peat moss +10% sand +10% soil (v/v/v) than in that of 80% pea moss +20% sand (v/v), Mineral contents of N, P, K, Ca, and Mg in clippings of three species of cool-season turfgrasses were significantly increased in treat-ment of root zone cooling but this was not found in Japanese lawngrass. Results showed that root zone cooling has a benefit in keeping good quality and growth of cool-season turfgrasses in sports field under supraoptimal ambient temperature during summer season.
